A Potential Regulatory Network of Selected Human Erythrocytic miRNAs with Plasmodium falciparum 3D7 mRNAs: A Computational Analysis.

BACKGROUND: miRNAs are small non-coding conserved RNA molecules (18-24 nts) that act as crucial gene regulators via post-transcriptional/translational modifications through interacting with the respective mRNAs during various pathophysiological conditions. Recent research has suggested that non-coding RNAs, particularly miRNAs, can be passed from one species to another to regulate gene expression. Since miRNA-mediated gene regulation has not yet been found in Plasmodia, it is hypothesized that erythrocytic miRNAs from Plasmodium falciparum (P. falciparum) could potentially migrate from the cyt

Mechanism of sepsis regulation by ELANE via macrophage polarization.

To identify the core immune regulatory gene ELANE in sepsis and explore its mechanism in regulating sepsis through macrophage polarization. Peripheral blood RNA sequencing and bioinformatics analysis were performed on 20 sepsis patients and 10 healthy controls. Differential gene expression (DEGs) screening, functional enrichment (GO/KEGG/GSEA), and protein-protein interaction (PPI) network analysis were conducted. Meta-analysis validated ELANE expression trends across GEO datasets, while survival analysis assessed its prognostic relevance. Single-cell sequencing localized ELANE expression, and

Sur7 mediates a novel pathway for PI4,5P2 regulation in C. albicans that promotes stress resistance and cell wall morphogenesis.

The human fungal pathogen Candida albicans can cause lethal systemic infections due to its ability to resist stress from the host and to undergo invasive hyphal growth. Previous studies showed that plasma membrane MCC/eisosome domains were important for virulence by promoting the ability of Sur7 to mediate normal cell wall morphogenesis and stress resistance. The sur7Δ mutant displayed abnormal clusters of PI4,5P2, suggesting that misregulation of this lipid underlies the sur7Δ phenotype. To test this, we increased PI4,5P2 levels by deleting combinations of the three PI4,5P2 5' phosphatase gen

Biodegradable cellulose scaffolds embedding kartogenin-loaded microspheres for cartilage regeneration.

Due to the extreme difficulty of cartilage, we endeavored to fabricate a novel biodegradable scaffold using natural polysaccharide cellulose and poly(lactic-co-glycolic acid) (PLGA) microspheres incorporating controllably released the cartilage inducer kartogenin (KGN) for enhancing regenerative healing in cartilage tissue. Initially, to facilitate the loading of KGN, PLGA/KGN microspheres were fabricated using the solvent volatilization method. These microspheres exhibited high sphericity, with particle sizes diminishing from 12.11 to 3.27 μm as the emulsification rate increased. Demonstratin

Gallbladder adenomas have molecular abnormalities different from those present in gallbladder carcinomas.

Although most gallbladder carcinomas evolve from dysplasia and carcinoma in situ, the role of gallbladder adenomas in the pathogenesis of gallbladder carcinoma is still controversial. A series of molecular changes including loss of heterozygosity (LOH) at 17p (TP53 gene), 13q (RB gene), 18q (DCC gene), and 9p21 (CDKN2a gene) chromosomal regions have been identified in dysplasias, carcinomas in situ, and invasive carcinomas of the gallbladder, whereas mutations in K- and N-ras genes are rare. To determine whether the molecular abnormalities of adenomas are similar to those found in carcinomas,

Integrated Bioinformatics Analysis of Shared Genes, miRNA, Biological Pathways and Their Potential Role as Therapeutic Targets in Huntington's Disease Stages.

Huntington's Disease (HD) is a progressive neurodegenerative disease caused by CAG repeat expansion in the huntingtin gene (HTT). The HTT gene was the first disease-associated gene mapped to a chromosome, but the pathophysiological mechanisms, genes, proteins or miRNAs involved in HD remain poorly understood. Systems bioinformatics approaches can divulge the synergistic relationships of multiple omics data and their integration, and thus provide a holistic approach to understanding diseases. The purpose of this study was to identify the differentially expressed genes (DEGs), HD-related gene ta

A Comprehensive multi-network analysis of ceRNAs and transcription factors for papillary thyroid carcinoma diagnosis and prognosis.

Papillary thyroid carcinoma (PTC) is a malignancy with an ambiguous etiology. The competitive endogenous RNA (ceRNA) hypothesis provides a framework for clarifying the molecular mechanisms that drive carcinogenesis. In this study, we constructed a novel ceRNA network to identify reliable diagnostic and prognostic indicators applicable across all stages of PTC. Transcriptome analysis was performed to identify stage-specific hub genes using the MCC, IVI, and MCODE algorithms. A novel five-layer ceRNA network and its associated regulatory network (DE-TF) were constructed. Receiver operating chara

Integration of network toxicology and transcriptomics reveals the potential regulatory role of the 'Arecoline/4-NQO-AKT1-m6A factor' axis in the development of OSCC.

OBJECTIVE: The synergistic carcinogenic effect of areca nut chewing and tobacco exposure is a major risk factor for oral squamous cell carcinoma (OSCC). However, the molecular mechanisms, particularly related to N6-methyladenosine (m6A) modification, remain unclear. This study explores the potential mechanisms by which arecoline and 4-nitroquinoline-1-oxide (4-NQO) induce OSCC, focusing on m6A regulatory factors and key targets. METHODS: Transcriptional data were obtained from Gene Expression Omnibus (GEO), with differentially expressed genes (DEGs) identified using the limma package. The int

Identification of potential autophagy-related genes in steroid-induced osteonecrosis of the femoral head via bioinformatics analysis and experimental verification.

