Evidence of oleuropein degradation by olive leaf protein extract.

The enzymatic activity of raw protein olive leaf extract has been investigated in vivo, on olive leaf homogenate and, in vitro with pure oleuropein and other phenolic substrates. At least two types of enzymes were found to be involved in the degradation of endogenous oleuropein in olive leaves. As for the in vitro experiments, the presence of active polyphenoloxidase and β-glucosidase was determined by HPLC and UV-Visible spectroscopy. Interestingly, both the enzymatic activities were found to change during the storage of olive leaves. Specifically, the protein extracts obtained from fresh lea

Identification and functional characterization of UDP-glycosyltransferases catalyzing the glycosylation of oleoside 11-methyl ester in Olea europaea.

MAIN CONCLUSION: Two UDP-glycosyltransferases from Olea europaea were identified and characterized. Both catalyze the glycosylation of oleoside 11-methyl ester to form 7-β-1-D-glucopyranosyl 11-methyl oleoside, a putative intermediate in the oleuropein biosynthetic pathway. INTRODUCTION: Oleuropein, a secoiridoid glycoside abundant in Oleaceae plants, is extensively utilized in the pharmaceutical, nutraceutical, and cosmetic industries due to its diverse pharmacological activities. The biosynthetic pathway from oleoside 11-methyl ester (OME) to oleuropein remains incompletely elucidated. In t

Antithrombotic effect of geniposide and genipin in the mouse thrombosis model.

Geniposide is one of the constituents of Gardenia fruit (Gardenia jasminoides Ellis, Rubiaceae), which has been used in traditional medicine. Although its anti-inflammatory and antithrombotic effects have been reported, the way it acts is still unclear. We have investigated the effects of geniposide and its metabolite genipin on thrombogenesis and platelet aggregation. In an in vivo model, geniposide and genipin significantly (P < 0.05) prolonged the time required for thrombotic occlusion induced by photochemical reaction in the mouse femoral artery. In an in vitro study, both geniposide and g
