Explaining the evolution of sex and recombination is particularly intriguing for some species of eusocial insects because they display exceptionally high mating frequencies and genomic recombination rates. Explanations for both phenomena are based on the notion that both increase colony genetic diversity, with demonstrated benefits for colony disease resistance and division of labor. However, the relative contributions of mating number and recombination rate to colony genetic diversity have never been simultaneously assessed. Our study simulates colonies, assuming different mating numbers, rec

Proteinase 3 (PR3) is the major autoantigen for anti-neutrophil cytoplasmic antibodies (ANCA) in patients with Wegener's granulomatosis. Little is known about the major antigenic sites on PR3. To facilitate epitope mapping, PR3 was cloned in insect cells using a baculovirus expression system. Four different sequences of the PR3 cDNA were amplified by PCR: two clones containing the pro-peptide of PR3 with or without a His-tag (rproPR3-his and rproPR3, respectively) and two clones without the pro-peptide and with or without a His-tag (rPR3-his and rPR3, respectively). The PR3 sequences were clon

Many cells in the thorax of Drosophila were found to stall during replication, a phenomenon known as underreplication. Unlike underreplication in nuclei of salivary and follicle cells, this stall occurs with less than one complete round of replication. This stall point allows precise estimations of early-replicating euchromatin and late-replicating heterochromatin regions, providing a powerful tool to investigate the dynamics of structural change across the genome. We measure underreplication in 132 species across the Drosophila genus and leverage these data to propose a model for estimating t

BACKGROUND: The mosquito Aedes aegypti is the primary global vector for dengue and yellow fever viruses. Sequencing of the Ae. aegypti genome has stimulated research in vector biology and insect genomics. However, the current genome assembly is highly fragmented with only ~31% of the genome being assigned to chromosomes. A lack of a reliable source of chromosomes for physical mapping has been a major impediment to improving the genome assembly of Ae. aegypti.
METHODOLOGY/PRINCIPAL FINDINGS: In this study we demonstrate the utility of mitotic chromosomes from imaginal discs of 4(th) instar lar

Insects experience a wide array of chemical pressures from plant allelochemicals and pesticides and have developed several effective counterstrategies to cope with such toxins. Among these, cytochrome P450 monooxygenases are crucial in plant-insect interactions. Flavin-dependent monooxygenases (FMOs) seem not to play a central role in xenobiotic detoxification in insects, in contrast to mammals. However, the previously identified senecionine N-oxygenase of the arctiid moth Tyria jacobaeae (Lepidoptera) indicates that FMOs have been recruited during the adaptation of this insect to plants that

Trypanosoma cruzi is the causative agent of Chagas disease, a neglected tropical disease, and one of the most important parasitic diseases worldwide. The first genome of T. cruzi was sequenced in 2005, and its complexity made assembly and annotation challenging. Nowadays, new sequencing methods have improved some strains' genome sequence and annotation, revealing this parasite's extensive genetic diversity and complexity. In this review, we examine the genetic diversity, the genomic structure, and the principal multi-gene families involved in the pathogenicity of T. cruzi. The T. cruzi genome

Macroparasites are among the most important agents of natural selection in their host populations, but anti-macroparasite immunity is poorly understood. Vinegar flies (Drosophilidae) and the parasitoid wasps that infect them are emerging models to study how animals defend against macroparasite attack. The canonical anti-parasitoid immune mechanism in insects is melanotic encapsulation, which involves cell-mediated encapsulation coupled with prophenoloxidase (PPO)-mediated melanization of parasitoid embryos. Recently, we discovered the horizontal transfer (HGT) of a bacterially-derived humoral

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

Insect wing polyphenism enables a single genome to produce distinct wing morphs in response to environmental cues, yet its underlying cellular determinants remain elusive. Here, we perform single-cell RNA sequencing of long-winged- and short-winged-destined wing buds of Pyrrhocoris apterus and Nilaparvata lugens, identifying six conserved cell types with comparable proportions between the two morphs. RNA interference-mediated silencing of 51 marker genes indicates that wing-patterning genes En (epithelial-like cells) and bs (tracheal cells), and cell-cycle genes Anln, CycB3, and cdk1 (neuron c

Triatomines are infamous as vectors of the parasite Trypanosoma cruzi, the causative agent of Chagas disease. However, climate-driven range expansion and urbanization adaptation of triatomine populations, coupled with their highly diverse feeding strategies (vertebrate haematophagy, kleptohaematophagy, and coprophagy), and has elevated interest in triatomines as potential arboviral vectors. Information on the triatomine virome is scant, with prior records including only eight insect-specific viruses: Triatoma virus (TrV) and Rhodnius prolixus viruses 1-7. Here, we leverage publicly available t

