Rtt107 cooperates with Rad55 to limit specific types of genome instability in Saccharomyces cerevisiae.

A range of genome maintenance factors respond to endogenous and exogenous DNA damage to prevent mutations and cell death. The scaffold protein, Rtt107, is important for the growth of cells exposed to DNA-damaging agents in the budding yeast Saccharomyces cerevisiae. Rtt107 binds to a diverse array of partner proteins and responds to DNA damage by localizing to phosphorylated histone H2A. Rad55-Rad57, a heterodimer involved in DNA repair, also binds to Rtt107, but the function of the Rtt107-Rad55-Rad57 complex remains unclear. In addition to their sensitivity to DNA-damaging agents, rtt107∆ mut

Tissue-specific TCF4 triplet repeat instability revealed by optical genome mapping.

BACKGROUND: Fuchs endothelial corneal dystrophy (FECD) is the most common repeat-mediated disease in humans. It exclusively affects corneal endothelial cells (CECs), with ≤81% of cases associated with an intronic TCF4 triplet repeat (CTG18.1). Here, we utilise optical genome mapping (OGM) to investigate CTG18.1 tissue-specific instability to gain mechanistic insights. METHODS: We applied OGM to a diverse range of genomic DNAs (gDNAs) from patients with FECD and controls (n = 43); CECs, leukocytes and fibroblasts. A bioinformatics pipeline was developed to robustly interrogate CTG18.1-spanning

Nuclear-receptor-mediated telomere insertion leads to genome instability in ALT cancers.

The breakage-fusion-bridge cycle is a classical mechanism of telomere-driven genome instability in which dysfunctional telomeres are fused to other chromosomal extremities, creating dicentric chromosomes that eventually break at mitosis. Here, we uncover a distinct pathway of telomere-driven genome instability, specifically occurring in cells that maintain telomeres with the alternative lengthening of telomeres mechanism. We show that, in these cells, telomeric DNA is added to multiple discrete sites throughout the genome, corresponding to regions regulated by NR2C/F transcription factors. The

EBV and genomic instability--a new look at the role of the virus in the pathogenesis of Burkitt's lymphoma.

Epidemiological and molecular evidence links Epstein-Barr virus (EBV) carriage to the pathogenesis of human malignancies of lymphoid and epithelial cell origin but the mechanisms of viral oncogenesis are poorly understood. Burkitt's lymphoma, a tumor occurring in both EBV-positive and -negative forms, provides a convenient model for analysis of the relative contribution of genetic changes and viral products that are expressed in the malignant cells. Here we review recent findings that highlight several mechanisms by which EBV could play an important role in oncogenesis by promoting genomic ins

Micronucleus cytome assay reveals genomic instability in premalignant cervical lesions among high-risk HPV-positive women.

Cervical cancer is a major health burden driven by persistent infection with high-risk human papillomavirus (HPV). Genomic instability biomarkers from the micronucleus cytome (MNcyt) assay may improve identification of women at risk of progression beyond cytology and HPV testing. A cross-sectional study of 184 women attending routine cervical cancer screening was conducted in Popayán, Colombia. Exfoliated cervical epithelial cells were collected for HPV DNA genotyping, liquid-based cytology, and MNcyt analysis using the Feulgen-Fast Green DNA-specific method. For each sample, 2,000 cells were

Genomic instability and biofilm determinants in Streptococcus mutans: insights from a sequence-defined arrayed transposon library.

Streptococcus mutans is a primary architect of dental caries, utilizing complex genetic networks to build resilient, acid-producing biofilms. While pooled screens (Tn-seq) have identified important fitness factors, they often fail to capture extracellular or moderate-effect determinants due to community-level masking. Therefore, to study biofilm phenotypes, we constructed a comprehensive arrayed library of 9,216 mutants and used Cartesian Pooling-Coordinate Sequencing (CP-CSeq) to establish a sequence-defined resource covering 51% of non-essential genes. By screening the entire collection in i

HRD Score: A Promising Biomarker for Genomic Instability Evaluation for Targeted Treatments Among Pancreatic Cancer Patients.

IntroductionA subgroup of pancreatic cancer with unstable genome, such as BRCA mutation, may be more sensitive to platinum-based chemotherapy. How to define the patients with homologous recombination deficiency (HRD) status other than BRCA mutation has been a clinical challenge and interest.MethodsIn this retrospective cohort study, we collected NGS data of 163 pancreatic cancer patients from July 2020 to April 2024. HRD score was calculated by 3DMed-HRD algorithm. The median of HRD score of our study population was used as potential cut-off value to determine HRD status. Cox regression was us

tRNA modification genes are associated with genomic instability, proliferative programs, and poor prognosis in breast cancer.

The modifications of transfer RNA (tRNA) add an extra layer of post-transcriptional gene regulation in cancer biology. While translation initiation and termination have been extensively studied, the role of tRNA modifications in elongation remains largely unexplored. These chemical changes, particularly in the anticodon loop, enhance codon–anticodon pairing and translation fidelity, but their impact on cancer, especially breast cancer, is not well defined. We analyzed 73 tRNA modification–related genes across 33 TCGA cancer types and breast cancer subtypes by integrating transcriptomic and pro

Assessment of cytotoxicity and genomic instability induced by coal-derived nanoparticles in V79 and HaCaT Cells.

