Aedes aegypti exhibits a distinctive mode of late ovarian development.

BACKGROUND: Insects live in almost every habitat on earth. To adapt to their diverse environments, insects have developed a myriad of different strategies for reproduction reflected in diverse anatomical and behavioral features that the reproductive systems of females exhibit. Yet, ovarian development remains largely uncharacterized in most species except Drosophila melanogaster (D. melanogaster), a high Diptera model. In this study, we investigated the detailed developmental process of the ovary in Aedes aegypti (Ae. aegypti), a major vector of various disease-causing pathogens that inhabits

MicroRNA-Transcription factor regulatory networks in the early strobilar development of Echinococcus granulosus protoscoleces.

BACKGROUND: Echinococcus granulosus sensu lato has a complex developmental biology with a variety of factors relating to both intermediate and final hosts. To achieve maximum parasite adaptability, the development of the cestode is dependent on essential changes in transcript regulation. Transcription factors (TFs) and miRNAs are known as master regulators that affect the expression of downstream genes through a wide range of metabolic and signaling pathways. In this study, we aimed to develop a regulatory miRNA-Transcription factor (miRNA-TF) network across early developmental stages of E. gr

Characterization of Aedes Dredd: a novel initiator caspase from the yellow fever mosquito, Aedes aegypti.

Caspases play an essential role during programmed cell death in all metazoans. These enzymes are cysteine proteases and comprise a multi-gene family with more than a dozen mammalian family members. Although caspases have been characterized in many animals, including Drosophila melanogaster, little is known about the caspases that exist in mosquitoes. Here we describe the identification and characterization of Aedes Dredd (AeDredd), a novel caspase in the yellow fever mosquito, Aedes aegypti. AeDredd contains two N-terminal death effector domains and the well conserved caspase catalytic domain.

Nuclear receptor E75 regulates moulting and ovarian development in Nilaparvata lugens.

E75, a member of the nuclear receptor gene family, regulates diverse biological processes, including ovarian development and fat body remodelling. The brown planthopper (Nilaparvata lugens Stål) (Hemiptera:Delphacidae), a major rice pest, poses a significant threat to rice production. However, the role of Nilaparvata lugens E75 (NlE75) in development, particularly ovarian development and its underlying mechanisms, remains largely unknown. In this study, we found that downregulation of NlE75 expression significantly reduced the eclosion rate, especially in the third-, fourth- and fifth-instar n

Broad complex Z2 functions as an upstream 20E-responsive regulator of the midgut chitinase gene HaCHT4 in Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae).

BACKGROUND: Chitinases and ecdysone are both crucial for insect growth and development, yet the regulatory interplay between them remains poorly understood. Our previous research demonstrated that the chitinase gene HaCHT4 critically regulates the chitin content and surface structure of peritrophic membrane (PM) in Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae). RESULTS: Here, a 1286 bp 5’-flanking region of HaCHT4 was cloned to investigate its regulatory mechanism. Bioinformatics analysis predicted the presence of several cis-regulatory elements (CREs) within its 5’-flanking region, i

Sulfasalazine-double stranded ribonucleic acid nanoparticle for controlling Aedes aegypti mosquitoes.

BACKGROUND OBJECTIVES: Aedes aegypti mosquitoes are vectors for dengue, Zika, chikungunya, and yellow fever viruses, which threaten over half the world's population. Therefore, new techniques are needed to control vector mosquitoes. The present study aimed to control female Ae. aegypti by targeting 3β-hydroxysteroid dehydrogenase (3β-HSD) and ecdysone receptor (EcR), which could potentially disrupt egg development, using sulfasalazine (SSZ)-double-stranded RNA (ds-RNA) nanoparticles (NPs). METHODS: The SSZ-dsRNA NPs are characterized by light microscopy, field emission scanning electron micro

Steroid regulation of terminal protein glycosyltransferase genes: molecular and functional homologies within sialyltransferase and fucosyltransferase families.

OBJECTIVE: Terminal glycosylations such as fucosylation and especially sialylation are crucial in maturation of proteins for their particular function. In cells and tissues where their function is under hormonal control, often terminal processes including glycosylations are tightly regulated by hormones. Therefore, it is not surprising that two key enzyme families involved in these steps, fucosyltransferases and sialyltransferases, are directly controlled by steroid hormones. METHODS: Gene structure and protein sequences have been analyzed by set of Unix-based complex sequence analysis progra

Altered coding for a strictly conserved di-glycine in the major bilirubin UDP-glucuronosyltransferase of a Crigler-Najjar type I patient.

The characterization (Ritter, J.K., Chen, F., Sheen, Y. Y., Tran, H.M., Kimura, S., Yeatman, M.T., and Owens, I. S. (1992) J. Biol. Chem. 267, 3257-3261) of the single-copy UGT1 gene complex locus encoding both bilirubin and phenol UDP-glucuronosyltransferases (transferase) has been critical to the determination of genetic defects in Crigler-Najjar patients. The complex (UGT1A-UGT1M) codes for at least two bilirubin, three bilirubin-like, and eight phenol transferase isozymes. In the 5' region, a minimum of 13 different exons 1, each with an upstream promoter, are arrayed in series with 4 comm

Phytoecdysteroids as modulators of the Toxoplasma gondii growth rate in human and mouse cells.

