The effect of genetic variation of the serotonin 1B receptor gene on impulsive aggressive behavior and suicide.

Impulsive-aggressive behaviors (IABs) are regarded as possible suicide intermediate phenotypes, mediating the relationship between genes and suicide outcome. In this study, we aimed to investigate the putative relationship between genetic variation at the 5-HT1B receptor gene, which in animal models is involved in impulse-aggression control, IABs, and suicide risk. We investigated the relationship of variation at five 5-HT1B loci and IAB measures in a sample of 696 subjects, including 338 individuals who died by suicide and 358 normal epidemiological controls. We found that variation at the 5-

Genetic and environmental influences on problematic masturbatory behavior in children: a study of same-sex twins.

Child sexual behavior problems, such as excessive or public masturbation, are often judged to result from environmental stress or trauma. We studied the relative importance of genetic and environmental factors for problematic masturbatory behavior among nonreferred prepubertal children. All twins born in Sweden in 1985-86 were identified from the Swedish Twin Registry. Parents, mainly mothers, completed Child Behavior Checklist (CBCL) ratings (Achenbach, 1991) for 401 monozygotic (MZ) and 248 dizygotic (DZ) same-sex twin pairs (male-male or female-female) at age 7-9 years. Scores of two CBCL i

Pharmacologic thresholds for self-injurious behavior in a genetic mouse model of Lesch-Nyhan disease.

Congenital deficiency of hypoxanthine-guanine phosphoribosyl transferase (HPRT) causes Lesch-Nyhan disease in humans, which is associated with severe and recurrent self-injurious behavior (SIB). The HPRT-deficient knockout mouse model, however, does not display this unusual behavior. The present studies tested whether these mice might be more vulnerable to pharmacologic agents known to cause SIB in normal rodents, including clonidine, Bay K 8644, GBR 12909, methamphetamine, pemoline and caffeine. The results provided three conclusions. First, normal mice did not display SIB using some drugs kn

Genetic association analysis of behavioral inhibition using candidate loci from mouse models.

Genes influence the development of anxiety disorders, but the specific loci involved are not known. Genetic association studies of anxiety disorders are complicated by the complexity of the phenotypes and the difficulty in identifying appropriate candidate loci. We have begun to examine the genetics of behavioral inhibition to the unfamiliar (BI), a heritable temperamental predisposition that is a developmental and familial risk factor for panic and phobic disorders. Specific loci associated with homologous phenotypes in mouse models provide compelling candidate genes for human BI. We conducte

Early, non-psychotic deviant behavior in schizophrenia: a possible endophenotypic marker for genetic studies.

Early non-psychotic deviance occurs in some, but not all, pre-schizophrenic patients and has been linked to the later course of the disorder, suggesting its relationship with the schizophrenia syndrome. However, early deviance has rarely been explored as an endophenotypic marker in large samples of schizophrenic patients. We characterized the early childhood behavior and syndrome history of 205 adults with DSM-IV schizophrenia. Sixty percent of our sample had poor socialization, extreme fears/chronic sadness, and/or attention impairment/learning disabilities beginning before age 10. The remain

Genetic and environmental causes of covariation in interview assessments of disruptive behavior in child and adolescent twins.

Multirater, face-to-face, interview data relating to conduct disorder (CD), oppositional-defiant disorder (ODD), and inattentive, impulsive, and hyperactive components of attention-deficit hyperactivity disorder (ADHD) in a population-based sample of 1376 pairs of 8- to 16-year-old MZ and DZ twins are analyzed to examine (1) the genetic and environmental causes of correlation among ratings of ODD and CD symptoms and (2) the pattern of genetic and environmental correlation among the three components of ADHD. Parental ratings of ADHD showed marked sibling contrast effects, specific within raters

The genetic liability to stress and postweaning isolation have a competitive influence on behavioral organization in rats.

Rats housed in social isolation postweaning (isolates) show profound behavioral and neurobiological differences when compared to socially housed rats (socials). Fischer rats (F344) relative to Lewis rats are hyperresponsive and significantly more susceptible to stressful stimuli. This investigation tested the hypothesis that the behavioral effects of postweaning isolation are more pronounced in a strain of rats with high susceptibility to stress compared to a strain with low susceptibility to stress. Seventy male Sprague-Dawley, Lewis, and F344 rats were housed individually or in groups at wea

Zinc-induced peripheral anosmia and behavioral responses to novelty in mice: a quantitative-genetic analysis.