GPCRs play crucial roles in the growth, development and reproduction of organisms. In insects, a large number of GPCRs have been reported for Holometabola but not Hemimetabola. The recently sequenced pea aphid genome provides us with the opportunity to analyze the evolution and potential functions of GPCRs in Hemimetabola. 82 GPCRs were identified from the representative model hemimetabolous insect Acyrthosiphon pisum, 37 of which have ESTs evidence, and 73 are annotated for the first time. A striking difference between A. pisum, Drosophila melanogaster and Tribolium castaneum is the duplicati

In anautogenous mosquitoes, egg development requires blood feeding and as a consequence mosquitoes act as vectors of numerous devastating diseases of humans and domestic animals. Understanding the molecular mechanisms regulating mosquito egg development may contribute significantly to the development of novel vector-control strategies. Previous studies have shown that in the yellow fever mosquito Aedes aegypti, nuclear receptors (NRs) play a key role in the endocrine regulation of reproduction. However, many mosquito NRs remain uncharacterized, some of which may play an important role in mosqu

Wolbachia is an obligate intracellular α-proteobacterium which commonly infects arthropods and filarial nematodes. Different strains of Wolbachia are capable of a wide range of regulatory manipulations in many hosts and modulate host cellular differentiation to influence host reproduction. The genetic basis for the majority of these phenotypes is unknown. The wWil strain from the neotropical fruit fly, Drosophila willistoni, exhibits a remarkably high affinity for host germline-derived cells relative to the soma. This trait could be leveraged for understanding how Wolbachia influences the host

Lucilia cuprina, a species of blowfly, consists of two recognized subspecies: L. cuprina cuprina and L. cuprina dorsalis. Although they are morphologically and molecularly similar to each other, they have very different ecological roles. In Australia, L. c. dorsalis is predominantly found in rural areas and is the primary causative agent of sheep myiasis (flystrike), while L. c. cuprina is necrophagous and not a significant pest of livestock in the Americas or elsewhere. Here, we present a chromosome-scale genome assembly for L. c. cuprina and an improved assembly for L. c. dorsalis, enabling

BACKGROUND: Hippobosca camelina (camel ked) is an obligate hematophagous ectoparasite that infests camels. Hematophagy inflicts painful bites leading to myiasis, anemia and pathogen transmission such as Candidatus Anaplasma camelii. A genome assembly for this biting flies is currently unavailable, limiting understanding of its genetics, including the chemosensory system.
RESULTS: The assembly size for Hippobosca camelina female is 135.6 Mb with 17.08% repeated regions, an N50 of 1.2 Mb, a total of 2,182 contigs, a GC content of 33.5% and compleasm (Busco) completeness of 95.38% with the dipte

Vector-borne diseases (VBDs) cause enormous health burden worldwide, as they account for more than 17% of all infectious diseases and over 700,000 deaths each year. A significant number of these VBDs are caused by RNA virus pathogens. Here, we used metagenomics and metabarcoding analysis to characterize RNA viruses and their insect hosts among biting midges from Kenya. We identified a total of 15 phylogenetically distinct insect-specific viruses. These viruses fall into six families, with one virus falling in the recently proposed negevirus taxon. The six virus families include Partitiviridae,

OBJECTIVE: The principal delivery method for CRISPR-based genome editing in insects is now based on microinjection into single cells or embryos. The direct protein transduction systems cannot be employed in aphids because oogenesis occurs without apparent vitellogenesis. Given the limited timing of injection into the embryonic stage in oviparous aphids, a protein delivery system from the hemolymph to the germline and embryos would be a useful tool for genome editing. This study reports a newly developed direct protein delivery system for aphids using cell-penetrating peptides (CPPs). CPPs are

Insulin-like peptides (ILPs) have been identified in several invertebrates, particularly insects, and work on these ILPs has revealed many roles including regulation of energy homeostasis, growth, development, and lifespan to name a few. However, information on arthropod ILPs outside of insects is sparse. Studies of Ixodid tick ILPs are particularly scarce, despite their importance as vectors of infectious agents, most notably Lyme disease. The recent publication of the genome of the black-legged tick, Ixodes scapularis, has advanced opportunities to study this organism from a molecular standp

BACKGROUND: Phlebotomus papatasi is a natural vector of Leishmania major, which causes cutaneous leishmaniasis in many countries. Simple sequence repeats (SSRs), or microsatellites, are common in eukaryotic genomes and are short, repeated nucleotide sequence elements arrayed in tandem and flanked by non-repetitive regions. The enrichment methods used previously for finding new microsatellite loci in sand flies remain laborious and time consuming; in silico mining, which includes retrieval and screening of microsatellites from large amounts of sequence data from sequence data bases using micros

Chagas disease is an illness caused by the protozoan Trypanosoma cruzi that is distributed in 21 countries of Latin America. The main way of transmission of T. cruzi is through the feces of triatomines infected with the parasite. With technological advances came new technologies called omics. In the pre-genomic era, the omics science was based on cytogenomic studies of triatomines. With the Rhodnius prolixus genome sequencing project, new omics tools were applied to understand the organism at a systemic level and not just from a genomic point of view. Thus, the present review aims to put toget
