Interferon signaling is required for early neutrophil recruitment after zebrafish heart injury.

Heart injury triggers a robust cellular response in zebrafish, characterized by neovascularization, immune cell activation, and the infiltration of proliferative cardiomyocytes that collectively lead to scarless regeneration. Upon injury, damage-associated molecular patterns are released by dying cells and injured extracellular matrix. These molecules bind to pattern recognition receptors on various cell types, promoting inflammation and immune cell recruitment by upregulating chemokines and pro-inflammatory cytokines. We previously identified the activation of injury-induced interferon signal

Acute kidney injury after heart transplantation: A systematic review and meta-analysis.

BACKGROUND: Acute kidney injury (AKI) after heart transplantation (HT) remains incompletely understood. This systematic review and meta-analysis aimed to estimate the pooled incidence of AKI and AKI requiring renal replacement therapy (RRT) after HT, assess associated mortality risks and identify characteristics associated with AKI requiring RRT. METHODS: A systematic literature search identified original studies reporting the incidence of AKI after HT using standardized definitions or reporting AKI requiring RRT. Meta-analyses were conducted using random-effects models. RESULTS: Forty-eight

Examining APOE ε4 and Longitudinal Vascular Brain Injury: The Strong Heart Study.

BACKGROUND AND OBJECTIVE: American Indians have a high population risk for cerebrovascular disease, vascular brain injury (VBI), and dementia. The apolipoprotein (APOE) ε4 allele is a strong risk factor for Alzheimer's disease but is also associated with white matter hyperintensities (WMH) and VBI. However, these association have not been thoroughly examined in the American Indian population. Our objective was to determine whether APOE ε4 carriers exhibited greater longitudinal changes in WMH or if the previously reported null effect from cross-sectional findings extended over time in American

The Feasibility of Ambulatory Heart Rate Variability Monitoring in Non-Suicidal Self-Injury.

The polyvagal theory proposes that the autonomic nervous system influences affective systems and top-down emotional regulation. Vagal tone, as indexed by heart rate variability (HRV), is a measure of emotion regulation capacity. It is possible that non-suicidal self-injury (NSSI) occurs at times of low vagal tone and that NSSI may increase it. Little is known about the feasibility of collecting ambulatory HRV data in the context of NSSI. This prospective observational study examined the feasibility of ambulatory HRV monitoring during NSSI. Ten participants wore a chest-based heart rate monitor

Formate Reduces Ischemic Injury in Female Hearts Lacking Alcohol Dehydrogenase 5.

Ischemic heart disease is a primary cause of death for men and women in the United States. Recent epidemiologic findings, however, suggest that premenopausal women have inherent protection from many cardiovascular pathologies compared to age-matched men, which is lost with menopause. We and others have documented similar protective signaling in animal models, with females exhibiting protection from ischemic injury that is lost with ovariectomy (OVX). Furthermore, in recent studies, we demonstrated that the loss of alcohol dehydrogenase 5 (ADH5) blocked sex-specific cardioprotection in females,

Diagnosis and Management of Inadvertent Iliac Vein Injury During Structural Heart Interventions.

Femoral venous access is increasingly used by structural interventional cardiologists in procedures such as patent foramen ovale, atrial septal defect, and left atrial appendage closure, as well as tricuspid and mitral interventions. Containing less muscular and elastic fibers than arteries, veins are fragile structures that are more easily pierced than arteries. In this case series, we present 2 cases of iliac vein perforation during structural cardiology interventions. Even mild resistance on advancement of a large sheath should prompt suspicion of venous perforation, in which case control a

Sonlicromanol Mitigates Sepsis-induced Heart Injury via Mitochondrial and Pyroptosis Modulation.

Sepsis-induced myocardial dysfunction (SIMD) is a severe consequence of systemic infection, primarily driven by mitochondrial dysfunction, inflammation, and pyroptosis. Sonlicromanol, a mitochondrial redox-modulating therapeutic agent, has shown promise in preserving mitochondrial function, but its role in sepsis-induced cardiac injury remains unclear. This study evaluates the protective effects of sonlicromanol in a rat model of sepsis-induced cardiac dysfunction, with a focus on mitochondrial dynamics, mitophagy, and inflammasome-pyroptosis pathways. Male Sprague-Dawley rats were subjected t

Myocardial lipin1 protects the heart against ischemic injury by preserving lipid homeostasis.

Impaired cardiac lipid metabolism has been reported to cause heart failure. Lipin1, a multifunctional protein, is a phosphatidate phosphatase that generates diacylglycerol from phosphatidic acid and a transcriptional cofactor that regulates lipid metabolism-related gene expression. Here, we investigated the roles of lipin1 in cardiac remodeling after myocardial infarction (MI). The expression levels of lipin1 significantly decreased in cardiomyocytes of the human failing heart and murine ischemic myocardium. Cardiomyocyte-specific Lpin1 knockout (cKO) mice showed left ventricle enlargement and

Sophocarpine alleviates doxorubicin-induced heart injury by suppressing oxidative stress and apoptosis.

