Generation of a ror2-EGFP transgenic Xenopus line for tracking Ror2-expressing cells during limb and intestinal development.

During Xenopus metamorphosis, thyroid hormone (TH) induces extensive remodeling of the intestine including the development of adult epithelial stem cells (ASCs). Ror2 (receptor tyrosine kinase-like orphan receptor 2), a receptor involved in non-canonical Wnt signaling, is expressed in a limited population of larval epithelial cells that undergo dedifferentiation into ASCs. To examine when and how the fate of Ror2-expressing larval epithelial cells is specified toward the ASC lineage, we successfully generated a new transgenic Xenopus expressing EGFP under the control of the ror2 promoter (Ror2

Digital-image analysis of the angiographic patterns of the popliteal artery in patients with aorto-iliac occlusive disease (Leriche syndrome).

Angiographic patterns of the popliteal artery are of great clinical relevance in vascular surgery below the knee. Using radiological, digital and statistical methods the variants and Luminal diameters of the popliteal artery branching in 46 men and 30 women with Lerich syndrome were studied. Statistical analysis did not reveal any gender or syntopic dimorphisms (P > or = 0.05). In subtype IA (87.5%) the anterior tibial artery and the short type of posterior tibioperoneal trunk were found. In subtype IB (2.63%) an arterial trifurcation was observed. In subtype IC (1.97%) the posterior tibial ar

The formation of somite compartments in the avian embryo.

The somites develop from the unsegmented paraxial mesoderm that flanks the neural tube. They form in an intrinsic process which lays down the primary segmental pattern of the vertebrate body. We review the processes of somitogenesis and somite differentiation as well as the mechanisms involved in these developmental events. Long before overt differentiation occurs, different compartments of the still epithelial somites give rise to special cell lines and to particular derivatives. By means of isotypic grafting between quail and chick embryos, it is possible to follow the fate of groups of somi

Defect in the maintenance of the apical ectodermal ridge in the Dactylaplasia mouse.

During vertebrate limb development the distal apex of the limb bud ectoderm is induced to form the apical ectodermal ridge (AER). The presence of the AER is required for the continued outgrowth of the limb bud. Classical embryological studies have led to the hypothesis that a secreted mesenchymal factor is required to maintain the AER. We have undertaken a detailed analysis of Dactylaplasia (Dac) mice, a semidominant mutant which displays missing central digits in the fore- and hindlimbs of heterozygous animals and monodactyly in homozygous animals. Our data show that Dac mice have a defect in

Research note: Zic5 alters digit morphology during avian limb development.

Avian limbs have evolved precise digit lengths and shapes that are optimized for flight and locomotion, but the transcriptional regulators that regulate this architecture remain poorly defined. Although anteriorly biased Zic3 expression constrains digit outgrowth, whether other zinc finger of the cerebellum (Zic) factors-particularly Zic5-shape avian distal limb morphology remains unknown. Here, we define the spatiotemporal dynamics and developmental function of Zic5 expression in the chicken limb and evaluate its role in shaping avian limb and digit morphogenesis. Whole-mount in situ hybridiz

Cdc42 is required for chondrogenesis and interdigital programmed cell death during limb development.

Cdc42, a member of the Rho subfamily of small GTPases, is known to be a regulator of multiple cellular functions, including cytoskeletal organization, cell migration, proliferation, and apoptosis. However, its tissue-specific roles, especially in mammalian limb development, remain unclear. To investigate the physiological function of Cdc42 during limb development, we generated limb bud mesenchyme-specific inactivated Cdc42 (Cdc42(fl/fl); Prx1-Cre) mice. Cdc42(fl/fl); Prx1-Cre mice demonstrated short limbs and body, abnormal calcification of the cranium, cleft palate, disruption of the xiphoid

Generation of mice with functional inactivation of talpid3, a gene first identified in chicken.

