Gb3 encodes a unique kinase fusion protein conferring greenbug resistance in wheat.

Understanding plant resistance to insects has lagged owing to a scarcity of cloned resistance genes. Here we report cloning of the greenbug resistance gene Gb3 in wheat (Triticum aestivum L.), initially introgressed into cultivars such as 'TAM112' from Aegilops tauschii. Gb3 encodes an intracellular kinase-pseudokinase protein, featuring a unique MAP3K-like domain. This gene probably originated through fusion, duplication and indel events in the Triticeae tribe, retaining its functionality in several Aegilops species. Transgenic Gb3 expression in the susceptible Fielder wheat enhanced resistan

A population genetic analysis of the nematode Strongyloides stercoralis in Asia shows that human infection is not a zoonosis from dogs.

Gut nematode worms are important parasites of people and other animals. The parasitic nematode Strongyloides stercoralis infects an estimated 600 million people worldwide and is one of the soil-transmitted helminthiases, a WHO-defined neglected tropical disease. It has long been suggested that human S. stercoralis infection may be a zoonosis from dogs. We investigated this by whole genome sequence analysis of S. stercoralis from sympatric human and dog populations in Asia. We find that human- and dog-derived S. stercoralis have genetically distinct nuclear genomes, but we also find evidence of

Use of the Serial Analysis of Gene Expression (SAGE) method in veterinary research: A concrete application in the study of the bovine trypanotolerance genetic control.

New postgenomic biotechnologies, such as transcriptome analyses, are now able to characterize the full complement of genes involved in the expression of specific biological functions. One of these is the Serial Analysis of Gene Expression (SAGE) technique, which consists of the construction of transcripts libraries for a quantitative analysis of the entire gene(s) expressed or inactivated at a particular step of cellular activation. Bioinformatic comparisons in the bovine genomic databases allow the identification of several up- and downregulated genes, expressed sequence tags, and unknown fun

Transposons drove the evolution of a male sex-determining gene from a female gene with deep introgression across cichlids.

Sex-determination mechanisms and their master regulators are diverse among vertebrates. Components of downstream regulatory pathways can sometimes be co-opted as new upstream triggers. Here, we report a notable finding of cross-pathway recruitment: A gene from the female differentiation cascade was repurposed as a male master sex-determining (MSD) gene. We identify figlaY, a Y-linked truncated duplicate of the female-associated figla, as the MSD gene in Mozambique tilapia. figlaY originated through duplication and translocation in an ancestral cichlid lineage and subsequently introgressed into

Low-gluten, nontransgenic wheat engineered with CRISPR/Cas9.

Coeliac disease is an autoimmune disorder triggered in genetically predisposed individuals by the ingestion of gluten proteins from wheat, barley and rye. The α-gliadin gene family of wheat contains four highly stimulatory peptides, of which the 33-mer is the main immunodominant peptide in patients with coeliac. We designed two sgRNAs to target a conserved region adjacent to the coding sequence for the 33-mer in the α-gliadin genes. Twenty-one mutant lines were generated, all showing strong reduction in α-gliadins. Up to 35 different genes were mutated in one of the lines of the 45 different g

Genetic Dissection of Wheat Heterosis Identifies Candidate Genes in Biparental and Diverse Hybrid Populations.

Understanding how numerous heterotic quantitative trait loci (hQTL) shape heterosis and shifting from hQTL to candidate genes are central challenges. To address these challenges, we performed a systematic, genome-wide mapping in a biparental population and a population of diverse hybrids. Leveraging both a triple testcross and immortalized F2 designs, we examined the genetic basis of the heterosis of the wheat hybrid Piko × Hermann. We detected a major hQTL on chromosome 4B, primarily driven by epistasis. This region contains the Green Revolution gene Rht-B1 as the primary candidate gene. Then

Phylogenetic position of the white-cheeked macaque (Macaca leucogenys), a newly described primate from southeastern Tibet.

