Molecular mechanisms of fluconazole resistance in Candida parapsilosis isolates from a U.S. surveillance system.

Candida parapsilosis is the second or third most common cause of candidemia in many countries. The Infectious Diseases Society of America recommends fluconazole as the primary therapy for C. parapsilosis candidemia. Although the rate of fluconazole resistance among C. parapsilosis isolates is low in most U.S. institutions, the resistance rate can be as high as 7.5%. This study was designed to assess the mechanisms of fluconazole resistance in 706 incident bloodstream isolates from U.S. hospitals. We sequenced the ERG11 and MRR1 genes of 122 C. parapsilosis isolates with resistant (30 isolates;

Cryptococcus neoformans: the yeast that likes it hot.

The ability of fungi to grow well at mammalian body temperatures is a fundamental characteristic of invasive human fungal pathogens. Cryptococcus neoformans, with its genetics, molecular biology, robust animal models and clinical importance, has become a premier fungal model system for molecular fungal pathogenesis studies. There has been over a half century of study into how C. neoformans grows at high temperatures. However, recently the understanding of high-temperature growth at a molecular level has dramatically accelerated. Many strategies have been used to identify genes and over a dozen

Induction of pan-azole resistance in Cryptococcus neoformans by agrochemical azole uniconazole through upregulation of efflux and chromosomal disomies.

Cryptococcus neoformans is a fungus naturally found in the environment, particularly in soil, bird droppings, and trees. Cryptococcosis, caused by Cryptococcus spp., primarily C. neoformans and C. gattii, poses a significant threat to human health. Agrochemicals are widely used worldwide, and most applied agrochemicals are dispersed into the environment, which can have direct and indirect effects on human health. This study investigates the impact of the plant growth regulator uniconazole (UCZ) on antifungal resistance in C. neoformans using the H99 laboratory strain. We found UCZ to have pote

CTG clade-specific proteins of the RSC chromatin-remodeling complex regulate cell cycle progression of a critical priority fungal pathogen, Candida albicans.

The remodels the structure of chromatin (RSC) and the homologous chromatin-remodeling complexes are known to regulate cell cycle progression in various organisms, including Saccharomyces cerevisiae, Drosophila, and Homo sapiens. In this work, we characterized the role of two novel CTG clade-specific proteins (Nri1 and Nri2) of the RSC complex in the regulation of cell cycle progression in a critical priority fungal pathogen, Candida albicans. We observed that Nri1, alone or along with Nri2, regulates cell cycle progression at multiple stages. The nri1Δ/Δ and nri1Δ/Δ nri2Δ/Δ mutants exhibited t

Wavelength-dependent regulation of Penicillium expansum: red and green light promote fungal fitness and patulin accumulation via metabolic homeostasis.

Light-based regulation is increasingly employed as an environmentally sustainable physical intervention for preserving postharvest agricultural commodities during storage and transit. Nevertheless, incident light can concurrently modulate the physiological and biochemical dynamics of phytopathogenic fungi, thereby introducing unintended risks to preservation outcomes. In this study, we constructed a customized illumination system integrating discrete wavelengths (green light: 540 nm; red light: 660 nm) and photoperiods (DD, control: continuous darkness; LD: 12 h light/12 h dark cycles; LL: con

N-acetylglucosamine sensing in the filamentous soil fungus Trichoderma reesei.

N-acetylglucosamine (GlcNAc) is involved in diverse signaling pathways in dimorphic yeasts and bacteria and is related to morphogenetic switching, mating, stress, virulence, and cell death. Recently, GlcNAc has been shown to promote plant growth by shaping the bacterial soil community. However, the role of GlcNAc sensing in filamentous soil fungi has not been investigated. By using Trichoderma reesei as a model organism, we show here that GlcNAc impacts the expression of around 2100 genes. Carbohydrate metabolism, amino acid metabolism, and secondary metabolism were the three most strongly aff

Methanol-induced transcription factor Mpp1 regulates the coordinated expression of multiple genes to achieve a balanced C1 metabolism in the methylotrophic yeast Candida boidinii.

