Three extracts (aqueous, methanolic and dichloromethane) of Manihot esculenta (Cassava) leaf were tested in vitro on four development stages of Haemonchus contortus using egg hatch assay (EHA), larval development assay (LDA), L3 migration inhibition assay (LMI) and adult worm motility assay (AWM). Compared to the negative control, significant effects (P<0.0001) were observed for the methanolic extract of leaf against larval development (57.6% +/-7.6), with a dose dependent effect. These results suggest that Cassava possess anthelmintic activity against H. contortus. The active principles respo

The current control of parasitic nematodes in small ruminants relies on the use of chemical anthelminthics, but the development of resistance and the problem of drug residues require research into alternatives. Acetone extract and solvent-solvent fractions of Vernonia amygdalina Del. (Compositae) were evaluated in vitro for potential anti-parasitic effects against the eggs and larvae of Haemonchus contortus. Significant effects were obtained with the extract and fractions but differences were observed depending on the parasitic stage. The acetone extract and fractions inhibited egg hatching an

Specific-pathogen-free Dorset and St. Croix lambs were placed on pasture contaminated with Haemonchus contortus third stage larvae and slugs carrying third stage larvae of Protostrongylus rufescens for an entire grazing season to evaluate breed differences in acquired resistance to these nematodes. Lambs were evaluated for clinical signs, clinical pathology and histopathologic lesions associated with these infections. Both breeds acquired natural infections with H. contortus and lungworm when allowed to graze contaminated pastures for 5 months during the summer and fall in central Maryland. Do

In contrast to observations in former years treatment of ewes after parturition with 5 to 7.5 mg/kg fenbendazole was not fully effective in removing Haemonchus contortus. Comparison of the H contortus populations in the lambs of these ewes and those of untreated ewes later in the year revealed that the treatment resulted in fenbendazole resistance and also in side resistance to other benzimidazoles. Though the faecal egg counts of the treated ewes were low, the prophylactic effect of the treatment on the population build up of H contortus in the lambs was very limited.

Haemonchus contortus is a highly pathogenic gastrointestinal nematode of domestic and wild ruminants whose epidemiological success is closely linked to high reproductive capacity. For female daily egg output, however, direct observations remain limited, and published estimates vary widely, ranging from fewer than 20 to 10,000-15,000 eggs per female per day. This study assessed in utero egg counts in female H. contortus recovered from experimentally infected lambs in order to evaluate their implications for fertility relative to values commonly cited in the literature. Archived female nematodes

BACKGROUND: Antiparasitic resistance in sheep necessitates non-chemical strategies for gastrointestinal strongylosis control. We have evaluated the potential of three predatory mite species-Macrocheles robustulus, Macrocheles muscaedomesticae and Rhabdocarpais consanguineus-to reduce transmission of the nematode Haemonchus contortus by preying on infective larvae.
METHODS: Under laboratory conditions, mites were exposed to mixed prey environments containing housefly Musca domestica eggs and H. contortus third-stage (L3) larvae. Predation was first monitored over 8 h. This was followed by a mo

Small ruminants are frequently infected with gastrointestinal nematode parasites (GIN) such as the highly pathogenic Haemonchus contortus, which severely impact animal health, welfare and production performance. The increasing prevalence of clinical anthelmintic resistance poses a global threat to effective parasite control and the productivity of livestock farming. This includes resistance to eprinomectin (EPR), the only anthelmintic currently approved for use in dairy production with a zero-withdrawal period. This study aims to link EPR therapeutic failure against H. contortus in dairy sheep

Parasitic infections with gastrointestinal nematodes are a serious problem for the health and welfare of domestic animals and negatively affect the economics of animal production. Haemonchus contortus is a haematophagous nematode of small ruminants responsible for significant mortality and morbidity. In addition, the widespread resistance to synthetic anthelmintic drugs emphasizes the urgent need of alternative treatment options against haemonchosis. This work aims to investigate the anthelmintic activity of an hydroethanolic Combretum mucronatum leaf extract (CMLE) against Haemonchus contortu

The objective of this study was to investigate associations between gastrointestinal microbiota and protein metabolism of lambs infected with Haemonchus contortus. Sixteen male Ujumqin lambs (initial body weight = 32.4 ± 3.9 kg) were dewormed and randomly assigned to 2 equal groups, to be infected or not infected with Haemonchus contortus (GIN and CON, respectively). The experiment lasted 77 days. The GIN lambs had lower packed cell volume (PCV) and increased wormegg count (WEC) after 14 days. Furthermore, in infected lambs, there were decreases in apparent digestibility of dry matter (P = 0.0

