Oral immunization with Lactococcus lactis secreting attenuated recombinant staphylococcal enterotoxin B induces a protective immune response in a murine model.

BACKGROUND: Staphylococcus aureus is unrestrictedly found in humans and in animal species that maintain thermal homeostasis. Inadequate cleaning of processing equipment or inappropriate handling can contaminate processed food and cause severe food poisoning. Staphylococcal enterotoxin B (SEB), a potent superantigenic exotoxin, is produced by 50% of clinical isolates of S. aureus and is associated with massive food poisoning and with the induction of toxic shock syndrome. RESULTS: A gene sequence encoding a recombinant SEB (rSEB), devoid of superantigenic activity, was successfully cloned and

Efficacy of Lactococcus lactis Strain Plasma in Patients with Mild COVID-19: A Multicenter, Double-Blinded, Randomized-Controlled Trial (PLATEAU Study).

INTRODUCTION: Coronavirus disease 2019 (COVID-19), caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), is still an ongoing public health threat. COVID-19 can be accompanied by prolonged symptoms, known as "long COVID", however, no pharmaceutical treatments are currently available for these symptoms. Lactococcus lactis strain Plasma (LC-Plasma; Lactococcus lactis subsp. lactis JCM 5805) directly activates human plasmacytoid dendritic cells (pDCs) and triggers antiviral immune responses. We hypothesized that LC-Plasma reduced SARS-CoV-2 viral load and eased symptoms in patien

Fermented camel milk by Lactococcus lactis subsp. cremoris attenuates erythrocytes oxidative stress-induced hematological and immunological damage in CCl4-intoxicated mice.

Fermented camel by Lactococcus lactis subsp. cremoris has been recently discovered to protect against the toxic effect of carbon tetrachloride (CCl4), but its beneficial effects in the presence of oxidative stress in the erythrocytes have not been established. In the present study, 28 mice were randomly divided into four groups: control group; CCl4 group: intoxicated by a single intraperitoneal injection (ip) of CCl4; group FCM-LLC + CCl4: pretreated with FCM-LLC daily during 14 days, and received a single dose of CCl4. FCM-LLC group received FCM-LLC alone. The occurrence of oxidative stress i

Euglena gracilis-derived β-1,3-glucan as a growth substrate for Lactococcus lactis-a snapshot of accompanying changes in transcriptomic and metabolomic profiles.

β-1,3-glucan from Euglena gracilis is an emerging microalgal prebiotic. However, its potential to reprogram the metabolic profile of lactic acid bacteria has not been reported previously. In the present study, the effects of insoluble β-1,3-glucan on growth dynamics, gene expression, and the extracellular metabolite profile of Lactococcus lactis DSM20175 were investigated using transcriptomic and untargeted metabolomic approaches. Real-time growth monitoring combined with flow cytometric cell enumeration showed that β-1,3-glucan supplementation significantly increased both growth rate and fina

Synbiotic Combination of Lactococcus lactis LB1022 and Fructo-Oligosaccharides Mitigates the Atopic March by Modulating the Microbiota-Gut-Skin-Lung Axis.

Dysregulated gut microbiota is increasingly recognized as a major contributor to allergic diseases and their progression. A key clinical manifestation of this progression is the atopic march, in which atopic dermatitis (AD) precedes the development of allergic airway disease. Although prebiotics and probiotics individually improve AD symptoms, their combined use as synbiotics, especially with regard to preventing the progression from cutaneous inflammation to airway hypersensitivity, has not been clearly established. In this study, we assessed the biological activity of a synbiotic composed of

[Construction of recombinant Lactococcus lactis strains expressing classical swine fever virus E2 antigen and evaluation of their immunogenicity in murine models].

Classical swine fever virus (CSFV) infects pigs primarily via the respiratory and digestive tracts and inflicts enormous economic losses on the global swine industry. To address the challenge of early CSFV invasion, this study developed an oral vaccine based on a Lactococcus lactis vector to induce sIgA-mediated mucosal immunity. This approach aims to provide robust early protection and a scalable, stress-free vaccination strategy for modern pig farming. In this study, we first assessed the colonization capacity of L. lactis NZ9000 in mice by in vivo luciferase imaging and fecal bacterial coun

Oral delivery of GLP-1 analogues by recombinant Lactococcus lactis restores pancreatic islet structure through intestinal mucosal absorption in diabetic mice.

