Helminth infections of urban hibernating Nyctalus noctula groups (Chiroptera, Vespertilionidae) in Belarus.

The city of Brest is one of the places in southwestern Belarus where common noctules Nyctalus noctula (Schreber, 1774) spend the winter. The aim of the study is to investigate the helminth infection of urban hibernating groups of this species of bats. The bat carcasses were subjected to a post-mortem examination. Organs and tissues compression was used to examine 26 of common noctules three groups that died in Brest city during the hibernation period from causes beyond our control. All of the examined common noctules were infected with helminths. Eight species of helminths were found: 4 specie

Suppression of mitochondrial respiration by hydrogen sulfide in hibernating 13-lined ground squirrels.

Hibernating mammals may suppress their basal metabolic rate during torpor by up to 95% to reduce energy expenditure during winter, but the underlying mechanisms remain poorly understood. Here we show that hydrogen sulfide (H2S), a ubiquitous signaling molecule, is a powerful inhibitor of respiration of liver mitochondria isolated from torpid 13-lined ground squirrels, but has a weak effect on mitochondria isolated during summer and hibernation arousals, where metabolic rate is normal. Consistent with these in vitro effects, we find strong seasonal variations of in vivo levels of H2S in plasma

Successful induction of deep hypothermia by isoflurane anesthesia and cooling in a non-hibernator, the rat.

The aim of the present study was to establish a novel method for inducing deep hypothermia in rats. Cooling rats anesthetized with isoflurane caused a time-dependent decrease in rectal temperature, but cardiac arrest occurred before their body temperature reached 20 °C when isoflurane inhalation was continued during the cooling process. Stopping inhalation of isoflurane when the rectal temperature reached 22.5 °C successfully induced deep hypothermia, although stopping the inhalation at 27.5 °C resulted in spontaneous recovery of rectal temperature. The hypothermic condition was able to be mai

Life history written in blood: erythrocyte parameters in a small hibernator, the edible dormouse.

The oxygen delivery system is one major determinant of the performance of vertebrates and responds sensitively to a variety of internal and environmental factors. To understand physiological mechanisms underlying variations of fitness, we investigated effects of demanding conditions associated with certain life-history events, food availability, and population density on the oxygen delivery system in free-ranging edible dormice (Glis glis). We sampled blood (n = 248) and urine (n = 319), performed an erythrocyte haemogram and visually determined the presence of haemoglobinuria. Reproduction le

Homocysteine homeostasis and betaine-homocysteine S-methyltransferase expression in the brain of hibernating bats.

Elevated homocysteine is an important risk factor that increases cerebrovascular and neurodegenerative disease morbidity. In mammals, B vitamin supplementation can reduce homocysteine levels. Whether, and how, hibernating mammals, that essentially stop ingesting B vitamins, maintain homocysteine metabolism and avoid cerebrovascular impacts and neurodegeneration remain unclear. Here, we compare homocysteine levels in the brains of torpid bats, active bats and rats to identify the molecules involved in homocysteine homeostasis. We found that homocysteine does not elevate in torpid brains, despit

Modulation of gene expression in heart and liver of hibernating black bears (Ursus americanus).

BACKGROUND: Hibernation is an adaptive strategy to survive in highly seasonal or unpredictable environments. The molecular and genetic basis of hibernation physiology in mammals has only recently been studied using large scale genomic approaches. We analyzed gene expression in the American black bear, Ursus americanus, using a custom 12,800 cDNA probe microarray to detect differences in expression that occur in heart and liver during winter hibernation in comparison to summer active animals. RESULTS: We identified 245 genes in heart and 319 genes in liver that were differentially expressed be

Identification and revascularization of hibernating myocardium in angina-free patients with left ventricular dysfunction.

