Changes in the ultrastructure and function of goldfish Mauthner neurons in the presence of 3,4-dihydro-2(1H)-pyrimidinethione.

The behavioral effects of 3,4-dihydro-2(1H)-pyrimidinethione (DPT, a pyrimidine derivative), which is used as a test system for detecting tumor growth, on the ultrastructure and function of Mauthner neurons (MN), were studied in goldfish. Application of DPT to MN was found to lead to increased resistance of neurons to exhaustive stimulation, which was accompanied by increases in the sizes of actin-containing membrane desmosome-like contacts, along with the formation of bundles of actin stress fibers; these effects are similar to those previously reported with dopamine. The similarity of the mo

Molecular characterization of E587 antigen: an axonal recognition molecule expressed in the goldfish central nervous system.

The E587 antigen (Ag) is a 200-Kd membrane glycoprotein originally identified by a monoclonal antibody on new and regenerating retinal ganglion cell axons in the adult goldfish. We report the isolation of cDNAs encoding the E587 Ag and identify it as a member of the L1 family of cell adhesion molecules (CAMs). The predicted amino acid sequence of E587 Ag shows an approximately equal identity (40%) to mouse L1, chick neuron-glia CAM, and chick neuron-glia-related CAM. Although the overall similarity is low, there is a high conservation of structural domains and specific sequence motifs. Wholemo

Inhibition of non-neuronal cell proliferation in the goldfish visual pathway affects the regenerative capacity of the retina.

Proliferating cells associated with the visual pathway were found in the present study to affect the regenerative capacity of the goldfish retina following optic nerve injury. The contribution of these cells to the process of regeneration was investigated in the goldfish visual system by reducing their proliferation in the optic tract and tecta, using X-irradiation. The regenerative ability of the retina was then evaluated by the following parameters: sprouting from retinal explants, protein synthesis in the retina and accumulation of radiolabeled transported components in the tectum. X-irradi

Trysinase positive oculocutaneous albinism in the goldfish, Carassius auratus l., and ultrastructural and biochemical study of the eye.

Ultrastructural studies, and cytochemical and biochemical determinations of tyrosinase activity were conducted on the pigment epithelium of albino and xanthic goldfish eyes. In eyes of xanthic goldfish, two types of melanosomes are present, spherical and elongated. Melanized melanosomes are absent in the eyes of the albino goldfish, but elongated lamellar premelanosomes are observed. Internal vesicles are present in both melanosome types in the pigment epithelium of the xanthic goldfish but are absent in premelanosomes of the albino. There are also differences in the distribution of lipid drop

Spontaneous polyploidy, gynogenesis and androgenesis in second generation (F2 ) koi Cyprinus carpio × goldfish Carassius auratus hybrids.

The objective of this study was to characterize the genetics of second generation (F2 ) koi Cyprinus carpio × goldfish Carassius auratus hybrids. Spermatozoa produced by a novel, fertile F1 male were found to be diploid by flow-cytometric analysis. Backcross (F1 female × C. carpio male and C. carpio female × F1 male) juveniles were triploid, confirming that female and male F1 hybrids both produced diploid gametes. The vast majority of surviving F2 juveniles was diploid and small proportions were aneuploid (2·1n-2·3n and 3·1n-3·9n), triploid (3n) and tetraploid (4n). Microsatellite genotyping s

A purine-sensitive pathway regulates multiple genes involved in axon regeneration in goldfish retinal ganglion cells.

In lower vertebrates, retinal ganglion cells (RGCs) can regenerate their axons and reestablish functional connections after optic nerve injury. We show here that in goldfish RGCs, the effects of several trophic factors converge on a purine-sensitive signaling mechanism that controls axonal outgrowth and the expression of multiple growth-associated proteins. In culture, goldfish RGCs regenerate their axons in response to two molecules secreted by optic nerve glia, axogenesis factor-1 (AF-1) and AF-2, along with ciliary neurotrophic factor. The purine analog 6-thioguanine (6-TG) blocked outgrowt

Integrated Sensory and Immune Cell Organization in the Lip Skin of the Goldfish (Carassius auratus, Linnaeus, 1758).

