Research on the application of nanopore sequencing technology in investigating microbial data deviation.

This study aimed to address a microbial contamination incident in which Achromobacter xylosoxidans was detected in two batches of different sterile products at the quality control (QC) laboratory of a pharmaceutical company. Using nanopore sequencing combined with environmental source tracing, we conducted a microbial data deviation (MDD) investigation to rapidly identify the contamination source and provide a reference for future investigations. Strain isolation and biochemical identification were first performed on the two contaminated samples, both confirming the presence of A. xylosoxidans

Dermal tolerance and effect on skin hydration of a new ethanol-based hand gel.

We studied the dermal tolerance (repetitive occlusive patch test; ROPT) and the skin hydrating properties of a new ethanol-based gel [85% (w/w)], Sterillium Gel. For the ROPT, 53 participants were studied. Gel was applied to one site on the back under an occlusive patch during an induction phase (nine applications over three weeks) and two weeks later to a virgin site on the back during a challenge phase (one application). Twenty-four hours after the removal of the patches (induction phase and challenge phase), then 48 and 72 h later (challenge phase) sites were graded for skin reactions using

Handwashing: what is the best solution?

The options for keeping hands clean in hospital include soap; antimicrobial solutions; iodine and iodophours; and alcohol solutions and rubs. Following the recent directive for alcohol rubs to be placed at hospital bedsides, this paper considers the pros and cons of these and other hand-hygiene methods.

Impact of different hand-drying methods on surrounding environment: aerosolization of virus and bacteria, and transfer to surfaces.

BACKGROUND: In recent years, hand drying has been highlighted as a key step in appropriate hand hygiene, as moisture on hands can increase the transfer of micro-organisms from hands to surfaces and vice versa. AIM: To understand bacterial and viral aerosolization following hand drying, and study the transfer of micro-organisms from hands to surfaces after drying using different methods. METHODS: Groups of five volunteers had their hands pre-washed with soap, rinsed and dried, then inoculated with a concentrated mixture of Pseudomonas fluorescens and MS2 bacteriophage. Volunteers entered an e

Performance of UV disinfection and the microbial quality of greywater effluent along a reuse system for toilet flushing.

This paper examines the microbial quality of treated RBC (Rotating Biological Contactor) and MBR (Membrane Bioreactor) light greywater along a continuous pilot-scale reuse system for toilet flushing, quantifies the efficiency of UV disinfection unit, and evaluates the regrowth potential of selected microorganisms along the system. The UV disinfection unit was found to be very efficient in reducing faecal coliforms and Staphylococcus aureus. On the other hand, its efficiency of inactivation of HPC (Heterotrophic Plate Count) and Pseudomonas aeruginosa was lower. Some regrowth occurred in the re

Surgical hand rub: Influence of duration of application on the immediate and 3-hours effects of n-propanol and isopropanol.

BACKGROUND: The recommended duration for surgical hand treatment has been changed from 10 over 5 to 3 minutes and even shorter. OBJECTIVES: Our objective was to study the impact of the length of surgical hand antisepsis with n-propanol 60% (vol/vol) or isopropanol 70% (vol/vol) applied for 1, 3, or 5 minutes on the reduction of resident hand flora in the setting of the microbiologic laboratory for experimental and applied testing of disinfectants and antiseptics at the Medical University Vienna, Austria, using a Latin Square design. METHODS: Our methods were according to the Austrian Guideli

Evaluating consumer exposure to disinfecting chemicals against coronavirus disease 2019 (COVID-19) and associated health risks.

Disinfection of surfaces has been recommended as one of the most effective ways to combat the spread of novel coronavirus (SARS-CoV-2) that causes coronavirus disease 2019 (COVID-19). However, overexposure to disinfecting chemicals may lead to unintended human health risks. Here, using an indoor fate and chemical exposure model, we estimate human exposure to 22 disinfecting chemicals on the lists recommended by various governmental agencies against COVID-19, resulting from contact with disinfected surfaces and handwashing. Three near-field exposure routes, i.e., mouthing-mediated oral ingestio

Limited efficacy of alcohol-based hand gels.