We examined 17 angina-free patients with left ventricular dysfunction, referred for surgical decision-making, who presented with no or few signs and symptoms of myocardial ischemia according to treadmill stress test. On cardiac catheterization they were affected by severe multi-vessel coronary artery disease; the mean left end-diastolic pressure of this population was 26.3 +/- 5.5 mm Hg (mean +/- SD) and their mean ejection fraction was 27.6 +/- 4.9% (mean +/- SD). They all were investigated for the presence of viable myocardium by the combined assessment of cardiac perfusion and metabolism us

Structure of the hibernating Francisella tularensis ribosome and mechanistic insights into its inhibition by antibiotics.

Francisella tularensis is the causative agent of tularemia, a zoonotic disease named after the Tulare County, California. Symptoms include sudden fever, chills, fatigue, and swollen lymph nodes, among others, and without treatment it is very serious or even fatal. In addition, F. tularensis is considered a potential bioterrorism threat due to its high infectivity and lethality. Ribosomes are key targets for many classes of antibiotics. In this study, we examined the F. tularensis ribosome and determined its structure at 2.5Å resolution using cryo-electron microscopy. Notably, we observed the s

Comparative Pathophysiology of Humans and Hibernating Bears: From Metabolic Failure to Adaptive Resilience.

Modern medicine has struggled to cope with the pathologies induced by sedentary lifestyles and excess calories. As hibernating bears are remarkably resilient to such complications, this review compares the pathophysiology of the two across three critical domains. While humans experience inflammatory adipose hypertrophy, bears control obesity by utilizing immunomodulatory lipids and the hyperplastic expansion of adipose tissue. Unlike catabolism and nitrogenous waste accumulation, which are typical of human muscles, bears employ a gut microbiota-based urea recycling system to preserve proteosta

Genome-Wide Association Reveals Signalling-Linked Infection Tolerance in Hibernating Bats.

Hibernation profoundly alters host-pathogen dynamics by suppressing metabolism and immune function, posing unique challenges for infection control. In this study, we examined how genomic variation modulates infection and physiological traits in temperate bats during hibernation. We combined infection screening, haematology, blood biochemistry, and whole-genome sequencing across five vespertilionid species, identifying over 170,000 single nucleotide variants (SNVs) and assessing their associations with 23 health-related variables. Using the phylogenetically informed treeWAS framework, we detect

Seasonal Changes and Phenotypic Flexibility in the Digestive System of the Hibernating Marsupial Dromiciops gliroides.

During hibernation, reduced metabolic activity enables to withstand periods of limited resources and harsh weather. Therefore, animals exhibit reduced activity and decreased feeding, which involves energy savings. Thus, a reduction in both physiological and morphological digestive capacities is expected due to the prolonged metabolic depression of hibernation, which involves phenotypic flexibility of the digestive system at several levels. We studied Dromiciops gliroides a heterothermic marsupial endemic of the temperate rainforests of southern South America, by comparing the morphology and ph

Transcriptome remodeling and adaptive preservation of muscle protein content in hibernating black bears.

Hibernation is an energy-saving adaptation associated with physical inactivity. In contrast to most mammals, hibernating bears demonstrate limited loss of muscle mass and protein content over the prolonged periods of immobility and fasting during winter. This suggests that bears have natural adaptive mechanisms preserving muscle mass and functionality. To identify transcriptional changes that underlie molecular mechanisms attenuating muscle loss, we conducted a large-scale gene expression profiling (14,199 genes) by transcriptome sequencing in quadriceps of adult black bears, comparing hiberna

The paddy frog genome provides insight into the molecular adaptations and regulation of hibernation in ectotherms.

Amphibians, like the paddy frog (Fejervarya multistriata), have played a critical role in the transition from water to land. Hibernation is a vital survival adaptation in cold environments with limited food resources. We decoded the paddy frog genome to reveal the molecular adaptations linked to hibernation in ectotherms. The genome contained 13 chromosomes, with a significant proportion of repetitive sequences. We identified the key genes encoding the proteins of AANAT, TRPM8, EGLN1, and VEGFA essential for circadian rhythms, thermosensation, and hypoxia during hibernation by comparing the hi

Identification of genes supporting cold resistance of mammalian cells: lessons from a hibernator.

