Multi-omics profiling reveals homologous recombination repair hyperactivation as a therapeutic vulnerability in neuroendocrine cervical carcinoma.

Neuroendocrine cervical carcinoma (NECC) is an aggressive and rare malignancy comprising 0.9-1.5% of cervical cancers. Its molecular basis remains poorly understood, limiting treatment options. A national multicenter cohort survival analysis showed that NECC patients had a 5.17-fold higher death risk than non-NECC patients and demonstrated broad chemotherapy resistance, which was further validated in both NECC cell lines and patient-derived organoids. To identify therapeutic targets, multi‑omics profiling of NECC tumors and matched normal adjacent tissues revealed pronounced DNA replication st

Interaction between ENPP1 and homologous recombination deficiency defines distinct pan-cancer signatures: A retrospective observational study.

Ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1), 1st identified in breast cancer and subsequently in multiple other cancer types, is an innate immune checkpoint regulator that recently emerged as a promising biomarker and therapeutic target. Homologous recombination deficiency (HRD) has gained clinical relevance with therapeutic vulnerability, particularly in breast and ovarian cancers. Despite the increasing significance of ENPP1 and HRD in cancer biology and treatment, their potential relationships have not yet been comprehensively investigated. We analyzed the relationship betwee

Germline homologous recombination deficiency influences TP53-mutant clonal hematopoiesis fitness during platinum and PARP inhibitor treatment.

Poly(ADP-ribose) polymerase inhibitors (PARPi) are commonly used in tumors with homologous recombination deficiency (HRD) but are associated with an increased risk of therapy-related myeloid neoplasms (tMN). Clonal hematopoiesis (CH) driven by DNA damage response (DDR) mutations is the origin of most tMN. Here, to better understand the causes of tMN following PARPi therapy, we studied the relationship between PARPi use and CH. We observed a high frequency of DDR CH following PARPi therapy, largely explained by prior carboplatin exposure. Among patients with serial blood sampling, DDR CH expand

Implications of Homologous Recombination Deficiency for Neoadjuvant Platinum-Based Chemotherapy in Pancreatic Cancer: A Narrative Review.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) remains a highly lethal malignancy, with a 5 year survival rate of approximately 13%. Survival is extended for the few patients who undergo surgical resection with curative intent, whereas most patients succumb to distant disease recurrence. Neoadjuvant platinum-based chemotherapy has emerged as a promising strategy to improve resectability rates and survival outcomes for PDAC patients. However, treatment-related toxicities, unpredictable clinical responses, and associated risk of tumor progression during neoadjuvant therapy may delay or prec

Genomic landscape and homologous recombination deficiency in malignant germ cell tumors reveals sex-specific therapeutic opportunities.

Malignant germ cell tumors (GCTs) are relatively rare tumors with limited therapeutic options. This study examined the homologous recombination deficiency (HRD) status and genomic characteristics in 14 patients with GCT who underwent next-generation sequencing. HRD was evaluated using the genomic instability score (GIS), which incorporates three components: loss of heterozygosity (LOH), telomeric allelic imbalance (TAI), and large-scale transition (LST). Our findings showed that 57.1% (8/14) of participants were HRD positive. Female patients presented a significantly higher prevalence of HRD p

Prevalence and spectrum of homologous recombination repair mutations in patients with metastatic prostate cancer from India.

BACKGROUND: Alterations in genes involved in homologous recombination repair (HRR) occur in approximately 20%-25% of patients with metastatic prostate cancer and are associated with aggressive biology, poor outcomes, and potential sensitivity to poly (ADP-ribose) polymerase inhibitors (PARPi). However, frequency and variations in somatic HRR mutations in the Indian population are unknown. METHODS: We analyzed somatic HRR alterations in patients at the All India Institute of Medical Sciences, New Delhi, between 2022 and November 2024. Targeted next-generation sequencing of 15 HRR genes was per

Mobilization of the AbGRI4 resistance island in Acinetobacter baumannii: IS26 action and homologous recombination both contribute.

UNLABELLED: Carbapenem-resistant Acinetobacter baumannii isolate HUMC1 (USA, 2009), which is hypervirulent and extensively antibiotic resistant, has been used in many experimental studies. The complete sequence of the chromosome and two plasmids was determined, and the causes of the resistance phenotype were investigated. HUMC1 (ST2:ST1839:KL22:OCL3) is an unusual global clone 2 (GC2) isolate that lacks an AbGRI1 type resistance island interrupting the comM gene. However, a version of AbGRI2, which is present together with AbGRI1 in the oldest known GC2 isolate, and versions of AbGRI3 and the

Homologous recombination deficiency testing implementation in the routine clinical setting: real-life experience and lessons learned.

BACKGROUND: Homologous recombination deficiency (HRD) is a critical biomarker for predicting response to poly (ADP-ribose) polymerase (PARP) inhibitors in high-grade serous ovarian cancer (HGSOC). Hence, genomic scar-based HRD testing must be implemented in routine clinical labs. This study aims to obtain a head-to-head comparison of some of the applications available for HRD status determination in a real-world clinical cohort to guide HRD testing standardization. MATERIALS AND METHODS: HRD scores were obtained from HGSOC formalin-fixed, paraffin-embedded samples from the Molecular Prescreen

Erbin destabilization by O-GlcNAcylation promotes 5-FU resistance via homologous recombination activation in colorectal cancer.

