Anorectic effect of fenfluramine, cholecystokinin and neurotensin in genetically obese (ob/ob) mice.

To investigate the satiety defect of hyperphagic genetically obese (ob/ob) mice, acute feeding responses to three differently acting anorectic agents were determined in 7-9 weeks old lean (+/+) and ob/ob mice habituated to a restricted (0900-1230 hr) daily feeding routine. Fenfluramine (10 mg/kg), cholecystokinin (100 U/kg) and neurotensin (500 micrograms/kg), administered intraperitoneally 15 min before feeding, each produced a rapid but transient suppression of food consumption in ob/ob mice, similar to lean controls. The results suggest that neural satiety mechanisms triggered via serotonin

Variability and habituation of nystagmic responses to hot caloric stimulation of normal subjects. Evidence that this test may be inapplicable to monitoring drug-induced vestibular toxicity.

Experiments were performed on 25 otoneurologically "normal' subjects to evaluate the hot caloric test as a screening test for aminoglycoside vestibular toxicity. Using portable equipment under non-ideal conditions, it was found that there was a large inter-subject variability in nystagmic response and that, instead of a random test-retest variability, a systematic variation in response occurred on repeated caloric stimulation with water at 44 degrees C. A response decline (habituation) evident in both the maximum slow phase velocity and the maximum frequency occurred at second test, although t

Primary headache pathophysiology in children: the contribution of clinical neurophysiology.

Although primary headaches are very prevalent also in pediatric age, most neurophysiologic studies in these diseases concerned only the adulthood. The neurophysiologic investigation of the pathophysiological mechanisms subtending migraine and tension-type headache in children and adolescents could be particularly interesting, since during the developmental age the migrainous phenotype is scarcely influenced by many environmental factors that can typically act on adult headache patients. The neurophysiologic abnormality most frequently found in adult migraineurs, that is the reduced habituation

Orbitofrontal cortex drives predictive filtering of sensory responses.

Habituation is a crucial sensory filtering mechanism whose dysregulation can lead to a continuously intense world in disorders with hypersensitivity. Although habituation is often termed the simplest form of learning, its circuit mechanisms remain elusive. Conventional peripheral explanations fail to fully account for long-term habituation in complex sensory environments, leading to theories proposing top-down regulation. Here we evaluated two competing top-down mechanistic explanations for habituation in mice (growth in predictive filtering and waning of novelty-driven amplification) and iden

Migraine and insufficient sleep: The effect of sleep restriction on nociceptive evoked potentials in migraine.

BackgroundThe association between insufficient sleep and migraine is largely unexplained. In this blinded cross-over study we investigate whether insufficient sleep disturbs processing of nociceptive signals more in individuals with migraine compared to those without migraine.MethodsEvoked potentials to nociceptive laser stimuli and high density electrical dermal stimuli were recorded in migraine and control subjects after two nights of habitual sleep and after two nights of sleep restriction (4 h sleep/night). Researchers conducting the measurements and data processing were blinded to diagnos

The distracting impact of repeated visible and invisible onsets on focused attention.

A sudden peripheral onset is a powerful attentional attractor. However, in real life potentially distracting events do not always occur as a single event, but rather they can occur in a repetitive fashion. Hence, one of the aims of the present study was to investigate how the attentional system reacts to multiple consecutive onsets within the same trial. The results, quite surprisingly, showed that repeated peripheral onsets do not have a negative impact on visual performance, while they confirmed that a single peripheral onset captures focused attention. We hypothesize the existence of a shor

Effects of abstinence from habitual involvement in regular exercise on feeling states: an ecological momentary assessment study.

Regular exercise was experimentally reduced to determine its effects on positive feeling states. Using ecological momentary assessments, 40 participants maintained their regular exercise routine on 3 days and were deprived of their scheduled exercise on 3 other days. They recorded their feeling states, using the Exercise-Induced Feeling Inventory, four times daily as well as prior to and following exercise. Multi-level modelling analyses controlling for diurnal variations in feeling states revealed that positive feeling states were elevated on days when exercise deprivation occurred compared w

Habituation to galvanic vestibular stimulation for analysis of susceptibility to carsickness.

OBJECTIVE: In a previous study we developed a standardized procedure for a reproducible quantification of galvanic-induced body sway (GBS). In line with other reports, GBS shows short-term (fast) habituation upon stimulus repetition. The aim of this study was to evaluate whether the degree of short-term habituation to galvanic vestibular stimulation (GVS) is correlated with susceptibility to carsickness. MATERIAL AND METHODS: A total of 24 female subjects underwent computer-controlled GVS as part of a prospective experimental study conducted in a tertiary referral center. A binaural 1-cosinus

Atrial natriuretic peptide levels in children with habitual epistaxis.

