A Highly Efficient Agrobacterium-Mediated Method for Transient Gene Expression and Functional Studies in Multiple Plant Species.

Although the use of stable transformation technology has led to great insight into gene function, its application in high-throughput studies remains arduous. Agro-infiltration have been widely used in species such as Nicotiana benthamiana for the rapid detection of gene expression and protein interaction analysis, but this technique does not work efficiently in other plant species, including Arabidopsis thaliana. As an efficient high-throughput transient expression system is currently lacking in the model plant species A. thaliana, we developed a method that is characterized by high efficiency

Integrative Analysis of Transcriptomic and Proteomic Changes Related to Cytoplasmic Male Sterility in Spring Stem Mustard (Brassica juncea var. tumida Tsen et Lee).

The development of flower and pollen is a complex biological process that involves multiple metabolic pathways in plants. In revealing novel insights into flower and pollen development underlying male sterility (MS), we conducted an integrated profiling of gene and protein activities in developing buds in cytoplasmic male sterile (CMS) mutants of mustard (Brassica juncea). Using RNA-Seq and label-free quantitative proteomics, 11,832 transcripts and 1780 protein species were identified with significant differential abundance between the male sterile line 09-05A and its maintainer line 09-05B at

mRNA Transcript abundance during plant growth and the influence of Li(+) exposure.

Lithium (Li) toxicity in plants is, at a minimum, a function of Li(+) concentration, exposure time, species and growth conditions. Most plant studies with Li(+) focus on short-term acute exposures. This study examines short- and long-term effects of Li(+) exposure in Arabidopsis with Li(+) uptake studies and measured shoot mRNA transcript abundance levels in treated and control plants. Stress, pathogen-response and arabinogalactan protein genes were typically more up-regulated in older (chronic, low level) Li(+)-treatment plants and in the much younger plants from acute high-level exposures. Integrated WGCNA and TO-GCN analysis reveals pathways and genes underlying salt tolerance in Sesbania cannabina.

Salt stress seriously threatens the growth and development of plants. To investigate how Sesbania cannabina responds to salt stress, we constructed a temporal regulatory response network under salt stress by integrating the physiological and transcriptomic data from 'Zhongke Jing No.1' at five time points (0, 6, 12, 24, and 48 h, n = 3) after salt treatment. The results indicated that salt stress increased the relative electrical conductivity (REC) and malondialdehyde (MDA) content, induced the accumulation of osmotic regulators, and enhanced antioxidant enzyme activities. A total of 688,424,7

Homoeologous exchange-associated ABA catabolism rewiring contributes to salinity tolerance in a synthetic tetraploid rice.

Altered ABA-related regulation is associated with transgressive salinity tolerance in a synthetic tetraploid rice carrying extensive post-polyploidy homoeologous exchanges. Plant polyploids often show higher environment resilience than their congeneric diploid progenitors. However, it remains unclear whether these traits arise directly from whole-genome duplication (WGD) or evolve through post-WGD changes. Here, we show that transgressive salt tolerance occurs in ca. 1.71% of an early-generation synthetic rice tetraploid from japonica-indica subspecies hybridization. Genome resequencing unrave

A four-dimensional spatial transcriptome atlas of barley caryopsis development and germination.

A 4-dimensional spatial gene expression atlas of Hordeum vulgare (barley) grain development and germination was generated using spatial transcriptomic analysis of serial sections to reconstruct transcript abundance in 3 physical dimensions and with temporal kinetics. We investigated the subtissue localizations of specific biological activities, using energy biology as an example, including genes encoding proteins involved in starch synthesis and degradation, sugar transport, and mitochondrial and chloroplast activity. This atlas revealed different patterns in gene expression across tissues and

Chlorophyll decomposition is accelerated in banana leaves after the long-term magnesium deficiency according to transcriptome analysis.

Magnesium (Mg) is an essential macronutrient for plant growth and development. Physiological and transcriptome analyses were conducted to elucidate the adaptive mechanisms to long-term Mg deficiency (MD) in banana seedlings at the 6-leaf stage. Banana seedlings were irrigated with a Mg-free nutrient solution for 42 days, and a mock control was treated with an optimum Mg supply. Leaf edge chlorosis was observed on the 9th leaf, which gradually turned yellow from the edge to the interior region. Accordingly, the total chlorophyll content was reduced by 47.1%, 47.4%, and 53.8% in the interior, ce

Transcriptome analysis reveals MYB and WRKY transcription factors involved in banana (Musa paradisiaca AA) magnesium deficiency.

