Anti-inflammatory glycoside withanolides from leaves of Athenaea velutina.

The phytochemical study of the CH2Cl2 and EtOAc soluble fractions from the leaf ethanol extract of Athenaea velutina resulted in the isolation of the new withanolides 1-3, two of which substituted at the C-3 by a disaccharide 3β-O-[α-L-rhamnopyranosyl-(1 → 4)-β-d-glucopyranosyl], and four others previously reported (4-7). The structures of the new withanolides were elucidated by detailed interpretation of their 1D- and 2D-NMR spectra and the HRESIMS analysis. An anti-inflammatory screening was performed using the two major withanolides (1 and 2), analyzing their effects on the viability of RAW

Synthesis of Andrographolide glycosides and evaluation of their antibacterial activity in vitro and in vivo.

Andrographolide, a diterpenoid lactone isolated from Andrographis paniculata, exhibits broad-spectrum biological activities but suffers from poor water solubility and limited oral bioavailability due to its native structure. In recent years, glycosylation has emerged as a promising strategy to improve the pharmacokinetic and pharmacodynamic properties of such compounds. In this study, a series of glycosylated derivatives of 14-deoxy-11,12-didehydroandrographolide, serving as aglycone scaffolds, were systematically synthesized for the first time. Their antibacterial activities were comprehensiv

Norwegian scabies in a patient treated with Tripterygium glycoside for rheumatoid arthritis.

We report an 80-year-old male patient with severe rheumatoid arthritis who was treated with tripterygium glycoside, an immunosuppressive agent made from the extract of a Chinese medicinal herb called Tripterygium wilfordii Hook F. The patient had no apparent skin lesions before the treatment, but he developed aggressive hyperkeratotic lesions with rapid progression after using tripterygium glycoside. He was repeatedly diagnosed with eczema, but treatment failed to achieve efficacy. Interestingly, a microscopic examination of the lesions revealed numerous scabies mites and eggs. Thus, we confir

Fatal cardiac glycoside poisoning due to mistaking foxglove for comfrey.

CONTEXT: Accidental ingestion of foxglove (Digitalis purpurea) can cause significant cardiac toxicity. We report a patient who ingested foxglove mistaking it for comfrey and developed refractory ventricular arrhythmias. The patient died despite treatment with digoxin-specific antibody fragments (DSFab) and veno-arterial extracorporeal membrane oxygenation (VA-ECMO). CASE DETAILS: A 55-year-old woman presented to the emergency department with nausea, vomiting and generalized weakness eight hours after drinking "comfrey" tea. She had bradycardia (54 beats/min) and hyperkalemia (7.6 mEq/L). Elec

Acutifoliside, a novel benzoic acid glycoside from Salix acutifolia.

Ultra high-performance liquid chromatography-mass spectrometry (UHPLC-MS) profiling of a polar solvent extract of juvenile stem tissue of Salix acutifolia Willd. identified a range of phenolic metabolites. Salicortin, 1, a well-known salicinoid, was the major compound present and the study identified young stem tissue of this species as a potential source of this compound for future studies. Three further known metabolites (salicin 2, catechin 3 and tremuloidin 4) were also present. The UHPLC-MS analysis also revealed the presence of a further, less polar, unknown compound, which was isolated

[What are the indications for digitalis glycosides in the current treatment of cardiac insufficiency?].

Digitalis was discovered over two centuries ago and has been in everyday use for 100 years: however nowadays it is not considered to be the almost exclusive treatment of heart failure as it used to be with diuretic therapy. In the last decade, our understanding of the physiopathology of heart failure and the body's mechanisms of adaptation have improved and this has lead to the use of new molecules and a different approach to the problem of left ventricular failure. The classical contra-indications and precautions of use of digitalis have been studied in detail. The use of digitalis is contest

Effects of basic drugs on the hepatic transport of cardiac glycosides in rats.

The effects of some basic and acidic drugs on the hepatic uptake of digoxin and ouabain were studied in isolated rat hepatocytes. Digoxin accumulated against a concentration gradient, and its initial uptake was energy- and temperature-dependent. Digoxin competitively inhibited the uptake of ouabain (Ki = 1.3 microM), which was reported to be transported by a carrier-mediated active transport system. All basic drugs tested (verapamil, dipyridamole, amiodarone, nifedipine, diltiazem, ajmaline, chlorpromazine, imipramine, disopyramide, quinidine, procainamide, propranolol and lidocaine: 50 microM

Toxicity of the anthraquinone glycoside P-1894B for human skin fibroblasts.

P-1894B inhibits prolyl hydroxylase in vitro and has been proposed as a topical treatment for dermal fibrosis. The drug had similar effects on two fibroblast lines from normal human skin and one line from a patient with lichen sclerosus et atrophicus. Exposure of logarithmically-growing cell monolayers for 72 h caused dose-dependent inhibition of proliferation at 0.05-0.5 microgram/ml but time-dependent cell death at 1-50 micrograms/ml. The epithelial cell line NCTC 2544 gave a similar result. Collagen lattices containing normal fibroblasts contracted more slowly in the presence of the drug at

Cardiac glycoside binding sites in cultured heart muscle cells.

Binding of (3H)-ouabain to cultured cardiac muscle and non muscle cells from chicken embryos and neonatal rats has been characterized and correlated with ouabain-induced inhibition of the sodium pump, as well as with the positive inotropic action of the drug. Cardiac muscle and non muscle cells from 10-12 day-old chicken embryos are characterized by a single class of ouabain binding sites (muscle cells: dissociation constant KD = 1.5 X 10(-7) M; binding capacity B = 2.6 pmoles/mg cell protein). Two classes of ouabain binding sites, however, have been found in cardiac muscle and non muscle cell

Pharmacokinetic and toxicological studies of antimony dextran glycoside (RL-712).

1. The absorption, tissue distribution and excretion of antimony dextran glycoside (RL-712) has been studied in normal rodents.2. Some organs in the body, especially liver and spleen, retain large amounts of antimony for considerable periods of time. Excretion of antimony in the urine was low and only about 10-12% of the dose administered was excreted within the first 48 hours.3. Blood levels were maintained for at least 3 days after a single intramuscular dose to rabbits, corresponding to 14 mg Sb/kg body weight.4. Toxicity studies and tests on foetal toxicity in mice and rats, respectively,

Bridging Pharmacopeial Standards and Chemical Reality of Triterpene Glycosides in Actaea racemosa.

Actaea racemosa L. (syn. Cimicifuga racemosa (L.) Nutt., black cohosh) rhizomes contain triterpene glycosides (TTGs) that serve as principal quality markers in the European Pharmacopoeia (Ph. Eur.) monograph. However, discrepancies between chromatographic profiles of the plant material and the herbal reference standard (HRS) dry extract complicate quality assessment. This study elucidated the chemical origin of these discrepancies and evaluated whether the extract-based HRS can represent the analytical fingerprint of the plant material. Thirteen TTGs were identified and characterized within th

Steroidal glycosides from Tylophora ovata with anti-inflammatory and radical scavenging activities.

A chemical investigation on 80% ethanol extract from the whole plants of Tylophora ovata afforded two new C21 steroidal glycosides, ovatacosides A and B (1 and 2). Structural elucidation of both compounds were performed by spectral methods such as 1D and 2D (1H-1H COSY, HMQC, and HMBC) NMR spectroscopy and high resolution mass spectrometry. The isolated components were evaluated in vitro for anti-inflammatory activities against COX-1 and COX-2 and radical scavenging potential using DPPH test. Consequently, 1 exhibited modest selective inhibition on COX-1 (61.9%) at the concentration of 100 μM
