The effect of carrageenan oligosaccharides on the muscle quality and myofibrillar protein stability of sea bass (Lateolabrax japonicus) during freezing storage under different freezing methods.

Frozen storage is widely used for the preservation of seafood quality, but protein denaturation and oxidative damage often lead to a decline in fish meat quality. This study investigated the effects of carrageenan oligosaccharides combined with different freezing treatments on the muscle quality and myofibrillar protein stability of sea bass (Lateolabrax japonicus) during 8 weeks of frozen storage. The results showed that the meat quality of fish stored after carrageenan oligosaccharide treatment was superior to that of the untreated group. Carrageenan oligosaccharides effectively alleviated q

Emerging Manufacturing Technologies in Freeze-Drying of Pharmaceuticals.

Freeze-drying of pharmaceutical products is an energy-intensive operation in pharmaceutical manufacturing. Emerging process-intensification and continuous-manufacturing technologies are now reshaping this traditionally static process. Among these, microwave-assisted freeze-drying, suspended-vial freeze-drying, and spin freeze-drying have demonstrated substantial reductions in cycle time, i.e., up to 80%-90%, while maintaining product stability and uniformity. These approaches enhance heat and mass transfer by replacing conduction through fixed shelves with volumetric or radiative energy delive

Principles Underlying Cryopreservation and Freeze-Drying of Cells.

Cryopreservation, vitrification, and freeze-drying can be used to preserve biomolecules, cells, or tissues for extended periods until they are needed. This chapter explores the underlying principles how to preserve biological systems in the frozen state at cryogenic temperatures or in the dried state at room temperature. Freezing and drying are different stress vectors that require different types of protective agents. Freezing injury results from solution effects and physical damage if intracellular ice is formed, while drying removes non-freezable water bound to biomolecules resulting in con

Freeze branding: a novel injurious mechanism for humans.

Freeze branding was first performed in 1966 to identify animals for agricultural purposes, and remains relevant today in veterinary medicine. In contrast, human branding for aesthetics, punishment, identification, and purported medicinal benefits is standardly performed through transfer of thermal energy via heat. Uniquely, this case report documents the first description of freeze branding as a mechanism of injury in humans resulting in a 0.25% total body surface area mixed partial thickness/full thickness forearm burn that was managed non-operatively with out-patient daily wound care over si

Freeze-drying of live virus vaccines: A review.

Freeze-drying is the preferred method for stabilizing live, attenuated virus vaccines. After decades of research on several aspects of the process like the stabilization and destabilization mechanisms of the live, attenuated viruses during freeze-drying, the optimal formulation components and process settings are still matter of research. The molecular complexity of live, attenuated viruses, the multiple destabilization pathways and the lack of analytical techniques allowing the measurement of physicochemical changes in the antigen's structure during and after freeze-drying mean that they form

Updating freeze: aligning animal and human research.

Freezing is widely used as the main outcome measure for fear in animal studies. Freezing is also getting attention more frequently in human stress research, as it is considered to play an important role in the development of psychopathology. Human models on defense behavior are largely based on animal models. Unfortunately, direct translations between animal and human studies are hampered by differences in definitions and methods. The present review therefore aims to clarify the conceptualization of freezing. Neurophysiological and neuroanatomical correlates are discussed and a translational m

Occupational asthma due to freeze-dried raspberry.

OBJECTIVE: To investigate the possibility that inhalation of raspberry powder at work can cause occupational asthma. METHODS: We report the case of a 35-year-old, nonsmoking woman who complained of hay fever symptoms, wheezing, and shortness of breath two or three times a month exclusively in association with coating chewing gum with raspberry powder. Prick test extracts with the five types of fruit powder were produced. Peak expiratory flow rate monitoring five times daily was performed for an 8-week period while the patient was at work. Specific IgE and histamine release tests for raspberry

Freeze-fracture replication technique of human skin.

The freeze-fracture replication technique represents one of the most advanced methods for studying the ultrastructure of biological tissues and has given a new dimension to the ultrastructure research. This technique makes it possible to obtain a cast of a fracture surface of frozen preparations. Apart from allowing samples to be examined without being dehydrated, i.e. under conditions very close to those of living state, it has proved especially useful for the analysis of many tissue functions concerned with membrane activity and exchanges between cells and their disturbance in specific disea

Freeze-fracture replication of cultured Pityrosporum orbiculare.

Skin scrapings from a patient with Pityriasis versicolor were plated on a bile salt medium containing 10% glycerol (for cryoprotection) and Pityrosporum orbiculare was harvested for freeze-fracture replication. Resting and budding cells were senn. The overall pattern of indentation of the plasma membrane towards the cytoplasm was well revealed as a spiral arrangement of major indentations with minor ones disposed more or less at right angles. Hexagonally-arranged particles were not observed. Mitochondria were elongated with constrictions at intervals, and the fracture face of the limiting memb

Aqueous-phase ice crystallization drives mayonnaise freeze-thaw failure: causal evidence from a non-freezing medium experiment.