Doxorubicin (DOX) is an effective anti-tumor drug accompanied with many side effects, especially heart injury. To explore what effects of sophocarpine (SOP) on DOX-induced heart injury, this study conducted in vivo experiment and in vitro experiment, and the C57BL/6J mice and the H9C2 cells were used. The experimental methods used included echocardiography, enzyme-linked immunosorbent assay (ELISA), dihydroethidium (DHE) staining, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining, western blotting and so on. Echocardiography showed that SOP alleviated DOX-induced ca

Irreversible myocardial injury attenuates the benefits of sacubitril/valsartan in heart failure patients.

BACKGROUND: Despite the established benefits of angiotensin receptor-neprilysin inhibitor (ARNI) in heart failure with reduced ejection fraction (HFrEF) across various etiologies, there are controversies regarding the effects of ARNI in patients with irreversible myocardial injury. The aim of this study is to investigate the impact of irreversible myocardial injury on the benefits of ARNI treatment in patients with HFrEF, consisted of both ischemic and non-ischemic etiologies. METHODS AND RESULTS: We conducted a retrospective single-center study including 409 consecutive patients with HFrEF t

Metabolomics analysis in rat hearts with ischemia/reperfusion injury after diazoxide postconditioning.

Background: Diazoxide is a selective mitochondrial-sensitive potassium channel opening agent that has a definite effect on reducing myocardial ischemia/reperfusion injury (MIRI). However, the exact effects of diazoxide postconditioning on the myocardial metabolome remain unclear, which might contribute to the cardioprotective effects of diazoxide postconditioning. Methods: Rat hearts subjected to Langendorff perfusion were randomly assigned to the normal (Nor) group, ischemia/reperfusion (I/R) group, diazoxide (DZ) group and 5-hydroxydecanoic acid + diazoxide (5-HD + DZ) group. The heart rate

Brain injury and neurodevelopmental outcomes in children undergoing surgery for congenital heart disease.

OBJECTIVES: Brain injury is commonly seen on magnetic resonance imaging in infants with complex congenital heart disease. The impact of perioperative brain injury on neurodevelopmental outcomes is not well understood. We evaluate the association of brain injury and other markers on neurodevelopmental outcomes in patients undergoing surgery for congenital heart surgery during infancy. METHODS: Term newborns with infant cardiac surgery performed between 2008 and 2019 at a single tertiary center, and both preoperative and postoperative brain magnetic resonance imaging were included. High-output heart failure following gunshot injury and traumatic arteriovenous fistula.

BACKGROUND: Heart failure due to high cardiac output rarely occurs. Few cases of post-traumatic arteriovenous fistula (AVF) as a cause of high-output failure were reported in the literature. METHODS: Herein, we report a case of 33-year-old male who was admitted to our institution due to symptoms of heart failure. He reported a gunshot injury of the left thigh 4 months earlier, when he had been shortly hospitalized and discharged 4 days later. Since the gunshot injury he had exertional dyspnea and left leg edema, so the diagnostic procedures were performed. RESULTS: Clinical examination revea

Assessing the Relationship Between Molecular Rejection and Parenchymal Injury in Heart Transplant Biopsies.

BACKGROUND: The INTERHEART study (ClinicalTrials.gov #NCT02670408) used genome-wide microarrays to detect rejection in endomyocardial biopsies; however, many heart transplants with no rejection have late dysfunction and impaired survival. We used the microarray measurements to develop a molecular classification of parenchymal injury. METHODS: In 1320 endomyocardial biopsies from 645 patients previously studied for rejection-associated transcripts, we measured the expression of 10 injury-induced transcript sets: 5 induced by recent injury; 2 reflecting macrophage infiltration; 2 normal heart t

The Molecular Microscope Diagnostic System: Assessment of Rejection and Injury in Heart Transplant Biopsies.

This review describes the development of the Molecular Microscope Diagnostic System (MMDx) for heart transplant endomyocardial biopsies (EMBs). MMDx-Heart uses microarrays to measure biopsy-based gene expression and ensembles of machine learning algorithms to interpret the results and compare each new biopsy to a large reference set of earlier biopsies. MMDx assesses T cell-mediated rejection (TCMR), antibody-mediated rejection (AMR), recent parenchymal injury, and atrophy-fibrosis, continually "learning" from new biopsies. Rejection-associated transcripts mapped in kidney transplants and expe

Myocardium-Targeted Micelle Nanomedicine That Salvages the Heart from Ischemia/Reperfusion Injury.

Cardioprotective medication is the common treatment to relieve myocardial ischemia/reperfusion (I/R) injury. However, limited by the low bioavailability of therapeutic drugs, the therapeutic outcome is barely satisfactory. Because the I/R injury can enhance the permeability of the vasculature and allow the extravasation of nanoparticles into the surrounding tissue, herein we formulate the cardiotonic drug olprinone (Olp) in cross-linked micelles as the nanomedicine to achieve myocardium-targeted delivery after systematic administration. As a result, the local concentration of Olp in the injure
