Fatal pneumonia among metalworkers due to inhalation exposure to Bacillus cereus Containing Bacillus anthracis toxin genes.

Bacillus cereus pneumonia is unusual in nonimmunocompromised hosts. We describe fatal cases in 2 metalworkers and the associated investigation. Anthrax toxin genes were identified in B. cereus isolates from both patients using polymerase chain reaction. Finding anthrax toxin genes in non-Bacillus anthracis isolates has, to our knowledge, only been reported once previously.

Pulmonary reactivity to vanadium pentoxide following subchronic inhalation exposure in a non-human primate animal model.

An experimental study was conducted to evaluate changes in pulmonary reactivity resulting from repeated vanadium pentoxide (V2O5) dust inhalation. The study assessed pulmonary reactivity to V2O5 through the use of provocation challenges, and compared V2O5 reactivity before and after subchronic V2O5 exposure. A total of 24 adult, male cynomolgus monkeys (Macaca fascicularis) were exposed by inhalation for 6 h per day, 5 days per week, for 26 weeks. Two V2O5-exposed groups (n = 8 each) received equal weekly V2O5 exposures (concentration x time) with different exposure profiles. One V2O5-exposed

Measurement of free radicals from smoke inhalation and oxygen exposure by spin trapping and ESR spectroscopy.

Research in smoke inhalation has established that free radicals are produced from gases released during combustion and these species impair lung function. Using spin traps and their adducts in an animal model free radicals were measured. Various hyperbaric oxygen regimens were tested in an attempt to attenuate pulmonary damage caused by free radical reactions. Our data demonstrated that persistent oxygen- and carbon-centered free radicals are detectable in intravascular fluids after smoke inhalation. The smoke inhalation model showed however, clearing of spin trap adducts one hour after smoke

Progressive lung injury over a one-year period after a single inhalation exposure to beryllium sulfate.

The chronic pulmonary toxicity of beryllium sulfate was examined in rats over a 1-yr period after a single, 1-h exposure. Male rats, exposed in a nose-only inhalation chamber to an aerosol of 4.05 micrograms Be/L, were evaluated for lung toxicity by the methods of bronchoalveolar lavage, lung cell kinetics, and histopathologic analysis. Bronchoalveolar lavage activities for alkaline phosphatase (Alk Pase) and acid phosphatase (Ac Pase) were elevated 3 wk after exposure; lactate dehydrogenase (LDH) and Alk Pase activities peaked 3 months after exposure. Histopathologic analysis revealed progres

Development of a microinstillation model of inhalation exposure to assess lung injury following exposure to toxic chemicals and nerve agents in Guinea pigs.

Respiratory disturbances due to chemical warfare nerve agents (CWNAs) are the starting point of mass casualty and the primary cause of death by these weapons of terror and mass destruction. However, very few studies have been implemented to assess respiratory toxicity and exacerbation induced by CWNAs, especially methylphosphonothioic acid S-(2-(bis(1-methylethyl)amino)ethyl)O-ethyl ester (VX). In this study, we developed a microinstillation technique of inhalation exposure to assess lung injury following exposure to CWNAs and toxic chemicals. Guinea pigs were gently intubated by placing a mic

Effects on the postnatal development of the mouse of preconception, postconception and/or suckling exposure to manganese via maternal inhalation exposure to MnO2 dust.

Female mice were exposed either to MnO2 dust (7 hours/day, 5 days/week) or filtered air (control group) for 16 weeks prior to conception. On day 1 of gestation, half of each of these groups was assigned randomly either to MnO2 dust or filtered air exposure until day 17 of gestation. To separate effects of prenatal maternal exposure to MnO2 from postnatal exposure of offspring to Mn via suckling, a complete fostering/-cross-fostering design was employed. Compared to control mothers, mothers exposed to MnO2 prior to conception produced significantly larger litters. Prenatal exposure resulted in

Inhalation exposure and uptake modeling of particle-associated phthalate: from respiratory deposition and clearance to organ toxicokinetics.

This study aimed to develop a novel hybrid computational fluid-particle dynamics (CFPD)-physiologically-based pharmacokinetics (PBPK) model to elucidate the toxicokinetics of particle-associated di(2-Ethylhexyl) phthalate (DEHP) in the human body via inhalation exposure. Initially, a steady mucus flow was simulated with constant rates for the upper and lower respiratory tracts. The desorption, dynamics, and tissue absorption of DEHP were simultaneously described by the Eulerian approach and the mucus-tissue-blood model. Mucociliary clearance effects were examined against the DEHP and its parti

The importance of exposure determination during Specific Inhalation Challenge (SIC) tests for the exclusion of nonspecific irritation.

Work-related asthma (WRA) comprises asthma caused by chemicals in the workplace (occupational asthma, or OA) and preexisting asthma made worse by chemicals and conditions (work-exacerbated asthma, or WEA). Distinguishing these two mechanisms is extremely challenging but imperative for effective health management and regulation under global chemical regulations. The Specific Inhalation Challenge (SIC) test is considered by clinicians as the gold standard for identifying OA. However, an exposure reconstruction exercise based on the case study of methyl methacrylate, currently undergoing harmoniz

Hepatotoxic alterations induced by subchronic exposure of rats to formulated fenvalerate (20% EC) by nose only inhalation.

