1-Azabicyclo[2.1.1]hexanes are valuable nitrogen-rich scaffolds with growing importance in medicinal chemistry. We report a Brønsted acid-catalyzed formal cycloaddition of N-substituted indoles with azabicyclobutane (ABB) ketones, providing a mild, transition-metal-free, and highly stereoselective route to aza-bicyclohexanes. Using pTSA·H2O in HFIP, the reaction proceeds at room temperature in up to 93% yield and 99:1 d.r. In light of the known neuroprotective properties of indole compounds and the promising potential of hydrazone derivatives, two series of aldehyde-heterocyclic hybrids combining those pharmacophores were synthesized as new multifunctional neuroprotectors. Under aerobic copper catalysis, an unprecedented direct synthesis of functionalized indoles via single electron oxidation induced coupling of diarylamines with 1,3-dicarbonyl compounds is presented. The protocol proceeds with good functional group and substrate compatibility, the use of readily available feedstocks and naturally abundant catalyst system, high step and atom efficiency, as well as selectivity, which offers a platform for accessing a new class of indoles with the potential for the discovery of functional molecules.

The combination of Co(acac)2 and AgOTf enables the bimetallic relay catalysis reaction of 2-ethynylanilines and isocyanides, allowing easy and low-cost access to new densely functionalized pyrrolo[2,3-b]indoles. The reaction pathway involves a Co(acac)2-catalyzed double isocyanide insertion followed by a silver-enabled 1,3-dipolar cycloaddition. The synthetic utility of these bicycloaddition reactions results in subsequent C-C and C-N bond-forming events to rapidly build up molecular complexity.
