Chemotherapy triggers immune evasion by fostering LEPR+ Kupffer cell differentiation in liver metastases.

Conventional chemotherapy achieves clinical efficacy not only through its cytotoxic effects but also by reactivating immune surveillance. However, whether chemotherapy can inversely suppress antitumor immunity remains largely unexplored. Here, we integrate cross-species single-cell and spatial transcriptomics to investigate how chemotherapy programs immune cell plasticity. Our findings reveal that chemotherapy-educated liver-resident Kupffer cells (KCs) promote immune evasion and chemoresistance in liver metastases. These reprogrammed KCs, characterized by leptin receptor expression (LEPR+), o

The Key Role of Complement Receptor CRIg in Kupffer Cell-Mediated Liver Disease Progression.

Liver diseases, ranging from chronic hepatitis and metabolic dysfunction to cirrhosis and hepatocellular carcinoma, represent a major global public health burden. As an immune-privileged organ, the liver harbors a unique and intricate immune microenvironment, which plays a dual role in pathogen clearance and chronicity. Kupffer cells (KCs), the primary resident macrophages in the liver, constitute the first line of defense in liver innate immunity and play complex and important roles in pathogen recognition, phagocytosis, and the regulation of liver inflammation and immune responses. The compl

Targeting Kupffer Cell Enolase 1 Attenuates Liver Inflammation and Injury in Hemorrhagic Shock.

Hemorrhagic shock (HS) is a type of hypovolemic shock and is a leading cause of mortality worldwide. Enolase 1 (ENO1), a key enzyme in glycolysis, has been implicated in the pathogenesis of inflammatory disorders. We hypothesize that Kupffer cell (KC) ENO1 contributes to liver inflammation and that inhibiting ENO1 with ENOblock protects the liver from HS-induced injury. HS was induced in mice by lowering mean arterial pressure to 25 mmHg for 90 min, followed by fluid resuscitation. Twenty-four hours later, KCs were isolated. To mimic HS in vitro, KCs were isolated from healthy mice and exposed

A novel Kupffer cell-targeting nanoparticle system to Mitigate alcohol-associated liver disease.

Alcohol-associated liver disease (ALD) is a major cause of cirrhosis-related deaths worldwide. Due to its asymptomatic progression, early diagnosis and treatment are challenging, often resulting in severe complications. To address challenges associated with conventional treatments, including non-specificity and toxicity, we developed a poly lactic-co-glycolic acid nanoparticle (PLGA NP)-based drug delivery system that specifically targets Kupffer cells (KCs), which are major drivers of chronic liver disease progression. The PLGA NPs were loaded with the anti-inflammatory agent dexamethasone, a

Polystyrene microplastics exposure aggravates acute liver injury by promoting Kupffer cell pyroptosis.

OBJECTIVE: To investigate the long-term effects of polystyrene (PS) exposure on acute liver injury. METHODS: The carbon tetrachloride-induced acute injury mouse model was subjected to long-term PS exposure. Pyroptosis was inhibited by knocking out Gsdmd in mice or treating with the Gsdmd inhibitor necrosulfonamide (NSA) to evaluate the effect of PS on liver injury. Kupffer cells were used as a cellular model to examine the effects of PS on cell pyroptosis, lactate dehydrogenase release rate, structural integrity (propidium iodide staining), and inflammatory factor levels. RESULTS: In mice, P

The surface carbohydrates of the Echinococcus granulosus larva interact selectively with the rodent Kupffer cell receptor.

The larvae of the cestodes belonging to the genus Echinococcus dwell primarily in mammalian liver. They are protected by the laminated layer (LL), an acellular mucin-based structure. The glycans decorating these mucins constitute the overwhelming majority of molecules exposed by these larvae to their hosts. However, their decoding by host innate immunity has not been studied. Out of 36 mammalian innate receptors with carbohydrate-binding domains, expressed as Fc fusions, only the mouse Kupffer cell receptor (KCR; CLEC4F) bound significantly to the Echinococcus granulosus LL mucins. The recepto

Kupffer cell blockade improves the endotoxin-induced microcirculatory inflammatory response in obstructive jaundice.