The white-cheeked macaque Macaca leucogenys is a recently described species that was only diagnosed based on photos, without any specimen measurements or molecular genetic diagnosis. Using DNA extracted from four newly collected skin specimens, we studied the genetic diversity and phylogenetic position of M. leucogenys using multilocus sequence data, including mitochondrial and Y chromosomal genes. Skin measurements of four individuals showed that the white-cheeked macaque is robust and larger than M. assamensis but is similar in body size to M. thibetana. Although the holotype male of M. leuc

Genetic analysis of X-linked hybrid sterility in the house mouse.

Hybrid sterility is a common postzygotic reproductive isolation mechanism that appears in the early stages of speciation of various organisms. Mus musculus musculus and Mus musculus domesticus represent two recently separated mouse subspecies particularly suitable for genetic studies of hybrid sterility. Here we show that the introgression of Chr X of M. m. musculus origin (PWD/Ph inbred strain, henceforth PWD) into the genetic background of the C57BL/6J (henceforth B6) inbred strain (predominantly of M. m. domesticus origin) causes male sterility. The X-linked hybrid sterility is associated w

Sex chromosomes and chromosomal rearrangements contribute to behavioral sexual isolation in Jaera albifrons marine isopods.

We focus on the evolution of behavioral sexual isolation through the empirical study of two marine isopod species. The males of Jaera albifrons and Jaera praehirsuta engage females in tactile courtship by brushing the female's back, but they do so with divergent sets of specialized setae, and female choice results in strong reproductive isolation. Using RADseq-derived genotypes of individuals from natural populations and controlled crosses, we found that secondary contacts between J. albifrons and J. praehirsuta resulted in different levels of heterospecific gene flow. Comparison of the genomi

Discovery and genomic analyses of hybridization between divergent lineages of Trypanosoma congolense, causative agent of Animal African Trypanosomiasis.

Hybrid populations and introgressive hybridization remain poorly documented in pathogenic micro-organisms, as such that genetic exchange has been argued to play a minor role in their evolution. Recent work demonstrated the existence of hybrid microsatellite profiles in Trypanosoma congolense, a parasitic protozoan with detrimental effects on livestock productivity in sub-Saharan Africa. Here, we present the first population genomic study of T. congolense, revealing a remarkable number of single nucleotide polymorphisms (SNPs), small insertions/deletions (indels) and gene deletions among 56 par

Genome-wide SNP evidence for the genetic uniqueness of indigenous cattle population.

India’s non-descript cattle, comprising 52% of the 193.46 million cattle population, are pivotal to the nation’s genetic diversity but contribute only 9.82% to milk production, highlighting a critical need for their genomic characterization so as to utilize this information in conservation and breeding strategies. This study uses the Bovine GGP50K SNP chip to determine the genetic uniqueness, population structure, ancestry proportion, and conservation status of two significant indigenous cattle populations from the Vidarbha region of Maharashtra, Umarda (UM) and Pahadi/Melghati (PAH), which fo

Extensive population subdivision of the cuttlefish Sepia officinalis (Mollusca: Cephalopoda) around the Iberian Peninsula indicated by microsatellite DNA variation.

The Atlantic Ocean-Mediterranean Sea junction has been proposed as an important phylogeographical area on the basis of concordance in genetic patterns observed at allozyme, mtDNA and microsatellite DNA markers in several marine species. This study presents microsatellite DNA data for a mobile invertebrate species in this area, the cuttlefish Sepia officinalis, allowing comparison of this relatively new class of DNA marker with previous allozyme results, and examination of the relative effects on gene flow of the Strait of Gibraltar and the Almería-Oran oceanographic front. High genetic diversity and demographic history of captive Siamese and Saltwater crocodiles suggest the first step toward the establishment of a breeding and reintroduction program in Thailand.

The Siamese crocodile (Crocodylus siamensis) and Saltwater crocodile (C. porosus) are two of the most endangered animals in Thailand. Their numbers have been reduced severely by hunting and habitat fragmentation. A reintroduction plan involving captive-bred populations that are used commercially is important and necessary as a conservation strategy to aid in the recovery of wild populations. Here, the genetic diversity and population structure of 69 individual crocodiles, mostly members of captive populations, were analyzed using both mitochondrial D-loop DNA and microsatellite markers. Uncovering the genetic architecture and evolutionary roots of androgenetic alopecia in African men.

