Human imprinting anomalies in fetal and childhood growth disorders: clinical implications and molecular mechanisms.

Genomic imprinting is among the most important epigenetic mechanisms whereby expression of a subset of genes is restricted to a single parental allele. Loss of imprinting (LOI) through hypo or hyper methylation is involved in various human syndromes. These LOI occur early during development and usually impair growth. Some imprinting syndromes are the consequences of genetic anomalies, such as uniparental disomies (UPD) or copy number variations (deletion or duplications) involving the imprinted domains; others are due to LOI at the imprinting control regions (ICR) regulating each domain. Impri

Urinary nerve growth factor levels in overactive bladder syndrome and lower urinary tract disorders.

Overactive bladder (OAB) is a syndrome based on self-reported symptoms of urgency and frequency with or without urge incontinence. Although urgency is the core symptom of OAB, patients might have difficulty to distinguish urgency from the urge to void. Urodynamic study is a useful diagnostic tool to discover detrusor overactivity (DO) in patients with OAB; however, not all OAB patients have DO. Therefore, a more objective and non-invasive way to diagnose and assess OAB including DO is needed. Recent research has focused on urinary biomarkers in assessment of OAB. Urinary nerve growth factor (N

[Relationship between bone mineral content and growth disorders in children with juvenile idiopathic arthritis].

UNLABELLED: In chronic inflammatory processes in children efforts are made to evaluate bone mineral content (BMC) with the use of densitometric parameters, which at the same time determine equivalent of lean body mass (LBM). This functional analysis of the musculoskeletal system by using DXA method seems to be particularly useful for the examination of bone mass in children suffering from juvenile idiopathic arthritis (JIA). THE AIM OF THE STUDY was to assess body mass content having regard of the BMC/LBM ratio in JIA children, depending on the degree of growth inhibition, disease advancement

Transmembrane helix heterodimerization in lipid bilayers: probing the energetics behind autosomal dominant growth disorders.

Here, we show that the energetics of transmembrane helix heterodimer formation can be characterized in liposomes using Förster resonance energy transfer (FRET). We present the theory and the protocol for measuring the free energy of heterodimerization, and the total (hetero and homo-dimeric) dimer fraction. We use the presented methodology to determine the propensity for heterodimer formation between wild-type fibroblast growth factor receptor 3 (FGFR3) transmembrane domain and the Ala391Glu mutant, linked to Crouzon syndrome with acanthosis nigricans.

Bone marrow stromal cell distribution of basic fibroblast growth factor in chronic myeloid disorders.

BACKGROUND AND OBJECTIVES: Basic fibroblast growth factor (bFGF) is a multifunctional cytokine that exerts positive regulation in hematopoiesis and that may also have a role in myelofibrosis and angiogenesis. We used bone marrow immunohistochemical stains to obtain additional insight into the cellular distribution of bFGF in both chronic myeloproliferative diseases (CMPD) and myelodysplastic syndrome (MDS). DESIGN AND METHODS: Bone marrow immunohistochemical stains were used to evaluate the cellular distribution of bFGF in 29 patients with CMPD, 31 patients with MDS, and 5 normal controls. R

Growth differences associated with compulsive and stereotyped behavior disorders in adults with mental retardation.

We have recently observed that compulsive behaviors in mentally retarded patients appear to be quite prevalent, can be reliably assessed, and have a high rate of co-occurrence with stereotyped and self-injurious behaviors in this population. As abnormal growth rate has been observed in obsessive-compulsive disorder (OCD) patients, we examined physical stature in adults with mental retardation who display repetitive movement disorders. Identification of cases with stereotypic movement disorder, and cases with compulsive behaviors was done using a symptom checklist and direct observation. Subjec

Nerve growth factor (NGF) in cerebrospinal fluid (CSF) from patients with various neurological disorders.

It has been recently shown that NGF is not only involved in the survival and development of sympathetic and neural crest-derived sensory neurons, but also in some mechanisms of the immune system. For this reason, we studied the content of NGF in CSF samples from patients with diseases in which neuroimmunological mechanisms seem to be involved (multiple sclerosis, amyotrophic lateral sclerosis, Alzheimer disease, chronic relapsing polyradiculoneuritis, Guillain-Barré syndrome, and tumors of the nervous system), as well as from a number of normal control subjects. We setup an ELISA aimed at the

Fetal Growth Disorders in Dichorionic Diamniotic IVF Twin Pregnancies: A Retrospective Longitudinal Cohort Study.

Growth disorders are common in in vitro fertilization (IVF)-conceived twins, but risk factors specific to dichorionic-diamniotic twins remain incompletely characterized. This study aimed to assess these risk factors. Data from in vitro fertilization pregnancies (December 2020 - November 2024) were retrospectively analyzed. Small for gestational age was defined as estimated fetal weight below the 10th percentile. Intertwin discordance greater than 15% was considered significant. Change in uterine artery pulsatility index was calculated by subtracting first-trimester from second-trimester values

Hypertensive disorders in pregnancies with early-onset fetal growth restriction: a retrospective cohort study.

OBJECTIVE: Early onset fetal growth restriction (FGR) shares pathophysiologic origins and may precede the development of Hypertensive Disorder of Pregnancy (HDP). The aim of this study was to evaluate risk factors associated with the development of HDP in individuals with early onset FGR. METHODS: This retrospective cohort included all consecutive patients who presented to our tertiary, university-affiliated medical center between 2011 and 2024 for suspected early-onset FGR, defined as onset before 32 weeks of gestation. Maternal and pregnancy information retrieved from hospital records inclu

Automated bone age assessment in rare pediatric growth disorders: a comparative study using Deeplasia.

OBJECTIVE: Bone age (BA) assessment is essential for monitoring growth and maturation and guiding therapeutic interventions. While deep learning (DL) models offer high-speed automated BA prediction, their generalizability to rare pathological and diagnostically complex populations remains a significant concern. This study aims to validate the open-source DL system Deeplasia on external data from pediatric patients with various syndromic, endocrine, and lysosomal storage disorders (LSDs) and to compare its accuracy and consistency against multiple expert human raters. Accuracy of skeletal scintigraphy for the evaluation of mandibular growth disorders: a systematic review.

Bone scans, reflecting blood flow and metabolic activity in a region of interest, are frequently used to evaluate mandibular growth disorders. Increased uptake is a non-specific finding and can occur as a result of multiple causes. The correlation between radioactive tracer uptake and growth activity has not been consistently demonstrated. The aim of this study was to assess the accuracy of planar skeletal scintigraphy (SS), single-photon emission computed tomography (SPECT), and SPECT with computed tomography (CT) images (SPECT/CT) in detecting abnormal mandibular growth activity compared to

White matter abnormalities in fetal alcohol spectrum disorders: Focus on axon growth and guidance.

Fetal Alcohol Spectrum Disorders (FASDs) describe a range of deficits, affecting physical, mental, cognitive, and behavioral function, arising from prenatal alcohol exposure. FASD causes widespread white matter abnormalities, with significant alterations of tracts in the cerebral cortex, cerebellum, and hippocampus. These brain regions present with white-matter volume reductions, particularly at the midline. Neural pathways herein are guided primarily by three guidance cue families: Semaphorin/Neuropilin, Netrin/DCC, and Slit/Robo. These guidance cue/receptor pairs attract and repulse axons an