BACKGROUND: The oral delivery of native glucagon like peptide-1 (GLP-1) using recombinant probiotics has shown hypoglycaemic effects and glucose tolerance improvement in diabetic mice. However, the pharmacological mechanisms remain incompletely understood. This study aimed to clarify whether GLP-1 analogues cross the intestinal mucosal barrier and exert systemic hypoglycaemic effects. METHODS: The recombinant Lactococcus lactis (L. lactis) was constructed for oral delivery of long-duration GLP-1 (LGLP) or exendin-4 (EX4). The hypoglycemic effects in vivo were investigated in db/db mice. The p

Lactococcus lactis and Bifidobacterium longum attenuate Clostridioides difficile- or Clostridium symbiosum-induced colitis and depression/anxiety-like behavior in male mice.

Clostridioides difficile causes severe colitis, which induces neuroinflammation and psychiatric disorder. In a preliminary study, we isolated Clostridium symbiosum from the stools of patients with ulcerative colitis. Therefore, we first examined whether oral infection with C. difficile or C. symbiosum could induce colitis and depression in male mice. Orally gavaged C. difficile or C. symbiosum caused diarrhea, bodyweight loss, depression/anxiety-like behavior, and tumor necrosis factor (TNF)-α and interleukin (IL)-6 overexpression in the colon and hippocampus in the pseudo-germ-free (PGF) and

Lactococcus lactis-HSP65 requires TLR2 to induce tolerogenic dendritic cells and confer protection against type 1 diabetes in a murine model.

Type 1 Diabetes (T1D) is characterized by the destruction of insulin-producing β cells, leading to hyperglycemia. New therapeutic strategies are crucial for preventing or managing the disease and its complications. Natural or recombinant probiotics, by modulating the gut microbiota and dictating the immunological tolerance, have been recently employed as tolerogenic immunotherapy for autoimmune diseases. Heat shock protein 65 (HSP65) exhibits regulatory and anti-inflammatory activity in autoimmune and chronic inflammatory conditions. Here, we show that dendritic cells from T1D patients exhibit

A sustainable solution for alleviating hexavalent chromium from water streams using Lactococcus lactis AM99 as a novel Cr(VI)-reducing bacterium.

Chromium, extensively used in various industries, poses significant challenges due to its environmental impact. The threat of Cr(VI) causes critical concerns in aquatic ecosystems as a consequence of the fluidity of water. The conventional approach for the treatment of effluents containing Cr(VI) is reducing Cr(VI) to low-noxious Cr(III). This research is related to a Gram positive bacterium newly isolated from tannery effluent under aerobic conditions. To characterize functional groups on the isolate, Fourier transform infrared spectroscopy was utilized. The effect of different factors on Cr(

Treatment with a Lactococcus lactis that chromosomally express E. coli cfaI mitigates salivary flow loss in a Sjögren's syndrome-like disease.

Sjögren's Syndrome (SjS) results in loss of salivary and lacrimal gland excretion due to an autoimmune attack on these secretory glands. Conventional SjS treatments address the symptoms, but not the cause of disease. Recognizing this deficit of treatments to reverse SjS disease, studies were pursued using the fimbriae from enterotoxigenic E. coli, colonization factor antigen I (CFA/I), which has anti-inflammatory properties. To determine if CFA/I fimbriae could attenuate SjS-like disease in C57BL/6.NOD-Aec1Aec2 (SjS) females, the Lactococcus lactis (LL) 301 strain was developed to chromosomall

Immune Responses to Orally Administered Recombinant Lactococcus lactis Expressing Multi-Epitope Proteins Targeting M Cells of Foot-and-Mouth Disease Virus.

Foot and mouth disease virus (FMDV), whose transmission occurs through mucosal surfaces, can also be transmitted through aerosols, direct contact, and pollutants. Therefore, mucosal immunity can efficiently inhibit viral colonization. Since vaccine material delivery into immune sites is important for efficient oral mucosal vaccination, the M cell-targeting approach is important for effective vaccination given M cells are vital for luminal antigen influx into the mucosal lymph tissues. In this study, we coupled M cell-targeting ligand Co1 to multi-epitope TB1 of FMDV to obtain TB1-Co1 in order
