Bioinspired proton relay on amino-functionalized PdCe metallene accelerates hydrogen oxidation kinetics and CO exclusion.

Inspired by [FeFe]-hydrogenases, a polyaniline-functionalized PdCe metallene mimics the enzyme's secondary coordination sphere, achieving triple synergy that weakens H* binding, relays protons via -NH- groups, and shields against CO. The catalyst surpasses Pt/C in HOR kinetics with only 1.2% activity loss under 100 ppm CO, establishing secondary-sphere engineering as a paradigm for CO-immune Pt-free HOR catalysts.

A De Novo Auto-Activated Solar-Driven Biohybrid System for Hydrogen Production in Methanotrophic Cells.

Climate change driving by greenhouse gas emissions from petroleum-based energy has garnered significant attention. Renewable energy production via a sustainable system that integrates the cell factory and visible-light-driven photocatalysts offers a novel approach for upcycling methane and addressing global energy challenges. Here, an auto-activated biohybrid system driven by solar energy is developed for converting methane into hydrogen fuel, which incorporated thienoviologen (S-MV2+) and genetically engineered methanotrophic bacteria. In this system, S-MV2+ functioned as photosensitizer and

Metabolic Regulation of Mesophilic Methanosarcina barkeri to Ammonium Inhibition.

Undesirable ammonium concentrations can lead to unstable anaerobic digestion processes, and Methanosarcina spp. are the representative methanogens under inhibition. However, no known work seems to exist for directly exploring the detailed metabolic regulation of pure cultured representative Methanosarcina spp. to ammonium inhibition. We used transcriptomics and proteomics to profile the metabolic regulation of Methanosarcina barkeri to 1, 4, and 7 g N/L of total ammoniacal nitrogen (TAN), where free ammonia concentrations were between 1.5 and 36.1 mg N/L. At the initial stages of ammonium inhi

Influence of a pendant amine in the second coordination sphere on proton transfer at a dissymmetrically disubstituted diiron system related to the [2Fe]H subsite of [FeFe]H2ase.

Studies of the protonation of [Fe2(CO)4(kappa2-PNP)(mu-pdt)] (1; PNP = (Ph2PCH2)2NCH3) by HBF4.Et2O showed that the nature of the reaction product depends on whether the reaction is conducted in acetone or in dichloromethane. In acetone, an N-protonated form, 2, is isolated. Tautomerization of 2 in CH2Cl2 gives rise to a mu-hydride species 3. Variable-temperature NMR experiments have been performed to clarify the processes involved.

Nickel-binding and accessory proteins facilitating Ni-enzyme maturation in Helicobacter pylori.

Helicobacter pylori colonizes the human gastric mucosa and this can lead to chronic gastritis, peptic and duodenal ulcers, and even gastric cancers. The bacterium colonizes over one-half of the worlds population. Nickel plays a major role in the bacteriums colonization and persistence attributes as two nickel enzyme sinks obligately contain the metal. Urease accounts for up to 10% of the total cellular protein made and is required for initial colonization processes, and the hydrogen oxidizing hydrogenase provides the bacterium a high-energy substrate yielding low potential electrons for energy

Multiple regulatory mechanisms in the chloroplast of green algae: relation to hydrogen production.

A complex regulatory network in the chloroplast of green algae provides an efficient tool for maintenance of energy and redox balance in the cell under aerobic and anaerobic conditions. In this review, we discuss the structural and functional organizations of electron transport pathways in the chloroplast, and regulation of photosynthesis in the green microalga Chlamydomonas reinhardtii. The focus is on the regulatory mechanisms induced in response to nutrient deficiency stress and anoxia and especially on the role of a hydrogenase-mediated reaction in adaptation to highly reducing conditions

Thematic minireview series: Metals in Biology 2012.