Cholestasis predisposes to hypersensitivity to LPS, leading to potential septic complications. We set out to characterize the involvement of Kupffer cell (KC) activation in the hepatic microcirculatory and structural consequences of obstructive jaundice in the presence and absence of acute endotoxemia. The hepatic microcirculatory consequences of 3-day extrahepatic bile duct ligation (BDL) were assessed in rats. The contributions of changes in hepatic perfusion, leukocyte influx, and proinflammatory cytokine release to the development of hepatic structural damage were also determined. Furtherm

Modeling inflammation-drug interactions in vitro: a rat Kupffer cell-hepatocyte coculture system.

Xenobiotic-inflammation interactions lead to hepatotoxicity in vivo. Selected xenobiotic agents (acetaminophen, APAP; chlorpromazine, CPZ; allyl alcohol, AlOH; monocrotaline, MCT) for which this occurs were evaluated for ability to elicit the release of Kupffer cell (KC)-derived inflammatory mediators and to modulate lipopolysaccharide (LPS)-stimulated release of these mediators. Using KCs and hepatocytes (HPCs) isolated from rat, KC/HPC cocultures were treated with either LPS, xenobiotic, vehicle or a combination. Six hours later, the release of inflammatory mediators was assessed. LPS alone

Neutrophil-Kupffer cell interaction: a critical component of host defenses to systemic bacterial infections.

Most bacteria that enter the bloodstream are taken up and eliminated within the liver. The specific mechanisms that underlie the role of the liver in the resolution of systemic bacterial infections remain to be determined. The vast majority of studies undertaken to date have focused on the function of resident tissue macrophages (Kupffer cells) that line the liver sinusoids. Indeed, it is often reported that Kupffer cells ingest and kill the bulk of organisms taken up by the liver. Recent studies indicate, however, that phagocytosis by Kupffer cells is not the principal mechanism by which orga

Dual role of Kupffer cell activation and endothelial cell damage in reperfusion injury to livers stored for transplantation surgery.

In rat models of liver preservation, the primary event leading to liver graft failure after cold storage is a reperfusion injury causing damage to sinusoidal endothelial cells and activation of Kupffer cells (KC). After storage for longer than 16 h in University of Wisconsin solution, reperfusion induces rapid endothelial cell killing. Kupffer cell activation also occurs as indicated by cell surface ruffling, degranulation, release of hydrolytic enzymes, generation of oxygen radicals, and increased phagocytosis. Down-regulation of KC activity with nisoldipine or pentoxifylline improves graft s

Clearance of pegylated interferon by Kupffer cells limits NK cell activation and therapy response of patients with HBV infection.

Pegylated interferon-α (PEG-IFN-α), where IFN-α is attached to polyethylene glycol (PEG), is an approved treatment for chronic hepatitis B virus (HBV) infection, a disease that causes liver-related morbidity and mortality in 257 million people worldwide. It is unknown why only a minority of patients respond to PEG-IFN-α. Using sequential blood samples and liver biopsies of patients with chronic HBV infection before, during, and after PEG-IFN-α treatment, we find that patients with early natural killer (NK) cell activation after PEG-IFN-α injection experienced greater liver inflammation, lysis

Inhibition of 5-lipoxygenase induces cell growth arrest and apoptosis in rat Kupffer cells: implications for liver fibrosis.

The existence of an increased number of Kupffer cells is recognized as critical in the initiation of the inflammatory cascade leading to liver fibrosis. Because 5-lipoxygenase (5-LO) is a key regulator of cell growth and survival, in the current investigation we assessed whether inhibition of the 5-LO pathway would reduce the excessive number of Kupffer cells and attenuate inflammation and fibrosis in experimental liver disease. Kupffer cells were the only liver cell type endowed with a metabolically active 5-LO pathway (i.e., expressed mRNAs for 5-LO, 5-LO-activating protein [FLAP], and leuko

Pancreatic Elastase Affects Liver Injury by Activating Proinflammatory Cytokines in Kupffer Cells via the JAK2/STAT3 Signaling Pathway.

INTRODUCTION: This study aimed to investigate the role of JAK2 (Janus kinase 2)/STAT3 (signal transducer and activator of transcription 3) signaling in liver injury during severe acute pancreatitis (SAP), focusing on pancreatic elastase- and lipopolysaccharide (LPS)-induced Kupffer cell (KC) activation. METHODS: A rat SAP model was established via retrograde taurocholic acid infusion into the biliopancreatic duct. Inflammatory cytokine levels and JAK2/STAT3 pathway activity were quantified in liver tissues. KCs were treated with elastase/LPS ± AG490 (JAK2 inhibitor). Proinflammatory cytokines

ZFP36L1-mediated PRDM1 mRNA degradation inhibits inflammatory activation of Kupffer cells and ameliorates DCD liver ischemic-reperfusion injury.

