Impact of increased Porphyromonas gingivalis peptidylarginine deiminase (PPAD) T2 variant allele on oral microbiota composition and severity of chronic periodontitis.

BACKGROUND: Porphyromonas gingivalis (Pg) is a keystone pathogen in periodontitis, encoding a unique peptidyl arginine deiminase (PPAD) linked to protein citrullination, a process associated with rheumatoid arthritis (RA). Recently, we identified a super-active PPAD variant (T2) in Pg isolates. Here, we evaluated if the presence of the super-active T2 variant of PPAD affects the salivary microbiome, the severity of chronic periodontitis (CP), and subsequently CP's causative association with RA onset/progression. PATIENTS/MATERIALS AND METHODS: We examined 56 CP patients and 36 healthy volunte

Quantitative detection of volatile sulfur compound- producing microorganisms in oral specimens using real-time PCR.

OBJECTIVE: It is well-known that some periodontopathic bacteria, especially Porphyromonas gingivalis, Fusobacterium nucleatum, Tannerella forsythia (formerly Bacteroides forsythus or Tan. forsythensis), and Treponema denticola, actively produce volatile sulfur compounds (VSCs), such as H2S and CH3SH. We previously reported a qualitative relationship between periodontopathic bacteria and VSCs; however, a quantitative analysis of periodontopathic bacteria in oral specimens is required for further characterization of the relationship between oral bacteria and VSCs. In this study, we report a real

Porphyromonas gingivalis and Treponema denticola synergistic polymicrobial biofilm development.

Chronic periodontitis has a polymicrobial biofilm aetiology and interactions between key bacterial species are strongly implicated as contributing to disease progression. Porphyromonas gingivalis, Treponema denticola and Tannerella forsythia have all been implicated as playing roles in disease progression. P. gingivalis cell-surface-located protease/adhesins, the gingipains, have been suggested to be involved in its interactions with several other bacterial species. The aims of this study were to determine polymicrobial biofilm formation by P. gingivalis, T. denticola and T. forsythia, as well