Androgenetic alopecia is a highly heritable trait. However, much of our understanding about the genetics of male-pattern baldness comes from individuals of European descent. Here, we examined a dataset comprising 2,136 men from Ghana, Nigeria, Senegal, and South Africa that were genotyped using the Men of African Descent and Carcinoma of the Prostate Array. We first tested how genetic predictions of baldness generalize from Europe to Africa and found that polygenic scores from European genome-wide association studies (GWASs) yielded area under the curve statistics that ranged from 0.513 to 0.5

Mitochondrial background can explain variable costs of immune deployment.

Organismal health and survival depend on the ability to mount an effective immune response against infection. Yet immune defence may be energy-demanding, resulting in fitness costs if investment in immune function deprives other physiological processes of resources. While evidence of costly immunity resulting in reduced longevity and reproduction is common, the role of energy-producing mitochondria on the magnitude of these costs is unknown. Here, we employed Drosophila melanogaster cybrid lines, where several mitochondrial genotypes (mitotypes) were introgressed onto a single nuclear genetic

Normal hearing in alpha-galactosidase A-deficient mice, the mouse model for Fabry disease.

Fabry disease (OMIM 301500) is a rare X-linked recessive disorder caused by mutations in the alpha-galactosidase gene (Gla). Loss of Gla activity leads to the abnormal accumulation of glycosphingolipids in lysosomes of predominantly vascular endothelial cells. Clinically the disorder presents with angiokeratomas, clouding of the cornea, and renal, cardiac, and cerebrovascular complications. In addition, there is an increased incidence of sensorineural hearing loss in Fabry patients. In this study, we investigated the loss of alpha-galactosidase A activity on hearing function in Gla-deficient m

Multilocus Characterization of Human Ascaris in Pakistan Indicates Ongoing Introgression and Shared Global Haplotypes.

BACKGROUND: Understanding the molecular epidemiology of Ascaris is essential for clarifying species boundaries and transmission dynamics within regions where human-animal interfaces may influence parasite dispersal. This study provides a comprehensive multilocus characterization of Ascaris populations in Pakistan using nuclear (partial 5.8 S, ITS2, partial 28 S) and mitochondrial (cox1) markers. METHODS: A total of 1675 children were enrolled across six districts of Khyber Pakhtunkhwa, and expelled adult worms were collected following single-dose anthelmintic treatment; genomic DNA from morph

Genomic Insights into Cold Adaptation and Demographic History of Amur-Heilong River Basin Populations.

Human adaptation to extreme cold has been studied predominantly in Arctic populations, leaving the genetic basis of cold resilience in non-Arctic high-latitude regions insufficiently understood. The Amur-Heilong River Basin (AR) in Northeast Asia represents one of the coldest inhabited regions in East Asia, yet its long-term resident populations remain underexplored at the genomic level. Here, we compiled high-coverage whole-genome sequences for 78 individuals representing three indigenous AR populations (Daur, Evenki, and Oroqen) to investigate their demographic history and genetic adaptation

Unveiling The Origin of Sacred Red Rice Preserved in Japanese Shrines.

Pericarp color is a key trait selected during rice domestication. Although most modern cultivars have a white pericarp, ancestral rice likely had a red pericarp, as extant wild rice retains this trait and the 14-bp deletion in the Rc gene, which confers white pericarp, is prevalent among modern cultivars. In Japan, several red rice accessions have been preserved in geographically separated shrines for ritual use. Here, we investigated the genomic origin and evolutionary history of the sacred red rice accessions preserved in four shrines across three regions of Japan, together with a red rice l

Genomic, morphological and physiological data support fast ecotypic differentiation and incipient speciation in an alpine diving beetle.

An intricate interplay between evolutionary and demographic processes has frequently resulted in complex patterns of genetic and phenotypic diversity in alpine lineages, posing serious challenges to species delimitation and biodiversity conservation planning. Here we integrate genomic data, geometric morphometric analyses and thermal tolerance experiments to explore the role of Pleistocene climatic changes and adaptation to alpine environments on patterns of genomic and phenotypic variation in diving beetles from the taxonomically complex Agabus bipustulatus species group.