The banana development was inhibited under the long-term magnesium deficiency (MD) stress, resulting in the leaf chlorosis. MYB108 and WRKY75 are involved in regulating the growth and development of banana leaves and roots under long-term MD. Magnesium deficiency (MD) causes plant growth inhibition, ageing acceleration, yield reduction and quality decline of banana (Musa paradisiaca AA), but the molecular regulatory mechanisms underlying the changes in response to long-term MD conditions remain unknown. In this study, a long-term MD experiment was performed with banana seedlings at the four-le

The degree and direction of subgenome expression bias in Brassica napus vary between cultivars, tissues and developmental stages.

Most extant plant lineages show evidence of past polyploidisation events in their genomes. Allopolyploids arise from hybridisation, resulting in polyploid genomes comprising subgenomes from different ancestors. Adaptation to their environment, pre- and post-hybridisation and selection for specific traits has led to several allopolyploids exhibiting an unequal contribution from their subgenomes to phenotypic traits. Associated regulatory changes may have given rise to cultivar-specific subgenome dominance that contributes to the diversity of phenotypes. Likewise, different tissues and developme

Chemical application improves stress resilience in plants.

In recent years, abiotic stresses, including droughts, floods, high temperatures, and salinity, have become increasingly frequent and severe. These stresses significantly hinder crop yields and product quality, posing substantial challenges to sustainable agriculture and global food security. Simultaneously, the rapidly growing global population exacerbates the need to enhance crop production under worsening environmental conditions. Consequently, the development of effective strategies to strengthen the resilience of crop plants against high temperatures, water scarcity, and extreme environme

Expression of BoNOL and BoHCAR genes during postharvest senescence of broccoli heads.

BACKGROUND: Chlorophyll is the most abundant pigment on Earth, essential for the capture of light energy during photosynthesis. During senescence, chlorophyll degradation is highly regulated in order to diminish toxicity of the free chlorophyll molecule due to its photoactivity. The first step in the chlorophyll degradation pathway is the conversion of chlorophyll b to chlorophyll a by means of two consecutive reactions catalyzed by enzymes coded by NYC1 (NON-YELLOW COLORING 1), NOL (NYC1-LIKE) and HCAR. RESULTS: In this work, we studied the expression of NOL and HCAR genes during postharvest

SDG721 and SDG705 are required for rice growth.

H3K4me3 plays important roles in development, transcription, and environmental responses. Here, we report that SDG721 (SET-domain group protein 721) and SDG705 are involved in regulating rice development. SDG721 and SDG705 encode TRITHORAX-like proteins, which appear to modulate H3K4 methylation levels. Loss of SDG721 and SDG705 function resulted in GA-deficient phenotypes, including semi-dwarfism, reduced cell length, and reduced panicle branching. The transcripts levels and H3K4me3 levels of GA biosynthesis genes and GA signaling pathway genes were downregulated in the sdg721 sdg705 plants.

Melatonin and Its Effects on Plant Systems.

Melatonin (N-acetyl-5-methoxytryptamine) is a nontoxic biological molecule produced in a pineal gland of animals and different tissues of plants. It is an important secondary messenger molecule, playing a vital role in coping with various abiotic and biotic stresses. Melatonin serves as an antioxidant in postharvest technology and enhances the postharvest life of fruits and vegetables. The application of exogenous melatonin alleviated reactive oxygen species and cell damage induced by abiotic and biotic stresses by means of repairing mitochondria. Additionally, the regulation of stress-specifi

Revisit and explore the ethylene-independent mechanism of sex expression in cucumber (Cucumis sativus).

This review provides a thorough and comprehensive perspective on the topic of cucumber sexual expression. Specifically, insights into sex expression mediated by pathways other than ethylene are highlighted. Cucumber (Cucumis sativus L.) is a common and important commercial crop that is cultivated and consumed worldwide. Additionally, this species is commonly used as a model for investigating plant sex expression. Cucumbers exhibit a variety of floral arrangements, comprising male, female, and hermaphroditic (bisexual) flowers. Generally, cucumber plants that produce female flowers are typicall

Auxin participates in regulating the leaf curl development of Wucai (Brassica campestris L.).

