Structure and expression of Rhodnius prolixus GH18 chitinases and chitinase-like proteins: Characterization of the physiological role of RpCht7, a gene from subgroup VIII, in vector fitness and reproduction.

Chitinases are enzymes responsible for the hydrolysis of glycosidic linkages within chitin chains. In insects, chitinases are typically members of the multigenic glycoside hydrolase family 18 (GH18). They participate in the relocation of chitin during development and molt, and in digestion in detritivores and predatory insects, and they control the peritrophic membrane thickness. Chitin metabolism is a promising target for developing vector control strategies, and knowledge of the roles of chitinases may reveal new targets and illuminate unique aspects of their physiology and interaction with

Molecular identification of Lutzomyia migonei (Diptera: Psychodidae) as a potential vector for Leishmania infantum (Kinetoplastida: Trypanosomatidae).

Visceral leishmaniasis (VL) in Brazil is caused by the protozoan Leishmania infantum. This parasite is transmitted by the bite of a female sand fly. The most important sand fly species in VL transmission is Lutzomyia longipalpis. In Fortaleza, the capital of Ceará State, Brazil, the simultaneous occurrence of Lutzomyia migonei and L. longipalpis was detected in localities where VL transmission is observed. The purpose of this study was to determine conclusively if L. migonei can be found naturally infected with L. infantum in key focus in Fortaleza. Using a CDC traps we performed phlebotomine

Clocks do not tick in unison: isolation of Clock and vrille shed new light on the clockwork model of the sand fly Lutzomyia longipalpis.

BACKGROUND: Behavior rhythms of insect vectors directly interfere with the dynamics of pathogen transmission to humans. The sand fly Lutzomyia longipalpis is the main vector of visceral leishmaniasis in America and concentrates its activity around dusk. Despite the accumulation of behavioral data, very little is known about the molecular bases of the clock mechanism in this species. This study aims to characterize, within an evolutionary perspective, two important circadian clock genes, Clock and vrille. FINDINGS: We have cloned and isolated the coding sequence of L. longipalpis' genes Clock

Cyanogenic glucosides in the biological warfare between plants and insects: the Burnet moth-Birdsfoot trefoil model system.

Cyanogenic glucosides are important components of plant defense against generalist herbivores due to their bitter taste and the release of toxic hydrogen cyanide upon tissue disruption. Some specialized herbivores, especially insects, preferentially feed on cyanogenic plants. Such herbivores have acquired the ability to metabolize cyanogenic glucosides or to sequester them for use in their own predator defense. Burnet moths (Zygaena) sequester the cyanogenic glucosides linamarin and lotaustralin from their food plants (Fabaceae) and, in parallel, are able to carry out de novo synthesis of the

Genome sequence of the saprophyte Leptospira biflexa provides insights into the evolution of Leptospira and the pathogenesis of leptospirosis.

Leptospira biflexa is a free-living saprophytic spirochete present in aquatic environments. We determined the genome sequence of L. biflexa, making it the first saprophytic Leptospira to be sequenced. The L. biflexa genome has 3,590 protein-coding genes distributed across three circular replicons: the major 3,604 chromosome, a smaller 278-kb replicon that also carries essential genes, and a third 74-kb replicon. Comparative sequence analysis provides evidence that L. biflexa is an excellent model for the study of Leptospira evolution; we conclude that 2052 genes (61%) represent a progenitor ge

The role of CYP4Gs involved in hydrocarbon formation in the survival, reproduction, and pyrethroid resistance of Triatoma infestans, a major Chagas disease vector.

Very long-chain normal and branched hydrocarbons are essential for insect physiology and ecology. The final metabolic step of hydrocarbon formation involves an oxidative decarbonylation carried out by cytochrome P450 from family 4, subfamily G (CYP4G). Triatoma infestans Klug (Hemiptera: Reduviidae: Triatominae) has two CYP4G genes, named CYP4G106 and CYP4G107. In this study, we have silenced in T. infestans the expression of both genes by RNA interference in order to assess the relevance of hydrocarbons to survival, reproductive physiology, and insecticide susceptibility. Gene knockdown led t

Parasitic-Plant Parasite Rewires Flowering Pathways to Induce Stem-Derived Galls.