BACKGROUND: Ischemia-reperfusion injury (IRI) represents a significant and unavoidable clinical challenge in donation after cardiac death (DCD) liver transplantation. Excessive sterile inflammation plays a critical role in the pathogenesis of liver IRI. This study, guided by bioinformatics insights, investigates the function of ZFP36 ring finger protein like 1 (ZFP36L1) in the activation of Kupffer cells (KCs) in IRI in DCD liver transplantation. METHODS: A rat DCD liver transplantation model and a lipopolysaccharide (LPS)-induced KCs inflammation model were established. Models were pretreate

Mannose-Conjugated Cholesterol Containing Lipid Nanoparticles for Active Targeted mRNA Delivery to Liver Sinusoidal Endothelial and Kupffer Cells.

Lipid nanoparticle (LNP) delivery of mRNA to specific cell types is a necessary task for the development of safe and effective medicines. LNP delivery to the liver is largely driven by the binding of serum ApoE to the LNP surface, followed by subsequent uptake in LDL receptor (LDL-R)-expressing hepatocytes, thereby reducing their utility in nonhepatocyte liver diseases. Herein, we developed an active targeting strategy to overcome this limitation by incorporating mannose-conjugated cholesterol into LNPs. Since cholesterol comprises about half of all molecules in LNPs, we reasoned that it could

Rubicon promotes the M2 polarization of Kupffer cells via LC3-associated phagocytosis-mediated clearance to improve liver transplantation.

BACKGROUND: Acute rejection (AR) after liver transplantation (LT) is closely related to the survival of patients after surgery. Enhancement of the ability of Kupffer cells (KCs) to eliminate apoptotic cells can effectively alleviate AR. METHODS: Rubicon lentivirus (LV) and Rubicon small interfering RNA (siRNA) were transfected into KCs extracted from the liver tissue of mice. Primary KCs were extracted and cocultured with zymosan and apoptotic T lymphocytes. The levels of CD86, CD163, IL-10, TNF-α, TGF-β, JAK1, STAT6, AKT1, mTOR and peroxisome proliferator-activated receptor-γ (PPARγ) were as

Gondoic acid alleviates LPS‑induced Kupffer cells inflammation by inhibiting ROS production and PKCθ/ERK/STAT3 signaling pathway.

Kupffer cells (KCs) is the main macrophage in liver, and its inflammation is related to liver diseases. It has been shown that inflammatory macrophages are accompanied by changes in monounsaturated fatty acid (MUFA) content. However, the effect of gondoic acid (GA) on inflammation and its underlying mechanism have not been described. In the current study, we demonstrated that GA significantly inhibited the expression of pro-inflammatory factors in LPS-exposed KCs. Further research found that GA reduced lipopolysaccharide (LPS)-stimulated reactive oxygen species (ROS) levels and enhanced the ex

Kupffer cells regulate liver recovery through induction of chemokine receptor CXCR2 on hepatocytes after acetaminophen overdose in mice.

Acetaminophen (APAP) is a widely used analgesic, but also a main cause of acute liver injury in the United States and many western countries. APAP hepatotoxicity is associated with a sterile inflammatory response as shown by the infiltration of neutrophils and monocytes. While the contribution of the immune cells to promote liver repair have been demonstrated, the direct interactions between macrophages or neutrophils with hepatocytes to help facilitate hepatocyte proliferation and tissue repair remain unclear. The purpose of this study was to investigate the relationship between resident macr

Inactivation of Kupffer Cells by Selenizing Astragalus Polysaccharides Prevents CCl4-Induced Hepatocellular Necrosis in the Male Wistar Rat.

Selenizing astragalus polysaccharides-3 (sAPS3) was prepared by nitric acid-sodium selenite method. The effects of sAPS3 on carbon tetrachloride (CCl4) induced hepatocellular necrosis, and its underlying mechanisms were studied in male Wistar rats. Hepatic damage was induced by intraperitoneal injection of CCl4 twice a week, for 3 weeks. Meanwhile, the rats in addition to CCl4 were also exposed to sodium selenite (SS), astragalus polysaccharides (APS), SS + APS or sAPS3, in parallel by oral gavage once a day for 3 weeks. At the end of 3 weeks, blood and liver tissue were taken. Serum was colle