This study examines the histological, ultrastructural, and immunohistochemical features of the lip skin of the goldfish (Carassius auratus, Linnaeus, 1758), a sensory-rich region that plays an essential role in feeding and environmental perception. Our findings highlight the coexistence and close association of immune, epithelial, and sensory cells within the epidermis and dermis. For the first time in goldfish, intraepidermal macrophages, eosinophilic granular cells, rodlet cells, Merkel cells, and several specialized sensory structures-neuromasts, taste buds, and tuberous-like sensory units-

The growth of developing oocytes and hypothalamus-pituitary-ovary functions in goldfish under absolute food deprivation.

Food deprivation (FD) seriously affects puberty onset and oogenesis in female fishes. It has been suggested that meal-responsive peptides interact with hypothalamus-pituitary-ovary (HPO) functions; however, current knowledge regarding the integration of metabolism with reproduction remains inconclusive. In the present study, we investigated the effects of 7 days absolute FD on the reproductive system of adult female goldfish (Carassius auratus). The number of nonvitellogenic oocytes was higher in the food-deprived goldfish than in fed goldfish, whereas the number of vitellogenic oocytes was lo

Prevalence of cyprinid herpesvirus-2 in goldfish aquarium shops in Tamil Nadu, India: a case report.

This study reports cyprinid herpesvirus-2 infection in farm-reared goldfish in Tamil Nadu during surveillance between 2022 and 2024. CyHV-2 is a temperature-dependent viral pathogen that causes mortality during temperature fluctuations of 18-24°C (post monsoon). Live goldfish showing hemorrhage, skin ulcers, pale gill color, and high mortality rates were collected from aquarium shops in Kolathur, Chennai District, Tamil Nadu, India. To examine pathogenesis, PCR and RT-PCR assays were performed on disease-suspected samples using primer sets for viral infections, such as CyHV-2, CyHV-3, CEV, VHS

Comparative efficacy of herbal extracts and separately bakuchiol against the monogenean parasite Gyrodactylus kobayashii in goldfish.

Species of Gyrodactylus are common monogenean ectoparasites infecting both freshwater and marine fish. Their rapid proliferation and high transmissibility make them a serious threat to aquaculture. Current chemical treatments have drawbacks of drug residues and high resistance, requiring the discovery of safe and environmentally friendly alternatives. In this study, anthelmintic efficacy against Gyrodactylus kobayashii of eight herbal medicines was evaluated. One of the components bakuchiol from Psoralea corylifolia showed the best anthelmintic efficacy. The in vitro assays showed a positive c

White spots amidst the gold: ultrastructural and histological aspects of the chronic inflammatory response of goldfish with ichthyophthiriasis.

Ichthyophthirius multifiliis, the causative agent of white spot disease, is a ciliated protozoan parasite that infects freshwater fish and induces high mortality. Outbreaks occur both in natural and production sites. The aim of the present study was to describe the lesions caused by chronic infection by I. multifiliis in goldfish (Carassius auratus) from an ornamental fish farm, highlighting important ultrastructural aspects of this protozoan. Damaged skin and gills, collected from fish with white or ulcerative skin lesions, were routinely processed for histological analysis and transmission e

Persistence of cyprinid herpesvirus 2 in asymptomatic goldfish Carassius auratus (L.) that survived an experimental infection.

Cyprinid herpesvirus 2 (CyHV-2) is the causative agent of herpesviral haematopoietic necrosis (HVHN) in goldfish, Carassius auratus, and Prussian carp, C. auratus gibelio. In this study, we investigated virus persistence in goldfish experimentally infected with CyHV-2. Virus DNA presence in organs was monitored in survivors reared at a virus permissive temperature and also in survivors treated with a non-permissive temperature for 4 days, initiated at three different time points post-infection in order to obtain fish with different virus loads. We detected virus DNA in all organs tested at 51

Expression analysis of immune genes in goldfish (Carassius auratus) infected with the monogenean parasite Gyrodactylus kobayashii.

