A normobaric gas exposure animal chamber and its validation in hyperoxia exposure.

We are living in an environment full of gases, and any change in the concentration of a component of the air or contaminants (usually toxic) in the air may significantly threaten human health. Thus, to investigate the influence of gases in animal models it is helpful to elucidate the pathogenesis of gas-related injury. Although there are devices used for gas exposure in animals, there are still limitations in the establishment of these animal models, such as the change in gas concentration during the refreshing of water, food and litter, and the contamination of toxic gases released by animals

Effects of hyperoxia and hypoxia on the physiological traits responsible for obstructive sleep apnoea.

Oxygen therapy is known to reduce loop gain (LG) in patients with obstructive sleep apnoea (OSA), yet its effects on the other traits responsible for OSA remain unknown. Therefore, we assessed how hyperoxia and hypoxia alter four physiological traits in OSA patients. Eleven OSA subjects underwent a night of polysomnography during which the physiological traits were measured using multiple 3-min 'drops' from therapeutic continuous positive airway pressure (CPAP) levels. LG was defined as the ratio of the ventilatory overshoot to the preceding reduction in ventilation. Pharyngeal collapsibility

Recovery effects of repeated exposures to normobaric hyperoxia on local muscle fatigue.

Reported recovery effects of hyeroxia are conflicted. This study aimed to identify the effects and the mechanisms of normobaric hyperoxia on the recovery of local muscle fatigue, which is the most commonly encountered form of fatigue both daily and in training and competitions. Twelve male subjects performed 3 × 3 × no less than 30 seconds of isometric quadriceps exercise at 70% of maximum voluntary isometric contraction (MVIC) separated by two 15-minute recovery sessions under 1 of 2 different atmospheric oxygen concentrations, one in normoxia (NOX; 20.9% O2) and another in hyperoxia (HOX; 30

Response of rat lung tissue to short-term hyperoxia: a proteomic approach.

An inspiratory oxygen fraction of 1.0 is often required to avoid hypoxia both in many pre- and in-hospital situations. On the other hand, hyperoxia may lead to deleterious consequences (cell growth inhibition, inflammation, and apoptosis) for numerous tissues including the lung. Whereas clinical effects of hyperoxic lung injury are well known, its impact on the expression of lung proteins has not yet been evaluated sufficiently. The aim of this study was to analyze time-dependent alterations of protein expression in rat lung tissue after short-term normobaric hyperoxia (NH). After approval of

[Effects of hyperoxia on erythropoietin receptor expression in lung development of neonatal rats].

OBJECTIVE: Oxygen toxicity is thought to be a major contributing factor in the pathogenesis of bronchopulmonary dysplasia (BPD). Animal experiments reveal that erythropoietin (EPO) may have protective effects against hyperoxic lung injury, but the mechanisms remain unknown. The aim of this study was to evaluate effects of hyperoxia on erythropoietin receptor expression in lung development of neonatal rats. METHODS: Several litters of Wistar pups were pooled together within 12 hours after birth and randomly divided into two groups (n = 24 in each): air-exposed control group and hyperoxia-expos

Placental vasculogenesis is regulated by keratin-mediated hyperoxia in murine decidual tissues.

The mammalian placenta represents the interface between maternal and embryonic tissues and provides nutrients and gas exchange during embryo growth. Recently, keratin intermediate filament proteins were found to regulate embryo growth upstream of the mammalian target of rapamycin pathway through glucose transporter relocalization and to contribute to yolk sac vasculogenesis through altered bone morphogenetic protein 4 signaling. Whether keratins have vital functions in extraembryonic tissues is not well understood. Here, we report that keratins are essential for placental function. In the abse

Fetal cerebral oxygenation: the role of maternal hyperoxia with supplemental CO2 in sheep.

OBJECTIVE: We tested the hypothesis that supplemental CO2 can enhance the effect of maternal oxygen administration on fetal cerebral oxygenation. STUDY DESIGN: In near-term fetal sheep (n = 6), we instrumented the cerebral cortex with tissue PO2-laser Doppler flow probes, and placed arterial catheters. Following a 30-minute control period, the ewe breathed 50% O2 for 15 minutes, followed by added 6% CO2 for 15 minutes. We examined fetal cortical tissue PO2, cerebral blood flow (CBF), and fetal and maternal blood gases and related variables. RESULTS: In response to maternal O2 administration,

Ventilatory response to hyperoxia in newborn mice heterozygous for the transcription factor Phox2b.

Heterozygous mutations of the transcription factor PHOX2B have been found in most patients with central congenital hypoventilation syndrome, a rare disease characterized by sleep-related hypoventilation and impaired chemosensitivity to sustained hypercapnia and sustained hypoxia. PHOX2B is a master regulator of autonomic reflex pathways, including peripheral chemosensitive pathways. In the present study, we used hyperoxic tests to assess the strength of the peripheral chemoreceptor tonic drive in Phox2b+/-newborn mice. We exposed 69 wild-type and 67 mutant mice to two hyperoxic tests (12-min a

Developmental plasticity of the hypoxic ventilatory response after perinatal hyperoxia and hypoxia.

