Selective autophagy, mediated by Atg8 binding proteins, has not been extensively studied in plants. Plants possess a large gene family encoding multiple isoforms of the Atg8 protein. We have recently reported the identification of two new, closely homologous Arabidopsis thaliana plant proteins that bind the Arabidopsis Atg8f protein isoform. These two proteins are specific to plants and have no homologs in nonplant organisms. The expression levels of the genes encoding these proteins are elevated during carbon starvation and also during late stages of seed development. Exposure of young seedli

BACKGROUND: Clostridium difficile is an anaerobic, spore-forming bacterium that is the most common cause of healthcare-associated diarrhea in developed countries. Control of C. difficile is challenging because the spores are resistant to killing by alcohol-based hand hygiene products, antimicrobial soaps, and most disinfectants. Although initiation of germination has been shown to increase susceptibility of spores of other bacterial species to radiation and heat, it was not known if triggering of germination could be a useful strategy to increase susceptibility of C. difficile spores to radiat

The Bacillus anthracis spore is the causative agent of the disease anthrax. The outermost structure of the B. anthracis spore, the exosporium, is a shell composed of approximately 20 proteins. The function of the exosporium remains poorly understood and is an area of active investigation. In this study, we analyzed the previously identified but uncharacterized exosporium protein ExsK. We found that, in contrast to other exosporium proteins, ExsK is present in at least two distinct locations, i.e., the spore surface as well as a more interior location underneath the exosporium. In spores that l

In order to evaluate the ability of three types of extracts to explain the ecotoxicological risk of treated municipal sewage sludges, the OECD 208A germination test was applied using three plants (Lolium perenne L., Brassica rapa L., and Trifolium pratense L.). Three equivalent batches of sludge, remained as dewatered sludge, composted with plant remains and thermally dried, from an anaerobic waste water treatment plant were separated. Samples from these three batches were extracted in water, methanol, and dichloromethane. Plant bioassays were performed and the Germination Index (GI) for the t

Inhalation of resting conidia is usually the first step of a systemic infection caused by the opportunistic fungal pathogen Aspergillus fumigatus. In the lung, the inhaled spores encounter an environment that permits germination. However, the relative importance of certain environmental conditions for conidial activation and subsequent hyphae formation has so far not been analyzed in detail. In this study, we studied the role of oxygen during germination. We found that inhibitors of the respiratory chain were nearly as efficient in blocking germination as cycloheximide, an inhibitor of protein

The developmental pattern of dark-grown Arabidopsis thaliana is dramatically shifted by exposure of the seedlings to light: inhibition of hypocotyl (stem) growth is one of the typical responses. Here, we show that the hypocotyl growth of dark-grown seedlings is reduced by exposure of the seeds to light. The light signal is perceived by phytochromes A and B during the hours immediately prior to seed germination. The effect is obviously selective, as other processes under phytochrome control were not equally affected by the pre-germination light cue. The hypocotyl response persists for two days

Observing that the conidia of Stachybotrys chartarum can germinate in the lung of infant rats, it became important to ascertain whether an infection can ensue. Viable conidia of S. chartarum were instilled into the lungs of 4 and 14 day-old rat pups. Germination was observed frequently in the lungs of 4 day-old but rarely in the 14 day-old pups. In the 4 day-old pups, pulmonary inflammation with hemorrhagic exudates was observed and resulted in about 15% mortality rate compared to 0% for the controls instilled with phosphate buffered saline. Acute neutrophilic inflammation and intense intersti

The role of Cl- transport across the plasma membrane was studied in an early step of pollen grain germination in tobacco Nicotiana tabacum L. The Cl- channel blockers, 5-nitro-2-(3-phenylpropylamino)benzoic acid (NPPB) and niflumic acid, completely suppress the germination with IC(50) approximately 8 micro M. At this concentration NPPB reduces the rate of Cl- efflux out of pollen grain by 1.8-fold in the interval 5-12 min, and niflumic acid reduces the rate 1.2-fold. 4,4;-Diisothiocyanatostilbene-2,2;-disulfonic acid, a known inhibitor of Cl- channels and antiporters, completely suppresses ger

Gene expression profiling has identified 2 major subclasses of diffuse large B-cell lymphoma (DLBCL). Cases resembling germinal center (GC) B cells (GCB-DLBCL) generally occur in younger patients, have a distinct molecular pathophysiology, and have improved outcomes compared with those similar to activated post-GC cells (activated B-cell DLBCL). We previously found that the ubiquitin hydrolase UCH-L1 is frequently overexpressed in mature B-cell malignancies and is a potent oncogene in mice. The cause for its overexpression in lymphoma, and whether it impacts the outcome of patients with DLBCL

The simultaneous nutrient germination of hundreds of individual wild-type spores of three Bacillus species and a number of Bacillus subtilis strains has been measured by two new methods, and rates of release of the great majority of the large pool of dipicolinic acid (DPA) from individual spores of B. subtilis strains has been measured by Raman spectroscopy with laser tweezers. The results from these analyses and published data have allowed a number of significant conclusions about the germination of spores of Bacillus species as follows. (i) The time needed for release of the great majority o