Metals are present in about one-half of the protein structures available and also have critical roles in nucleic acid biochemistry. This prologue introduces the fourth of the Thematic Minireview Series on Metals in Biology, which deals with several topics involving iron, manganese, copper, and other metals. The six minireviews discuss metal transport and intracellular homeostasis, including chaperones and siderophores, maturation of the diiron active sites in hydrogenases, the balance between manganese and iron, and copper homeostasis relevant to pathogens.

Sustained hydrogen delivery enhances microbiome-associated cadmium and copper immobilization in co-contaminated soils.

Molecular hydrogen (H₂) is an important energy source for soil microbes and has been implicated in biogeochemical regulation; however, whether-and how-exogenous H₂ inputs promote Cd and Cu immobilization in co-contaminated soils remains unclear. We engineered MgH₂-biochar granules capable of sustained H₂ release for up to 448 h. The granules exhibited high adsorption capacities in aqueous Cd-Cu systems (Cu: 59.76 mg g⁻¹; Cd: 35.76 mg g⁻¹). In soil, MgH₂-biochar treatment was associated with enhanced microbial hydrogenase activity, reduced oxidative stress, and enrichment of functional taxa, to

A 3D-printed algal-bacterial microreactor for photobiological hydrogen production coupled with actual wastewater treatment.

Microalgae-driven photosynthetic hydrogen production has emerged as a promising approach to tackle the global energy crisis and reduce carbon emissions. Simultaneously, microalgae can remove phosphorus and nitrogen from wastewater while sequestering CO2 during hydrogen production. However, challenges such as the intrinsic oxygen sensitivity of hydrogenases and the relatively low hydrogen productivity remain major hurdles that constrain the process viability of microalgae for combined wastewater treatment and hydrogen production. A 3D-bioprinted algal-bacterial microreactor (ABMR) with a core-s

Functional role of the MrpA- and MrpD-homologous protein subunits in enzyme complexes evolutionary related to respiratory chain complex I.

NADH:quinone oxidoreductase or complex I is a large membrane bound enzyme complex that has evolved from the combination of smaller functional building blocks. Intermediate size enzyme complexes exist in nature that comprise some, but not all of the protein subunits in full size 14-subunit complex I. The membrane spanning complex I subunits NuoL, NuoM and NuoN are homologous to each other and to two proteins from one particular class of Na(+)/H(+) antiporters, denoted MrpA and MrpD. In complex I, these ion transporter protein subunits are prime candidates for harboring important parts of the pr

Inverted Regulation of Multidrug Efflux Pumps, Acid Resistance, and Porins in Benzoate-Evolved Escherichia coli K-12.

Benzoic acid, a partial uncoupler of the proton motive force (PMF), selects for sensitivity to chloramphenicol and tetracycline during the experimental evolution of Escherichia coli K-12. Transcriptomes of E. coli isolates evolved with benzoate showed the reversal of benzoate-dependent regulation, including the downregulation of multidrug efflux pump genes, the gene for the Gad acid resistance regulon, the nitrate reductase genes narHJ, and the gene for the acid-consuming hydrogenase Hyd-3. However, the benzoate-evolved strains had increased expression of OmpF and other large-hole porins that

Occurrence and Genetic Diversity of Trichomonas gallinae in Captive Synanthropic Birds in Southeastern Brazil.

Avian trichomonosis is caused by protozoa of the genus Trichomonas, mainly Trichomonas gallinae, which infects the upper digestive tract of birds and is commonly associated with Columbiformes, the main reservoirs of the parasite. This study aimed to investigate the occurrence and genetic diversity of Trichomonas spp. in captive synanthropic birds from southeastern Brazil. Oropharyngeal swabs were collected from 281 birds belonging to 13 avian orders and analyzed using Diamond medium culture, Giemsa-stained smears, and molecular assays. Of the 262 samples submitted to culture analysis, 72 (27.4

Di-, tri- and tetraphosphine-substituted Fe/Se carbonyls: synthesis, characterization and electrochemical properties.

