INTRODUCTION: It is widely assumed that sepsis is a life-threatening systemic inflammation caused by a dysregulated host response to infection mediated by an increase in multiple proinflammatory cytokines. The levels of key proinflammatory cytokines tumour necrosis factor, interleukin-1β and interferon γ are poorly characterised during sepsis. We believe this project will produce a 'gold-standard' document to which other reports on cytokine levels will be compared. The objective of this systematic review will be to identify key cytokine circulating levels in patients with sepsis and assess the

BACKGROUND: Excessive weight gain during pregnancy is common in developed countries and increases the risk of complications during pregnancy, delivery and the postpartum period, which can affect both maternal and fetal outcome. Interventions to reduce excessive gestational weight gain have previously not been systematically evaluated using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) system.
OBJECTIVES: To determine whether published trials of interventions to reduce excessive gestational weight gain are of sufficient quality and provide sufficient data to en

BACKGROUND: Protein Energy Malnutrition is an important cause of child morbidity and mortality in middle- and low-income countries. It has been suggested that excessive free radical activity may be responsible for the clinical manifestation of kwashiorkor. Antioxidants may be able to curb excessive free radical activity and prevent the development of kwashiorkor in susceptible children.
OBJECTIVES: To evaluate the benefits of supplementation of vitamin E, selenium, cysteine and riboflavin (alone or in combination) in preventing kwashiorkor.
SEARCH STRATEGY: We conducted searches of CENTRAL 2

INTRODUCTION: Invasive mechanical ventilation (IMV) may contribute to the development and progression of acute kidney injury (AKI) in critically ill patients. Given the expanding evidence-base and the immense variation in the association described, particularly following the COVID-19 pandemic era, a rigorous synthesis is warranted. This systematic review and meta-analysis will evaluate this association and identify potential risk-mitigation strategies.
METHODS AND ANALYSIS: We will conduct a comprehensive search of databases, including Ovid MEDLINE, Ovid Embase, CINAHL, Scopus, Web of Science

Hepatitis B virus (HBV) infection continues to pose a significant threat to global public health. Current therapies, including nucleoside analogs and interferon-α, suppress viral replication but fail to eliminate covalently closed circular DNA (cccDNA) - a viral minichromosome essential for HBV persistence. Targeting cccDNA is critical for achieving a functional cure, yet few effective inhibitors have been reported. Building on our previously established HepG2-HBV/loxP cell model, we applied a high-throughput screening workflow to identify cccDNA-targeting agents from marine natural compounds.

BACKGROUND: Neurodegenerative diseases (NDs) such as Parkinson's disease (PD), Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), Huntington's disease (HD) are set apart by progressive neuronal loss and concomitant functional decline. Traditional therapies are equipped with only symptomatic relief, devoid of neurorestorative properties. Stem-cell-based therapies have the potential to revolutionize neurological care by replenishing lost cells, mitigating inflammation, and fostering a neuroprotective environment.
OBJECTIVES: This narrative review aims to appraise the treatment poten

BACKGROUND AND AIMS: Metachronous gastric lesions (MGLs) are a significant concern after endoscopic resection (ER) for precancerous lesions and early gastric cancers (EGCs). The follow-up strategy for the development of MGLs after ER remains unclear. The aim of this study was to compare the trend in the cumulative MGL incidence after ER and to identify risk factors of MGLs.
METHODS: We searched four databases (PubMed, Embase, Web of Science Core Collection and Cochrane Library) for studies reporting on patients who underwent ER, and performed meta-analysis. A dose-response analysis was perfor

INTRODUCTION: Nosocomial infections in Neonatal Intensive Care Units (NICU) are a major concern due to the vulnerability of premature and immunocompromised infants. Serratia marcescens is an opportunistic pathogen often involved in these infections, contributing significantly to morbidity and mortality. Integrating Next-Generation Sequencing (NGS) into infection control programs can enhance detection, surveillance, and prevention efforts. This study aimed to develop a mapping-based pipeline for strain typing and phylogenetic analysis of nosocomial infections, enabling detailed comparison of mi

