Mycotoxin exposure from food occurs globally but is more common in hot humid environments, especially in low-income settings, and might affect pregnancy outcomes. This study aimed to synthesize the evidence from epidemiological studies on the relationship between maternal or fetal exposure to different mycotoxins and the occurrence of adverse pregnancy outcomes. Multiple databases were systematically searched up to December 2018 to identify studies that assessed the association between mycotoxin exposure in pregnant women or fetuses and at least one pregnancy outcome. Studies were appraised an

OBJECTIVE: Neutrophils promote experimental abdominal aortic aneurysm (AAA) formation via a mechanism that is independent from MMPs (matrix metalloproteinases). Recently, we reported a dominant role of IL (interleukin)-1β in the formation of murine experimental AAAs. Here, the hypothesis that IL-1β-induced neutrophil extracellular trap formation (NETosis) promotes AAA was tested.
APPROACH AND RESULTS: NETs were identified through colocalized staining of neutrophil, Cit-H3 (citrullinated histone H3), and DNA, using immunohistochemistry. NETs were detected in human AAAs and were colocalized wit

Some venom components are known to present potential biological activities that are useful as tools in therapeutics. In this study anti-tumoral activity of Androctonus australis hector (Aah) venom and its purified fraction on early step of hepato-carcinogenesis initiated by Fumonisin (FB1), was tested. Initiated hepatic tumor was assessed in mice by decreased doses of Fumonisin B1 associated to phenobarbital. Scorpion venom was used to investigate its activity on initiated tumor by FB1. Evaluation of oxidative unbalance, enzymatic activities and DNA quantification in the liver were correlated

There is a critical need to better understand the patterns, levels and combinatory effects of exposures we are facing through our diet and environment. Mycotoxin mixtures are of particular concern due to chronic low dose exposures caused by naturally contaminated food. To facilitate new insights into their role in chronic disease, mycotoxins and their metabolites are quantified in bio-fluids as biomarkers of exposure. Here, we describe a highly sensitive urinary assay based on ultra-high performance liquid chromatography - tandem mass spectrometer (UHPLC-MS/MS) and 13C-labelled or deuterated i

Four water-soluble Fusarium metabolites (fumonisin B1, fusaric acid, butenolide and moniliformin), water-insoluble pigment (8-O-methylbostrycoidin), and an Alternaria metabolite (AAL-toxin) were tested for relative cytotoxicity to five established mammalian cell lines. Butenolide was the most cytotoxic to all five cell lines. LC50s were; 1 microgram/ml to rat hepatoma (RH) (tumors derived from parenchymal cells), 7 micrograms/ml to baby hamster kidney (BHK-21) fibroblast cells, and 15 micrograms/ml to McCoy mouse (MM) fibroblast cells: LC100s were 1 microgram/ml to Chinese hamster ovary (CHO)

Here, we present a protocol to describe the bacteriome of the intestinal content of toxin-exposed fallow deer. We describe steps for measuring fecal mycotoxin (deoxynivalenol, zearalenone, fumonisin B1, and aflatoxin B1) levels using liquid chromatography-mass spectrometry, as well as serum glyphosate. We then detail a short-read shotgun DNA sequencing-based bioinformatic pipeline for the toxin level-associated analysis of the bacteriome and resistome and the construction of metagenome-assembled bacterial genomes. This protocol has potential applications in further toxin level-associated metag

Analysis of melatonin (MEL) in pineal cell cultures by enzyme linked immunosorbent assay showed its concentration was increased by fusaric acid (FA), a mycotoxin produced by Fusarium species and associated with toxic duck and ostrich feeds. Subsequent cell culture studies demonstrated the precursors of MEL, 5-hydroxytryptophan (5HTP) and serotonin (5HT), were also affected by FA as well as other Fusarium mycotoxins. Herein we describe a technique for the analysis of 5HTP and 5HT in pineal cell cultures using HPLC with electrochemical detection (EC), and report on the effects of FA alone and in

In Brazil, in the last 20 years, dietary risk assessments have been conducted on pesticides, mycotoxins, food additives, heavy metals (mainly mercury), environmental contaminants (mainly DDT) and acrylamide, a compound formed during food processing. The objectives of this paper were to review these studies, discuss their limitations and uncertainties and identify the most critical chemicals that may pose a health risk to Brazilian consumers. The studies have shown that the cumulative intake of organophosphorus and carbamate pesticides by high consumers of fruits and vegetables may represent a

Fungal keratitis (FK) is a vision-threatening infection in which Fusarium species not only invade the cornea but may also release mycotoxins that exacerbate tissue injury. Here, we established a corneal exposure model to delineate the direct effects of Fusarium-derived mycotoxins independent of live fungal infection, using deoxynivalenol (DON), fumonisin B1 (FB1), and T-2 toxin as representative compounds. Hstological analyses revealed that all three mycotoxins markedly impaired corneal epithelial repair, induced stromal disorganization, and promoted inflammatory cell infiltration. At the mole