The emergence of 5-fluorouracil (5FU) resistance critically compromises chemotherapy efficacy in colorectal cancer (CRC). While O-GlcNAcylation-a dynamic post-translational modification linked to tumor progression-has been widely investigated, its functional role in 5FU chemoresistance remains poorly defined. Through mass spectrometry and tissue microarray analysis, we identified aberrantly elevated levels of O-GlcNAcylation and its catalytic enzyme O-GlcNAc transferase (OGT) in CRC tissues. Genetic silencing of OGT significantly reduced global O-GlcNAcylation, suppressing CRC cell proliferati

Decrotonylation of cGAS K254 prompts homologous recombination repair by blocking its DNA binding and releasing PARP1.

Cyclic GMP-AMP synthase (cGAS), a cytosolic DNA sensor, also exhibits nuclear genomic localization and is involved in DNA damage signaling. In this study, we investigated the impact of cGAS crotonylation on the regulation of the DNA damage response, particularly homologous recombination repair, following exposure to ionizing radiation (IR). Lysine 254 of cGAS is constitutively crotonylated by the CREB-binding protein; however, IR-induced DNA damage triggers sirtuin 3 (SIRT3)-mediated decrotonylation. Lysine 254 decrotonylation decreased the DNA-binding affinity of cGAS and inhibited its intera

Relationship between homologous recombination deficiency and clinical features of breast cancer based on genomic scar score.

BACKGROUND: Homologous recombination deficiency (HRD) phenotype will sensitize tumors to poly (ADP-ribose) polymerases inhibitors and platinum. However, previous studies did not focus on the prevalence of HRD among Chinese breast cancer (BC) patients. METHODS: One hundred and forty-seven BC patients were included in this study. Their HRD status was assessed by Genomic Scar Score (GSS), which was determined according to the length, site, and type of copy number. HRD was defined as positive when a harmful BRCA1/2 mutation was detected or GSS ≥50. RESULTS: Our data revealed that 9.5% of the 147

Limiting homologous recombination at stalled replication forks is essential for cell viability: DNA2 to the rescue.

The disease-associated nuclease-helicase DNA2 has been implicated in DNA end-resection during DNA double-strand break repair, Okazaki fragment processing, and the recovery of stalled DNA replication forks (RFs). Its role in Okazaki fragment processing has been proposed to explain why DNA2 is indispensable for cell survival across organisms. Unexpectedly, we found that DNA2 has an essential role in suppressing homologous recombination (HR)-dependent replication restart at stalled RFs. In the absence of DNA2-mediated RF recovery, excessive HR-restart of stalled RFs results in toxic levels of abo

Tumor infiltrating lymphocytes and homologous recombination deficiency are independently associated with improved survival in ovarian carcinoma.

OBJECTIVE: The presence of tumor infiltrating lymphocytes (TIL) and defects in homologous recombination (HR) are each important prognostic factors in ovarian carcinoma (OC). We characterized the association between HR deficiency (HRD) and the presence of TILs in a cohort of OC patients and the relative contribution to overall survival. METHODS: Patients with carcinoma of the ovary, fallopian tube, or peritoneum were prospectively enrolled. Malignant neoplasm and serum samples were collected. Immunohistochemistry for CD3+ T cells and CD68+ tumor associated macrophages (TAMs) was performed on s

Isoorientin triggers apoptosis of hepatoblastoma by inducing DNA double-strand breaks and suppressing homologous recombination repair.

Hepatoblastoma (HB) is the most common malignant liver tumor in children. DNA and DNA-associated processes are one of the most important targets of chemotherapeutic agents. Isoorientin (Iso), a natural flavonoid compound, can be extracted from several plant species. The effects of Iso and its molecular mechanisms on hepatic malignancies remain unclear. Herein, the anti-tumor effects of Iso in HB and its underlying mechanisms were explored. We found that Iso significantly inhibited the proliferation of HB cells both in vitro and in vivo. Mechanistic studies showed that Iso triggered cell apopto

MLL self fusion mediated by Alu repeat homologous recombination and prognosis of AML-M4/M5 subtypes.

Fifty-six patients with de novo acute myeloid leukemia M4/M5 subtypes were studied for rearrangements of the mixed lineage leukemia gene, MLL (also called HRX, Htrx-1, or ALL-1). Ten patients (18%) showed rearrangements of the MLL gene, 9 in a major breakpoint cluster region within a centromeric 8.3-kb BamHI fragment, whereas rearrangement in one patient was the result of a direct tandem duplication of exons 2-6 of MLL. Analysis of sequences at the duplication junction revealed that the points of MLL fusion within introns 6 and 1 both lie within Alu elements. This suggests the involvement of A

Role of homologous recombination/recombineering on human adenovirus genome engineering: Not the only but the most competent solution.

Adenoviruses typically cause mild illnesses, but severe diseases may occur primarily in immunodeficient individuals, particularly children. Recently, adenoviruses have garnered significant interest as a versatile tool in gene therapy, tumor treatment, and vaccine vector development. Over the past two decades, the advent of recombineering, a method based on homologous recombination, has notably enhanced the utility of adenoviral vectors in therapeutic applications. This review summarizes recent advancements in the use of human adenoviral vectors in medicine and discusses the pivotal role of rec