Specification of digit number and identity is central to digit pattern in vertebrate limbs. The classical talpid(3) chicken mutant has many unpatterned digits together with defects in other regions, depending on hedgehog (Hh) signalling, and exhibits embryonic lethality. The talpid(3) chicken has a mutation in KIAA0586, which encodes a centrosomal protein required for the formation of primary cilia, which are sites of vertebrate Hh signalling. The highly conserved exons 11 and 12 of KIAA0586 are essential to rescue cilia in talpid(3) chicken mutants. We constitutively deleted these two exons t

Two nonsense GLI3 variants are associated with polydactyly and syndactyly in two families by affecting the sonic hedgehog signaling pathway.

BACKGROUND: Polydactyly and syndactyly are congenital limb deformities, segregating in an autosomal-dominant fashion. The variants in the GLI3 gene are closely related to congenital limb malformations. However, the causes underlying polydactyly and syndactyly are not well understood. METHODS: We conducted a whole-exome sequencing on two four-generation Chinese families with polydactyly and syndactyly. Then c.2374C>T and c.1728C>A mutant plasmids were transfected to HEK293T cells and mice limb bud cells to explore the functional consequences of these variants. Western blot and real-time quanti

Unusual pattern of Sonic hedgehog expression in the polydactylous mouse mutant Hemimelic extra-toes.

We have examined the dynamic expression of Sonic hedgehog (Shh) in limb buds of the Hemimelic extra-toes (Hx) mutant. An ectopic domain of expression appears in the limb bud at embryonic day 11.5, which is not restricted to the anterior mesenchyme as in other polydactylous mutants, but extends along the entire apical ectodermal ridge. No difference in expression was observed between heterozygotes and homozygotes. This ectopic expression domain forms later and is maintained longer than the normal one. We verified that the Shh signal is properly transduced in the ectopic expression domain by ana

Human limb abnormalities caused by disruption of hedgehog signaling.

Human hands and feet contain bones of a particular size and shape arranged in a precise pattern. The secreted factor sonic hedgehog (SHH) acts through the conserved hedgehog (Hh) signaling pathway to regulate the digital pattern in the limbs of tetrapods (i.e. land-based vertebrates). Genetic analysis is now uncovering a remarkable set of pathogenetic mutations that alter the Hh pathway, thus compromising both digit number and identity. Several of these are regulatory mutations that have the surprising attribute of misdirecting expression of Hh ligands to ectopic sites in the developing limb b

Splenogonadal fusion with limb deficiency and micrognathia.

Splenogonadal fusion (SGF) is a rare abnormality with two known types. In the continuous type, the spleen is connected to the gonad, and there are often limb defects, micrognathia, or other congenital malformations such as ventricular septal defect, anal atresia, microgastria, spina bifida, craniosynostosis, thoracopagus, diaphragmatic hernia, hypoplastic lung and abnormal lung fissures, polymicrogyria, deficient coccyx, and bifid spine C6-T3. The discontinuous type is usually not associated with congenital defects, and the gonad that fused with an accessory spleen has no connection with the n

[Sirenomelia as a part of VACTERL association: a study of three cases].

Sirenomelia, characterized by a fusion of lower limb buds, is rare. Moreover, the coexistence of this malformation with a VACTERL sequence is exceptional. We report, here, three new observations associating these two diseases on fetuses from 14 to 26 weeks gestation. With these three new cases associating sirenomelia and VACTERL, observed in our unit and examined in light of data from the medical literature, we discuss the embryologic origin of such malformations and the nosologic frontiers between these two diseases.

Dextrocardia and ventricular septal defect in the Möbius syndrome.

A case of Möbius syndrome is reported with the additional features of dextrocardia and a single ventricle, neither of which have been previously reported in the literature with this syndrome. Correlation of the period in embryogenesis of cardiac partitioning and rotation with the appearance of the cranial nuclei and limb buds would support the proposal that the fourth to the seventh week of gestation is the critical period of embryogenesis, wherein an intrauterine insult could give rise to the Möbius syndrome.

Unraveling the transcriptional regulation of TWIST1 in limb development.

