Unique Cytologic Features of Thyroiditis Caused by Immune Checkpoint Inhibitor Therapy for Malignant Melanoma.

Blockade of immune checkpoint molecules to reverse cancer-induced immune suppression can improve anti-tumor immune responses in cancer patients. Monoclonal antibodies targeting two such molecules, Programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte associated protein 4 (CTLA-4) have shown clinical benefit in the treatment of advanced malignancies, including metastatic melanoma. Adverse effects of these immune checkpoint inhibitors include immune-related adverse events (irAE) and the inducing of new autoimmunity, of which one of the most common is autoimmune thyroiditis. Though th

Self-assembled protein-aptamer nanoparticles for targeted cancer immunotherapy.

Developing effective delivery methods for therapeutic proteins poses a major challenge in oncology. Protein drugs offer targeted mechanisms against tumor cells, minimizing off-target effects and potential for personalized therapy. Our study proposes a controllable in situ self-assembly strategy to construct FimH protein-based nanoparticles incorporating mIFP protein and PD-L1 aptamer, demonstrating potential for targeted protein delivery and tumor therapy. The PD-L1 aptamer exerts its effect by disrupting immune checkpoints, thereby modulating immune checkpoints and promoting immune cell activ

A PD-1/PD-L1-Sensitive Co-culture-Based Primary T-Cell Activation Assay.

Programmed cell death protein 1 (PD-1) is crucial in inhibiting immune responses by modulating the activity of T cells. We present an in vitro assay that is based on a co-culture of primary immune cells represented by human peripheral blood mononuclear cells (PBMCs), isolated from healthy donors with Chinese hamster ovary-derived cell line (CHO-K1) overexpressing human PD-L1 protein (hPD-L1) and an artificial TCR-activator construct (TCRAct). CHO-K1/TCRAct/hPD-L1 cells mimic antigen-presenting cells by activating T cells via the T-cell receptor (TCR) and providing a ligand for the negative imm

Immune checkpoint inhibitors and the orbit; two cases of reactive dacryoadenitis.

Immune checkpoints refer to mechanisms entrusted with the modulation of immune responses in peripheral tissues and are required for minimising collateral damage. Immune checkpoint inhibitors (ICPi) work through numerous pathways, including the anti-CTLA-4 (cytotoxic T-lymphocyte-associated protein 4), anti-PD-1 (programmed cell death protein 1) and the PD-L1 (protein cell death protein-ligand-1) pathways. They are proving to be an exciting therapeutic avenue in the attempt to activate anti-tumour activity. Ipilimumab is a fully human monoclonal antibody working on the anti-CTLA-4 pathway, whil

ctDNA Dynamics Identifies Pseudoprogression in a Metastatic Melanoma Patient Treated With Nivolumab/Relatlimab.

Pseudoprogression is known to occur in patients with metastatic melanoma treated with immune checkpoint inhibitors and complicates clinical decision-making. Better methods of distinguishing pseudoprogression from true progression are necessary. In this case report, we show how circulating tumor DNA (ctDNA) plays a role in identifying pseudoprogression on early interval radiologic response assessment in a patient with metastatic melanoma treated with combination anti-programmed cell death protein 1 (anti-PD-1) and anti-lymphocyte activation gene 3 (anti-LAG-3). Circulating tumor DNA is a promis

Immune checkpoint activity regulates polycystic kidney disease progression.

Innate and adaptive immune cells modulate the severity of autosomal dominant polycystic kidney disease (ADPKD), a common kidney disease with inadequate treatment options. ADPKD has parallels with cancer, in which immune checkpoint inhibitors have been shown to reactivate CD8+ T cells and slow tumor growth. We have previously shown that in PKD, CD8+ T cell loss worsens disease. This study used orthologous early-onset and adult-onset ADPKD models (Pkd1 p.R3277C) to evaluate the role of immune checkpoints in PKD. Flow cytometry of kidney cells showed increased levels of programmed cell death prot

Update on Immunotherapy Cardiotoxicity: Checkpoint Inhibitors, CAR T, and Beyond.

