Vestibular Function and Gross Motor Development in 195 Children With Congenital Hearing Loss-Assessment of Inner Ear Malformations.

OBJECTIVES: To evaluate the relationship between vestibular function and gross motor development in children with inner ear malformations. METHODS: A total of 195 children (average age: 2.4 ± 1.6 yr) with profound hearing loss, defined as a hearing threshold greater than 80 dB, participated in this study. Evaluation of vestibular function was assessed in all children using a damped rotational chair test. Gross motor development was assessed by examining the age of acquisition of head control and independent walking. Temporal bone computed tomography scans were conducted in 190 of the 195 chil

Intrinsic network changes associated with cognitive impairment in patients with hearing loss and tinnitus: a resting-state functional magnetic resonance imaging study.

BACKGROUND: Hearing loss and tinnitus often occur concurrently and play a vital role in the development and progression of cognitive impairment (CI). However, the exact mechanism remains unclear. This study aimed to investigate the changes in intrinsic brain connectivity in patients with hearing loss and tinnitus accompanied by CI. METHODS: A total of 24 hearing loss and tinnitus patients with CI, 23 hearing loss and tinnitus patients with cognitive normality (CN), and 20 healthy controls (HCs) underwent resting-state functional magnetic resonance imaging (rs-fMRI). Resting-state networks (RS

Ossicular Bone Damage and Hearing Loss in Rheumatoid Arthritis: A Correlated Functional and High Resolution Morphometric Study in Collagen-Induced Arthritic Mice.

Globally, a body of comparative case-control studies suggests that rheumatoid arthritis (RA) patients are more prone to developing hearing loss (HL). However, experimental evidence that supports this hypothesis is still lacking because the human auditory organ is not readily accessible. The aim of this study was to determine the association between bone damage to the ossicles of the middle ear and HL, using a widely accepted murine model of collagen-induced arthritis (RA mice). Diarthrodial joints in the middle ear were examined with microcomputer tomography (microCT), and hearing function was

Vestibular function analyzed by a video head impulse test in patients with idiopathic sudden sensorineural hearing loss associated with vertigo.

BACKGROUND: The vestibular-organ site that is responsible for vertigo-associated idiopathic sudden sensorineural hearing loss (ISSNHL) has not been identified clearly. AIMS/OBJECTIVES: To analyze the function of each semicircular canal (SCC) separately in patients with vertigo-associated ISSNHL. PATIENTS/METHODS: ISSNHL patients with or without vertigo (n = 19 each) underwent a video head impulse test. Their vestibulo-ocular reflex (VOR) and catch-up saccades (CUS) were recorded. RESULTS: As for the affected side, the VOR gains of the lateral and posterior SCCs were lower in the vertigo gro

Sentinels or saboteurs, the Janus face of cochlear-resident macrophages in hearing loss: spatiotemporal distribution, pathophysiological functions, and translational potential.

BACKGROUND: Neuroinflammation is a critical mechanism underlying various forms of sensorineural hearing loss (SNHL). Contrary to the conventional view that cochlea is an immune-privileged organ shielded by the blood-labyrinth barrier, it is now evident that it hosts a diverse immune landscape dominated by cochlear-resident macrophages (CoRMs). These CoRMs share a common developmental origin with central nervous system microglia. Despite this phylogenetic homology, CoRMs have evolved distinct phenotypic and functional profiles to adapt to the unique peripheral sensory environment of cochlea. Re

Inner Hair Cell Loss Disrupts Hearing and Cochlear Function Leading to Sensory Deprivation and Enhanced Central Auditory Gain.

There are three times as many outer hair cells (OHC) as inner hair cells (IHC), yet IHC transmit virtually all acoustic information to the brain as they synapse with 90-95% of type I auditory nerve fibers. Here we review a comprehensive series of experiments aimed at determining how loss of the IHC/type I system affects hearing by selectively destroying these cells in chinchillas using the ototoxic anti-cancer agent carboplatin. Eliminating IHC/type I neurons has no effect on distortion product otoacoustic emission or the cochlear microphonic potential generated by OHC; however, it greatly red

Bioconversion of Scutellaria baicalensis extract can increase recovery of auditory function in a mouse model of noise-induced hearing loss.

In noise-induced hearing loss (NIHL), noise exposure damages cochlear sensory hair cells, which lack the capacity to regenerate. Following noise insult, intense metabolic activity occurs, resulting in a cochlear free radical imbalance. Oxidative stress and antioxidant enzyme alterations, including lipoxygenase upregulation, have been linked to chronic inflammation, which contributes to hearing impairment. We previously proposed Scutellaria baicalensis (SB) extract as an alternative therapeutic for preventing NIHL and attributed its pharmacological effects to baicalein. Although baicalein was m

Functional variants of MIF, INFG and TFNA genes are not associated with disease susceptibility or hearing loss progression in patients with Ménière's disease.

Variability in acute immune response genes could determine susceptibility or prognosis for Ménière's disease (MD). The cytokines tumor necrosis factor α (TNFα), macrophage migration inhibitory factor (MIF) and interferon γ (INFγ) are proinflammatory cytokines of the innate immune response. These cytokines mediate inflammation and have been previously associated with the inflammatory process in several autoimmune diseases. We investigated the association between functional allelic variants of MIF (rs35688089), IFNG (rs2234688) and TNFA (rs1800629) in patients with MD. In addition to testing the

Perilymphatic ATP plays a critical role in modulating cochlear function to protect from loud sound induced hearing loss.

