Four new triterpenoids from Ganoderma lucidum inhibit fibroblast-myofibroblast differentiation: Ganolanoid a identified as an ESR1 ligand.

Four previously unreported triterpenoids, ganolanoids A-D (1-4), were isolated from Ganoderma lucidum. Their structures, including relative and absolute configurations, were elucidated by IR, UV, HRESIMS, 1D/2D NMR, and X-ray crystallography. The antifibrotic potential of these compounds was evaluated in a TGF-β1-induced activation model using primary mouse lung fibroblasts (MLFs). Compounds 1-3 significantly suppressed the upregulation of the fibrosis marker α-SMA. To explore the underlying mechanism of the most active compound, ganolanoid A (1), an integrated computational approach combining

Ganoderma lucidum polysaccharides ameliorates D-galactose-induced aging salivary secretion disorders by upregulating the rhythm and aquaporins.

Longer-term deterioration in saliva secretion has been observed to occur in response to aging. The functional deterioration of the salivary gland damages swallowing and chewing abilities and consequently reduces life quality of the elderly. There are, however, only a few proven effective treatments for aging salivary secretion disorders. Ganoderma lucidum polysaccharide (GLP) has been applied to treat various diseases because of its safety, efficacy, and low cost. We investigated the protective effect of GLP on the submandibular gland (SMG) during aging. D-galactose (D-gal) was used to treat t

Z16b, a natural compound from Ganoderma cochlear is a novel RyR2 stabilizer preventing catecholaminergic polymorphic ventricular tachycardia.

Catecholaminergic polymorphic ventricular tachycardia (CPVT) is an inherited, lethal ventricular arrhythmia triggered by catecholamines. Mutations in genes that encode cardiac ryanodine receptor (RyR2) and proteins that regulate RyR2 activity cause enhanced diastolic Ca2+ release (leak) through the RyR2 channels, resulting in CPVT. Current therapies for CPVT are limited. We found that Z16b, a meroterpenoid isolated from Ganoderma cochlear, inhibited Ca2+ spark frequency (CaSF) in R2474S/ + cardiomyocytes in a dose-dependent manner, with an IC50 of 3.2 μM. Z16b also dose-dependently suppressed

Enzyme-Assisted Extraction of Polysaccharides from Steam-Exploded Ganoderma lucidum and Its Yield, Structural Characterisation, and Immunomodulatory Activity.

Polysaccharides are the primary bioactive constituents of Ganoderma lucidum. Nevertheless, the industrial potential of this species is constrained by the fact that yields are relatively low. The present study developed a technology and method of extracting polysaccharides from steam-exploded Ganoderma lucidum by enzymatic extraction, with the aim of increasing the yield. The yield of crude polysaccharide is 15.39%, which is 6.96 times that of the traditional water method (2.21%). A purified, homogeneous polysaccharide fraction, designated GLP-L, was isolated with a molecular weight of 5.59 × 1

Ganoderma lucidum low molecular weight polysaccharide promotes the repair of spinal cord injury through anti-inflammatory and antioxidant.

Oxidative stress after spinal cord injury (SCI) significantly promotes neuronal apoptosis. The imbalance of flora leads to the destruction of the intestinal barrier, so that pro-inflammatory cytokines enter the blood and further aggravate neuroinflammation. The change of microglia phenotype can regulate neuroinflammation. The purpose of this study is to explore the characteristics of Ganoderma lucidum low molecular weight polysaccharides peptide (GL-LPP) in regulating intestinal flora, anti-inflammatory, anti-oxidation and anti-apoptosis, and their role and mechanism in the treatment of SCI.
