A Reusable Capillary Flow-Driven Microfluidic System for Abscisic Acid Detection Using a Competitive Immunoassay.

Point-of-care (PoC) devices offer a promising solution for fast, portable, and easy-to-use diagnostics. These characteristics are particularly relevant in agrifood fields like viticulture where the early detection of plant stresses is crucial to crop yield. Microfluidics, with its low reagent volume requirements, is well-suited for such applications. Self-driven microfluidic devices, which rely on capillary forces for fluid motion, offer an attractive alternative by eliminating the need for external pumps and complex fluid control systems. However, traditional microfluidic prototyping material

μLAMP-LFA: A nucleic acid analysis platform for the field diagnosis of pathogens in aquatic animals.

BACKGROUND: Aquatic animal diseases cause significant economic losses in aquaculture. Effective pathogen diagnostics enable targeted treatment measures. Traditional detection methods primarily rely on polymerase chain reaction. However, due to the bulky and expensive nature of the equipment, on-site testing is impractical. Furthermore, the process requires low-temperature preservation, transportation to centralized laboratories, and analysis by trained personnel. The combination of loop-mediated isothermal amplification (LAMP) and lateral flow assay (LFA) has promoted on-site detection with gr

A urine detection chip for the analysis of urinary cells and extracellular vesicles for bladder cancer screening.

Bladder cancer is the most prevalent malignant neoplasm of the urinary system. However, current techniques for bladder cancer diagnosis and prognosis are associated with significant patient discomfort and potential risk of urinary tract infection. To address these issues, we propose a non-invasive urine detection chip for isolating and identifying bladder cancer-related cells and extracellular vesicles. This chip enables simultaneous capture of exfoliated cells and blood cells for analysis, followed by microparticle-based filtration to remove cells and cell debris before capturing and analyzin

A millifluidic esophagus on-a-chip model for evaluating a natural remedy-mediated mucosal repair in GERD.

Gastroesophageal reflux disease (GERD) is a common gastrointestinal disorder, characterized by the retrograde flow of gastric contents into the esophagus, causing symptoms such as heartburn and regurgitation. Although many patients with symptomatic GERD show no visible mucosal injury, impairment of its integrity at microscopic level is considered a relevant factor contributing to symptom generation and inflammation. In this scenario, protection of esophageal mucosa by topical agents has become a potential therapeutic approach. We investigated the protective and regenerative effects of a new na

Joint-on-a-Chip: The Future of Preclinical Modelling of Joint Diseases.

Joint diseases, including osteoarthritis (OA) and rheumatoid arthritis (RA), are leading causes of disability worldwide and pose a major socioeconomic burden. Effective treatments remain limited, partly due to difficulties in understanding and replicating complex joint structures as well as the dynamic interactions among multiple tissues and immune components. Traditional in vitro and animal models are restricted in their ability to accurately simulate the human joint microenvironment, limiting their predictive value for drug discovery and therapeutic development. Joint-on-a-Chip (JOC) is emer

Human Cervix Chip: A Preclinical Model for Studying the Role of the Cervical Mucosa and Microbiome in Female Reproductive Health.

Advancements in women's reproductive health have been hindered by insufficient knowledge and the underrepresentation of women in research, leading to symptom-focused care with poor outcomes. Modeling female reproductive biology and disease pathophysiology has been challenging due to the complexity and dynamic nature of the female organs. Here, we briefly review recent advancements made with a new in vitro microfluidic organ-on-a-chip model of the human cervix (Cervix Chip) that faithfully mimics key features of the cervix, including mucus production and physiological responses to hormonal, env

Translation of a Human-Based Malaria-on-a-Chip Phenotypic Disease Model for In Vivo Applications.

In 2023 malaria claimed ≈600000 lives, with 90% of those deaths attributed to the Plasmodium falciparum parasite. This resurgence in mortality emphasizes the necessity of adopting alternative models to accelerate therapeutic development. The Malaria-on-a-Chip model used here incorporated human liver, spleen, and endothelium with P. falciparum-infected blood, and was maintained for 7 days using serum-free medium. This model sustained all stages of the intraerythrocytic life cycle and allowed for organ-organ interaction, providing advantageous preclinical insight into malaria pathophysiology. Ch

Sample-sparing multiplexed antibody Fc biomarker discovery using a reconfigurable integrated microfluidic platform.

