Teaching Contemporary Lactate Physiology: Why It Is Time to Retire "Lactic Acid".

The terminology used to teach physiology should reflect contemporary scientific understanding. Although decades of research have fundamentally transformed our understanding of lactate biology, the term lactic acid remains widely used throughout physiology education, textbooks, coaching education, certification programs, and public communication. Under physiological conditions, however, lactate-not lactic acid-is the predominant molecular species, and modern evidence recognizes lactate as an oxidative fuel, signaling molecule, and regulator of metabolism rather than a metabolic waste product. Interface Control of Poly(L-lactic Acid)/Hydroxyapatite Composites by Using Bisphosphonate.

Poly-(L-lactic acid)/hydroxyapatite (PLLA/HAp) composites have attracted significant attention as environmentally friendly tough materials due to their excellent biocompatibility and mechanical strength. However, the poor organic-inorganic interfacial adhesion between PLLA and HAp has been a significant challenge, often resulting in the premature elution of PLLA and a reduction in the mechanical integrity. Here, we prepared PLLA/HAp composites with a controllable interface by using etidronic acid, a bisphosphonate compound, as an interface modifier for HAp. Etidronic acid modification of HAp i

Fructophilic lactic acid bacteria as a window into multi-scale convergent evolution.

Fructophilic lactic acid bacteria (FLAB) are a group of lactic acid bacteria with unique growth characteristics, that is, poor growth on glucose. Their growth is enhanced in the presence of fructose or external electron acceptors. These organisms inhabit fructose-rich environments such as flowers, fruits, and pollinating insects, particularly honey bees. Apilactobacillus spp. and Fructobacillus spp. are representatives of FLAB, although they belong to phylogenetically distant clades. These organisms commonly possess markedly small genomes with a low number of coding DNA sequences. Furthermore,

Lactic acid promotes metastasis of papillary thyroid carcinoma by enhancing CPT1A lactylation.

Papillary thyroid carcinoma (PTC), the most prevalent thyroid malignancy, exhibits aggressive behavior in subsets with metastasis. Despite advances in risk stratification, biomarkers predicting metastatic potential remain limited. Here, we identify lactate as a critical driver of PTC metastasis through lactylation of carnitine palmitoyltransferase 1 A (CPT1A), the rate-limiting enzyme in fatty acid β-oxidation (FAO). Multi-omics profiling of 27 paired PTC tissues revealed elevated lactate levels and FAO activation, corroborated by TCGA data. Functional assays demonstrated that exogenous lactat

A Stereospecific Lactic Acid Exclusion Biosensor for Grass Silage Fed Green Biorefinery.

Grass silage fed green biorefineries require high quality grass silage leachates to produce high quality products. Real-time, cost-effective methods to monitor key leachate analytes are needed for small-scale decentralised systems. In this study, Escherichia coli mutants, designated JSP0090 and JSP0094, were created to quantify d- and l-lactic acid concentrations in grass leachate samples using an oxygen probe. The genes encoding either d- or l-lactate dehydrogenase were expressed in an exclusion biosensor strain (JSK0115). This strain required successive gene deletions to prevent interference

Development of metformin-encapsulated poly(lactic-co-glycolic acid)-polyethylene glycol nanoparticles.

BACKGROUND: Thyroid cancer is a common endocrine malignancy. Conventional treatments, such as surgery, radioiodine therapy, radiotherapy, and chemotherapy, often result in significant side effects and toxicity. Nanoparticle-based drug delivery systems have emerged as a promising strategy to overcome these challenges. In this study, we investigated the effects of free metformin, a PLGA-PEG copolymer, and PLGA-PEG-MET nanoparticles on FTC-133 thyroid cancer cells. METHODS AND RESULTS: PLGA-PEG-MET nanoparticles were synthesized using the double-emulsion method and characterized using FT-IR and

A rare case of lethal lactic acid poisoning: an autopsy case report.

More than 700,000 people die due to suicide every year, and ingestion of pesticides, hanging, and firearms are among the most commonly used suicide methods globally. Although corrosives are among the least preferred agents for suicide due to the extreme pain caused and the possibility of prolonged suffering instead of immediate death, cases of ingestion continue to be observed. Commonly ingested corrosive substances include acids such as hydrochloric and sulfuric acid, alkalis such as sodium and potassium hydroxide, heavy metal salts, formalin, and iodine tincture. However, previous literature

Bioremoval of lead (pb) salts from synbiotic milk by lactic acid bacteria.

The present study investigated the effects of five species of indigenous probiotic bacteria (Lactobacillus sp. and Bifidobacterium sp.) and prebiotic inulin on the decontamination of two Pb2+ salts (lead nitrate and lead acetate) in food models (low-fat, semi-fat, and high-fat milk). The average of the lowest and highest pH values was related to samples containing L. paracasei and L. acidophilus bacteria, as determined by pH change results. In most instances, adding inulin accelerated the pH-decreasing process, but the results were not particularly significant (p < 0.05). All probiotic strains

Production of L-lactic acid from methanol by engineered yeast Pichia pastoris.

Lactic acid (LA) serves as a widely used platform compound and has received significant attention as a raw material for synthesis of biodegradable polylactic acid. Currently, LA is mainly produced through microbial fermentation, but its high costs undermine its competitive advantage against other materials, necessitating the development of novel production routes. Methanol bioconversion represents an emerging low-carbon circular economy, where LA could become an outstanding representative product. This study successfully established an efficient methanol-based LA synthesis route in Pichia past

Intratumoral lactic acid neutralization strategy for boosting chemoimmunotherapy using liposomal sodium bicarbonate.