OBJECTIVE: Fenvalerate (20% EC) is a synthetic pyrethroid, which is commonly used in India by farmers for the protection of many food and vegetable crops against a wide variety of insects. However, its inhalation toxicity data is very limited in the literature due to the fact that the exposure levels associated with these effects were usually not reported. Hence, inhalation exposure was carried out to investigate the hepatotoxic effects. METHOD: Adult male rats were exposed to fen for 4 h/day, 5 days a week for 90 days by using Flow Past Nose Only Inhalation Chamber. Sham treated control rats

Inhalation toxicokinetics of 1,2,4-trimethylbenzene in volunteers: comparison between exposure to white spirit and 1,2,4-trimethylbenzene alone.

The objective of this study was to compare the toxicokinetics of inhaled 1,2,4-trimethylbenzene (1,2,4-TMB) in man after exposure to white spirit with that observed after exposure to 1,2,4-TMB alone. TMBs occur mainly in petroleum products and the TMBs or their metabolites have been suggested as suitable biomarkers of exposure to white spirit and other distillation products. The toxicokinetics were studied in 9 male, healthy volunteers exposed to solvent vapours in an exposure chamber for 2 h during a work load of 50 W. The subjects were exposed to 11 mg/m3 of 1,2,4-TMB on two occasions; durin

Low flow anaesthesia reduces occupational exposure to inhalation anaesthetics. Environmental and biological measurements in operating room personnel.

In the present study we evaluated the occupational exposure to N2O and isoflurane during open circuit (OC) (fresh gas flow > or = minute volume) and low flow (LF) (fresh gas flow = 1.5 l/min) anaesthesia. The effects of active scavenging and of a charcoal filter positioned on the exhausting branch of the ventilator on environmental and urinary concentrations of inhalation anaesthetics were also investigated. The study was carried out in the same operating room provided with non-recirculating air changes (10 per hour). It involved anaesthetists and nurses during routine activity. N2O and isoflu

Effects of prenatal alcohol exposure in mice: influence of an ADH inhibitor and a chronic inhalation study.

In order to examine the effects of an alcohol dehydrogenase inhibitor on the embryotoxic effects of ethanol, pregnant ICR mice were treated with 100 mg/kg pyrazole prior to ethanol injection. I.p. treatment with 2 or 4 g/kg ethanol on day 7 of gestation increased the prenatal mortality rate and produced external and skeletal malformations in the offspring, and the embryotoxic effects were potentiated by pyrazole pretreatment, suggesting that ethanol rather than its metabolites is mainly responsible for the embryotoxicity. In the second experiment, we housed pregnant mice in an ethanol-vapor bo

Polybrominated Diphenyl Ethers (PBDEs) in PM1 of Residential Indoor Air: Levels, Seasonal Variability, and Inhalation Exposure Assessment.

Indoor exposure to polybrominated diphenyl ethers (PBDEs), particularly those bound to fine particulate matter (PM1, particles < 1 µm), may pose a health concern, especially in light of prolonged indoor occupancy and the capacity of ultrafine particles to reach the lower respiratory tract. This study investigates indoor exposure to PBDEs associated with PM1 in residential homes in Zagreb, Croatia, across warm and cold seasons. BDE-47 was consistently detected in all samples, while BDE-183 was consistently absent. Elevated concentrations and increased detection frequencies of BDE-99 and BDE-100

Regional and population-scale trends in human inhalation exposure to airborne microplastics: Implications for health risk assessment.

Growing evidence shows that breathing microplastics (MPs)-polluted air increases the risk of pulmonary health effects. However, a complete understanding of how inhaled MPs distribute within the human respiratory tract (HRT) remains insufficient. This study developed a physiologically-based kinetic HRT model to evaluate the deposition and clearance of MPs over time and at varying concentrations based on their aerodynamic diameter (AD). We quantified the contributions of AD-specific MPs to inhalation exposure trends using literature-based atmospheric MP pollution data from 2015 to 2022. Exposure

Inhalation exposure to a tire-derived pollutant (6-PPD) triggers lung injury and mitochondrial dysfunction in mice.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6-PPD)-a commonly used rubber antioxidant-is primarily released into the environment through tire wear particles, raising concerns about its potential respiratory health effects. However, little is known of its direct effects. This study investigated the pulmonary toxicity of 6-PPD in mice administered 5 or 10 mg/kg via intratracheal instillation for 7 or 14 days. No treatment-related mortality or significant changes in body weight were observed. However, lung weight significantly increased in all 6-PPD-treated groups, indicating lung tissue

Maternal nano-titanium dioxide inhalation exposure alters placental cyclooxygenase and oxidant balance in a sexually dimorphic manner.

The placenta plays a critical role in nutrient-waste exchange between the maternal and fetal circulation, and thus impacts fetal growth and development. We have previously shown that nano-titanium dioxide (nano-TiO2) inhalation exposure during gestation decreased fetal female pup and placenta mass [1], which persists in the following generation [2]. In utero exposed females, once mated, their offspring's placentas had increased capacity for H2O2 production. Generation of oxidants such as hydrogen peroxide (H2O2), have been shown to impact cyclooxygenase activity, specifically metabolites such