BACKGROUND: Searching for new effective drugs against human and animal toxoplasmosis we decided to test the anti-Toxoplasma potential of phytoecdysteroids (α-ecdysone and 20-hydroxyecdysone) characterized by the pleiotropic activity on mammalian organisms including the enhancement of host's anti-parasitic defence. This objective was accomplished by the in vitro evaluation of T. gondii growth in phytoecdysteroid-treated immunocompetent cells of selected hosts: humans and two strains of inbred mice with genetically determined different susceptibility to toxoplasmosis. METHODS: Peripheral mononu

Neuroendocrine transcriptomics in Eriocheir sinensis: Photoperiod-induced modulation of gonadal development and circadian clock genes in neural tissues.

Photoperiod is a critical environmental cue that orchestrates reproductive physiology and circadian biology in crustaceans, yet the neural gene networks linking photoperiod to ovarian maturation and clock regulation remain largely unknown. To elucidate the molecular mechanisms underlying photoperiod regulation of ovarian development and associated circadian rhythmicity, the eyestalk (SG) and brain ganglion (BG) of female Chinese mitten crab (Eriocheir sinensis) from two photoperiod groups (Light (L): Dark (D) = 0 h: 24 h (L0) and L: D = 18 h: 6 h (L18)) were used for transcriptome analysis. Efficacy of the herbal pair, Radix Achyranthis Bidentatae and Eucommiae Cortex, in preventing glucocorticoid-induced osteoporosis in the zebrafish model.

OBJECTIVE: In traditional Chinese medicine, the herbal pair, Radix Achyranthis Bidentatae (RAB) and Eucommiae Cortex (EC), is widely used to treat osteoporosis. Herein, we determined whether this herbal pair can be used to ameliorate glucocorticoid (GC)-induced osteoporosis (GIOP) and find its optimal dosage in zebrafish. METHODS: The characteristics of the aqueous extract of RAB and EC were separately characterized using high-performance liquid chromatography. Osteoporosis was induced in 5-day post-fertilization zebrafish larvae by exposing them to 10 μmol/L dexamethasone (Dex) for 96 h. Sev

Juglans regia and Pfaffia paniculata extracts: implications for periodontal disease treatment and correlation with Alzheimer's risk.

Periodontal disease (PD) is a significant global health concern, affecting approximately 19% of the world's population. It is one of the most prevalent diseases today, causing substantial socio-economic impacts and diminished quality of life. Recent research has also revealed a potential link between PD and Alzheimer's disease. This study investigated the antimicrobial effects of Juglans regia and Pfaffia paniculata extracts against P. endodontalis and P. gingivalis, bacteria that cause PD and are related to Alzheimer's risk. The study also assessed the impact of these extracts on macrophage m

Overproduction of DNA polymerase eta does not raise the spontaneous mutation rate in diploid human fibroblasts.

Telomerase-immortalized lines of diploid xeroderma pigmentosum variant (XP-V) fibroblasts (XP115LO and XP4BE) were complemented for constitutive or regulated expression of wild-type human DNA polymerase eta (hpol eta). The ectopic gene was expressed from a retroviral LTR at a population average of 34- to 59-fold above the endogenous (mutated) mRNA and high levels of hpol eta were detected by immunoblotting. The POLH cDNA was also cloned downstream from an ecdysone-regulated promoter and transduced into the same recipient cells. Abundance of the wild-type mRNA increased approximately 10-fold by

Methoprene-Tolerant (Met) Acts as Methyl Farnesoate Receptor to Regulate Larva Metamorphosis in Mud Crab, Scylla paramamosain.

The conserved role of juvenile hormone (JH) signals in preventing larvae from precocious metamorphosis has been confirmed in insects. Crustaceans have different metamorphosis types from insects; we previously proved that methyl farnesoate (MF) can prohibit larvae metamorphosis in mud crabs, but the molecular signal of this process still needs to be elucidated. In this study, methoprene-tolerant (Met) of Scylla paramamosain was obtained and characterized, which we named Sp-Met. Sp-Met contains a 3360 bp ORF that encodes 1119 amino acids; the predicted protein sequences of Sp-Met include one bHL

Gemin3 is an essential gene required for larval motor function and pupation in Drosophila.

The assembly of metazoan Sm-class small nuclear ribonucleoproteins (snRNPs) is an elaborate, step-wise process that takes place in multiple subcellular compartments. The initial steps, including formation of the core RNP, are mediated by the survival motor neuron (SMN) protein complex. Loss-of-function mutations in human SMN1 result in a neuromuscular disease called spinal muscular atrophy. The SMN complex is comprised of SMN and a number of tightly associated proteins, collectively called Gemins. In this report, we identify and characterize the fruitfly ortholog of the DEAD box protein, Gemin

Developing new drugs that activate the protective arm of the renin-angiotensin system as a potential treatment for respiratory failure in COVID-19 patients.

COVID-19, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has reached pandemic proportions with negative impacts on global health, the world economy and human society. The clinical picture of COVID-19, and the fact that Angiotensin converting enzyme 2 (ACE2) is a receptor of SARS-CoV-2, suggests that SARS-CoV-2 infection induces an imbalance in the renin-angiotensin system (RAS). We review clinical strategies that are attempting to rebalance the RAS in COVID-19 patients by using ACE inhibitors, angiotensin receptor blockers, or agonists of angiotensin-II receptor type 2

The role of autophagy in bone metabolism and clinical significance.

The skeletal system is the basis of the vertebral body composition, which affords stabilization sites for muscle attachment, protects vital organs, stores mineral ions, supplies places to the hematopoietic system, and participates in complex endocrine and immune system. Not surprisingly, bones are constantly reabsorbed, formed, and remodeled under physiological conditions. Once bone metabolic homeostasis is interrupted (including inflammation, tumors, fractures, and bone metabolic diseases), the body rapidly initiates bone regeneration to maintain bone tissue structure and quality.