Glycolysis-related lactic acid overproduction creates an "ion-trapping" barrier and immunosuppressive tumor microenvironment that compromise effective intratumoral drug delivery and therapy. Therefore, normalization of tumor microenvironment via lactic acid neutralization can be a promising avenue for overcoming this therapeutic hurdle. In this study, the flexible liposomes loaded with sodium bicarbonate (NaHCO3@Flip) were used as a nano-adjuvant to boost chemoimmunotherapy. Their effects on assisting DOXIL and anti-programmed cell death protein 1 (PD-1) therapy were investigated. NaHCO3@Flip

Mathematical modeling of vancomycin release from Poly-L-Lactic Acid-Coated implants.

This study aimed to develop a mathematical model to predict the release profile and antibacterial efficacy of a vancomycin delivery system integrated with poly(L-lactic acid)-coated bone implants specifically designed for bone plates. Using Fickian diffusion principles within an ANSYS-CFX computational fluid dynamic model, we validated the model against our in vitro vancomycin release and agar diffusion studies, as well as previously published in vivo data, confirming the reliability of the model. The model predictions demonstrated the effectiveness of the system in inhibiting bacterial growth

Rat microbial biogeography and age-dependent lactic acid bacteria in healthy lungs.

The laboratory rat emerges as a useful tool for studying the interaction between the host and its microbiome. To advance principles relevant to the human microbiome, we systematically investigated and defined a multi-tissue full lifespan microbial biogeography for healthy Fischer 344 rats. Microbial community profiling data was extracted and integrated with host transcriptomic data from the Sequencing Quality Control (SEQC) consortium. Unsupervised machine learning, Spearman's correlation, taxonomic diversity, and abundance analyses were performed to determine and characterize the rat microbia

Lactic acid bacteria secreted proteins as potential Listeria monocytogenes quorum sensing inhibitors.

Listeria monocytogenes is an important human and animal pathogen able to cause an infection named listeriosis and is mainly transmitted through contaminated food. Among its virulence traits, the ability to form biofilms and to survive in harsh environments stand out and lead to the persistence of L. monocytogenes for long periods in food processing environments. Virulence and biofilm formation are phenotypes regulated by quorum sensing (QS) and, therefore, the control of L. monocytogenes through an anti-QS strategy is promising. This study aimed to identify, by in silico approaches, proteins s

Preparation of Hydrogarnet/Poly(Lactic Acid) Composite Adsorbents for Humic Substance Removal.

Humic substances are constituents of organic matter that require removal from water environments because of their adverse ecological and sanitation effects. A mixture of hydrogarnet and poly(lactic acid) dissolved in chloroform was electrospun to prepare a composite as a adsorbent for humic substance removal. Here, humic acid was used as the model substance for evaluating the adsorbent's water remediation efficiency. Despite the hydrogarnet particles being embedded in its poly(lactic acid) fibers, the composites demonstrated a higher humic acid removal ability than the pure poly(lactic acid) s

Lactic Acid Bacteria: A Promising Tool for Menopausal Health Management in Women.

Menopause is a period during which women undergo dramatic hormonal changes. These changes lead to physical and mental discomfort, are greatly afflictive, and critically affect women's lives. However, the current safe and effective management measures for women undergoing menopause are insufficient. Several probiotic functions of lactic acid bacteria (LAB) have been recognized, including alleviation of lactose intolerance, protection of digestive tract health, activation of the immune system, protection against infections, improvement of nutrient uptake, and improvement of the microbiota. Mild Lactic Acid Stress Causes Strain-Dependent Reduction in SEC Protein Levels.

Staphylococcal enterotoxin C (SEC) is a major cause of staphylococcal food poisoning in humans and plays a role in bovine mastitis. Staphylococcus aureus (S. aureus) benefits from a competitive growth advantage under stress conditions encountered in foods such as a low pH. Therefore, understanding the role of stressors such as lactic acid on SEC production is of pivotal relevance to food safety. However, stress-dependent cues and their effects on enterotoxin expression are still poorly understood. In this study, we used human and animal strains harboring different SEC variants in order to eval

Probiotic potential of lactic acid bacteria obtained from fermented curly kale juice.

The aim of the paper was to analyse changes in lactic acid bacteria (LAB) populations during spontaneous fermentation of green curly kale juice (Brasicca oleracea L. var. acephala L.) and to determine the probiotic potential of LAB isolates. The analyses revealed that changes in LAB populations were specific for spontaneously fermented vegetable juices. The initial microbiota, composed mostly of Leuconostoc mesenteroides bacteria, was gradually replaced by Lactobacillus species, mainly Lactobacillus plantarum, Lactobacillus sakei, and Lactobacillus coryniformis. Screening tests for the antimic

Disease managing capacities and mechanisms of host effects of lactic acid bacteria.

Consumption of lactic acid bacteria (LAB) has been suggested to confer health-promoting effects on the host. However, effects of LABs have been reported to be species- and strain-specific and the mechanisms involved are subjects of discussion. Here, the possible mechanisms by which LABs induce antipathogenic, gut barrier enhancing and immune modulating effects in consumers are reviewed. Specific strains for which it has been proven that health is improved by these mechanisms are discussed. However, most strains probably act via several or combinations of mechanisms depending on which effector