Susceptibility of human cells to cold stress restricts the use of therapeutic hypothermia and long-term preservation of organs at low temperatures. In contrast, cells of mammalian hibernators possess remarkable cold resistance, but little is known about the molecular mechanisms underlying this phenomenon. In this study, we conducted a gain-of-function screening of genes that confer cold resistance to cold-vulnerable human cells using a cDNA library constructed from the Syrian hamster, a mammalian hibernator, and identified Gpx4 as a potent suppressor of cold-induced cell death. Additionally, g

Translational large animal model of hibernating myocardium: characterization by serial multimodal imaging.

Nonrevascularizable coronary artery disease is a frequent cause of hibernating myocardium leading to heart failure (HF). Currently, there is a paucity of therapeutic options for patients with this condition. There is a lack of animal models resembling clinical features of hibernating myocardium. Here we present a large animal model of hibernating myocardium characterized by serial multimodality imaging. Yucatan minipigs underwent a surgical casein ameroid implant around the proximal left anterior descending coronary artery (LAD), resulting in a progressive obstruction of the vessel. Pigs under

The effects of neurectomy and hibernation on bone properties and the endocannabinoid system in marmots (Marmota flaviventris).

Hibernators have adapted a physiological mechanism allowing them to undergo long periods of inactivity without experiencing bone loss. However, the biological mechanisms that prevent bone loss are unknown. Previous studies found meaningful changes, between active and hibernating marmots, in the endocannabinoid system of many tissues, including bone. Cannabinoid receptors (CB1 and CB2) have divergent localization in bone. CB1 is predominately found on sympathetic nerve terminals, while CB2 is more abundant on bone cells and their progenitors. This study aimed to determine the contribution of in

Hibernating Little Pocket Mice Show Few Seasonal Changes in Bone Properties.

Periods of disuse or physical inactivity increases bone porosity and decreases bone mineral density, resulting in a loss of bone mechanical competence in many animals. Although large hibernators like bears and marmots prevent bone loss during hibernation, despite long periods of physical inactivity, some small hibernators do lose bone during hibernation. Little pocket mice (Perognathus longimembris) remain underground during winter hibernation and undergo bouts of torpor and interbout arousals, but the torpor bout duration is shorter than other rodent hibernators. Additionally, little pocket m

The effects of hibernation and forced disuse (neurectomy) on bone properties in arctic ground squirrels.

Bone loss is a well-known medical consequence of disuse such as in long-term space flight. Immobilization in many animals mimics the effects of space flight on bone mineral density. Decreases in metabolism are also thought to contribute to a loss of skeletal mass. Hibernating mammals provide a natural model of disuse and metabolic suppression. Hibernating ground squirrels have been shown to maintain bone strength despite long periods of disuse and decreased metabolism during torpor. This study examined if the lack of bone loss during torpor was a result of the decrease in metabolic rate during

Metabolic depression in hibernation and major depression: an explanatory theory and an animal model of depression.

Metabolic depression, an adaptive biological process for energy preservation, is responsible for torpor, hibernation and estivation. We propose that a form of metabolic depression, and not mitochondrial dysfunction, is the process underlying the observed hypometabolism, state-dependent neurobiological changes and vegetative symptoms of major depression in humans. The process of metabolic depression is reactivated via differential gene expression in response to perceived adverse stimuli in predisposed persons. Behavior inhibition by temperament, anxiety disorders, genetic vulnerabilities, and e

Brain gangliosides in hibernating dormice (Glis glis) and cold-exposed laboratory mice.

The concentration of proteins, sialo-glycoproteins and gangliosides and the ganglioside composition of 8 brain regions from normothermic and hibernating fat dormice (Glis glis) and from laboratory mice being acclimated to 6, 22 and 28 degrees C were investigated. During hibernation the concentration of sialo-glycoproteins and gangliosides decreased significantly in brain of dormice; the protein content remained uninfluenced.