Immunotherapy is an innovative approach to cancer treatment that involves using the body's immune system to fight cancer. The landscape of immunotherapy is constantly evolving, as new therapies are developed and refined. Some of the most promising approaches in immunotherapy include immune checkpoint inhibitors (ICIs): these drugs target proteins on the surface of T-cells that inhibit their ability to attack cancer cells. By blocking these proteins, checkpoint inhibitors allow T-cells to recognize and destroy cancer cells more effectively. CAR T-cell therapy: this therapy involves genetically

CSNK2B Expression Aligns with a GDF15-Associated Immune Molecular State in Hepatocellular Carcinoma.

Background: Hepatocellular carcinoma (HCC) remains a major cause of cancer-related mortality, and responses to immune checkpoint blockade are often limited. Casein kinase 2 beta (CK2beta), encoded by CSNK2B, has established tumor-intrinsic functions, but its relationship with immune states in HCC remains unclear. Methods: We integrated TCGA-LIHC, six GEO cohort-platform combinations, GSE149614 single-cell RNA sequencing data, CIBERSORTx deconvolution, broad-compartment CellChat inference, and public proteomic resources. We evaluated CSNK2B expression, overall survival, a prespecified GDF15-ass

Role of mitochondrial translation in modulating inflammatory disease outcome: Current knowledge and future perspectives.

Mitochondrial translation is crucial for maintaining cellular respiration, energy balance, calcium signaling, apoptosis, immune surveillance, and the regulation of inflammatory responses. This specialized process, involving mitochondrial rRNAs, tRNAs, mitoribosomes, and nuclear-encoded translation factors, ensures the synthesis of mitochondrially encoded proteins that support oxidative phosphorylation. The mitochondrial translation cycle is tightly regulated by RNA-binding proteins, mitochondrial unfolded protein response, and stress-responsive pathways such as mTOR, particularly during metabo

Inhibitory TIGIT signaling is dependent on T cell receptor activation.

The immune checkpoint receptor T cell immunoreceptor with Ig and ITIM domains (TIGIT) can signal via cytoplasmic motifs to regulate T cell function. The signaling molecules that mediate inhibitory TIGIT signaling and its regulation in T cells remain poorly defined. Here, proximity proteomics is employed to define TIGIT-proximal proteins upon CD155 engagement, identifying several that are ligation-specific, including those involved in signaling (Grb2 and SOS1), cytoskeletal regulation (CD2AP and SdcBP), and endocytosis (IST1 and SNX3). Through crosslinking followed by immunoprecipitation, we sh

Targeting LAG-3, TIM-3, and TIGIT for cancer immunotherapy.

In one decade, immunotherapy based on immune checkpoint blockades (ICBs) has become a new pillar of cancer treatment following surgery, radiation, chemotherapy, and targeted therapies. However, not all cancer patients benefit from single or combination therapy with anti-CTLA-4 and anti-PD-1/PD-L1 monoclonal antibodies. Thus, an increasing number of immune checkpoint proteins (ICPs) have been screened and their effectiveness evaluated in preclinical and clinical trials. Lymphocyte activation gene-3 (LAG-3), T cell immunoglobulin and mucin-domain-containing-3 (TIM-3), and T cell immunoreceptor w

Takotsubo syndrome as an outcome of the use of checkpoint inhibitor therapy in patients with COVID-19.

Takotsubo Syndrome (TS) is a heart disease caused by extreme exposure of the body to physical or psychological stress. In the context of COVID-19, the virus can be a significant source of stress, with particular attention being paid to the cytokine storm as a cause of damage to the body. New research shows that the production of specific cytokines is linked to the activation of immune checkpoint proteins such as PD-1, PD-L1, and CTLA-4 on T cells. Although initially beneficial in combating infections, it can suppress defense and aid in disease progression. Therefore, checkpoint inhibitor thera

PD-1 /PD-L1 checkpoint in hematological malignancies.

