Endoplasmic Reticulum Stress and Ethanol Neurotoxicity.

Ethanol abuse affects virtually all organ systems and the central nervous system (CNS) is particularly vulnerable to excessive ethanol exposure. Ethanol exposure causes profound damages to both the adult and developing brain. Prenatal ethanol exposure induces fetal alcohol spectrum disorders (FASD) which is associated with mental retardation and other behavioral deficits. A number of potential mechanisms have been proposed for ethanol-induced brain damage; these include the promotion of neuroinflammation, interference with signaling by neurotrophic factors, induction of oxidative stress, modul

Wernicke encephalopathy and ethanol-related syndromes.

Ethanol causes diverse neurologic conditions caused by acute and chronic brain damage. This review provides an overview of Wernicke encephalopathy and other ethanol-related brain changes, such as chronic brain atrophy, Marchiafava-Bignami disease, osmotic demyelination syndrome, chronic hepatic encephalopathy, and acute alcohol withdrawal. As clinical symptoms of this spectrum of diseases have nonspecific neurologic alterations, radiologists should have current radiologic information and understand the imaging findings pertaining to the pathophysiology to support diagnosis.

Ethanol cytotoxic effect on trophoblast cells.

Prenatal ethanol exposure may cause both, altered fetal neurodevelopment and impaired placental function. These disturbances can lead to growth retardation, which is one of the most prevalent features in Fetal Alcohol Syndrome (FAS). It is not known whether there is a specific pattern of cytotoxicity caused by ethanol that can be extrapolated to other cell types. The aim of this study was to determine the cytotoxic effects caused by sustained exposure of trophoblast cells to ethanol. The cytotoxic effect of sustained exposure to standard doses of ethanol on an in vitro human trophoblast cell l

Fomepizole for severe disulfiram-ethanol reactions.

Ingestion of ethanol in the presence of disulfiram may cause a histamine-like reaction due to accumulation of acetaldehyde. These disulfiram-ethanol reactions (DERs) are manifested by hypotension, tachycardia, gastritis, and angioedema. Fomepizole, an inhibitor of alcohol dehydrogenase, may halt progression of this reaction by blocking ethanol metabolism to acetaldehyde. We present 2 cases of disulfiram and alcohol overdose leading to severe reactions unresponsive to fluid resuscitation and treated with a single dose of fomepizole. Case 1: A 20-year-old woman presented after ingestion of vodka

Lithium-mediated protection against ethanol neurotoxicity.

Lithium has long been used as a mood stabilizer in the treatment of manic-depressive (bipolar) disorder. Recent studies suggest that lithium has neuroprotective properties and may be useful in the treatment of acute brain injuries such as ischemia and chronic neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, Huntington's disease and amyotrophic lateral sclerosis. One of the most important neuroprotective properties of lithium is its anti-apoptotic action. Ethanol is a neuroteratogen and fetal alcohol spectrum disorders (FASD) are caused by maternal ethanol exposure d

Ethanol embolotherapy of hand arteriovenous malformations.

PURPOSE: Ethanol embolotherapy is one of the established methods in the treatment of extremity arteriovenous malformations (AVMs). The purpose of this study was to report the application of this method to hand AVMs and to assess retrospectively the therapeutic outcomes and complications. PATIENTS AND METHODS: From December 1998 to March 2009, we treated 31 patients with hand AVMs (16 women, 15 men, age range, 5-51 years; mean age, 27 years). With the patients under general anesthesia, they underwent staged ethanol embolotherapy (range, 1-11 sessions; mean, 2.8 sessions) by direct puncture and

Autophagy and ethanol-induced liver injury.

The majority of ethanol metabolism occurs in the liver. Consequently, this organ sustains the greatest damage from ethanol abuse. Ethanol consumption disturbs the delicate balance of protein homeostasis in the liver, causing intracellular protein accumulation due to a disruption of hepatic protein catabolism. Evidence indicates that ethanol or its metabolism impairs trafficking events in the liver, including the process of macroautophagy, which is the engulfment and degradation of cytoplasmic constituents by the lysosomal system. Autophagy is an essential, ongoing cellular process that is high

Ethanol neurotoxicity and dentate gyrus development.

Maternal alcohol ingestion during pregnancy adversely affects the developing fetus, often leading to fetal alcohol syndrome (FAS). One of the most severe consequences of FAS is brain damage that is manifested as cognitive, learning, and behavioral deficits. The hippocampus plays a crucial role in such abilities; it is also known as one of the brain regions most vulnerable to ethanol-induced neurotoxicity. Our recent studies using morphometric techniques have further shown that ethanol neurotoxicity appears to affect the development of the dentate gyrus in a region-specific manner; it was found

Ethanol sclerotherapy of peripheral venous malformations.

BACKGROUND: Venous malformations are congenital lesions that can cause pain, decreased range of movement, compression on adjacent structures, bleeding, consumptive coagulopathy and cosmetic deformity. Sclerotherapy alone or combined with surgical excision is the accepted treatment in symptomatic malformations after failed treatment attempts with tailored compression garments. OBJECTIVES: To report our experience with percutaneous sclerotherapy of peripheral venous malformations with ethanol 96%. PATIENTS AND METHODS: 41 sclerotherapy sessions were performed on 21 patients, aged 4-46 years, 1

Hydrocephalus following prenatal exposure to ethanol.