Gall-inducing insects manipulate host plant development, redirecting cellular fate and physiological processes to form novel structures. This phenomenon is even more intriguing when the host itself is a holoparasitic plant with minimal photosynthetic capacity. In the stem of Cuscuta campestris, the weevil Smicronyx madaranus forms galls that unexpectedly activate photosynthesis, in contrast to the typical suppression of photosynthetic activity observed in leaf-derived galls. This reversal of the usual source-to-sink transition highlights a unique form of insect-induced organogenesis. To elucid

Laccase2-Mediated Mandibular Development Is Crucial for Larval Feeding Behavior in the Silkworm, Bombyx mori.

The phenoloxidase gene Laccase2 (Lac2) is an indispensable survival gene in insects, playing key roles in cuticle tanning, exoskeleton development and pigment deposition. However, in lepidopteran insects, the roles of Lac2 in mandibular development and feeding behavior remain largely unexplored. In this study, we demonstrated that Lac2 is essential for mandibular development in early larvae of the model lepidopteran, the silkworm, Bombyx mori. We cloned and characterized BmLac2 and found that it is evolutionarily conserved in lepidopteran insects. Disruption of BmLac2 via CRISPR/Cas9 system in

Microbiome-Epigenome Interplay Impacts Microbial Symbiosis and Stress Adaptations in the Brown Planthopper (Nilaparvata lugens).

The gut microbiota and epigenetic processes both contribute to insect survival and adaptation; however, the relationship between these two systems remains largely unexplored. In this study, we used the brown planthopper (Nilaparvata lugens; BPH) to explore microbiome-epigenome interactions and evaluate its impact on BPH survivability under environmental stress. Disruption of the gut microbiome using antibiotics significantly altered the epigenetic profile of various stress-responsive genes in the BPH. Similarly, perturbations in the epigenome induced by 5-azacytidine resulted in an altered mic

Development and applications of transgenesis in the yellow fever mosquito, Aedes aegypti.

Transgenesis technology has been developed for the yellow fever mosquito, Aedes aegypti. Successful integration of exogenous DNA into the germline of this mosquito has been achieved with the class II transposable elements, Hermes, mariner and piggyBac. A number of marker genes, including the cinnabar(+) gene of Drosophila melanogaster, and fluorescent protein genes, can be used to monitor the insertion of these elements. The availability of multiple elements and marker genes provides a powerful set of tools to investigate basic biological properties of this vector insect, as well as the materi

Ultrabithorax regulates Leptinotarsa decemlineata wing development along with insulin and ecdysteroid signaling pathways.

Leptinotarsa decemlineata (Say) (Colorado potato beetle, CPB) is a notorious pest of potatoes. Its hardened forewings, known as elytra, protect the hindwings and the abdomen against enemies and chemical pesticides. Ubx plays a crucial role in differentiating insect wings, but the molecular mechanism remains unclear. Our research showed that Ubx is highly expressed in the T3 thoracic segment (T3) and appendages, while Ubx is absent in the forewings. RNA interference for Ubx resulted in the dorsal plate of the T3 being shifted toward the T2, hindwings were sheathed from the membranous in adults,

The Genome Sequence of the Rugose Spiraling Whitefly (Aleurodicus rugioperculatus Martin): Insights on Biology of an Invasive Agricultural Insect Pest and Implications for Pest Control.