PURPOSE: Steroid-induced osteonecrosis of the femoral head (SONFH) is a refractory orthopaedic hip joint disease that occurs in young- and middle-aged people. Previous experimental studies have shown that autophagy might be involved in the pathological process of SONFH, but the pathogenesis of autophagy in SONFH remains unclear. We aimed to identify and validate the key potential autophagy-related genes involved in SONFH to further illustrate the mechanism of autophagy in SONFH through bioinformatics analysis. METHODS: The GSE123568 mRNA expression profile dataset, including 10 non-SONFH (fol

Prognostic Score for Myelodysplastic Syndromes Based on Molecular Evolution.

BACKGROUND: Myelodysplastic syndromes are clonal hematopoietic stem cell disorders characterized by multistep molecular evolution and a variable risk of leukemic transformation. Given this prognostic heterogeneity, accurate risk stratification is essential for clinical decision-making. We developed ProgEvo, a proprietary framework that infers molecular evolutionary trajectories and integrates them with clinical data to improve prognostic accuracy. METHODS: ProgEvo was trained on 2519 patients in cBioPortal (https://www.cbioportal.org) and validated using two external cohorts: Genomed4All (204

Dental Pulp Stem Cell-Derived Intracellular Vesicles Promote Cartilage Regeneration and Alleviate Pain in Temporomandibular Joint Osteoarthritis.

INTRODUCTION AND AIMS: Temporomandibular joint osteoarthritis (TMJ-OA) is characterized by cartilage destruction and pain. Although mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have shown therapeutic efficacy, their clinical translation is hampered by low yield and batch-to-batch variability. Recent studies indicate that intracellular vesicles (IVs) can be harvested at markedly higher quantities than EVs while displaying comparable biological profiles. Therefore, this study directly compared high-yield dental pulp stem cell-derived intracellular vesicles (DPSC-IVs) with conve

Disruption of ETV6 in intron 2 results in upregulatory and insertional events in childhood acute lymphoblastic leukaemia.

We describe four cases of childhood B-cell progenitor acute lymphoblastic leukaemia (BCP-ALL) and one of T-cell (T-ALL) with unexpected numbers of interphase signals for ETV6 with an ETV6-RUNX1 fusion probe. Three fusion negative cases each had a telomeric part of 12p terminating within intron 2 of ETV6, attached to sequences from 5q, 7p and 7q, respectively. Two fusion positive cases, with partial insertions of ETV6 into chromosome 21, also had a breakpoint in intron 2. Development and validation of a multiple myeloma diagnostic model based on systemic lupus erythematosus-associated genes and identification of specific genes.

BACKGROUND: Monoclonal immunoglobulins are commonly found in multiple myeloma (MM), a prevalent hematologic malignancy that is currently incurable. In recent years, the association between systemic lupus erythematosus (SLE), an autoimmune disease, and MM has garnered increasing attention. However, there remains a lack of in-depth research regarding the interactions between these two conditions and their potential pathogenic mechanisms. Therefore, in order to improve the identification of MM associated with SLE, this work attempts to clarify the pathogenic pathways that are shared by MM and SLE

Integrative Transcriptomic Analysis Identifies a Key Candidate Gene Bridging Inflammation and DNA Repair in Endometriosis.

BACKGROUND: Endometriosis is a chronic, estrogen-dependent inflammatory disorder characterized by the ectopic implantation of endometrial-like tissue. Although retrograde menstruation is highly prevalent, only a subset of women develops the disease. This epidemiologic paradox suggests that intrinsic molecular alterations in the eutopic endometrium may precondition refluxed cells to survive under inflammatory and oxidative stress. METHODS: Eutopic endometrial transcriptomes from the GSE6364 dataset (21 endometriosis patients and 16 controls) were analyzed across menstrual phases. Differential

Onabotulinumtoxin-A intradetrusorial injections modulate bladder expression of NGF, TrkA, p75 and TRPV1 in patients with detrusor overactivity.

How onabotulinumtoxinA (onab/A) injected in the detrusor muscle improves detrusor overactivity (DO) is still a matter of debate. Nerve growth factor (NGF) seems to play a role in determining urgency and DO. Recent studies showed that NGF decreases in patients with DO who respond to onab/A treatment. We investigated onab/A-induced changes on gene expression of NGF, TRPV1, TrkA and p75 in bladder wall tissue of patients affected by neurogenic and idiopathic DO. Twenty-five patients (18 with neurogenic DO and 7 with idiopathic DO) received onab/A injections into the detrusor muscle. Urodynamic st

Genetic models of human cancer as a multistep process. Paradigm models of colorectal cancer, breast cancer, and chronic myelogenous and acute lymphoblastic leukaemia.

Tumour formations arise as a consequence of alterations in the control of cell proliferation as well as with disorders in interactions between cells and their environment that result in invasion and metastasis. Recent advances in understanding the genetic basis of malignant diseases have been dominated by research in colorectal cancer. Genetic alterations of several proto-oncogenes and tumor-suppressor genes (e.g. APC/MCC, RAS, DCC, p53 mutations and/or allelic losses, hyperexpression of c-MYC and RB genes), as well as other genomic alterations, appear at characteristic stages of tumor develop