The active sites of [FeFe]-hydrogenase promoted by Fe/E (E = S, Se) clusters have attracted considerable interest due to their significance in understanding the interconversion of hydrogen with protons and electrons. As an extension of the study on Fe/Se clusters related to H-cluster model compounds of [FeFe]-hydrogenase, a series of tertiary phosphine substituted Fe/Se carbonyls were successfully prepared. The treatment of Fe2(μ-SePh)2(CO)6 (A) and excess PR3 resulted in the ferrous bis(selenolate) carbonyls Fe(SePh)2(CO)2(PR3)2 (PR3 = PPhMe2, 1; PMe3, 2) in moderate yields. In striking contr

Preferential use of carbon central metabolism and anaerobic respiratory chains in porcine extraintestinal pathogenic Escherichia coli during bloodstream infection.

Porcine extraintestinal pathogenic Escherichia coli (ExPEC) is occurring with increasing frequency in China, and leads to significant economic and welfare costs in the swine industry. The underlying mechanisms of porcine ExPEC in blood colonization during systematic infection is poorly understood. Here we measured the gene expression of porcine ExPEC in infected animal bloodstream in vivo and fresh swine blood in vitro. Using comparisons with P values of ≤ 0.01, we identified 354 and 313 genes as being significantly up- or down-regulated at least 2-fold change during bloodstream infection, res

Nar1 deficiency results in shortened lifespan and sensitivity to paraquat that is rescued by increased expression of mitochondrial superoxide dismutase.

Saccharomyces cerevisiae Nar1p is an essential Fe/S protein that exhibits striking similarity to bacterial iron-only hydrogenases. Nar1p is required for the maturation of cytosolic and nuclear, but not of mitochondrial Fe/S proteins, and plays a role in modulating sensitivity to oxygen in both yeast and Caenorhabditis elegans through unknown mechanisms. Here we report that Nar1 deficiency results in shortened lifespan and sensitivity to paraquat that is rescued by increased expression of mitochondrial superoxide dismutase. These data suggest that Nar1p promotes protection against oxidative str

trans-[Fe(CN)4 (CO)2 ]2- , a 21st Century [Fe(CN)(CO)] Compound.

In 1887, more than a century after the synthesis of [FeII (CN)6 ]4- , the complex [FeII (CN)5 (CO)]3- , in which a CN- ligand is replaced by CO, was reported. It has taken an additional century for the synthesis of the complex in which a second CN- ligand is substituted by CO: trans-[FeII (CN)4 (CO)2 ]2- (structure shown). Remarkably, this anion is prepared by the simple reaction of FeCl2 with NaCN and CO in water. [Fe(CN)x (CO)y ] complexes are of interest as models for the active site of hydrogenase enzymes.

Effects of elemental sulfur on the metabolism of the deep-sea hyperthermophilic archaeon Thermococcus strain ES-1: characterization of a sulfur-regulated, non-heme iron alcohol dehydrogenase.

The strictly anaerobic archaeon Thermococcus strain ES-1 was recently isolated from near a deep-sea hydrothermal vent. It grows at temperatures up to 91 degrees C by the fermentation of peptides and reduces elemental sulfur (S(o)) to H2S. It is shown here that the growth rates and cell yields of strain ES-1 are dependent upon the concentration of S(o) in the medium, and no growth was observed in the absence of S(o). The activities of various catabolic enzymes in cells grown under conditions of sufficient and limiting S(o) concentrations were investigated. These enzymes included alcohol dehydro

Chemoautotrophic growth of hydrogen-uptake-positive strains of Rhizobium japonicum.

Recently reported research from this laboratory has demonstrated the autotrophic growth of certain hydrogen-uptake-positive strains of Rhizobium japonicum and defined minimal conditions for such growth. Ribulose 1,5-bisphosphate carboxylase has been detected in autotrophically growing cells, but at low specific activity. Moreover, growth rates were low, and growth ceased at low cell densities. We report here improved autotrophic growth rates of R. japonicum SR through the use of a modified mineral salts/vitamins medium and a programmed increase in oxygen tension as autotrophic growth proceeds.