OBJECTIVE: By virtue of no identifiable causes in the majority of children with habitual epistaxis, it continues to be problematic in pediatric clinical practice. The purpose of this study is to explore the possible change of atrial natriuretic peptide (ANP) levels in the children with epistaxis. METHODS: Both the plasma and nasal mucus ANP levels have been determined in 30 sick children by a sensitive radioimmunoassay (RIA) technique. RESULTS: Our results revealed that the plasma and nasal mucus ANP levels were considerably decreased in 24 children with habitual epistaxis when compared with

Stimulus control of maternal responsiveness to Norway rat (Rattus norvegicus) pup ultrasonic vocalizations.

Mother rats (Rattus norvegicus; 6 to 8 days postpartum) approach and maintain proximal orientation to a pup that is emitting ultrasonic vocalizations (USVs) far more than do virgin females (W. J. Farrell & J. R. Alberts, 2002). We used a playback regimen to examine the roles of acoustic and nonacoustic cues in regulating maternal proximal orientation toward vocalizing pups. When presented with recorded USVs, mothers of 6- to 8-day-old pups and nulliparous virgin females exhibited equivalent levels of proximal orientation toward the playback speaker. Mothers did show enhanced proximal orientati

Abnormalities of the acoustic startle reflex and reaction time in gilles de la tourette syndrome.

OBJECTIVE: To study the startle reflex and the effect of the startle reflex stimulus over reaction time (start-react effect) in Gilles de la Tourette syndrome (GTS). METHOD: Ten GTS patients and ten matched healthy volunteers underwent a simple RT paradigm (4 blocks of 50 trials). Forty acoustic startle reflex stimuli (110 dB) were randomly delivered with a 20% occurrence probability and presented unexpectedly at the same time as the imperative stimuli of the RT. Variables of interest were: amplitude, onset latency, degree of spread and rate of habituation of the startle response, and RT and

The influence of age on susceptibility to motion sickness in monkeys.

A longitudinal study on the effects of age on the susceptibility to motion sickness in the squirrel monkey was carried out over a 10-year period (1982 to 1991). The typical life span of squirrel monkeys is 15 years. Ten mature male squirrel monkeys of the Bolivian subspecies were found to be susceptible to motion sickness induced by a combination of vertical oscillation at 0.5 Hz and rotation in the horizontal plane at 25 rotations per minute (RPM) in a visually unrestricted environment. Signs of motion sickness were quantified according to a rating scale based on Graybiel's diagnostic criteri

[Habituation in the cortical division of the visceral analyzer].

Reactions of neurons without background activity ("silent" neurons) on long-lasting stimulation of mesenteric nerves were studied in cats anesthetized with chloralose 65-70 mg/kg) and paralyzed with flaxedile. It is shown that the "silent" neurons of the parietal and first projections areas are characterized by habituation during long-lasting stimulation of visceral nerves. Habituation developed faster in the parietal cortex and in the focus of maximal activity of the first somatosensory areas as compared to the second somatosensory area. The particularities of neuronal habituation in the cort

Tactile prepuff inhibition of startle in children with Tourette's syndrome: in search of an "fMRI-friendly" startle paradigm.

BACKGROUND: Functional magnetic resonance imaging (fMRI) studies in neuropsychiatric populations will be enhanced by "on-line" tasks that assess brain activation linked to neurocognitive and psychophysiological functions. In some cases, task modifications may be required for use in an fMRI environment. Prepulse inhibition (PPI) of the startle reflex is an operational measure of sensorimotor gating that is deficient in specific neuropsychiatric disorders, including schizophrenia, Huntington's disease, and Tourette's syndrome (TS). This study examined whether a modified "fMRI-friendly" PPI parad

Ethanol alters mechanosensory habituation in C. elegans by way of the BK potassium channel through a novel mechanism.

In this research, we investigated how alcohol modulates the simplest form of learning, habituation, in Caenorhabditis elegans. We used our high throughput Multi-Worm Tracker to conduct a large scale study of more than 21,000 wild-type worms to assess the effects of different concentrations of alcohol on habituation of the well-characterized tap withdrawal response. We found that the effect of alcohol on habituation of this reversal response to a repeated mechanosensory stimulus (taps) differed depending on the component of the reversal response assessed. Interestingly, when we examined habitua

Prenatal ethanol exposure impairs sensory processing and habituation to visual stimuli, effects normalized by enrichment of postnatal environmental.

BACKGROUND: Individuals with fetal alcohol spectrum disorders (FASD) often show processing deficits in all sensory modalities. Using an operant light reinforcement model, we tested whether prenatal ethanol exposure (PE) alters operant responding to elicit a contingent sensory stimulus-light onset (turning on the light) and habituation to this behavior in rats. We also explored whether postnatal environmental enrichment could ameliorate PE-induced deficits. METHODS: Pregnant Sprague Dawley rats were gavaged twice/day with 0 or 3 g/kg/treatment ethanol (15% w/v) during gestational days 8-20, mi