Adult male mice were made anosmic by intranasal flushing with a 5% zinc sulfate solution. Twelve behavioral variables were measured in treated as well as saline-irrigated control animals placed in a novel environment. The genetic underpinnings and the genotype-treatment interactions with regard to these behaviors were analyzed in a classical Mendelian cross between the inbred strains C57BL/6 and DBA/2 and in a full 4 X 4 diallel cross, replicated five times, between these strains and strains C3H/St an CPB-K. Co-occurrences of substance use and other potentially addictive behaviors: Epidemiological results from the Psychological and Genetic Factors of the Addictive Behaviors (PGA) Study.

BACKGROUND AND AIMS: Changes in the nomenclature of addictions suggest a significant shift in the conceptualization of addictions, where non-substance related behaviors can also be classified as addictions. A large amount of data provides empirical evidence that there are overlaps of different types of addictive behaviors in etiology, phenomenology, and in the underlying psychological and biological mechanisms. Our aim was to investigate the co-occurrences of a wide range of substance use and behavioral addictions. METHODS: The present epidemiological analysis was carried out as part of the P

Separating Family-Level and Direct Exposure Effects of Smoking During Pregnancy on Offspring Externalizing Symptoms: Bridging the Behavior Genetic and Behavior Teratologic Divide.

Maternal smoking during pregnancy (MSDP) has been robustly associated with externalizing problems and their developmental precursors in offspring in studies using behavioral teratologic designs (Wakschlag et al., Am J Public Health 92(6):966-974, 2002; Espy et al., Dev Psychol 47(1):153-169, 2011). In contrast, the use of behavior genetic approaches has shown that the effects commonly attributed to MSDP can be explained by family-level variables (D'Onofrio et al., Dev Psychopathol 20(01):139-164, 2008). Reconciling these conflicting findings requires integration of these study designs. We util

Both Genetic Manipulation and Acupuncture Alleviate Depressive-Like Behavior Mediated by Astrocytic P2X7 Receptors.

Major depressive disorder (MDD) has been associated with P2X7 receptors (P2X7Rs), yet the contribution of astrocytic P2X7Rs to neuronal dysfunction remains unclear. We investigated the role of astrocytic P2X7Rs in depression-like behaviors using a chronic unpredictable mild stress (CUMS) mouse model and further examined whether electroacupuncture (EA) exerts antidepressant effects by modulating astrocytic P2X7Rs. Astrocytic P2X7Rs in the medial prefrontal cortex (mPFC) were selectively knocked down or overexpressed via gene manipulation. Depression-like behaviors were assessed after CUMS expos

Behavior of neural networks in culture suggest that sporadic and genetic forms of Alzheimer's disease may not be equivalent.

INTRODUCTION: Alzheimer's disease (AD) research often assumes that familial and sporadic forms share a common pathogenesis. However, the cellular impacts of amyloid precursor protein (APP) mutations compared with age-related overexpression of APP or the deposition of amyloid beta (Aβ) are likely different. METHODS: Using high-density multi-electrode arrays, we compared neural activity in cultured neurons subjected to Aβ exposure or APP overexpression (via lentiviral delivery or genetic models). Effects on neuronal firing, synaptogenesis, axonal branching, and network connectivity were assesse

Multimodal neuroimaging changes and their behavioral, genetic, and neurotransmitter correlates in electroconvulsive therapy for major depressive disorder.

Electroconvulsive therapy (ECT) is an effective treatment for major depressive disorder (MDD), yet its underlying mechanisms remain unclear. This study investigated the antidepressant effects of ECT through a multimodal neuroimage meta-analysis combined with functional, genetic, and neurotransmitter assessments. Resting-state functional magnetic resonance imaging (fMRI) and voxel-based morphometry (VBM) data were analyzed using seed-based d mapping with permutation of subject images (SDM-PSI) to identify changes in spontaneous brain activity and gray matter volume (GMV) before and after ECT. Effects of adverse childhood experiences on behavior in adolescence: Insights from brain imaging and genetic risk.

