Nerve growth factor, interoception, and sympathetic neuron: lesson from congenital insensitivity to pain with anhidrosis.

Nerve growth factor (NGF) is a well-known neurotrophic factor essential for the survival and maintenance of sensory and sympathetic neurons. Congenital insensitivity to pain with anhidrosis (CIPA) is a genetic disorder due to loss-of-function mutations in the NTRK1 (also known as TRKA) gene encoding TrkA, a receptor tyrosine kinase for NGF. Patients with CIPA provide us a rare opportunity to explore the developmental and physiological function of the NGF-dependent neurons in behavior, cognitive, and mental activities that are not available in animal studies. Here, I discuss the significance of

Emotional and Social Dimension of Abstract Concepts Meet with Interoception in Right Anterior Insula.

According to the embodied cognition theory, which claims that concepts' representation is grounded in sensory and motor components, abstract concepts are grounded in interoception, which is processed in the Anterior Insula (AIns). However, it is not clear whether interoception and abstract concepts share common anatomical substrates, and if yes, whether AIns is one candidate. In this study, we used repetitive transcranial magnetic stimulation (rTMS) on healthy human participants (N = 25, 19 females) to examine whether left and right AIns play a role in both abstract concepts and interoception.

Emotional profiles across physical activity: exploring interoception, emotional regulation and attitudes toward physical effort.

UNLABELLED: While the health impacts of physical activity are well-established, the contribution of internal regulatory processes to these behaviors remains unclear. This study adopts an integrative perspective to examine how interoceptive sensibility and emotion regulation jointly differentiate physical activity levels and predict attitudes toward effort. METHOD: 1073 adults (557 males, mean age = 35.4) completed the Beck Depression Inventory, State-Trait Anxiety Inventory, Cognitive Emotion Regulation Questionnaire, Multidimensional Assessment of Interoceptive Awareness, and Physical Effort

The Critical Role of Pdyn -Lineage Enteric Neurons in Colonic Motility and Visceral Interoception.

Summary/Abstract Sensory neurons play well defined roles in the regulation of intestinal motility, digestion, and interoception, but transcriptional dissection of intrinsic and extrinsic sensory neurons innervating the intestines has been challenging because these cells share many genetic markers. However, we cross-referenced transcriptional profiles of intrinsic and extrinsic intestinal neurons and the cells that surround them and identified Pdyn, the gene encoding Prodynorphin, as a marker of putative sensory enteric neurons of the mouse intestines. In a Pdyn lineage-reporter mouse, we ident

Regulation of addiction by the insular cortex: The interplay between interoception and cognitive function.

The insular cortex (IC) functions as a pivotal core for interoception. Functional magnetic resonance imaging reveals how the IC integrates internal and external stimuli by observing its neural activation patterns. It facilitates interactions among sensory, emotional, and cognitive inputs via specific neural circuits. The finding that IC injury can reduce nicotine intake and the pioneering research on the involvement of the IC in addiction. Subsequently, human studies discovered that the IC amplifies drug-derived pleasure, strengthening the link between cravings and drug use, thus increasing co

Targeting skeletal interoception: a novel mechanistic insight into intervertebral disc degeneration and pain management.

Despite being a leading cause of chronic pain and disability, the underlying mechanisms of intervertebral disc (IVD) degeneration (IVDD) remain unclear. Emerging evidence suggests that mechanosensation (the ability of the skeletal system to perceive mechanical and biochemical signals) mediated by abnormal mechanical loading plays a critical role in the regulation of IVD health. This review examines the complex interactions amongIVDs, intraosseous sensory mechanisms, and the central nervous system (CNS), with a particular focus on the roles of pathways such as PGE2/EP4, Wnt/β-catenin, and NF-κB

Involvement of physiological reactivity and interoception in emotional experience after a traumatic brain injury.

OBJECTIVE: Emotional experience is based, among other factors, on physiological reactivity (PR) and the awareness of this reactivity corresponding to interoception. After a traumatic brain injury (TBI), patients exhibit reduced PR and interoception, raising questions about the integrity of their emotional experience. METHOD: To examine this issue, 26 men with moderate to severe TBI (age: 37 ± 11) and 26 healthy male controls (age: 35 ± 14) watched emotional films (amusement, tenderness, anger, disgust). PR was measured via electrodermal activity (EDA) and heart rate variability (HRV). After e

Gut sensing of food ingredients and interoception-mediated regulation of feeding and glucose metabolism.

Food intake not only provides pleasure through exteroceptive sensations such as taste and smell but also elicits beneficial physiological effects via interoceptive signals arising from the gastrointestinal tract and beyond. Among these interoceptive pathways, vagal sensory (vagal afferent) nerves play a central role in transmitting food-derived information to the brain. This review first outlines the anatomical and functional characteristics of vagal sensory nerves. It then examines how food-related signals, including mechanical stretching of the gastrointestinal wall, gastrointestinal and pan

Neuroimaging Meta-Analyses Reveal Convergence of Interoception, Emotion, and Social Cognition Across Neurodegenerative Diseases.

BACKGROUND: Simultaneous interoceptive, emotional, and social cognition deficits are observed across neurodegenerative diseases. Indirect evidence suggests shared neurobiological bases underlying these impairments, termed the allostatic-interoceptive network (AIN). However, no study has yet explored the convergence of these deficits in neurodegenerative diseases or examined how structural and functional changes contribute to cross-domain impairments. METHODS: A Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) activated likelihood estimate meta-analysis encompassed s

Interoception, emotion regulation strategies and skin-picking behaviors - Results of an intensive longitudinal study.

