Development and evaluation of a TaqMan® real-time PCR assay for species-specific detection of Ehrlichia canis.

The aim of this work was to develop a real-time PCR assay with a TaqMan® probe that detects a species-specific part of the 16S rDNA gene of Ehrlichia canis. Canine blood samples (n = 207), collected and tested by a conventional PCR assay within a study conducted by De Salvo et al., were simultaneously analyzed with the novel designed real-time PCR, and the results of both assays were compared. The agreement between the two methods was 97.6 % with a kappa value of 0.92186. Hereby, the standard error was 0.034416 and the 95 % confidence interval from 0.8544 to 0.98931. While the conventional PCR

Characterization of serum protein fractions of dogs naturally infected with Ehrlichia canis or Anaplasma platys associated with uveitis.

Uveitis associated with Ehrlichia canis or Anaplasma platys infections were reported in dogs. However, only two E. canis-infected dogs with hypergammaglobulinemia showed acute blindness were reported. There were limited data of the species of Ehrlichia or Anaplasma and the alteration of serum protein fractions in infected dogs. Thus, the species of causative pathogen were investigated and compared the serum protein fractions between infected dogs associated with anterior uveitis and panuveitis in clinical situations. All 103 studied dogs were brought into the ophthalmology clinic which each do

Molecular assessment of the transplacental transmission of Toxoplasma gondii, Neospora caninum, Brucella canis and Ehrlichia canis in dogs.

Given the fact that numerous microbial species can be detected in pregnant female dogs, the objective of this study was to assess the transplacental transmission of Brucella canis, Ehrlichia canis, Neospora caninum and Toxoplasma gondii in stillborn puppies. This study involved 41 stillborn puppies, 78.6% of which were positive for T. gondii, 52.4% for N. caninum and 59.5% for B. canis. E. canis was not detected in any of the analyzed puppies. Pregnancy is an important physiological condition for the transmission of infectious agents to puppies and transplacental transmission may be epidemiolo

A survey of antibodies against Rickettsia rickettsii and Ehrlichia chafeensis in domestic animals from a rural area of Colombia.

In a rural area of Colombia endemic for Rocky Mountain spotted fever, we performed indirect immunofluorescent antibody assays for Rickettsia spp. and Ehrlichia spp. using sera from randomly sampled dogs and horses to test the use of domestic animals as possible sentinels. Antibodies against Ehrlichia spp. were detected in 8 dogs (31.8%). Antibody titers against Rickettsia rickettsii antigen were positive in 4 dogs (18.2%) and 26 horses (16.3%). These values, albeit not directly comparable, are lower than those previously reported for humans in this region. A systemic approach to understanding

High prevalence of granulocytic Ehrlichiae and Borrelia burgdorferi sensu lato in Ixodes ricinus ticks from Bulgaria.

Bulgarian Ixodes ricinus ticks were examined for Ehrlichia and Borrelia coinfection: 34 and 32% of adult ticks and at least 2 and 10% of nymphs were positive for these infections, respectively. Coinfections and dual or triple Borrelia infections were frequent, although Ehrlichia phagocytophila heterogeneity was minimal. Multiple tick-borne bacteria coexist in I. ricinus ticks in southeastern Europe.

Human endothelial cells as an alternative to DH82 cells for isolation of Ehrlichia chaffeensis, E. canis, and Rickettsia rickettsii.

Ehrlichia chaffeensis, etiologic agent of human ehrlichiosis, and Rickettsia rickettsii, etiologic agent of Rocky Mountain spotted fever (RMSF), are both tick-borne agents that cause nonspecific symptoms that may be indistinguishable from each other early in the course of infection. E. canis is a canine pathogen closely related to E. chaffeensis and was initially suspected of being the causative agent of human ehrlichiosis. If a febrile illness is reported, after tick exposure, neither ehrlichiosis nor RMSF can be immediately ruled out. When attempts are made to isolate the agent from blood, a

First report of Providencia rettgeri, Colpodella spp., Ehrlichia spp., and Rickettsia hoogstraalii in ticks infesting goats of Pakistan.

Ticks are the second most important vector of infectious diseases, after mosquitoes, and can transmit several diseases of concern for both human and veterinary health. This study molecularly barcoded ticks collected from goats in Pakistan and screened for associated pathogens. From July 2023 to June 2024, examination of 253 goats (Capra hircus) in the 7th district of Khyber Pakhtunkhwa found 170 goats infested with 1,305 ticks, equating to a mean abundance of 5.15 ticks per goat. A phenol-chloroform technique was used to extract DNA and subsequently amplify the presence of pathogen DNA targeti

Molecular detection and phylogenetic characterization of Ehrlichia- and Anaplasma-related genotypes in free-ranging anteaters from southeastern Brazil.