Alcohol-based gels have been introduced recently in many hospitals worldwide for hand antisepsis. We investigated the antimicrobial efficacy of ten gels and four rinses according to European standards (EN 1500). No gel met the EN 1500 requirements within 30 s of application, whereas all hand rinses did. The tested hand gels should be considered a retrograde step for hand hygiene because the application time in clinical practice is often shorter than 30 s; they should not replace alcohol-based liquid hand disinfectants currently used in hospitals or be implemented as first choice agents.

The effect of sample type and location on industrial workplace sink and hand dryer microbiomes.

One major issue in tackling antimicrobial resistance (AMR) is the ability to effectively track resistance spread in environments where surveillance is limited. Such environments include those experiencing high volumes of hand washing and drying from multiple users. This study characterised the microbial populations and antimicrobial resistomes of two different sample types from a pharmaceutical industrial site as part of an AMR environmental surveillance programme. Paired samples were collected from hand dryers and adjacent sinks in distinct sampling locations: from toilets adjacent to 'wet' l

Comparison of the efficacy of two hand decontamination methods: Alcoholic septicidine and povidone-iodine solutions.

BACKGROUND AND AIM: Surgical site infections (SSIs) are a critical concern in ophthalmic surgical settings, where rigorous hand hygiene is essential to prevent postoperative complications. This study aimed to compare the efficacy of two antiseptic agents, Septicidine and Betadine, in reducing bacterial contamination on the hands of surgical staff. METHODS: Thirty ophthalmic surgeons and operating room personnel participated in the study. The participants scrubbed their hands with either Septicidine (alcohol-based antiseptic) or Betadine (iodine-based antiseptic). Bacterial cultures were obtai

Tailored positioning and number of hand rub dispensers: the fundamentals for optimized hand hygiene compliance.

BACKGROUND: Availability of alcohol-based hand rub (ABHR) dispensers at positions adapted to the work flow of healthcare workers (HCWs) is decisive in order to carry out indication-based hand rubbing. Although requirements and guidelines regarding the positioning of ABHR dispensers are in place, scientific evidence is often lacking. METHODS: In order to analyse the impact of the location and number of ABHR dispensers on hand hygiene performance, additional dispensers were systematically placed in patient rooms in a surgical 38-bed ward at Marburg University Hospital, Germany to complement the

Improving Access to WHO Formulations of Alcohol-Based Hand Rub in Healthcare Facilities: A District-Wide Approach.

Alcohol-based hand rub (ABHR) is an effective hand hygiene measure to mitigate and prevent infectious disease transmission in healthcare facilities (HCFs); however, availability and affordability in low- and middle-income countries are limited. We sought to establish centralized local production of ABHR using a district-wide approach to increase provider access at all public HCFs in Kabarole and Kasese Districts in Western Uganda. Partner organizations worked with district governments to adapt and implement the WHO protocol for local ABHR production at the district scale. These groups identifi

Dirty hands: bacteria of faecal origin on commuters' hands.

Although many studies have investigated bacteria on the hands of health-care workers and caregivers, few have looked at microbiological contamination on the hands of the general adult public. This study investigated faecal bacteria on the hands of commuters in five UK cities. Of the 404 people sampled 28% were found to have bacteria of faecal origin on their hands. A breakdown by city showed that the proportion of people with contaminated hands increased the further north the city of investigation (P<0.001), an effect which was due in large part to a significant trend in men but not in women.

The flip side of frequent sanitising and hand washing.

The stratum corneum is the outermost layer of the epidermis. It acts as an interface with the external environment and functions as a barrier that prevents microorganisms and allergens from penetrating the skin, while preventing bodily fluids, electrolytes and proteins from being lost in a process aimed at maintaining homeostasis. With the novel coronavirus disease 2019 (COVID-19) outbreak, there has been an increase in hygiene practice, particularly hand washing and the use of hand sanitisers. These practices have undoubtedly assisted a great deal in combatting the rate of transmission and co

Efficacy of chlorine-based disinfectants to control Legionella within premise plumbing systems.

Legionella is an opportunistic waterborne pathogen that causes Legionnaires' disease. It poses a significant public health risk, especially to vulnerable populations in health care facilities. It is ubiquitous in manufactured water systems and is transmitted via inhalation or aspiration of aerosols/water droplets generated from water fixtures (e.g., showers and hand basins). As such, the effective management of premise plumbing systems (building water systems) in health care facilities is essential for reducing the risk of Legionnaires' disease. Chemical disinfection is a commonly used control