BACKGROUND: Extracellular adenosine triphosphate (ATP) signalling via purinergic receptors plays a key role in cochlear adaptation to loud sound. Traditionally, ATP activation of purinergic receptor P2X receptors has been proposed to induce a cation shunt, reducing the endolymphatic potential and the driving force for sound transduction. However, direct evidence for this protective mechanism remains limited. Here, we provide direct experimental evidence identifying a distinct, compartment-specific ATP signalling pathway that modulates cochlear function. METHODS: In mature Dunkin-Hartley guine

Targeted genome editing restores auditory function in adult mice with progressive hearing loss caused by a human microRNA mutation.

Mutations in microRNA-96 (MIR96) cause autosomal dominant deafness-50 (DFNA50), a form of delayed-onset hearing loss. Genome editing has shown efficacy in hearing recovery through intervention in neonatal mice, yet editing in the adult inner ear is necessary for clinical applications, which has not been done. Here, we developed a genome editing therapy for the MIR96 mutation 14C>A by screening different CRISPR systems and optimizing Cas9 expression and the sgRNA scaffold for efficient and specific mutation editing. AAV delivery of the KKH variant of Staphylococcus aureus Cas9 (SaCas9-KKH) and

Evaluation of the Cognitive Function of Adults with Severe Hearing Loss Pre- and Post-Cochlear Implantation Using Verbal Fluency Testing.

Hearing loss is a major public health problem with significant evidence correlating it with cognitive performance. Verbal fluency tests are commonly used to assess lexical access. They provide a great deal of information about a subject's cognitive function. The aim of our study was to evaluate phonemic and semantic lexical access abilities in adults with bilateral severe to profound hearing loss and then to re-evaluate a cohort after cochlear implantation. 103 adult subjects underwent phonemic and semantic fluency tests during a cochlear implant candidacy evaluation. Of the total 103 subjects

Visual Impairment, Hearing Loss and Cognitive Function in an Older Population: Longitudinal Findings from the Blue Mountains Eye Study.

The presence of visual impairment (VI) and hearing loss (HL) with may be a marker for subsequent cognitive decline over time in older people. A prospective, longitudinal population-based study of the 3654 participants of the Blue Mountains Eye Study were assessed for the associations between VI and HL and a decline in mini-mental state examination (MMSE) scores over a duration of 10 years from the 5-year (baseline of this report) to the 15-year follow-up visits. MMSE was assessed at the 5-, 10- and 15-year follow-up visits. A decline ≥3 scores from 5-year to 10- or 15-year visits indicated pos

Intermittent exposure with moderate-level sound impairs central auditory function of mature animals without concomitant hearing loss.

Long-term, passive, continuous exposure of mature animals to moderate-level, band-limited sounds can profoundly decrease neural activity in primary auditory cortex (AI) to sounds in the exposure frequency range, and increase activity to sounds outside the exposure range. The resulting reorganization of the AI tonotopic map resembles that following a restricted lesion of the cochlear epithelium. Here we show qualitatively similar effects of passive exposure when it is limited to 12 h/day, simulating a noisy-work/quiet-living environment, albeit at substantially lower intensity levels (68 dB SPL

Functional characterization of the first missense variant in CEP78, a founder allele associated with cone-rod dystrophy, hearing loss, and reduced male fertility.

Inactivating variants in the centrosomal CEP78 gene have been found in cone-rod dystrophy with hearing loss (CRDHL), a particular phenotype distinct from Usher syndrome. Here, we identified and functionally characterized the first CEP78 missense variant c.449T>C, p.(Leu150Ser) in three CRDHL families. The variant was found in a biallelic state in two Belgian families and in a compound heterozygous state-in trans with c.1462-1G>T-in a third German family. Haplotype reconstruction showed a founder effect. Homology modeling revealed a detrimental effect of p.(Leu150Ser) on protein stability, whic

Assessment of age-related hearing loss and related everyday functional and psychosocial consequences in older adults: A systematic review of self-report instruments.

To detect age-related hearing loss (presbycusis), beyond objective audiometric measurements, self-report instruments are commonly employed. However, clear evidence regarding the methodological and measurement quality of self-report instruments for assessing hearing loss in older adults is lacking. This systematic review, for the first time, aims to identify and evaluate self-report instruments for assessing age-related hearing loss and related functional and psychological consequences in everyday life among older adults. Following PRISMA and COSMIN guidelines, we preregistered in PROSPERO (CRD

Targeted Next Generation Sequencing Revealed a Novel Homozygous Loss-of-Function Mutation in ILDR1 Gene Causes Autosomal Recessive Nonsyndromic Sensorineural Hearing Loss in a Chinese Family.

Hereditary hearing impairment is one of the major and common birth defects in Chinese population. Non-syndromic sensorineural hearing loss (NSHL) is the most common types of hereditary hearing impairment. Genotypically and phenotypically NSHL is extremely heterogenous and follow either autosomal dominant or autosomal recessive or X-linked mode of inheritance. Presently, 127 genes have been identified to be associated with both syndromic and (NSHL). Here, we studied a Chinese family with moderate and profound hearing impairment. The proband is a 30-year old Chinese man. The proband was born wit