Mycotoxins threaten food safety, public health, and trade in Africa, where cereals are staple foods largely produced and consumed through informal systems. This scoping review synthesizes evidence on pre- and post-harvest mycotoxin management in African cereal systems and translates it into an integrated adaptive framework for policy and practice. A structured literature search conducted in Google Scholar, Web of Science, and Scopus identified 9,817 records, of which 33 primary studies met the eligibility criteria. The included studies covered interventions across the cereal value chain, inclu

BACKGROUND: Outbreaks of liver disease in horses are common but the etiology of most remains unknown. Forage mycotoxins have been suspected to be a cause.
OBJECTIVES: To examine the association between outbreaks of liver disease and the presence of mycotoxins in forage stored on the same premises.
ANIMALS: Premises were identified where ≥4 horses were contemporaneously affected by liver disease, and a control group was formed from premises where ≥4 horses had been examined and found to have no evidence of liver disease.
METHODS: Forage was collected from 29 case and 12 control premises. The

Consumption of fruit juice is becoming trendy for consumers seeking freshness and high vitamin and low caloric intake. Mycotoxigenic moulds may infect fruits during crop growth, harvest, and storage leading to mycotoxin production. Many mycotoxins are resistant to food processing, which make their presence in the final juice product very likely expected. In this way, the presence of 30 mycotoxins including aflatoxin B1 (AFB1), aflatoxin B2 (AFB2), aflatoxin G1 (AFG1), aflatoxin G2 (AFG2), alternariol (AOH), alternariol monomethyl ether (AME), Ochratoxin A (OTA), fumonisin B1 (FB1), fumonisin B

Maize is susceptible to mycotoxin contamination throughout harvesting, storage, and processing, which poses potential risks to human health and livestock production. This review summarizes the occurrence, toxicological mechanisms, and contamination dynamics of major mycotoxins commonly detected in maize, including aflatoxins (AFs), ochratoxin A (OTA), zearalenone (ZEN), fumonisins (FBs), and deoxynivalenol (DON). We further examine their distribution and accumulation patterns across the maize production chain, from pre-harvest to processing. During the pre-harvest period, toxin accumulation is

Dietary exposure to both processing contaminants and natural toxins in fried foods poses a potential health risk, but analytical methods for their simultaneous monitoring are lacking. To address this gap, a rapid and efficient method based on QuEChERS extraction and UPLC-MS/MS detection was developed for the simultaneous determination of two β-carboline heterocyclic amines (harman and norharman) and seven mycotoxins (fumonisins B1, B2, B3 and aflatoxins B1, B2, G1, G2). The optimized extraction utilized a 30:20:50 (v/v/v) acetonitrile/methanol/water mixture. The method was rigorously validated

Mycotoxins are toxic and carcinogenic metabolites produced by fungi that colonize food crops. The most agriculturally important mycotoxins known today are aflatoxins, which cause liver cancer and have also been implicated in child growth impairment and acute toxicoses; fumonisins, which have been associated with esophageal cancer (EC) and neural tube defects (NTDs); deoxynivalenol (DON) and other trichothecenes, which are immunotoxic and cause gastroenteritis; and ochratoxin A (OTA), which has been associated with renal diseases. This review describes the adverse human health impacts associate

The European corn borer (ECB) plays an important role in promoting Fusarium verticillioides infections and in the consequent fumonisin contamination in maize grain in temperate areas. The objective of this study was to evaluate whether the ECB feeding activity could also affect the occurrence of emerging mycotoxins in maize kernels. During the 2008-10 period, natural infestation of the insect was compared, in field research, with the protection of infestation, which was obtained by using an entomological net. The ears collected in the protected plots were free from ECB attack, while those subj

Fusarium verticillioides and Aspergillus parasiticus are typical fungi of stored maize. To prevail over other organisms when competing for a substrate, fungi emit volatile organic compounds (VOCs) with different bioactivities. The aim of this study was to evaluate A. parasiticus and F. verticillioides interaction in maize grains stored in silo bags at different initial humidities (iH; 7.5% or 28%) for 28 days. For each treatment, dry grain weight loss, maize fungal infection, filamentous fungi count, ergosterol content, FB1 and AFB1 content, and VOCs were determined. In silo bags, A. parasitic

Mycotoxin contamination in feed poses a major threat to animal health and productivity, particularly in pigeons. This study evaluated the protective effects of three mycotoxin biodegradation agents (MBAs) in pigeons subchronically co-exposed to a mycotoxin-contaminated (MYC) diet for 60 days. The MYC diet was prepared by mixing 3% naturally mold-contaminated corn with 97% basal diet, yielding final dietary concentrations of fumonisin B1 (7829.41 µg/kg), aflatoxin B1 (201.16 µg/kg), zearalenone (1675.46 µg/kg), T-2 toxin (7721.41 µg/kg), and deoxynivalenol (1211.77 µg/kg).