Adolescence is a critical developmental stage, and adverse childhood experiences (ACEs) are important environmental factors influencing behavior. However, the relationship between ACEs, individual behavior, brain mechanisms, and the role of genetic risk remains underexplored. In this study, data from 9796 youths aged 9-13 years in the Adolescent Brain Cognitive Development (ABCD) study were analyzed, including 67 ACE-related variables and 30 behavioral outcomes. Using sparse canonical correlation analysis, we identified two distinct patterns: a parental mental health-child psychopathology, lin

Decomposing genetic effects of social connectedness and depression on youth behaviors and long-term clinical outcomes.

IMPORTANCE: Lacking social connectedness is associated with broad morbidity. Yet the underlying mechanisms remain obscures, especially given its entanglement with depressive symptoms. Clarifying whether social connectedness confers unique risk beyond depression can guide clinical care and social infrastructure interventions. OBJECTIVE: To decompose shared genetic architecture between social connectedness and depressive affect, derive independent polygenic liabilities, and test their differential associations with youth behaviors, digital engagement, and adult clinical outcomes. Polygenic overlap of substance use behaviors and disorders with externalizing and internalizing problems independent of genetic correlations.

BACKGROUND: Externalizing and internalizing pathways may lead to the development of substance use behaviors (SUBs) and substance use disorders (SUDs), which are all heritable phenotypes. Genetic correlation studies have indicated differences in the genetic susceptibility between SUBs and SUDs. We investigated whether these substance use phenotypes are differently related to externalizing and internalizing problems at a genetic level. METHODS: We analyzed data from genome-wide association studies (GWAS) of four SUBs and SUDs, five externalizing traits, and five internalizing traits using the b

Editorial: Genetic Predisposition to Attention-Deficit/Hyperactivity Disorder Increases the Risk of Emotional and Behavioral Difficulties.

Our understanding of the clinical impact and the long-term risks of having attention-deficit/hyperactivity disorder (ADHD) has improved recently, with large-scale epidemiological studies showing that individuals with ADHD are at increased risk of suicidal behavior, educational underachievement, substance misuse, and car accidents,1 as well as at increased risk of psychiatric disorders such as depression, bipolar disorder, and anxiety disorder.2 It may be that having ADHD predisposes to educational underachievement and adversity later in life and thus predisposes to the development of other psy

Common genetic variation is associated with longitudinal decline and network features in behavioral variant frontotemporal degeneration.

The T allele in rs1768208 located in or near the myelin oligodendrocyte basic protein gene (MOBP) is a risk factor for frontotemporal degeneration pathology. We evaluated the hypothesis that the presence of a T allele in rs1768208 will be associated with rate of cognitive decline in behavioral variant frontotemporal degeneration (bvFTD) related to compromised frontal networks. We studied 81 individuals clinically diagnosed with bvFTD who were genotyped for rs1768208 and coded using a dominant model reflecting the presence (i.e., MOBP +) or absence (MOBP -) of the T risk allele. Longitudinal Links Between Callous-Unemotional Behaviors and Parenting in Early Childhood: A Genetically Informed Design.

OBJECTIVE: Most research on callous-unemotional behaviors (CU) and parenting does not focus on directions of effect, and work that does so has not been genetically informed. The present study is the first to examine potential reciprocal effects between parenting and CU in a community sample of early childhood. Use of a twin sample also allows us to distinguish child-based genetic effects from environmentally driven effects, which is necessary before translating this research to interventions. METHOD: The present study used biometric cross-lagged models to investigate the relation between CU a

Genetic Suppression of mTOR Rescues Synaptic and Social Behavioral Abnormalities in a Mouse Model of Pten Haploinsufficiency.

Heterozygous mutations in PTEN, which encodes a negative regulator of the mTOR and β-catenin signaling pathways, cause macrocephaly/autism syndrome. However, the neurobiological substrates of the core symptoms of this syndrome are poorly understood. Here, we investigate the relationship between cerebral cortical overgrowth and social behavior deficits in conditional Pten heterozygous female mice (Pten cHet) using Emx1-Cre, which is expressed in cortical pyramidal neurons and a subset of glia. We found that conditional heterozygous mutation of Ctnnb1 (encoding β-catenin) suppresses Pten cHet co
