Calycosin-7-O-β-D-glucoside alleviates nonalcoholic fatty liver disease by regulating short-chain fatty acids metabolism and metabolic regulators.

Nonalcoholic fatty liver disease (NAFLD) is a metabolic stress liver injury disease caused by excessive fat deposition in hepatocytes. Astragalus memeranaceus Fisch., mainly consists of flavonoids, is considered to have good liver protection effects. This study aimed to explore the active ingredients and possible mechanisms of Astragalus flavone in the treatment of NAFLD. Network pharmacology and molecular docking were used to screen the active ingredients and targets of Astragalus flavone in the treatment of NAFLD, and were verified in vivo high-fat diet-induced mouse models. There were 89 pr

Inhibitory effect of flavonoids on sulfo- and glucurono-conjugation of acetaminophen in rat cultured hepatocytes and liver subcellular preparations.

A large group of flavonoids was investigated for inhibitory effects on sulfo- and glucurono-conjugation of acetaminophen when added to rat cultured hepatocytes and liver subcellular preparations. The flavonoids inhibited the production of both sulfate and glucuronide conjugates in the cultured cells, with potencies that depended on the specific flavonoid. Among the flavonols, quercetin, kaempferol and galangin were much more effective than myricetin and morin. Flavones including luteolin, apigenin and chrysin were as effective as the corresponding three flavonols above. The inhibition of conju

Spasmolytic activity of the flavonoids from Thymus vulgaris.

Flavonoids from Thymus vulgaris L. have been studied in vitro for spasmolytic activity on the smooth muscles of the guinea-pig ileum and trachea and of the rat vas deferens. The flavones and thyme extracts inhibit responses to agonists which stimulate specific receptors (acetylcholine, histamine, L-noradrenaline) as well as to agents whose actions are not mediated via specific receptors (BaCl2). Cumulative dose-response curves point to a non-competitive antagonism. Inhibition of Ca2+ contractions on K+ depolarized smooth muscles suggest inhibition of availability of Ca2+ for muscle contraction

Mechanisms involved in the chemoprevention of flavonoids.

Flavonoids, widespread in edible plants, have been studied extensively for their anticarcinogenic properties. However, only few studies have been done with these constituents being administered by the dietary route. In our research, the effects of feeding rats with flavone, flavanone, tangeretin, and quercetin were investigated on two steps of aflatoxin B1 (AFB1)-induced hepatocarcinogenesis (initiation and promotion). Nonpolar flavonoids such as flavone, flavanone and tangeretin administered through the initiation period, decreased the number of -gamma-glutamyl transpeptidase-preneoplastic fo

Activation of acetaminophen-reactive metabolite formation by methylxanthines and known cytochrome P-450 activators.

Previous in vitro studies suggested that caffeine enhanced acetaminophen (APAP) oxidation to N-acetyl-p-benzoquinone imine (NAPQI) by selectively activating the male-specific constitutive cytochrome P-450IIIA2. Monomethylxanthine and dimethylxanthine analogs of caffeine (also metabolites) were studied for their potential effect to accelerate NAPQI formation in various preparations of rat liver microsomes. In contrast to caffeine, none of the mono- and dimethylxanthines (2.5 mM) activated P-450. Rather, the analogs either inhibited NAPQI formation or had no effect; 1-methylxanthine (2.5 mM) was

Antifungal and antibiofilm activities of flavonoids against Candida albicans: Focus on 3,2'-dihydroxyflavone as a potential therapeutic agent.

Effective management of microbial biofilms holds significance within food and medical environments. Candida albicans, an opportunistic fungus, forms mucosal biofilms closely linked to candidiasis and drug-resistant infections due to their drug tolerance. Morphologic change from yeast to filamentous cells is a key virulence factor and a prerequisite for biofilm development. This study investigated the anti-fungal and antibiofilm activities of 20 flavonoids against C. albicans. With their known antioxidant capabilities, flavonoids hold promise in combating infections associated with biofilms. Am

Secondary metabolites of the invasive tree, Callery pear (Pyrus calleryana), provide support for the empty niche theory of invasion.

Invasive woody species like Callery pear (Pyrus calleryana Decne.) alter ecosystems directly and indirectly through effects on arthropod communities as well as chemical alterations of the soil. Evidence suggests that the aggressive spread and negative impacts are due to allelopathic chemicals present throughout plant tissues which reduce herbivory and add unique allochthonous inputs to the soil, thereby reducing germination of native species and furthering Callery pear's domination on the landscape. To assess the allelopathic potential of Callery pear, we collected leaf tissue from Callery pea

Intake of dietary flavonoids in relation to bone loss among U.S. adults: a promising strategy for improving bone health.

Dietary flavonoids have been recommended for improving bone health due to their antioxidant, anti-inflammatory and osteogenic properties. However, the effectiveness of each flavonoid subclass in the prevention and treatment of osteoporosis remains controversial. The objective of the current study was to examine the association between the intake of flavonoid subclasses and bone loss in 10 480 U.S. adults in the National Health and Nutrition Examination Survey. We employed a multinomial logistic regression model to calculate the odds ratios (OR) and 95% confidence intervals (95% CI). The intake

Genome-wide characterization of the AP2/ERF gene family in Scutellaria baicalensis Georgi reveals root-preferential and MeJA-responsive candidate genes.

APETALA2/ethylene-responsive factor (AP2/ERF) transcription factors play central roles in plant growth, development and environmental responses. Scutellaria baicalensis is a medicinal plant that predominantly accumulates pharmacologically important flavones in its roots, and methyl jasmonate (MeJA) induced these flavones accumulation. However, the AP2/ERF family has not been systematically characterized in S. baicalensis, and members showing root-preferential expression and MeJA responsiveness remain unknown. Here, we identified 137 SbAP2/ERF genes at the genome-wide level and investigated the

An integrative multi-omics approach reveals metabolic mechanism of flavonoids during anaerobic fermentation of de'ang pickled tea.

Anaerobic fermentation (AF) is critical process for Yunnan De'ang pickled tea production. Therefore, widely targeted metabolomics and metagenomics were integrated to reveal the AF mechanism. Lactic acid bacteria (LAB) (e.g. Lactiplantibacillus plantarum, Lactobacillus vaccinostercus and Lactobacillus paracollinoides) and yeasts like Candida metapsilosis and Cyberlindnera fabianii dominated in the AF. Based on bacterial community succession and metabolites variation, the whole AF processes were divided into two phases, i.e., before and after four months. A total of 327 characteristic metabolite

Sedative and sleep-enhancing properties of linarin, a flavonoid-isolated from Valeriana officinalis.

We have recently reported the presence of the anxiolytic flavone 6-methylapigenin (MA) and of the sedative and sleep-enhancing flavanone glycoside 2S (-) hesperidin (HN) in Valeriana officinalis and Valeriana wallichii. MA, in turn, was able to potentiate the sleep-inducing properties of HN. The present paper reports the identification in V. officinalis of the flavone glycoside linarin (LN) and the discovery that it has, like HN, sedative and sleep-enhancing properties that are potentiated by simultaneous administration of valerenic acid (VA). These effects should be taken into account when co