The Rugose Spiraling Whitefly (RSW) (Aleurodicus rugioperculatus Martin), a pest native to Central America, infests coconut palms and has been introduced to other regions of the world including North America (e.g., Florida) and Southeast Asia. In India, RSW was first reported in 2016, and rapidly expanded to multiple states nationwide. Currently, RSW has growing global relevance as an agricultural insect pest. In addition to coconut, the RSW exhibits a broad host range, causing damage to various palms, fruit crops such as guava, vegetables, and ornamental shrubs. In this study, we present a hi

The brain atlas of a subsocial bee reflects that of eusocial Hymenoptera.

The evolutionary transition from solitary life to group-living in a society with cooperative brood care, reproductive division of labor and morphological castes is associated with increased cognitive demands for task-specialization. Associated with these demands, the brains of eusocial Hymenoptera divide transcriptomic signatures associated with foraging and reproduction to different populations of cells and also show diverse astrocyte and Kenyon cell types compared with solitary non-hymenopteran insects. The neural architecture of subsocial bees, which represent evolutionary antecedent states

OdTx12/GNA, a chimeric variant of a β excitatory toxin from Odontobuthus doriae, reveals oral toxicity towards Locusta migratoria and Tenebrio molitor.

OdTx12, a beta excitatory toxin from Odontobothus doriae had previously been identified and characterized. It had been shown that OdTx12 causes significant lethal effects on insects by injection but does not show any toxicity on mice. Due to the natural ineffectiveness of scorpion toxins to act as oral toxins, OdTx12 was fused to Galanthus nivalis agglutinin (GNA), a protein with the potential to cross the insect gut. The sequence of OdTx12/GNA gene was chemically synthesized, cloned in Escherichia coli, and expressed. The effect of the purified fusion protein (OdTx12/GNA) was assessed on the

ABHD5 regulates midgut-specific lipid homeostasis in Bombyx mori.

Lipids are an important energy source and are utilized as substrates for various physiological processes in insects. Comparative gene identification 58 (CGI-58), also known as α/β hydrolase domain-containing 5 (ABHD5), is a highly conserved and multifunctional gene involved in regulating lipid metabolism and cellular energy balance in many organisms. However, the biological functions of ABHD5 in insects are poorly understood. In the current study, we describe the identification and characterization of the ABHD5 gene in the lepidopteran model insect, Bombyx mori. The tissue expression profile i

Role of ocular albinism type 1 (OA1) GPCR in Asian gypsy moth development and transcriptional expression of heat-shock protein genes.

The ocular albinism type 1 gene, named OA1, is a coding pigment cell-specific G protein-coupled receptor exclusively localized in intracellular organelles. However, the function of OA1 in insects remains generally unknown. In the present study, we explore for the first time the function of LdOA1 in the Asian gypsy moth, Lymantria dispar. To identify the function of LdOA1 gene in the development and growth of the Asian gypsy moth, the LdOA1 gene in third instar larvae was knocked down by RNAi. Compared with the controls, the knockdown of LdOA1 increased larval mortality but did not significantl

A virus plays a role in partially suppressing plant defenses induced by the viruliferous vectors.

Herbivorous attack induces plant defenses. There is evidence that some pests suppress these defenses by interfering with signaling pathways. We here report that infestation by the white-backed planthopper, Sogatella furcifera, induces defense responses in rice and infection of the southern rice black-streaked dwarf virus in the planthoppers partially suppresses the planthopper-induced plant defenses. Salicylic acid (SA) levels generally showed a temporal increase pattern while jasmonic acid (JA) levels generally exhibited a decrease pattern in the planthopper-infested plants, irrespective of v

Paratransgenic control of vector borne diseases.

Conventional methodologies to control vector borne diseases with chemical pesticides are often associated with environmental toxicity, adverse effects on human health and the emergence of insect resistance. In the paratransgenic strategy, symbiotic or commensal microbes of host insects are transformed to express gene products that interfere with pathogen transmission. These genetically altered microbes are re-introduced back to the insect where expression of the engineered molecules decreases the host's ability to transmit the pathogen. We have successfully utilized this strategy to reduce car