The transcription factor TWIST1 plays a vital role in mesoderm development, particularly in limb and craniofacial formation. Accordingly, haploinsufficiency of TWIST1 can cause limb and craniofacial malformations as part of Saethre-Chotzen syndrome. However, the molecular basis of TWIST1 transcriptional regulation during development has yet to be elucidated. Here, we characterized active enhancers in the TWIST1-HDAC9 locus that drive transcription in the developing limb and branchial arches. Using available p300 and H3K27ac ChIP-seq data, we identified 12 enhancer candidates, located both with

The pathogenic mechanism of syndactyly type V identified in a Hoxd13Q50R knock-in mice.

Syndactyly type V (SDTY5) is an autosomal dominant extremity malformation characterized by fusion of the fourth and fifth metacarpals. In the previous publication, we first identified a heterozygous missense mutation Q50R in homeobox domain (HD) of HOXD13 in a large Chinese family with SDTY5. In order to substantiate the pathogenicity of the variant and elucidate the underlying pathogenic mechanism causing limb malformation, transcription-activator-like effector nucleases (TALEN) was employed to generate a Hoxd13Q50R mutant mouse. The mutant mice exhibited obvious limb malformations including

How to make a zone of polarizing activity: insights into limb development via the abnormality preaxial polydactyly.

Early in vertebrate limb development, a program initiates that polarizes the limb along the antero-posterior axis. The mesenchyme at the posterior margin is ultimately responsible for the asymmetry due to a region called the zone of polarizing activity (ZPA). The ZPA produces and secretes the molecule SHH, which coordinates the patterning of the resulting digits. Preaxial polydactyly (PPD) is a commonly occurring limb abnormality; investigating the genetic basis of this defect has provided insights into our understanding of digit patterning. PPD disrupts limb asymmetry by producing an ectopic

Ulnar dimelia--a limb without anteroposterior differentiation.

A case of ulnar dimelia with radial aplasia and polydactyly and the main features of its treatment are described. I believe that this abnormality results from impairment of the mechanisms responsible for organization and differentiation of the limb bud in the anteroposterior direction. This deformity is not the result of duplication of any of the parts of the limb, either compensatory or because of a primary duplicated limb field. The anomaly should be called "anteroposteriorly undifferentiated limb with polydactylism" because the long bones in the forearm are probably not ulnae. Similarities

Tissue-specific requirement of sodium channel and clathrin linker 1 (Sclt1) for ciliogenesis during limb development.

Primary cilia have essential roles as signaling centers during development and adult homeostasis. Disruption of ciliary structure or function causes congenital human disorders called ciliopathies. Centriolar distal appendage (DAP) proteins are important for anchoring cilia to the membrane. However, the exact functions of DAP during in vivo ciliogenesis and animal development remain poorly understood. Here, we showed that the DAP component sodium channel and clathrin linker 1 (Sclt1) mutant mice had abnormal craniofacial and limb development with postnatal lethality. In mutant embryos, most of

Fetal Amelia With Hypoplastic Tibia and Terminal Fibular Hemimelia: A Case Report With Review of the Literature.

Congenital limb deformities, with a birth frequency of 0.55 per 1,000, are extremely rare prenatal defects that can present with either partial or complete lack of a limb or a specific portion of a limb. Amelia is a sporadic anomaly that is defined by the complete absence of a limb's skeletal elements, whereas hypomelia is defined by the incomplete development of a limb's skeletal elements. We present the case of a neonate with gross facial deformities in the form of the absence of both external ears and a saddle-shaped nose. The absence of the right lower limb bud was seen. The left lower lim

Polydactylous limbs in Strong's Luxoid mice result from ectopic polarizing activity.

Strong's Luxoid (1stD) is a semidominant mouse mutation in which heterozygotes show preaxial hindlimb polydactyly, and homozygotes show fore- and hindlimb polydactyly. The digit patterns of these polydactylous limbs resemble those caused by polarizing grafts, since additional digits with posterior character are present at the anterior side of the limb. Such observations suggest that 1stD limb buds might contain a genetically determined ectopic region of polarizing activity. Accordingly, we show that mutant embryos ectopically express the pattern-determining genes fibroblast growth factor 4 (fg