Bacteria of the family Anaplasmataceae are tick-borne pathogens of recognized veterinary and zoonotic relevance, widely distributed among domestic and wild vertebrate hosts. Although increasingly reported in Brazilian wildlife species, molecular data on these agents in xenarthrans remain scarce, particularly in regions undergoing environmental transformation. This study investigated the occurrence of Anaplasma spp. and Ehrlichia spp. in free-ranging giant anteaters (Myrmecophaga tridactyla) and southern tamanduas (Tamandua tetradactyla) from the central-western region of São Paulo State, Brazi

Detection of Ehrlichia spp., Candidatus Neoehrlichia sp. (FU98) and Anaplasma phagocytophilum in red foxes (Vulpes vulpes) in Germany.

Vector-borne diseases pose a growing challenge to public health. Effective surveillance, interdisciplinary cooperation in line with the One Health approach, and prevention and control strategies are therefore essential to reduce the risk of future outbreaks and burden of disease in the long term. Identifying and investigating sentinels can help in this regard. In this study, 537 red fox (vulpes) spleen samples from three regions in Germany (Berlin, Brandenburg, Arnsberg administrative region) were tested for Anaplasmataceae and Rickettsia spp. using PCR. Only 3% of the samples (n = 15) tested

Absence of serological evidence of Rickettsia spp., Bartonella spp., Ehrlichia spp. and Coxiella burnetii infections in American Samoa.

Little is known about the epidemiology of zoonotic diseases in American Samoa (Pacific). A review of literature did not identify any published information on human Rickettsia spp., Bartonella spp., Ehrlichia spp. or Coxiella burnetii infections in this country. To determine the presence of these diseases, we conducted a serosurvey of American Samoans. The presence of immunoglobulin G antibodies against Rickettsia felis, Rickettsia typhi, Rickettsia conorii, C. burnetii, Bartonella henselae, Bartonella quintana, and Ehrlichia chaffeensis was evaluated by indirect immunofluorescence assay in ser

Surveillance for Ehrlichia canis, Anaplasma phagocytophilum, Borrelia burgdorferi, and Dirofilaria immitis in Dogs From Three Cities in Colombia.

OBJECTIVE: Emerging infectious and zoonotic diseases are made up in large proportion by vector-borne diseases (VBD). Dogs are parasitized by disease vectors such as ticks and mosquitoes, making dogs adequate reservoirs for zoonoses. Risk of exposure to VBD exists for the U.S. military personnel and Military Working Dogs (MWD) when deployed globally. The importance of canine VBD surveillance relates to veterinary and public health significance for the host nations as well as for the U.S. troops and MWDs. The objective of this work was to survey dogs from the cities of Medellin, Barranquilla, an

Molecular-based detection of Ehrlichia spp. in stray dogs-infesting Rhipicephalus ticks in high-altitude of northern Pakistan.

Although ticks are recognized as carriers of zoonotic pathogens, the risks of tick-borne infections associated with dogs have received limited attention. The close interaction between humans and dogs increases the zoonotic potential of pathogens, increasing the urgency of addressing this overlooked vector-borne health risk, especially in developing regions. This study sought to identify tick-borne zoonotic pathogens by collecting ticks from dogs and screening for Ehrlichia species, a group known to pose significant yet often neglected health risks to both humans and animals, particularly in th

Changes in the Seroprevalence of Tick-Borne Rickettsia and Ehrlichia Among Soldiers-Fort Liberty, North Carolina, 1991-2019.

We obtained samples from the Department of Defense Serum Repository from soldiers who were stationed at Fort Liberty, North Carolina, between 1991 and 2019 to assess temporal trends in tick-borne rickettsiosis and ehrlichiosis. Serological evidence of infection was common, with nearly 1 in 5 (18.9%) demonstrating antibodies. We observed significant decreases in Rickettsia seroprevalence (adjusted odds ratio [aOR], 0.42 [95% CI, .27-.65], P = .0001) while over the same period Ehrlichia seroprevalence, albeit less common, nearly doubled (aOR, 3.61 [95% CI, 1.10-13.99], P = .048). The increase in

PCR detection of Ehrlichia ruminantium and Babesia bigemina in cattle from Kwara State, Nigeria: unexpected absence of infection.

Ticks and tick-borne diseases (TBDs) continue to pose an insidious and ever-present threat to livestock and livelihoods across the globe. Two of the most significant TBDs of cattle in Africa are heartwater and babesioisis, caused by Ehrlichia ruminantium and Babesia bigemina respectively. Both pathogens are endemic in Nigeria. However, to date, little data has been published regarding the number of cattle infected. In this study, blood samples were collected from cattle of the Kwara State, north-central Nigeria. Probe-based quantitative PCR (qPCR) and semi-nested PCR were used to investigate t