Programmed cell death protein 1 (PD-1), is a cell surface receptor with an important role in down-regulating the immune system and promoting self-tolerance by suppressing T cell inflammatory activity. PD-1/PDL1 axis represents a checkpoint to control immune responses and it is often used as a mechanism of immune escaping by cancers and infectious diseases. Many data demonstrate its important role in solid tumors and report emerging evidences in lymphoproliferative disorders. In this review, we summarized the available data on the role of PD-1/PD-L1 checkpoint in lymphoproliferative diseases an

B7-H3 (CD276): an actionable therapeutic target and prognostic biomarker across human malignancies.

Immune checkpoint protein B7-H3 (CD276) has garnered increasing attention as a cancer immunotherapy target. Despite the absence of a definitively identified receptor, B7-H3 is implicated in multiple mechanisms of tumor progression. As a cancer biomarker, it is overexpressed in numerous cancer types and is often associated with poor prognosis. B7-H3 is currently the target of numerous immunotherapeutic strategies, and clinical trials are underway in various cancer types. This review provides an updated summary of the current body of knowledge regarding immune checkpoint protein B7-H3 and cancer

NU-1025 modulates immunogenic cell death in bladder cancer: A multi-omics insight.

Immune checkpoint blockade has shown limited efficacy in bladder cancer, underscoring the need for therapeutic strategies that enhance tumor immunogenicity. Immunogenic cell death (ICD) can stimulate adaptive immunity and remodel the tumor microenvironment, yet its comprehensive landscape and druggable potential in bladder cancer remain unexplored. Here, we performed integrative bioinformatics analysis of transcriptomic data from TCGA and GEO bladder cancer cohorts to characterize ICD-associated subtypes, construct a prognostic risk model, and analyze immune infiltration. Connectivity Map (CMa

Checkpoint Blockade Toxicity and Immune Homeostasis in the Gastrointestinal Tract.

Monoclonal antibodies targeting the regulatory immune "checkpoint" receptors CTLA-4, PD-1, and PD-L1 are now standard therapy for diverse malignancies including melanoma, lung cancer, and renal cell carcinoma. Although effective in many patients and able to induce cures in some, targeting these regulatory pathways has led to a new class of immune-related adverse events. In many respects, these immune toxicities resemble idiopathic autoimmune diseases, such as inflammatory bowel disease, autoimmune hepatitis, rheumatoid arthritis, and vitiligo. Understanding the pathogenesis of these immune tox

Intratumor delivery of a novel plasmid combination induces high levels of metastatic melanoma regression.

Direct intratumoral administration of immune-stimulating agents can generate systemic immunity following a tumor-localized immune response. Interleukin 12 (IL-12) activates T cells and reverses immunosuppression, generating a localized patient-antigen-specific immune response. Changes in the tumor microenvironment in response to this cytokine facilitate the function of checkpoint inhibitors. In this study, a plasmid (p)-based approach was used to generate intratumor expression of IL-12 protein and of a polypeptide designed to inhibit PD-1/PD-L1 binding, potentially to function as a checkpoint

Blockade of colon cancer metastasis via single and double silencing of PCSK7/PCSK9: enhanced T cells cytotoxicity in mouse and human.

BACKGROUND: Immunotherapy approaches based on T cells provided breakthroughs in cancer treatment but could cause many immune-related adverse events, and their efficacy is limited for many cancers with an acquired dysfunction/exhaustion of T cells. The present study presents a novel role in immunity for proprotein convertase subtilisin-kexin 7 (PCSK7), the seventh proprotein convertase of the 9-membered secretory proprotein convertase subtilisin-kexin (PCSK)-family. METHODS: We analyzed cell surface levels of various immune checkpoint proteins in human and mouse cell models in the presence or
