Genetic and molecular factors associated with changes in structural-functional coupling in medication-free obsessive-compulsive disorder.

Obsessive-compulsive disorder (OCD) is a complex psychiatric disorder. While existing studies have revealed abnormalities in brain structure and function associated with OCD, there is a paucity of research integrating these two aspects, and the transcriptional patterns underlying these abnormalities remain unclear. This study is a multiscale, exploratory investigation designed to generate hypotheses rather than to test causal mechanisms. We aimed to investigate aberrations in brain structure-function coupling (SFC) in OCD patients and, by integrating gene expression profiles and neurotransmitt

Patient Perceptions of Sensitive Genetic and Other Medical Information: Findings from a Cancer Survivors Survey in Japan.

INTRODUCTION: Our previous findings indicated that hospitals frequently restrict access to genetic information, while access to other types of sensitive information-such as psychological counseling records or infectious disease diagnoses-is limited in a smaller proportion of facilities (Suzuki et al., 2023). This practice, grounded in a paternalistic medical framework, highlights the need to incorporate patients' perspectives into future information governance. Therefore, this study aimed to explore how individuals with a history of cancer perceive the sensitivity of various types of medical i

Prevalence and genetic characteristics of Staphylococcus aureus isolates from cell phones of medical students from Iran.

Although mobile phones as a rapid communication vehicle can lead to improved quality of healthcare, they can also facilitate the transmission of pathogens to patients. This current research focuses on genetic diversity, and genes involved in resistance and biofilm production of Staphylococcus aureus isolates from mobile phones of medical students. Antibiotic resistance profiling and polymerase chain reaction (PCR) amplification of antibiotic resistance and biofilm-related genes were investigated and statistically analyzed. Staphylococcal cassette chromosome mec (SCCmec) types were analyzed by

Comprehensive genetic testing improves the clinical diagnosis and medical management of pediatric patients with isolated hearing loss.

PURPOSE: Genetic testing is widely used in diagnosing genetic hearing loss in patients. Other than providing genetic etiology, the benefits of genetic testing in pediatric patients with hearing loss are less investigated. METHODS: From 2018-2020, pediatric patients who initially presented isolated hearing loss were enrolled. Comprehensive genetic testing, including GJB2/SLC26A4 multiplex amplicon sequencing, STRC/OTOA copy number variation analysis, and exome sequencing, were hierarchically offered. Clinical follow-up and examinations were performed. RESULTS: A total of 80 pediatric patients

Rates of Actionable Genetic Findings in Individuals with Colorectal Cancer or Polyps Ascertained from a Community Medical Setting.

As clinical testing for Mendelian causes of colorectal cancer (CRC) is largely driven by recognition of family history and early age of onset, the rates of such findings among individuals with prevalent CRC not recognized to have these features is largely unknown. We evaluated actionable genomic findings in community-based participants ascertained by three phenotypes: (1) CRC, (2) one or more adenomatous colon polyps, and (3) control participants over age 59 years without CRC or colon polyps. These participants underwent sequencing for a panel of genes that included colorectal cancer/polyp (CR

The Change of a Medically Important Genus: Worldwide Occurrence of Genetically Diverse Novel Brucella Species in Exotic Frogs.

The genus Brucella comprises various species of both veterinary and human medical importance. All species are genetically highly related to each other, sharing intra-species average nucleotide identities (ANI) of > 99%. Infections occur among various warm-blooded animal species, marine mammals, and humans. Until recently, amphibians had not been recognized as a host for Brucella. In this study, however, we show that novel Brucella species are distributed among exotic frogs worldwide. Comparative recA gene analysis of 36 frog isolates from various continents and different frog species revealed

Effects of naltrexone on alcohol sensitivity and genetic moderators of medication response: a double-blind placebo-controlled study.