UNLABELLED: Methylotrophic yeasts can grow on methanol as the sole carbon and energy source. They have been used as hosts for heterologous protein production, which is achieved by their strongly and tightly regulated methanol-induced gene promoters. The expression levels of methanol-induced genes, such as AOD1 and DAS1, are regulated by concentration-regulated methanol induction (CRMI). In this study, we identified CbMpp1 as a methanol-induced transcription factor necessary for the expression of multiple genes in the methylotrophic yeast Candida boidinii and identified the upstream activating

A unique metabolic gene cluster regulates lactose and galactose metabolism in the yeast Candida intermedia.

Lactose assimilation is a relatively rare trait in yeasts, and Kluyveromyces yeast species have long served as model organisms for studying lactose metabolism. Meanwhile, the metabolic strategies of most other lactose-assimilating yeasts remain unknown. In this work, we have elucidated the genetic determinants of the superior lactose-growing yeast Candida intermedia. Through genomic and transcriptomic analyses, we identified three interdependent gene clusters responsible for the metabolism of lactose and its hydrolysis product galactose: the conserved LAC cluster (LAC12, LAC4) for lactose upta

Analysis of Paracoccidioides lutzii mitochondria: a proteomic approach.

The genus Paracoccidioides is composed of thermal dimorphic fungi, causative agents of paracoccidioidomycosis, one of the most frequent systemic mycoses in Latin America. Mitochondria have sophisticated machinery for ATP production, which involves metabolic pathways such as citric acid and glyoxylate cycles, electron transport chain and oxidative phosphorylation. In addition, this organelle performs a variety of functions in the cell, working as an exceptional metabolic signalling centre that contributes to cellular stress responses, as autophagy and apoptosis in eukaryotic organisms. The aim

Therapeutic potential of thiazolidinedione-8 as an antibiofilm agent against Candida albicans.

Candida albicans is known as a commensal microorganism but it is also the most common fungal pathogen in humans, causing both mucosal and systemic infections. Biofilm-associated C. albicans infections present clinically important features due to their high levels of resistance to traditional antifungal agents. Quorum sensing is closely associated with biofilm formation and increasing fungal pathogenicity. We investigated the ability of the novel bacterial quorum sensing quencher thiazolidinedione-8 (S-8) to inhibit the formation of, and eradication of mature C. albicans biofilms. In addition,

Involvement of transposon-like elements in penicillin gene cluster regulation.

Subtelomeric secondary metabolite (SM) gene clusters are frequently surrounded by DNA repeats and transposon-like elements. The Aspergillus nidulans penicillin cluster, 30kb from the telomere of chromosome VI, is bordered by such elements. Deletions of penicillin telomere proximal and distal border regions resulted in decreased penicillin production. A 3.7kb distal region called PbIa, consisting of the putative transposable element DNA-2, was examined further where its replacement by a pyrG marker presented a similar phenotype as loss of the global SM regulator LaeA, resulting in a decrease in

Upregulation of glyoxylate cycle genes upon Paracoccidioides brasiliensis internalization by murine macrophages and in vitro nutritional stress condition.

Paracoccidioides brasiliensis, the etiologic agent of paracoccidioidomycosis, is a facultative intracellular human pathogen that can persist within macrophage phagolysosomes, indicating that the fungus has evolved defense mechanisms in order to survive under nutritionally poor environments. The analysis of P. brasiliensis transcriptome revealed several virulence factor orthologs of other microorganisms, including the glyoxylate cycle genes. This cycle allows the utilization of two-carbon (C2) compounds as carbon source in gluconeogenesis. Semiquantitative RT-PCR analyses revealed that these ge

Krüppel-like Factor 4 modulates interleukin-6 release in human dendritic cells after in vitro stimulation with Aspergillus fumigatus and Candida albicans.

Invasive fungal infections are associated with high mortality rates and are mostly caused by the opportunistic fungi Aspergillus fumigatus and Candida albicans. Immune responses against these fungi are still not fully understood. Dendritic cells (DCs) are crucial players in initiating innate and adaptive immune responses against fungal infections. The immunomodulatory effects of fungi were compared to the bacterial stimulus LPS to determine key players in the immune response to fungal infections. A genome wide study of the gene regulation of human monocyte-derived dendritic cells (DCs) confron

Yeast-secreted compounds with antifungal activity-screening, genetic parts, biosynthetic pathways, and regulation.