Malignant pleural mesothelioma (MPM) is a rare, aggressive tumor with a poor prognosis. In view of the poor survival benefit from first-line chemotherapy and the lack of subsequent effective treatment options, there is a strong need for the development of more effective treatment approaches for patients with MPM. This review will provide a comprehensive state of the art of new investigational approaches for mesothelioma. In an introductory section, the etiology, epidemiology, natural history, and standard of care treatment for MPM will be discussed. This review provide an update of the major c

Functional genomics and forward genetics seek to assign function to all known genes in a genome. Entamoeba histolytica is a protozoan parasite for which forward genetics approaches have not been extensively applied. It is the causative agent of amoebic dysentery and liver abscess, and infection is prevalent in developing countries that cannot prevent its fecal-oral spread. It is responsible for considerable global morbidity and mortality. Given that the E. histolytica genome has been sequenced, it should be possible to apply genomic approaches to discover gene function. We used a genome-wide o

Tear fluid is a promising minimally invasive source of biomarkers for ocular surface diseases (OSDs). However, variability in sampling and pre-analytical processing remains a major limitation for reproducibility and cross-study comparability. Here, we quantitatively evaluated two commonly used tear collection methods, microcapillary tubes (CT) and Schirmer strips (SCH), and assessed the effect of elution buffer composition on protein recovery from SCH. In Study I, tears were collected from the same healthy donors using CT and SCH. In Study II, SCH from additional donors were longitudinally spl

Acute kidney injury (AKI) is a global healthcare burden attributable to high mortality and staggering costs of dialysis. The underlying causes of AKI include hypothermia and rewarming (H/R), ischemia/reperfusion (I/R), mitochondrial dysfunction and reactive oxygen species production. Inspired by the mechanisms conferring organ protection in hibernating hamster, 6-chromanol derived compounds were developed to address the need of effective prevention and treatment of AKI. Here we report on the pre-clinical screening of 6-chromanol leads that confer protection during I/R to select compounds with

Melanoma represents ~5% of all cutaneous malignancies, yet accounts for the majority of skin cancer deaths due to its propensity to metastasise. To develop new therapies, novel target molecules must to be identified and the accessibility of cell surface proteins makes them attractive targets. Using CRISPR activation technology, we screened a library of guide RNAs targeting membrane protein-encoding genes to identify cell surface molecules whose upregulation enhances the metastatic pulmonary colonisation capabilities of tumour cells in vivo. We show that upregulated expression of the cell surfa

Nature's library of venoms is a vast and untapped resource that has the potential of becoming the source of a wide variety of new drugs and therapeutics. The discovery of these valuable molecules, hidden in diverse collections of different venoms, requires highly specific genetic and proteomic sequencing techniques. These have been used to sequence a variety of venom glands from species ranging from snakes to scorpions, and some marine species. In addition to identifying toxin sequences, these techniques have paved the way for identifying various novel evolutionary links between species that w

To prevent epidemic and pandemic cholera disease, an indispensible approach is to develop cholera vaccines based on comprehensive epitope information of this pathogen. This study aimed to utilize our previously raised monoclonal antibody IXiao3G6, which can recognize an epitope in lipopolysaccharide (LPS) sites of Ogawa, to identify mimetic peptides, which may represent Ogawa LPS's epitope information. A phage display library screening using IXiao3G6 antibody resulted in identification of a mimic peptide (MP) with high avidity. A recombinant protein, containing one cholera toxin subunit B (CTB

BACKGROUND: The domesticated apple (Malus domestica) is widely considered an admixture primarily involving M. sieversii, M. sylvestris, and M. orientalis, with possible contributions from tiny-fruited species such as M. baccata. Although this origin theory is well supported by genomic studies, the relative contributions of each progenitor species to cultivar genomes remain unclear, and taxonomic classifications of some Malus species are conflicting. The availability of new genomic tools and resources enables probing of these topics.
RESULTS: A panel of wild Malus accessions was genotyped usin
