Facile synthesis of laccase mimic Cu/H3BTC MOF for efficient dye degradation and detection of phenolic pollutants.

Herein, we report an effectual method for designing a novel form of nanozyme laccase mimic namely Cu/H3BTC, using copper ions and 1,3,5-benzene tricarboxylic acid (1,3,5-H3BTC). This Cu-based metal-organic framework (MOF) was synthesized through a simple procedure of mixing of two usual reagents at room temperature. Amido Black 10B (AB-10B) was chosen as a model dye for degradation consequences. Results showed that Cu/H3BTC MOF revealed significantly higher catalytic efficacy under certain conditions like high pH, extreme temperature and high salt conditions and it has long-term storage stabil

Enhancing Electricity Generation Using a Laccase-Based Microbial Fuel Cell with Yeast Galactomyces reessii on the Cathode.

The fungi associated with termites secrete enzymes such as laccase (multi-copper oxidase) that can degrade extracellular wood matrix. Laccase uses molecular oxygen as an electron acceptor to catalyze the degradation of organic compounds. Owing to its ability to transfer electrons from the cathodic electrode to molecular oxygen, laccase has the potential to be a biocatalyst on the surface of the cathodic electrode of a microbial fuel cell (MFC). In this study, a two-chamber MFC using the laccase-producing fungus Galactomyces reessii was investigated. The fungus cultured on coconut coir was plac

Laccase-catalyzed removal of the antimicrobials chlorophene and dichlorophen from water: Reaction kinetics, pathway and toxicity evaluation.

As active agents in cleaning and disinfecting products, antimicrobials have been widely spread in the environment and have drawn extensive attention as potential threats to the ecological system and human health. In this study, the laccase-catalyzed removal of two emerging antimicrobials, chlorophene (CP) and dichlorophen (DCP), was investigated under simulated environmental conditions. Intrinsic reaction kinetics showed that the removal of CP and DCP followed second-order reaction kinetics, first-order with respect to both the enzyme and the substrate concentration. It was also found that ful

Immobilization of laccase on gold, silver and indium tin oxide by zirconium-phosphonate-carboxylate (ZPC) coordination chemistry.

In this paper we present a simple method allowing for stable laccase immobilization on various conducting surfaces that retains the activity of the enzyme. The strategy for laccase immobilization presented in this paper relies on Zr(4+) ion coordination chemistry that involves -COO- terminal groups present on the protein. Using a host of techniques, including surface plasmon resonance (SPR), quartz crystal microbalance (QCM) gravimetry, atomic force microscopy (AFM), surface enhanced Raman scattering (SERS), resonance Raman scattering (RR) and electrochemical techniques, we show that laccase b

Enhanced Inhibition of Trametes versicolor by Structurally Modified Medicarpin: In Vitro Evaluation and In Silico Insights into Laccase Binding.

Medicarpin, a natural pterocarpan phytoalexin, contributes to tree defense against microbial decay, particularly from the aggressive white-rot fungus Trametes versicolor, an ASTM standard for wood durability testing. To improve upon the inhibitory effect of medicarpin against this fungus (150 mg/L), eleven derivatives were synthesized and evaluated. The acetylated analog demonstrated superior activity, achieving complete growth inhibition at 100 mg/L. To establish a structure-activity relationship, molecular docking was performed on the copper cluster on fungal laccase, the primary oxidative e

A novel colorimetric method for nitrite detection in food based on histidine-manganese nanozyme with laccase-mimicking activity.

BACKGROUND: Excessive nitrite in food poses a serious threat to public health. This compound not only induces methemoglobinemia by interfering with oxygen transport in the blood but also serves as a precursor to carcinogenic N-nitrosamines, posing long-term cancer risks. Thus, the development of rapid and highly sensitive methods for nitrite detection is crucial for ensuring food safety. Laccase mimics, which have been widely used for the degradation and detection of phenolic compounds, have recently emerged as promising tools for sensing various food contaminants. However, their application f

Theoretical Insights into the Catalytic Oxidation of Phenols and Arylamines by Laccases via the Proton-Coupled Electron Transfer Mechanism.

Laccases play a vital role in the degradation of toxic phenolic and aromatic amine compounds, generating considerable attention in ecological pollution remediation. However, the distinct mechanism of the laccase-catalyzed oxidation of phenols and arylamines remains unclear. Here, we examined the catalytic oxidation mechanisms of phenols and arylamines by Trametes versicolor (TvL) and Melanocarpus albomyces (MaL) laccases using molecular docking, quantum mechanics (QM), and QM/molecular mechanics (QM/MM) calculations. We docked four phenolic substrates, including 1,2-benzenediol, 2-propenylphen

Biotransformation of bisphenol A in vivo and in vitro by laccase-producing Trametes hirsuta La-7: Kinetics, products, and mechanisms.