Compulsive skin-picking is associated with emotion regulation difficulties, whose origins remain unclear. Interoception, plays an important role in effective emotion regulation. This study examined the relationship between interoception, emotion regulation strategies, and skin-picking in 136 individuals (85% women, aged 18-41), including 71 engaging in skin-picking and 65 psychologically healthy controls. We were interested in between-group differences in maladaptive and adaptive emotion regulation and aspects of interoception as well as associations of the latter with skin-picking symptoms an

Neurobiology of pain, interoception and emotional response: lessons from nerve growth factor-dependent neurons.

Although nerve growth factor (NGF) is a well-known neurotrophic factor, it also acts as a mediator of pain, itch and inflammation. Congenital insensitivity to pain with anhidrosis (CIPA) is an autosomal recessive genetic disorder caused by loss-of-function mutations in NTRK1, the gene encoding a receptor tyrosine kinase for NGF, TrkA. Mutations in NTRK1 cause the selective loss of NGF-dependent neurons in otherwise intact systems. NGF-dependent primary afferents are thinly myelinated Aδ or unmyelinated C-fibers that are dependent on the NGF-TrkA system during development. In CIPA, the lack of

The impact of threat- and safety-enhancing information on the acquisition of fear of interoceptive threat.

Social learning, such as learning through verbal instruction, has been proposed to play a crucial role in the development pathological fear of bodily sensations. Despite the ubiquity of verbal information in daily life, the effects of verbal information on subsequent interoceptive fear learning remain empirically untested. Therefore, we aimed to examine how verbal information about the threatening vs. benign nature of bodily sensations influences subsequent interoceptive fear conditioning. 77 healthy participants were randomly assigned to receive either threat- or safety-enhancing information

Testing a learning-based account of interoceptive hunger using an illusory induction.

Physiological changes cause many interoceptive cues (e.g., a rumbling stomach), and so it is often assumed they also cause their motivational consequences (e.g., hunger). However, there is grounds to suspect that the meaning of interoceptive cues are learned and it is their meaning that causes their motivational consequences. We tested this possibility by psychologically inducing an illusory interoceptive hunger cue to see if it stimulated hunger and food intake. Participants (n = 130) received either a stomach rumbling or control sound (electrical arcing/buzzing), while judging liking for non

The Emotional Experience of Time: Effects of Anxiety and Interoceptive Ability on Temporal Perception.

Our experience of time is deeply shaped by emotional and bodily states. Anxiety is known to distort our temporal perception, yet the underlying mechanisms remain poorly understood. Here, we tested whether interoception links anxiety to variability in timing. Thirty participants underwent fMRI while performing a temporal reproduction task within a modified Threat of Scream paradigm. Safe and Threat blocks alternated, with aversive screams unpredictably delivered during Threat blocks. Screams reliably increased subjective anxiety and engaged salience and limbic regions, including the amygdala, h

The cognitive neurophysiological mechanisms underlying the impact of Interoceptive attention on cognitive control.

Influential theories and extensive research suggest that interoception significantly affects an individual's cognition and emotions, yet the neural mechanisms involved remain incompletely understood. This study aims to clarify the neurophysiological dynamics of how interoceptive attention modulates cognitive control. Participants completed a classic Stroop task preceded by cue stimuli designed to bias processing toward cardiac/interoceptive information (normal vs. abnormal heart-rate cues) or externally salient threat-related information (fearful faces). Neural electrophysiological data elicit

Regulatory architecture controlling terminal differentiation of an interoceptive paraneuron in C. elegans.

Interoceptive paraneurons are neuron-like cells located within internal epithelial cell surfaces that sense internal stimuli to evoke specific behavioral or physiological responses. The elucidation of terminal differentiation programs of paraneurons is expected to provide insights into how epithelial cells acquire neuron-like feature during development and possibly also over evolutionary time. We define here transcriptional programs that control the terminal differentiation of an interoceptive paraneuron class in the nematode C. elegans, called uv1. The uv1 cells sense mechanosensory inputs in

Interoceptive and metacognitive contributors to fatigue in multiple sclerosis: network and mediation analyses.

BACKGROUND: Although fatigue is one of the most prevalent symptoms in patients with multiple sclerosis (MS), its underlying mechanisms remain insufficiently understood. Emerging evidence suggests that interoceptive sensibility and maladaptive metacognitive beliefs may play a central role in the development and maintenance of fatigue. The present study examined the interplay between interoceptive sensibility, maladaptive metacognitive beliefs, affective symptoms, and fatigue in individuals with MS. METHODS: In this cross-sectional study, 240 patients with relapsing-remitting MS completed valid

Synchronization of Cardiac and Musical Signals Improves Interoceptive, Cardiac, and Emotional Functioning.

The ability to seamlessly integrate sensory information from the environment (exteroception) and physiological states (interoception) is a key aspect of our awareness and well-being. Alterations in these processes often result in uncertainty about bodily states and dysregulation of physiological and emotional processes, as observed in clinical conditions. In this study, we employed an interactive music system as a novel method to improve interoceptive, cardiac, and emotional functioning, combining cardiac biofeedback, mindfulness, and music listening approaches. A sample of 24 healthy particip

Predictive role of exteroceptive and interoceptive bodily dimensions to schizotypal personality traits.

The phenomenological approach to schizophrenia emphasizes the role of bodily experiences in the onset and manifestation of positive, negative and disorganized psychotic symptoms. According to the dimensional approach to psychosis, there exists a continuum ranging from individuals with low levels of schizotypy to diagnosed schizophrenia patients, with schizotypy encompassing positive-like, negative-like, and disorganized-like symptoms of schizophrenia. Empirical evidence suggests that along this continuum, both exteroceptive (external sensory) and interoceptive (internal bodily) dimensions migh