Wucai (Brassica campestris L. ssp. chinensis var. rosularis Tsen) belongs to the Brassica genus of the Cruciferae family, and its leaf curl is a typical feature that distinguishes Wucai from other nonheading cabbage subspecies. Our previous research found that plant hormones were involved in the development of the leaf curl in Wucai. However, the molecular mechanisms and the hormones regulating the formation of leaf curl in Wucai have not yet been reported. This study aimed to understand the molecular functions related to hormone metabolism during the formation of leaf curl in Wucai. A total o

Genome-Wide Identification and Functional Characterization of the TIFY Gene Family Associated with Abiotic Stress and Hormone Response in Carthamus tinctorius L.

Jasmonate-ZIM domain (JAZ) proteins act as core negative regulators of jasmonic acid signaling and mediate plant stress adaptation, developmental progression, and secondary metabolism. Safflower (Carthamus tinctorius L.) is an economically important Asteraceae crop with medicinal, edible oil, and ornamental values; however, the evolutionary characteristics and molecular functions of its TIFY gene family remain poorly characterized. In the present study, we performed a genome-wide identification of the safflower CtTIFY family, followed by systematic analyses of phylogenetic relationships, chrom

Ultra-low concentration of chlorine dioxide regulates stress-caused premature leaf senescence in tobacco by modulating auxin, ethylene, and chlorophyll biosynthesis.

Exploring novel growth regulators for premature senescence regulation is important for tobacco production. In the present study, chlorine dioxide (ClO2) was explored as a novel plant growth regulator for tobacco growth, particularly its effect on leaf senescence and root development. The results showed that 0.15 μM ClO2 maintained the lushness of detached leaves and whole plants. Also, the leaves of ClO2-treated plants exhibited a chlorophyll content of 58% higher than in CK (control) plants (P < 0.05). Besides, ClO2 treatment increased the biomass of roots and aboveground parts by 54 and 16%,

Exogenous spermidine, arsenic and beta-aminobutyric acid modulate tobacco resistance to tobacco mosaic virus, and affect local and systemic glucosylsalicylic acid levels and arginine decarboxylase gene expression in tobacco leaves.

The polyamine spermidine and the metalloid arsenic increased resistance responses in the well-known pathosystem NN tobacco/tobacco mosaic virus (TMV). Both the hypersensitive response to TMV in a leaf disk model system (inoculated disks floating in the 0.1mM treatments) and systemic acquired resistance (SAR) in whole plants were significantly affected. In the latter case, 1mM foliar sprays of spermidine and arsenic were as effective as TMV and dl-beta-aminobutyric acid (BABA), both taken as positive controls, in improving the plant's response to subsequent challenge inoculation with TMV. Comparative Transcriptomic Analysis Provides Insights into PSK-α-Mediated Regulation of Diosgenin Biosynthesis in Trigonella foenum-graecum.

Diosgenin is an important steroidal sapogenin widely used for the industrial production of steroidal drugs. Phytosulfokine-α (PSK-α) is a sulfated pentapeptide hormone known to regulate plant growth, development, and stress responses. However, its role in the regulation of diosgenin biosynthesis remains unclear. Fenugreek seedlings were treated with PSK-α to investigate its effects on diosgenin biosynthesis through metabolite profiling and comparative transcriptomic analyses. PSK-α induced a dynamic response in diosgenin accumulation, characterized by an initial decrease during the early stage

Comparative transcriptomic and co-expression analyses enable the discovery of key enzymes responsible for oleuropein biosynthesis in olive (Olea europaea).

Olive (Olea europaea) is one of the most important crop trees, with olive oil being a key ingredient of the Mediterranean diet. Oleuropein, an oleoside-type secoiridoid, is the major determinant of olive oil flavor and quality. Iridoid biosynthesis has been elucidated in Catharanthus roseus, which produces secologanin-type secoiridoids, but iridoid biosynthesis in other species remains unresolved. In this work, we sequenced RNA from the fruit mesocarp of six commercial olive cultivars with various oleuropein contents during maturation and ripening. Using these data, we discovered three polyphe