Control of endemic infectious diseases is often impeded by the lack of sensitive and specific yet easy-to-obtain biomarkers. Antibody fragment crystallizable (Fc) regions, such as Fc glycosylation, which are modulated in a pathogen-specific and disease-state-specific manner have emerged as potential such biomarkers. However current methods to perform large-scale antigen-specific antibody Fc feature screening for biomarker discovery often require too much sample volume, cost and expertise to be realistically realizable in many disease contexts. Here we present a simple, flexible and reconfigura

Construction of in vitro liver-on-a-chip models and application progress.

The liver is the largest internal organ of the human body. It has a complex structure and function and plays a vital role in drug metabolism. In recent decades, extensive research has aimed to develop in vitro models that can simulate liver function to demonstrate changes in the physiological and pathological environment of the liver. Animal models and in vitro cell models are common, but the data obtained from animal models lack relevance when applied to humans, while cell models have limited predictive ability for metabolism and toxicity in humans. Recent advancements in tissue engineering,

Rapid automated production of tubular 3D intestine-on-a-chip with diverse cell types using coaxial bioprinting.

Despite considerable animal sacrifices and investments, drug development often falters in clinical trials due to species differences. To address this issue, specific in vitro models, such as organ-on-a-chip technology using human cells in microfluidic devices, are recognized as promising alternatives. Among the various organs, the human small intestine plays a pivotal role in drug development, particularly in the assessment of digestion and nutrient absorption. However, current intestine-on-a-chip devices struggle to accurately replicate the complex 3D tubular structures of the human small int

Modeling Healthy and Dysbiotic Vaginal Microenvironments in a Human Vagina-on-a-Chip.

Women's health, and particularly diseases of the female reproductive tract (FRT), have not received the attention they deserve, even though an unhealthy reproductive system may lead to life-threatening diseases, infertility, or adverse outcomes during pregnancy. One barrier in the field is that there has been a dearth of preclinical, experimental models that faithfully mimic the physiology and pathophysiology of the FRT. Current in vitro and animal models do not fully recapitulate the hormonal changes, microaerobic conditions, and interactions with the vaginal microbiome. The advent of Organ-o

Digital recombinase polymerase amplification chip based on asymmetric contact angle composite interface.

BACKGROUND: Digital recombinase polymerase amplification (dRPA) is an effective tool for the absolute quantification of nucleic acids and the detection of rare mutations. Due to the high viscosity or other physical properties of the reagent, this can compromise the accuracy and reproducibility of detection results, which limits the broader adoption and practical application of this technology. In this study, we developed an asymmetric contact angle digital isothermal detection (ACA-DID) chip and optimized the ACA-DID chip structure to achieve rapid digital recombinase polymerase amplification.

Development of a microfluidic device to observe dynamic flow around the villi generated by deformation of small intestinal tissue.

The mucosal immune system in the small intestine is crucial for human health. For the immune response, mutual contact between gut bacteria and intestinal epithelial cells is important because there are unique epithelial cells specialized in gut bacteria sampling on the villi. The travel of gut bacteria to the villi is led by a complex dynamic flow in the small intestine. However, the complex dynamic flow around the villi has not yet been explored at a micro scale. In this study, we proposed a microfluidic device to observe the flow around the villi generated by the dynamic deformation of small

Self-assembled innervated vasculature-on-a-chip to study nociception.

Nociceptor sensory neurons play a key role in eliciting pain. An active crosstalk between nociceptor neurons and the vascular system at the molecular and cellular level is required to sense and respond to noxious stimuli. Besides nociception, interaction between nociceptor neurons and vasculature also contributes to neurogenesis and angiogenesis. In vitromodels of innervated vasculature can greatly help delineate these roles while facilitating disease modeling and drug screening. Herein, we report the development of a microfluidic-assisted tissue model of nociception in the presence of microvas

Reaching the Tumor: Mobility of Polymeric Micelles Inside an In Vitro Tumor-on-a-Chip Model with Dual ECM.

Degradable polymeric micelles are promising drug delivery systems due to their hydrophobic core and responsive design. When applying micellar nanocarriers for tumor delivery, one of the bottlenecks encountered in vivo is the tumor tissue barrier: crossing the dense mesh of cells and the extracellular matrix (ECM). Sometimes overlooked, the extracellular matrix can trap nanoformulations based on charge, size, and hydrophobicity. Here, we used a simple design of a microfluidic chip with two types of ECM and MCF7 spheroids to allow "high-throughput" screening of the interactions between biologica