The increasing prevalence of anthelmintic resistant strains of helminths, the drug residues in animal products and the high cost of conventional anthelmintics has created an interest in studying medicinal plants as an alternative source of anthelmintics. Thymus capitatus (Lamiales: Lamiaceae) is used traditionally by people as spices and reported to possess some biological effects. The objective of this study is to evaluate the anthelmintic efficacy of T. capitatus in comparison to albendazole against the gastrointestinal nematodes of sheep. To fulfil the objectives, in vitro anthelmintic acti

Ethanol extracts of 25 plant species were screened for anthelmintic effects against Haemonchus contortus. Ethanol extracts of each plant were used at various concentrations (10, 20 and 30%) to treat 10-day faecal cultures, incubated at 27°C with control cultures which were treated with ethanol for 48 h. Five plants with high efficacies (Ananas comosus, Aloe ferox, Allium sativum, Lespedeza cuneata and Warburgia salutaris) were selected from the first screening for further investigation using ethanol, dichloromethane and water extracts at four concentrations (2.5, 5, 10 and 20%). Haemonchus contortus (H. contortus), a major gastrointestinal parasitic nematode threatening ruminants worldwide, exhibits albendazole resistance that is primarily associated with single nucleotide polymorphisms (SNPs) at specific codons of the β-tubulin isotype I gene, particularly at codons 167 (F167Y), 198 (E198A), and 200 (F200Y). Among these, mutations at codons 198 and 200 are the most frequently detected in resistant isolates in China. To enable rapid and accurate resistance detection, this study aimed to develop a duplex real-time quantitative PCR (duplex qPCR) assay targeting SNPs at

A cross-sectional descriptive study was carried out on a study population of 81 goat herds to investigate the prevalence and frequency of two single nucleotide polymorphisms (SNPs; E198A and F200Y) associated with resistance to benzimidazoles (BZ) in Haemonchus contortus populations of goats in Poland. The distribution of gastrointestinal nematode (GIN) genera/species in a pooled fecal sample from each herd was determined in larval cultures and the presence of H. contortus DNA in larval cultures was confirmed using real-time PCR. SNPs were detected and their frequency was quantified using pyro

BACKGROUND: Rising drug resistance necessitates genomic selection to enhance parasite resistance in sheep. Copy number variations (CNVs) constitute structural genomic alterations that influence livestock traits by modifying gene dosage. This study aimed to identify candidate CNVs that differentiate between Haemonchus contortus-resistant and susceptible Rideau Arcott ewe lambs through comprehensive whole genome sequencing (WGS) analysis.
RESULTS: Fecal egg count (FEC) analysis of 32 inoculated animals across five time points (days 21, 28, 35, 42, and 56 post-infection) identified 16 individual

A complex interaction of gastrointestinal parasites with sheep hosts may involve bacteria communities, parasite genera, parasitic genes, and biological pathways. Haemonchus contortus presents a global challenge for ruminants, and the bacterial community can influence sheep's resistance and susceptibility to these parasites. Thus, a better understanding of this complex interaction could contribute to the development of a new approach to parasite control. This study evaluated the bacterial community of Corriedale sheep naturally infected with H. contortus based on the fecal egg counts over ten m

This study evaluated the effect of partial and total replacement of Haemonchus contortus in sheep breeds. Pregnant ewes of White Dorper (DO), Santa Inês (SI) and Texel (TX) breeds were allocated into three groups: Control (C), Partial Replacement (PR) and Total Replacement (TR). PR and TR ewes received anthelmintics (AH), were artificially infected with H. contortus-susceptible isolate and grazed on resistant-infested or worm-free paddocks, respectively. Control animals were untreated and naturally infected. 106 lambs were born and kept in the paddocks of their respective mothers. Their egg co

Haemonchus contortus poses a global challenge as a parasite affecting small ruminants, yet the problem of absence of an effective vaccine against H. contortus infection still exists. This investigation sought to appraise the immunological reaction induced by recombinant H. contortus excretory/secretory-24 (rHcES-24) in combination with complete Freund's adjuvant (CFA) and bio-polymeric nanoparticles (NPs) within a murine model. In this study, rHcES-24 was encapsulated in poly(d, l-lactide-co-glycolide) (PLGA) and chitosan (CS) NPs, administered subcutaneously to mice. Researchers analyzed the