Pregnant rats were administered a liquid diet containing 5% (w/v) ethanol between gestational days 10 and 21, and the brains of nine offspring were examined at 8 weeks of age. Two ethanol-treated offspring showed obvious hydrocephalus characterized by markedly enlarged lateral ventricles. Most of the other ethanol-treated rats only showed a slight enlargement of the lateral ventricles. An ethanol-treated offspring showed no neuropathological signs of hydrocephalus. Histological observation of the hydrocephalic brains revealed dilation of the lateral ventricles, loosely bundled corpus callosum,

[Ethanol-induced injury and GI bleeding].

Ethanol-induced injury of digestive organs closely linked to the GI bleeding. The gastric or esophageal mucosal hemorrhage evoked by extra-amounts of alcohols is initiated by the microcirculatory damage of gastrointestinal mucosa. Many investigators focused on the ethanol-induced gastric ulcer formation as well as the ethanol-induced gastroesophageal reflux injury. Although the most important treatment for such a damage is of course to stop the drinking, it is also important to protect the gastrointestinal mucosa by potentiate the mucosal defense systems.

Ethanol inhibits sphincter of Oddi motility.

Patients with alcohol-induced liver disease are at increased risk for pigment gallstones, which are known to be particularly associated with biliary stasis. Although the effects of ethanol on the sphincter of Oddi are thought to contribute to alcoholic pancreatitis, the precise effects of ethanol on the biliary component of the sphincter of Oddi are unclear. In the prairie dog the common bile and pancreatic ducts enter the duodenum separately, facilitating pressure measurement in the sphincter choledochus in isolation. We therefore used this model to test the hypothesis that ethanol administra

Ethanol production from spent cherry brine.

Spent cherry brine is an acidic byproduct of maraschino cherry processing and typically consists of variable amounts of glucose and fructose of up to 11% fermentable solids, 0.5-1.5% CaCl2, up to 0.4% sulfur dioxide, sorbitol, and lesser amounts of other cherry constituents. Disposal of brine represents a significant cost to processors because of its high biological oxygen demand. As an alternative, brine was tested as a substrate for ethanol production. Initially, the toxic level of sulfur dioxide was reduced by raising brine pH to 8.0 to precipitate calcium sulfite. Because alkalinization wa

Intravenous pyridoxine in acute ethanol intoxication.

Intravenous pyridoxine was evaluated as an agent for the reversal of ethanol-induced central nervous depression in a randomised double blind controlled study of 108 patients presenting with a clinical diagnosis of acute ethanol intoxication to two accident and emergency departments. Level of consciousness, measured by a modified Glasgow coma scale, showed no significant change after a single 1 g dose of intravenous pyridoxine when compared to controls given saline. The mean fall in blood alcohol concentration after one hour was 33 mg dl-1 (7.2 mmol l-1) in both groups suggesting that pyridoxin

Cross-tolerance between ethanol and chlordiazepoxide.

Drug-induced hypothermia was used to investigate drug tolerance and cross-tolerance. C57BL/6J mice, which were injected with a single dose of chlordiazepoxide (CDP; 30 mg/kg) one day before and reinjected with an equivalent dose of CDP the next day, did not develop tolerance to the drug. However, ethanol-pretreated (3.5 g/kg, 24 hr earlier) mice, when injected with CDP (30 mg/kg), showed cross-tolerance to CDP. The cross-tolerance was short-lived (less than 48 hr). On the other hand, CDP-pretreated mice (30 mg/kg, 24 hr earlier) did not show cross-tolerance to ethanol. The lack of a reciprocat

Non-ethanol components of Baijiu alleviate ethanol-induced energy metabolism disorder and gut microbiota dysbiosis in mice.

Energy and gut-liver metabolic disorders are critical events in alcohol-related diseases. However, the specific mechanisms by which they mediate the differential effects between Chinese Baijiu and ethanol remain largely unknown. As a traditional Chinese fermented beverage, Baijiu contains abundant non-ethanol components in addition to ethanol. In this study, we conducted a multi-dimensional assessment of the ameliorative effects and underlying mechanisms of non-ethanol components from Baijiu against ethanol-induced liver injury using animal experiments, serum metabolomics, 16S rRNA sequencing

Impact of two different rodent diets on maternal ethanol consumption, serum ethanol concentration and pregnancy outcome measures.

Recent studies report varying levels of ethanol consumption by rodents maintained on different commercially available laboratory diets. As varied ethanol consumption by dams may impact offspring outcome measures in prenatal ethanol exposure paradigms, we compared ethanol consumption by rats maintained on the Envigo 2920 diet, used in our vivarium, with an isocalorically equivalent PicoLab 5L0D diet used in some alcohol consumption studies. Compared to 5L0D diet, female rats maintained on 2920 diet consumed 14% less ethanol during daily 4-h drinking sessions prior to pregnancy and 28% less etha