Heavy infection with the ectoparasite Gyrodactylus kobayashii commonly leads to high mortality of ornamental goldfish Carassius auratus. To understand the underlying mechanisms of goldfish against infection of gyrodactylids, transcription responses of immune-related genes including IL-1β2, TNFα1, TNFα2, IFN-γ, TGFβ, MHC II β, TCRβ1 and complement factor C3 were studied by real-time quantitative PCR analysis. Significant increases of expression of inflammatory genes such as IL-1β2, TNFα1, TNFα2 and TGFβ were detected at days 7 and 14 post-infection (pi). No significant differences of G. kobayas

Effects of goldfish (Carassius auratus) population size and body condition on the transmission of Gyrodactylus kobayashii (Monogenea).

Field surveys indicate that host population size, rather than density, is the most important determinant of monogenean infection dynamics. To verify this prediction, epidemic parameters were monitored for 70 days at five host population sizes held at constant density using a goldfish - Gyrodactylus kobayashii laboratory model. During the first 20 days, the rate of increase of prevalence and mean abundance was faster in small host populations. Total mean prevalence and total mean abundance throughout the experiment were not significantly affected by host population sizes. Higher transmission ra

Differential modulation of ghrelin-induced GH and LH release by PACAP and dopamine in goldfish pituitary cells.

Ghrelin (GRLN) participates in multiple physiological processes, including the regulation of growth hormone (GH) and luteinizing hormone (LH) release. In the goldfish, neuroendocrine control of GH and LH release are multifactorial. In this system, pituitary adenylate cyclase-activating polypeptide (PACAP)-stimulated GH and LH secretion, as well as dopamine (DA)-induced GH release, are mediated by protein kinase A (PKA)-dependent, but protein kinase C (PKC)-independent, mechanisms. In addition, DA inhibits LH secretion by actions at sites along both PKA and PKC signaling pathways. Recently, gol

Retina and optic tectum interact to modulate taurine effect on goldfish and rat retinal explants outgrowth.

A role for nitric oxide in hypoxia-induced activation of cardiac KATP channels in goldfish (Carassius auratus).

Hypoxia-induced shortening of cardiac action potential duration (APD) has been attributed in mammalian hearts to the activation of ATP-sensitive potassium (KATP) channels. Since KATP channels are also present at high densities in the hearts of vertebrate ectotherms, speculation arises as to their function during periods of reduced environmental oxygen. The purpose of the present study was to determine whether nitric oxide (NO) plays a role in cardiac sarcolemmal KATP channel activation during hypoxia in a species with a high degree of tolerance to low oxygen environments: the goldfish (Carassi

[Effect of polypeptides from scorpion venom on morpho-functional changes of the incubated Mauthner neurons in goldfish].

Using the electron microscopical method of negative staining it was shown that one of the fractions obtained from scorpion venom directly interacts with monomeric chromatographically pure actin causing its polymerization and transformation from globular into fibrillar form. The effect of long-term orthodromic stimulation on evoked electric activity and on the ultrastructure of Mauthner neurons (MN) in the goldfish hindbrain slices incubated in vitro was studied after the application of this venom fraction. The peptides of this fraction were shown to stabilize the amplitude of MN evoked electri

Histochemical and immunocytochemical localization of nitric oxide synthase in the central nervous system of the goldfish, carassius auratus.

The distribution of the neuronal type of nitric oxide synthase in the goldfish brain and spinal cord was investigated via NADPH-diaphorase histochemistry and immunocytochemistry using an antiserum raised against the purified mammalian enzyme. Many structures, including magnocellular neurosecretory cells, motoneurons, mesencephalic trigeminal neurons, and radial glial fibers, were stained by the NADPH-diaphorase reaction but were not immunoreactive. This nonspecific NADPH-diaphorase activity was strongly reduced after preincubation of the sections. Therefore, when sections were first reacted fo
