Fragrance material review on 3,7-dimethyl-7-methoxyoctan-2-ol.

A toxicologic and dermatologic review of 3,7-dimethyl-7-methoxyoctan-2-ol when used as a fragrance ingredient is presented. 3,7-Dimethyl-7-methoxyoctan-2-ol is a member of the fragrance structural group branched chain saturated alcohols. The common characteristic structural elements of the alcohols with saturated branched chain are one hydroxyl group per molecule, and a C(4)-C(12) carbon chain with one or several methyl side chains. This review contains a detailed summary of all available toxicology and dermatology papers that are related to this individual fragrance ingredient and is not inte

A safety assessment of branched chain saturated alcohols when used as fragrance ingredients.

The Branched Chain Saturated Alcohol (BCSA) group of fragrance ingredients was evaluated for safety. In humans, no evidence of skin irritation was found at concentrations of 2-10%. Undiluted, 11 materials evaluated caused moderate to severe eye irritation. As current end product use levels are between 0.001% and 1.7%, eye irritation is not a concern. The materials have no or low sensitizing potential. For individuals who are already sensitized, an elicitation reaction is possible. Due to lack of UVA/UVB light-absorbing structures, and review of phototoxic/photoallergy data, the BCSA are not ex

Visible light photopatch testing of common photocontactants in female filipino adults with and without melasma: a cross-sectional study.

BACKGROUND: Although consistently associated with sun exposure, melasma is common among sun-shy Filipino women who generally prefer to have lighter skin, use skin lighteners, regularly practice sun avoidance, and are more exposed to indoor lights. OBJECTIVE: To determine presence/absence of photocontact dermatitis in melasma/no-melasma patients using photopatch testing (PhPT) with standard photocontactants and an indoor visible light (VL) source. DESIGN: Cross-sectional study: random population of 40 female patients aged 30-55 years, 20 with and 20 without melasma. The PhPT included 67 photo

Afloqualone photosensitivity: immunogenicity of afloqualone-photomodified epidermal cells.

Afloqualone (AQ) is an oral muscle relaxant and evokes ultraviolet A (UVA)-induced photosensitivity dermatitis as a side effect. Histologic observations of the skin eruption suggest that AQ photosensitivity is mediated not only by phototoxic but also by photoallergic mechanisms. To address the immunological mechanisms of AQ photosensitivity, we examined the immunogenicity of AQ-photomodified epidermal cells in mice. Afloqualone was covalently coupled with bovine serum albumin by irradiation with UVA but not UVB. Because of this ability of AQ to photobind to protein, murine epidermal cells were

Study on the mechanism of photosensitive dermatitis caused by ketoprofen in the guinea pig.

To investigate the mechanism on photosensitive dermatitis caused by ketoprofen (KP) in humans, the following experiments were performed by topical application on guinea pigs. The phototoxicity study involving treatment with 10% solution of KP, its enantiomers (R-KP and S-KP), loxoprofen, and flurbiprofen revealed no phototoxic reactions. In the photoallergenicity study, KP and its enantiomers (0.5-2% solution) induced skin reaction at all dosages; however, loxoprofen and flurbiprofen (1-5% solution) did not induce such a photoallergenic reaction. These results suggest that the chemical structu

Evaluation and management of the patient with photosensitivity.

Evaluation of patients with photosensitivity includes a detailed history, physical examination, phototests, photopatch tests, and other laboratory tests as appropriate. The epidemiology, clinical features, diagnosis, and management of the more common idiopathic photodermatoses, namely, polymorphous light eruption, chronic actinic dermatitis, and solar urticaria will be reviewed. A brief overview of phototoxicity, photoallergy, and photoprotection is discussed with further elaboration upon the principles of phototherapy and its utility in treating idiopathic photodermatoses.

Dermal Safety of Tapinarof Cream 1%: Results From 4 Phase 1 Trials.

BACKGROUND: Tapinarof (VTAMA®; Dermavant Sciences, Inc.) is a novel, non-steroidal, topical, aryl hydrocarbon receptor agonist, FDA approved for psoriasis treatment and under investigation for atopic dermatitis treatment as a 1% cream formulation for once-daily (QD) application. OBJECTIVE: Evaluate cumulative skin irritation, sensitization, and photoallergic and phototoxic potential of tapinarof cream 1% across a range of dosing frequencies and conditions. METHODS: We conducted 4 randomized, controlled, phase 1 trials of topical tapinarof cream 1% vs vehicle or other appropriate controls

Anticancer treatments and photosensitivity.

Drug-induced photosensitivity is associated with a wide range of anticancer treatments, including conventional chemotherapeutic agents, targeted anticancer therapies, and immune checkpoint inhibitors. These dermatologic adverse events can have a major impact on the well-being and quality of life of cancer patients, leading to dose modifications and interruption or discontinuation of anticancer treatments in severe cases. However, the heterogeneous nature of the photosensitive reactions induced by these agents, as well as the common concomitant use of other potentially photosensitizing drugs (a

Photodermatoses, including phototoxic and photoallergic reactions (internal and external).

Photodermatoses are caused by an abnormal reaction mainly to the ultraviolet component of sunlight. Photodermatoses can be broadly classified into four groups: immunologically mediated photodermatoses, chemical- and drug-induced photosensitivity, photoaggravated dermatoses, and DNA repair-deficiency photodermatoses. In this review, we focus mainly on chemical- and drug-induced photosensitivity, namely, phototoxicity and photoallergy.