Both genetic and environmental factors influence the normal development of the respiratory control system. This review examines the role perinatal O2 plays in the development of normoxic breathing and the hypoxic ventilatory response in mammals. Hyperoxia and hypoxia elicit plasticity in respiratory control that is unique to development and may persist weeks to years after return to normoxia. Specifically, both hyperoxia and hypoxia during early postnatal development attenuate the adult hypoxic ventilatory response, but the underlying mechanisms for this plasticity differ. Hyperoxia attenuates

Histological changes in the alveolar structure of the rat lung after exposure to hyperoxia.

Oxygen therapy is an important powerful tool in the management of critically ill patients; however, it carries the hazards of pulmonary toxicity if not properly monitored. The aim of the present study was to evaluate the effect of acute and subacute exposure to normobaric hyperphysiologic concentrations of oxygen (O2) on the alveolar structure of rat lung. Thirty adult male albino rats were used in the present work. They were divided equally into 5 groups. Group I included unexposed rats and was considered a control group, group II included rats exposed to 95% O2 for 24 hours, group III includ

Hyperoxia activates the ATR-Chk1 pathway and phosphorylates p53 at multiple sites.

Hyperoxia has been shown to cause DNA damage resulting in growth arrest of cells in p53-dependent, as well as p53-independent, pathways. Although H2O2 and other peroxides have been shown to induce ataxia telangiectasia-mutated (ATM)-dependent p53 phosphorylation in response to DNA damage, the signal transduction mechanisms in response to hyperoxia are currently unknown. Here we demonstrate that hyperoxia phosphorylates the Ser15 residue of p53 independently of ATM. Hyperoxia phosphorylated p53 (Ser15) in DNA-dependent protein kinase null (DNA-PK-/-) cells, indicating that it may not depend on

Effects of hyperoxia on neonatal myocardial energy status and response to global ischemia.

BACKGROUND: This study examines the effect of neonatal exposure to clinically relevant hyperoxia levels on both in vivo myocardial metabolism and the subsequent metabolic response to global ischemia. METHODS: Three-day-old pigs were ventilated to normoxia (80 mm Hg, 2 or 5 hours, n = 11), mild hyperoxia (250 mm Hg, 2 hours, n = 9), or severe hyperoxia (500 mm Hg, 5 hours, n = 14). Ventricular biopsies obtained at the end of the ventilation period, and at early and late ischemia were analyzed for ATP, ADP, AMP, creatine phosphate, glycogen, and lactate. RESULTS: Hyperoxia did not significantl

Lung inflammation in hyperoxia can be prevented by antichemokine treatment in newborn rats.

Hyperoxia may contribute to lung disease in newborns through effects on alveolar neutrophils which predominate in respiratory distress syndrome and other acute lung injuries. Neutrophil chemokines such as interleukin-8 (IL-8) regulate chemoattraction, and are elevated in tracheal aspirates of newborns who develop bronchopulmonary dysplasia (BPD). Blockade of neutrophil chemokines may reduce hyperoxia-induced inflammatory lung injury and BPD. We therefore tested the hypothesis that hyperoxia contributes to elevations of rat neutrophil chemokines, cytokine-induced neutrophil chemoattractant-1 (C

Effect of chronic hyperoxia on young and old rat carotid body ultrastructure.

Morphologic, physiological, and biochemical changes occur in the carotid body (CB) during postnatal development in relation to physiological requirements. Chronic normobaric hyperoxia attenuates the carotid chemosensory response to hypoxia. During aging there is less of a CB response to hypoxia, which results in a reduced ventilatory adaptation and chemosensory discharge. To test if the oxygen-sensitive mechanism is affected by chronic hyperoxia in an age-dependent fashion, we have studied structural and ultrastructural aspects of young and old rat CBs. Four groups of six male Wistar rats were

Ventilatory stability to transient CO2 disturbances in hyperoxia and normoxia in awake humans.

Modarreszadeh and Bruce (J. Appl. Physiol. 76: 2765-2775, 1994) proposed that continuous random disturbances in arterial PCO2 are more likely to elicit ventilatory oscillation patterns that mimic periodic breathing in normoxia than in hyperoxia. To test this hypothesis experimentally, in nine awake humans we applied pseudorandom binary inspired CO2 fraction stimulation in normoxia and hyperoxia to derive the closed-loop and open-loop ventilatory responses to a brief CO2 disturbance in terms of impulse responses and transfer functions. The closed-loop impulse response has a significantly higher

Histamine-induced reflex tracheal constriction is attenuated by hyperoxia and exaggerated by hypoxia.

The effect of vagal reflexes on bronchomotor tone can be altered by their interaction with other bronchomotor factors, such as the prevailing state of oxygenation. In anesthetized, paralyzed, and artificially ventilated dogs, a reflex constriction was induced in an isolated tracheal segment by administration of aerosolized histamine to the lungs. When the challenge was repeated during hypoxic conditions (PaO2, 45 mmHg), the magnitude of the reflex response was significantly larger than during normoxia. In contrast, during hyperoxia (PaO2, 344 mmHg), the response was significantly smaller than

A critical assessment of the mechanism by which hyperoxia attenuates exercise-induced asthma.

Recent data demonstrate that the magnitude of the heat loss that occurs from the respiratory tract during exercise correlates with the degree of post-exertional obstruction that develops in asthmatics. Respiratory heat loss relates directly to the minute ventilation and heat capacity of the inspired gas and inversely to its water content and temperature. Because it has been shown that inhaling 100% oxygen during exercise blunts the obstructive response, we wondered if this effect could be accounted for by differing values of heat exchange with air and oxygen breathing. To examine this question