Mayonnaise, a protein-stabilized oil-in-water emulsion, undergoes irreversible destabilization during freeze-thaw (FT) cycling, yet the relative contributions of aqueous-phase ice formation, protein denaturation, and fat crystallization remain debated. Here, we investigated FT destabilization using cryo-microscopy, differential scanning calorimetry (DSC), particle size analysis, interfacial protein assays, and a non-freezing control experiment. Cryo-microscopy revealed heterogeneous ice distribution, with an ice area fraction of 83% at the surface compared with 65% internally, indicating mecha

Pharmacologically Modulating the Noradrenergic Arousal System to Reduce Freezing of Gait in Parkinson's Disease: Study Protocol of the International AnTi-FREEZE Study.

Freezing of gait (FOG) is a burdensome Parkinson's disease (PD) symptom. Gait control in people with PD relies on compensation from cognitive, sensory and limbic networks. Their integration is influenced by the noradrenergic ascending arousal system, modulated by the locus coeruleus. In PD, locus coeruleus degeneration has been linked to increased FOG severity. Atomoxetine, a selective noradrenaline reuptake inhibitor, may provide a novel therapeutic approach for FOG. However, to date, studies on atomoxetine's effect on FOG have been inconsistent and had small sample sizes. This study will eva

Effects of co-supplementation of trehalose with fetuin and taxifolin in freezing extender on quality parameters of bull spermatozoa following freeze-thaw process.

Glycerol is commonly used as a cryoprotectant in bovine sperm cryopreservation; however, its cytotoxic effects can negatively impact sperm quality. This study aimed to reduce glycerol concentration in semen extenders by supplementing with natural antioxidants (taxifolin and fetuin) and the disaccharide trehalose. Twelve ejaculates were collected from three bulls and initially evaluated, then diluted in a tris-egg yolk (TEY) extender containing either 7 % or 4 % glycerol. The control group received 7 % glycerol (TEY), while experimental groups included G7F (TEY + fetuin), G7T (TEY + taxifolin),

Ultrastructural study of glomerular capillary loops at different perfusion pressures as revealed by quick-freezing, freeze-substitution and conventional fixation methods.

Wistar rat kidneys were perfused with some fixatives at different pressures of 100, 150, 200 or 250 cm H2O via the aorta and were studied by the quick-freezing and freeze-substitution (QF-FS) or deep-etching (QF-DE) methods, in addition to the conventional immersion or perfusion fixation method. Different parts of glomerular capillary walls were selected for morphometric analyses. It was demonstrated that the widths of glomerular slit diaphragms and the spaces between foot processes were more widely dilated at higher perfusion pressures (200 and 250 cm H2O) than those seen at both normal perfu

Numerical investigation of radiation-assisted freezing in nanomaterial-enhanced porous enclosures.

This investigation examines the application of a wavy porous enclosure as an effective approach for improving cold energy storage, which is of significant importance in both industrial and environmental contexts. The thermal behavior of the system is numerically evaluated using Galerkin method to enhance the efficiency of cold energy conservation. The study emphasizes three principal enhancement mechanisms: the incorporation of porous structures, the addition of hybrid nanomaterials, and the inclusion of thermal radiation effects. To simplify the numerical formulation, assumptions such as homo

Effect of freezing and cryostorage on follicular fluid vitamin D levels.

In the studies assessing the relationship between vitamin D levels and pregnancy outcomes in in vitro fertilization, doctors sometimes collected and froze the follicular fluid (fluid around eggs in a woman's ovaries) so they can test it later for research. This study aimed to find out if freezing and storage changes the level of vitamin D in that fluid, which could affect how reliable the tests are. We collected follicular fluid of 20 women going through in vitro fertilization. The samples were split into portions and were frozen at two different temperatures: -20 and -80°C. Vitamin D levels w

Molecular mechanism of cold acclimation regulating freezing tolerance in Prunus mume.

The Prunus mume species has undergone a long process of domestication and introduction in China, resulting in marked differences in freezing tolerance among populations across a latitudinal gradient of approximately 1,000 km. However, the introduction and domestication process is time-consuming, constraining the efficiency of species introduction. In this study, P. mume samples with the same genetic background spanning over 1,000 km from north to south were selected as experimental materials. The molecular mechanisms underlying cold-acclimation-mediated freeze tolerance in P. mume were investi

Deciphering Freezing of Gait: What Neuropathology Reveals About an Episodic Phenomenon.

Freezing of gait (FoG) is an episodic, debilitating motor phenomenon defined as paroxysmal episodes wherein there is an inability to step effectively, despite attempting to do so. Although FoG is most commonly associated with Parkinson's disease, it manifests across a spectrum of progressive neurodegenerative and potentially transient non-neurodegenerative conditions that affect overlapping locomotor circuits. The episodic nature of FoG implies transient disruptions of a distributed locomotor network rather than fixed structural lesions, rendering its neuropathological basis particularly chall

Observation of Topological Chirality Switching Induced Freezing of a Skyrmion Crystal.

Magnetic skyrmions are topologically protected quasi-particles with a well-defined chirality. Control over their chirality is proposed as an additional feature for encoding data bits or as qubits in quantum computing due to their high efficiency and stability against achiral magnetic textures. Here it is shown that an in-plane magnetic field can be utilized to reshape the energy barriers between different skyrmionic bubbles (e.g., Bloch type, type-II) enabling spontaneous chirality fluctuations with a frequency that increases with the strength of the in-plane field. The insulating van der Waal
