Reversible static-site PCR via dual-phase confinement in digital microfluidics.

Digital microfluidics (DMF) enables programmable manipulation of discrete droplets for multi-step biochemical workflows; however, integrating polymerase chain reaction (PCR) into reconfigurable DMF systems remains challenging due to evaporation, surface fouling, and loss of droplet mobility during PCR. Here, we present a dual-phase pinning system (DPPS) for stable and reversible static-site PCR in DMF. In this approach, an aqueous PCR droplet is locally encapsulated within an immiscible lubricating phase and stabilized through combined mechanical confinement and continuous electrowetting actua

Review of CNN-Based Approaches for Preprocessing, Segmentation and Classification of Knee Osteoarthritis.

Osteoarthritis (OA) is a prevalent joint disorder characterized by symptoms such as pain and stiffness, often leading to loss of function and disability. Knee osteoarthritis (KOA) represents the most prevalent type of osteoarthritis. KOA is usually detected using X-ray radiographs of the knee; however, the classification of disease severity remains subjective and varies among clinicians, motivating the need for automated assessment methods. In recent years, deep learning-based approaches have shown promising performance for KOA classification tasks, particularly when applied to structured imag

A Comparison of mRNA Sequencing with Random Primed and 3'-Directed Libraries.

Creating a cDNA library for deep mRNA sequencing (mRNAseq) is generally done by random priming, creating multiple sequencing fragments along each transcript. A 3'-end-focused library approach cannot detect differential splicing, but has potentially higher throughput at a lower cost, along with the ability to improve quantification by using transcript molecule counting with unique molecular identifiers (UMI) that correct PCR bias. Here, we compare an implementation of such a 3'-digital gene expression (3'-DGE) approach with "conventional" random primed mRNAseq. Given our particular datasets on

Raman spectroscopy which is based upon inelastic scattering of photons has a potential to emerge as a noninvasive bedside in vivo or ex vivo molecular diagnostic tool. There is a need to improve the sensitivity and predictability of Raman spectroscopy. We developed a grid matrix-based tissue mapping protocol to acquire cellular-specific spectra that also involved digital microscopy for localizing malignant and lymphocytic cells in sentinel lymph node biopsy sample. Biosignals acquired from specific cellular milieu were subjected to an advanced supervised analytical method, i.e., cross-correlat

Digital Twin-Based Virtual Sensor Data Prediction and Visualization Techniques for Smart Swine Barns.

To address the limitations of sensor deployment and high maintenance costs in smart swine barns, this study proposes a digital twin (DT)-based virtual sensor prediction and visualization method. Spatial constraints and harsh barn environments often cause sensor blackout zones, hindering precise environmental monitoring. To overcome these challenges, a virtual sensor was defined at the central position between Zone 1 and Zone 2, and its data were generated using a hybrid model that combines inverse distance weighting (IDW)-based spatial interpolation with long short-term memory (LSTM)-based tim