To assess the impact of the germinal center (GC) reaction on viral spread in Epstein-Barr virus (EBV) infection, we isolated EBV(+) GC B cells from the tonsils of two infectious mononucleosis patients, sequenced their rearranged V genes, and determined expression of the EBV latency genes EBV nuclear antigen 2 and latent membrane protein 1. Most EBV(+) GC B cells belonged to clones of cells harboring somatically mutated V gene rearrangements. Ongoing somatic hypermutation, the hallmark of the GC reaction, was seen only in uninfected GC B cell clones, not in EBV(+) B cell clones. Thus, in infect

Plants integrate multiple environmental cues to decide the timing of seed germination. Light promotes germination primarily through photoreceptor phytochrome B (phyB), which eliminates the action of transcription factor PHYTOCHROME-INTERACTING FACTOR 1 (PIF1). Conversely, abiotic stresses typically elevate cytosolic Ca2+ and inhibit germination. However, the coordination of light and calcium signaling during germination under stress conditions remains poorly understood. Here, we demonstrate that salt and osmotic stresses rapidly increase cytosolic Ca2+ in imbibed seeds, and that the calcium se

BACKGROUND: Strigolactones are root-exuded plant metabolites acting as germination stimulants for several parasitic weeds. While strigolactone bioactivity in soil is assumed to be temporally and spatially restricted, quantitative estimates of their signaling distance are lacking. This study aimed to quantify strigolactone signaling distance in soils of contrasting texture, as an estimate of physical transport distance, and to assess the influence of soil biological activity and water content.
RESULTS: Phelipanche aegyptiaca seed germination served as a biological indicator in a soil column ex

Programmed cell death (PCD) is a pivotal biological process that occurs at various stages of plant development, including embryogenesis and seed germination. This study investigated whether the absence of PCD in endosperm cells is connected to the poor germination of Viola odorata seeds. Seeds of poorly germinating V. odorata and well-germinating V. × wittrockiana were either cold-stratified for 10 days or left untreated. Germination tests, tetrazolium viability tests, Western blot analyses for caspase-like proteases, and Terminal deoxynucleotidyl transferase (TdT) dUTP nick end labeling (TUNE

Seed dormancy and germination are tightly regulated by complex signaling networks that integrate internal and external cues, including the endogenous phytohormone abscisic acid (ABA). ABA HYPERSENSITIVE GERMINATION 1 (AHG1), a group A type 2C protein phosphatase (PP2C), is thought to modulate the activity of transcription factors such as ABA INSENSITIVE 5 (ABI5) in seeds and during germination. AHG1 is regulated by DELAY OF GERMINATION 1 (DOG1), a key regulator of seed dormancy, through physical interaction. We previously reported that AHG1 also interacts with ABI FIVE BINDING PROTEIN 2 (AFP2)

BACKGROUND AND AIMS: Fire-related cues can influence germination and seedling establishment in fire-prone ecosystems by breaking physical dormancy (PY) or stimulating germination in species with water-permeable seeds. However, most studies rely on fresh seeds and overlook how soil seed bank storage may change germination responses to fire cues. We investigated whether in situ seed storage affects germination responses to heat shock and smoke in Cerrado species.
METHODS: Seeds of 13 Cerrado species, dispersed at the beginning of either the dry or rainy season, were collected and exposed to hea

Sesame (Sesamum indicum) seeds accumulate specialized lignans, including (+)-sesamin, (+)-sesamolin, and (+)-sesaminol triglucoside (SL-TG). Although lignan biosynthesis during seed development is well characterized-with SiCYP92B14 recognized as a (+)-sesamin-specific oxygenase-the molecular basis of the metabolic transition during germination, where lipophilic lignans are fully converted into glucosides, remained unclear. Herein, we identify a set of (+)-sesamin oxygenases, SiCYP706V12-V14, cytochrome P450 enzymes (CYPs) that exhibit a broader substrate range than SiCYP92B14. These enzymes ox

Pollen viability is a key parameter for understanding the functioning of plant reproductive systems and for supporting strategies aimed at genetic resource conservation and plant breeding. Accurate assessments of this attribute are essential for identifying genotypes with a higher reproductive rate, monitoring prezygotic barriers, testing compatibility between individuals, and planning controlled crosses, especially in cases of floral asynchrony or spatial isolation. Accordingly, this chapter compiles the main methods used for this purpose, providing detailed and up-to-date protocols. It cover

Food security is one of the world's top challenges, specifically considering global issues like climate change. Seed priming is one strategy to improve crop production, typically via increased germination, yields, and/or stress tolerance. Hydropriming, or soaking seeds in water only, is the simplest form of seed priming. However, the addition of certain seed priming agents has resulted in a variety of modified strategies, including osmopriming, halopriming, hormonal priming, PGR priming, nutripriming, and others. Most current research has focused on hormonal and nutripriming. This review will

Moringa stenopetala is a rapidly growing, unappreciated tree regarded as the "miracle tree" for its food, feed, and medicinal benefits. It appears to be a versatile and promising species for use under changing conditions. However, the biological response of Moringa stenopetala, a valuable but underutilized species, to drought remains unclear during its early growth stages. Therefore, the main aim of this study was to investigate the morphological, physiological, and biochemical responses of M. stenopetala seedlings to different levels of drought stress induced by PEG-6000. Four concentrations