Awareness is rising that antifungal resistance poses a threat to agriculture, food safety, biodiversity, and human health. There is a limited number of antifungals available and resistance to all of them has been reported. The development of novel antifungals is complex, as eukaryotic organisms have very few selective drug targets that distinguish them from the infected plant, human, or animal host. Yeasts produce different compounds with antifungal activity, ranging from small molecules such as iron chelators, biosurfactants, and volatile organic compounds, to proteins like myocins and hydrol

Moh1 coordinates ROS-dependent apoptosis in genotoxic stress response of Candida albicans.

Candida albicans employs apoptosis to maintain genomic stability under genotoxic stress, yet its regulatory mechanisms remain poorly defined. Here, we characterize the role of a putative pro-apoptotic factor Moh1 in C. albicans. Our findings reveal that MOH1 transcription is significantly upregulated under genotoxic stress and in DNA repair-deficient backgrounds (e.g., rad52Δ). Paradoxically, MOH1 deletion enhances resistance to genotoxic agents, improving cellular survival and reducing apoptosis, as evidenced by decreased nuclear condensation. Mechanistically, moh1Δ cells exhibit reduced intr

Expanded Gene Regulatory Network Reveals Potential Light-Responsive Transcription Factors and Target Genes in Cordyceps militaris.

Cordyceps militaris, a fungus widely used in traditional Chinese medicine and pharmacology, is recognized for its abundant bioactive compounds, including cordycepin and carotenoids. The growth, development, and metabolite production in various fungi are influenced by the complex interactions between regulatory cascades and light-signaling pathways. However, the mechanisms of gene regulation in response to light exposure in C. militaris remain largely unexplored. This study aimed to identify light-responsive genes and potential transcription factors (TFs) in C. militaris through an integrative

Antifungal Activity of Methyl 2-Methylbutyrate Against Botrytis cinerea and the Induced Physiological, Transcriptional, and Metabolic Responses.

Gray mold, caused by the necrotrophic fungus Botrytis cinerea, is a devastating disease responsible for substantial global crop losses. The escalating threat of fungicide resistance and environmental concerns necessitates the development of eco-compatible control alternatives. In this study, we investigated the antifungal efficacy of methyl 2-methylbutyrate (M2M), a microbial volatile organic compound (mVOC), against B. cinerea, and elucidated the underlying physiological, transcriptomic, and metabolomic responses. Our results demonstrated that M2M significantly inhibited mycelial growth in a

Effects and Mechanisms of Sodium Nitroprusside, Spermidine, and Coumarin Addition In Vitro on Epichloë sinensis.

Epichloë sinensis forms mutualistic symbiosis with the Chinese native grass Festuca sinensis. However, the mechanisms by which E. sinensis responds to host-derived defense signals remain unclear. To explore these responses, three compounds, sodium nitroprusside, spermidine, and coumarin, were added to a potato dextrose broth (PDB) medium to mimic host defense-related signals. Their effects on the growth, antioxidant capacity, and gene expression of three E. sinensis strains (1, 2, and 84F) isolated from different ecotypes of F. sinensis were identified. The results showed that in PDB, certain

Small RNA Isolation and Library Construction for Expression Profiling of Small RNAs from Neurospora crassa and Fusarium oxysporum and Analysis of Small RNAs in Fusarium oxysporum-Infected Plant Root Tissue.

Due to crucial roles in gene regulation, noncoding small RNAs (sRNAs) of 20-30 nucleotides (nt) have been intensively studied in mammals and plants and are implicated in significant diseases and metabolic disorders. Elucidation of biogenesis mechanisms and functional characterization of sRNAs is often achieved using tools such as separation of small-sized RNA and deep sequencing. Although RNA interference pathways, such as quelling and meiotic silencing, have been well-described in Neurospora crassa, knowledge of sRNAs in other filamentous fungi is still limited compared to other eukaryotes. Nitrogen Catabolite Repression in members of Paracoccidioides complex.

Paracoccidioides complex is a genus that comprises pathogenic fungi which are responsible by systemic disease Paracoccidioidomycosis. In host tissues, pathogenic fungi need to acquire nutrients in order to survive, making the uptake of nitrogen essential for their establishment and dissemination. Nitrogen utilization is employed by the alleviation of Nitrogen Catabolite Repression (NCR) which ensures the use of non-preferential or alternative nitrogen sources when preferential sources are not available. NCR is controlled by GATA transcription factors which act through GATA binding sites on pro