Bisphenol A (BPA) is a ubiquitous endocrine disruptor that poses adverse human health risks. Herein, biotransformation kinetics, products, and mechanisms of BPA undergoing a laccase-producing Trametes hirsuta La-7 metabolism were for the first time reported. Strain La-7 could completely biotransform ≤0.5 mmol·L-1 BPA within 6 d in vivo. Notably, its extracellular crude laccase solution (ECLS) and intracellular homogenized mycelium (HM) only required 6 h to convert 85.71% and 84.24% of 0.5 mmol·L-1 BPA in vitro, respectively. The removal of BPA was noticeably hampered by adding a cytochrome P-4

Biodegradation of sulfonamide antibiotics through the heterologous expression of laccases from bacteria and investigation of their potential degradation pathways.

In this study, seven laccase genes from different bacteria were linked with the signal peptides PelB, Lpp or Ompa for heterologous expression in E. coli. The recombinant strains were applied for the removal of sulfadiazine (SDZ), sulfamethazine (SMZ), and sulfamethoxazole (SMX). The results obtained for different signal peptides did not provide insights into the removal mechanism. The removal ratios of SDZ, SMZ, and SMX obtained with the recombinant strain 6#P at 60 h were around 92.0%, 89.0%, and 88.0%, respectively. The degradation pathways of sulfonamides have been proposed, including SO2 e

Laccase isoform diversity on basal medium in Cyathus bulleri and role in decolorization/detoxification of textile dyes and effluent.

Laccases (EC 1.10.3.2) are multi-copper oxidases that can degrade several xenobiotics, including textile dyes. Present study investigated the nature of laccase isoforms induced by 2,6-dimethylaniline in Cyathus bulleri cultivated on basal salt medium. Two isoforms, LacI and LacII were identified and purified by a combination of ultrafiltration and ion-exchange chromatography. The MS spectrum of the two proteins displayed a number of non-identical and identical molecular peaks (m/z), and, the latter were mapped to protein originating from the previously reported Laccase (Lcc) 1 gene. The LacI i

Ecotoxicological test based on inhibition of fungal laccase activity: Application to agrochemicals and the monitoring of pesticide degradation processes.

Ecotoxicological evaluations require the use of assays with several bioindicators from different trophic levels. Only a few ecotoxicological tests using fungi have been developed, reason why, detection of adverse effects from compounds that exert fungicide action may be overlooked. This work developed a toxicity test based on the inhibition of laccase enzymatic activity in the fungus Trametes versicolor. The test was applied to several fungicides and succeeded to determine inhibition values (half maximum effective concentration, EC50) for most of them (flusilazole, imazalil, pyrimethanil, tetr

Isolation, characterization, and optimization of extracellular laccase-producing bacteria from dye-contaminated textile wastewater for dye decolonization.

Textile industries are important in both the global economy and our daily lives. They are, however, simultaneously becoming one of the main sources of environmental pollution in the world. It needs a greater amount of water for wet textile operations such as dyeing, and as a result, produces significant dosages of wastewater. Textile wastewater is highly colorful and contains dyes that are highly harmful to living creatures, and is discharged to the environment every day. The objective of the present study was to isolate and characterize extracellular laccase-producing bacteria from textile dy

A novel electrochemical sensor based on process-formed laccase-like catalyst to degrade polyhydroquinone for tumor marker.

Methods to improve the sensitivity of electrochemical sensors based on catalytic reactions generally require adscititious or pre-modified catalysts, which make the sensitive detection of sensors extremely challenging. This is because the activity of the catalyst is susceptible to the storage and modification process, such as aggregation during storage or loss of active sites during multi-step modification, which impairs the performance of the sensor. To solve this thorny issue, a novel electrochemical sensor based on a process-formed laccase-like catalyst was constructed for sensitive detectio

Carbon disulfide mediated self-assembly of Laccase and iron oxide nanoparticles on gold surfaces for biosensing applications.

A simple one-step methodology was explored to prepare enzyme-modified nanostructured electrodes for the development of biosensing interfaces. Magnetite type nanoparticles conjugated with Laccase were immobilized on gold surfaces. This approach relies on the reaction between carbon disulfide and amine groups of biomolecules to form dithiocarbamate (DTC) moieties, as well as on the strong affinity between sulfur species and metals. Special emphasis was given to demonstrate DTC formation in aqueous solution and further attachment to iron oxide nanoparticles and to gold electrodes. UV-visible spec