Coal is a major global energy resource, but its extraction raises significant environmental and health concerns. Mining activities release large amounts of particulate matter, including nanoparticles (NPs), into the environment. These NPs are hazardous due to their content of polycyclic aromatic hydrocarbons (PAHs), metals, oxides, and other compounds capable of disrupting cellular and molecular processes. This study evaluated the cytotoxic and genotoxic effects of coal-derived NPs on V79 and HaCaT cell lines, focusing on their impact on DNA stability and the mechanisms responsible for cellula

Young Aggressive Prostate Tumors Exhibit Increased Genomic Instability and an Exploratory RNA-CNA-Concordant Transcriptomic Pattern.

Prostate cancer (PCa) is a highly heterogeneous disease, and the molecular features underlying differences in tumor aggressiveness remain incompletely understood. This study aimed to characterize the molecular differences between young aggressive (YA) and old indolence-associated (OI) prostate tumors using an integrative multi-omics approach. RNA sequencing, copy number alteration (CNA), and clinical data from The Cancer Genome Atlas Prostate Adenocarcinoma (TCGA-PRAD) cohort were analyzed. YA tumors (age ≤ 58 years, Gleason score ≥ 8; n = 30) were compared with OI tumors (age ≥ 67 years, Glea

POLE Deficiency Exacerbates Diesel Engine Exhaust-Induced Genomic Instability and Malignant Transformation of Bronchial Epithelial Cells.

Diesel engine exhaust (DEE) is classified as a Group I carcinogen, yet direct experimental evidence that DEE induce carcinogenesis is lacking. Here, it is innovatively discovered that 120 µg mL-1 DEE exposure for 20 weeks, original bronchial epithelial cells exhibit abnormal morphology, form colonies in soft agar, and readily develop the lumps under the subcutaneous tissue of immunodeficient mice, which are pathologically confirmed as lung squamous cell carcinoma. Whole genome sequencing and RNA sequencing identify DEE-induced mutational signatures (DBS3, ID1/ID2), associated with polymerase e

Heat stress-induced genomic instability in neural stem cells and its association with neuronal developmental deficits.

BACKGROUND: Maternal hyperthermia, an increasingly prevalent environmental stressor linked to global climate change, is epidemiologically associated with a higher risk of neurodevelopmental disorders, including autism and schizophrenia. However, the molecular mechanisms underlying this neurotoxicity, particularly those leading to long-term neurological deficits, remain poorly understood. This study tested the novel hypothesis that hyperthermia acts not only as an acute physiological stressor but also as a potent genotoxic agent, inducing persistent genomic alterations in neural progenitor cell

ABL1 kinase plays an important role in spontaneous and chemotherapy-induced genomic instability in multiple myeloma.

AbstractGenomic instability contributes to cancer progression and is at least partly due to dysregulated homologous recombination (HR). Here, we show that an elevated level of ABL1 kinase overactivates the HR pathway and causes genomic instability in multiple myeloma (MM) cells. Inhibiting ABL1 with either short hairpin RNA or a pharmacological inhibitor (nilotinib) inhibits HR activity, reduces genomic instability, and slows MM cell growth. Moreover, inhibiting ABL1 reduces the HR activity and genomic instability caused by melphalan, a chemotherapeutic agent used in MM treatment, and increase

Arsenite Impairs BRCA1-Dependent DNA Double-Strand Break Repair, a Mechanism Potentially Contributing to Genomic Instability.

BRCA1 is a key player in maintaining genomic integrity with multiple functions in DNA damage response (DDR) mechanisms. Due to its thiol-rich zinc-complexing domain, the protein may also be a potential target for redox-active and/or thiol-reactive (semi)metal compounds. The latter includes trivalent inorganic arsenic, which is indirectly genotoxic via induction of oxidative stress and inhibition of DNA repair pathways. In the present study, we investigated the effect of NaAsO2 on the transcriptional and functional DDR. Particular attention was paid to the potential impairment of BRCA1-mediated

Effects of Curcumin and Soy Isoflavones on Genomic Instability of Human Colon Cells NCM460 and SW620.

Curcumin (CUR) and soy isoflavones (SIs) are two plant-based polyphenols that have attracted much attention, because of their extensive anticancer and health maintenance effects. However, the relevant molecular mechanisms are still uncertain. Genomic instability (GIN) refers to a combination of gene abnormal amplification, sequence deletion, ectopic, and other types of gene damage in cells, and it is one of the main factors causing cells to lose normal physiological functions. Therefore, we used the cytokinesis-block micronucleus cytome (CBMN-Cyt) assay as the main research method to analyze t

High Dietary Folic Acid Intake Is Associated with Genomic Instability in Peripheral Lymphocytes of Healthy Adults.

Mandatory fortification of food with synthetic folic acid (FA) was instituted in 1998 to reduce the incidence of neural tube defects. Adequate folate status is correlated with numerous health benefits. However, elevated consumption of FA is controversially associated with deleterious effects on health. We previously reported that excess FA mimicked folate depletion in a lymphoblastoid cell line. To explore the impact of FA intake from fortified food, we conducted an observational human study on 33 healthy participants aged 18-40 not taking any supplements. Food intake, anthropomorphic measurem

Obesity, oxidative DNA damage and vitamin D as predictors of genomic instability in children and adolescents.

BACKGROUND/OBJECTIVES: Epidemiological evidence indicates obesity in childhood and adolescence to be an independent risk factor for cancer and premature mortality in adulthood. Pathological implications from excess adiposity may begin early in life. Obesity is concurrent with a state of chronic inflammation, a well-known aetiological factor for DNA damage. In addition, obesity has been associated with micro-nutritional deficiencies. Vitamin D has attracted attention for its anti-inflammatory properties and role in genomic integrity and stability. The aim of this study was to determine a novel