CONTEXT: Clinical trials have suggested a modest effect of naltrexone as a pharmacotherapy for alcoholism, and a recent study has suggested that the effects may be moderated by variations in the mu-opioid receptor gene (OPRM1). However, the mechanism by which naltrexone may be differentially effective as a function of the OPRM1 genotype is unclear. OBJECTIVES: (1) To replicate and expand on the association between the A118G single nucleotide polymorphism(SNP) of the OPRM1 gene and alcohol sensitivity, (2) to examine the effects of naltrexone on alcohol sensitivity, and (3) to test the A118G S

Genetic markers for the identification and characterization of Opisthorchis viverrini, a medically important food borne trematode in Southeast Asia.

The liver fluke, Opisthorchis viverrini, is one of the major food borne trematodes in Southeast Asia, where infection causes hepatobiliary disease and subsequent development of cholangiocarcinoma. In Thailand, O. viverrini is most prevalent in the northeast where there is marked regional variation in the rate of infection in humans at provincial, district and village levels. To date, the roles of genetic variation of O. viverrini on this observed variability in infection, transmission and associated disease are not known. We have applied multilocus enzyme electrophoresis (MEE), specifically al

The National Association of Medical Examiners (NAME) Position Paper: Postmortem Genetic Testing in Forensic Pathology Cases.

Individuals who die suddenly and unexpectedly will often fall under the jurisdiction of a Medical Examiner or Coroner (MEC). Forensic pathologists may therefore be the first physicians to diagnose a genetic disease. Identifying these conditions at autopsy improves the accuracy of death certification and provides biological relatives the opportunity to seek diagnosis and intervention. Accessibility and diagnostic capabilities of postmortem genetic testing (PMGT) have rapidly expanded over the last decade since the previous National Association of Medical Examiners position paper on this topic.

Barriers and facilitators for medical oncologists in the further implementation of mainstream genetic testing in breast cancer care in the Netherlands.

Only 67% of eligible patients with breast cancer currently undergo genetic testing. Mainstream genetic testing (i.e. healthcare professionals (HCPs) from outside the genetics field providing genetic counselling), also referred to asmainstreaming, leads to increased uptake of genetic testing. Unlike surgical oncologists, medical oncologists have not yet widely adopted mainstreaming. This study aims to identify barriers and facilitators for HCPs, in particular medical oncologists, in advancing the implementation of mainstreaming. Structured interviews were conducted with HCPs (predominantly medi

Analysis of early diagnosis methods for asymmetric dementia in brain MR images based on genetic medical technology.

Alzheimer's disease (AD) is a relatively common senile neurodegenerative disease and the main manifestation of senile dementia. In the pathological changes of AD, the asymmetry of the brain also changes. Therefore, finding an early diagnosis method of AD based on asymmetry is the key to the treatment of Alzheimer's. Magnetic resonance (MR) imaging can quantitatively reflect the structural and functional changes of various tissues in the brain. It has the advantages of non-invasive, high spatial resolution, and non-radiation, and has been widely used in the early diagnosis of AD. In this work,

Genetic Testing in Patients with Neurodevelopmental Disorders: Experience of 511 Patients at Cincinnati Children's Hospital Medical Center.

Our institution developed and continuously improved a Neurodevelopmental Reflex (NDR) algorithm to help physicians with genetic test ordering for neurodevelopmental disorders (NDDs). To assess its performance, we performed a retrospective study of 511 patients tested through NDR from 2018 to 2019. SNP Microarray identified pathogenic/likely pathogenic copy number variations in 27/511 cases (5.28%). Among the 484 patients tested for Fragile X FMR1 CGG repeats, a diagnosis (0.20%) was established for one male mosaic for a full mutation, a premutation, and a one-CGG allele. Within the 101 normoce

The Genetic Association Between Prescription Pain Medication Use and the Risk for Fibromyalgia: A Bidirectional and Multivariable Mendelian Randomization Analysis.

INTRODUCTION: Increasing evidence has indicated that prescription opioid use is associated with poor prognosis in patients with fibromyalgia. However, it remains unknown whether there are causal associations between prescription opioid and fibromyalgia. METHODS: We performed a bidirectional two-sample MR analysis using genetic variants associated with prescription pain medication use and fibromyalgia from genome-wide association studies (GWAS). Multivariable Mendelian randomization (MVMR) was further employed to ascertain the causal relationship, incorporating confounders such as major depres
