Glucagon-like peptide-1 analog, liraglutide, alters blood-testis barrier permeability in juvenile healthy rats.

Glucagon-like peptide-1 (GLP-1) analogue liraglutide has recently been approved for treating obese children. However, little is known about liraglutide's effect on testicular function during the juvenile developmental period, particularly regarding the blood-testis barrier (BTB) formation. This study analyses the impact of liraglutide treatment on BTB function in young rats and its consequences in adulthood. Rats received a daily dose of liraglutide from postnatal day (Pnd) 20 to 33, the period when a functional BTB is formed. At Pnd 34, liraglutide promoted a significant increase in permeable

Liraglutide and Weight Loss Among Suboptimal Responders to Metabolic Bariatric Surgery: A Randomized Clinical Trial.

IMPORTANCE: Suboptimal weight loss after metabolic bariatric surgery (MBS) is a common indication for conversion surgery. The use of obesity management medications may improve weight loss, reducing the need for further surgery. OBJECTIVE: To examine whether liraglutide improves weight loss and health outcomes after MBS in patients who have had suboptimal weight loss. DESIGN, SETTING, AND PARTICIPANTS: This prospective, double-blinded, single-center randomized clinical trial compared the effect of daily liraglutide with placebo in adults treated at an academic surgical department. Patients wi

Liraglutide preserves endothelial function in ophthalmic arteries of septic mice via prevention of oxidative stress.

We hypothesized that the glucagon-like peptide-1 (GLP-1) receptor agonist liraglutide enhances endothelial functions in ophthalmic arteries of mice with polymicrobial sepsis. The study involved three groups of mice. Two groups underwent cecal ligation and puncture (CLP) to induce polymicrobial sepsis, with one of these groups receiving liraglutide treatment. The third group underwent sham surgery as a control. 24 h after the CLP procedure, the mice were euthanized, and ophthalmic arteries were isolated either for in vitro analysis of vascular function using videomicroscopy or for quantificatio

Liraglutide attenuates intestinal ischemia/reperfusion injury via NF-κB and PI3K/Akt pathways in mice.

AIMS: Previous studies have reported that glucagon-like peptide-1 (GLP-1) may play a critical role in the development of intestinal ischemia-reperfusion (I/R) injury. The present study aimed to investigate whether liraglutide (GLP-1 analog) protects against intestinal I/R injury and reveals the possible underlying mechanism. MAIN METHODS: Temporary superior mesenteric artery occlusion was performed to establish an intestinal I/R injury mouse model. Different doses of liraglutide were administered in vivo. Then, the survival rate of mice exposed to different ischemia times, the histopathology,

GLP-1 agonist liraglutide improves ouabain-induced mania and depressive state via GSK-3β pathway.

Bipolar disorder (BD) is a severe mental illness characterized by aberrant mood changes between hypomania and mania or mixed states and depression. Metabolic changes also accompany disease progression and cause significant morbidity. Symptomatic treatment options are available, but asymptomatic patients and poor drug responders are significant problems. Based on the most common pharmacological agent that is used in the treatment, lithium and its main mechanisms of action, oxidative stress, and glycogen synthase kinase-3β (GSK-3β) signaling are extensively investigated. However, knowledge about

Sitagliptin and Liraglutide Modulate L-dopa Effect and Attenuate Dyskinetic Movements in Rotenone-Lesioned Rats.

L-dopa is still considered as the gold standard therapy for Parkinson's disease (PD); however, L-dopa-induced dyskinesia (LID) is a serious complication of long-term L-dopa treatment. The present study investigated the therapeutic potential of sitagliptin and liraglutide in comparisons with L-dopa against PD. In addition, their capacity to modulate L-dopa dose and/or side effects was investigated, too. Rats were injected with rotenone (3 mg/kg/day, s.c.) for 10 consecutive days to induce the experimental PD. The rotenone-treated rats were administered sitagliptin (30 mg/kg/day, p.o.) and lirag

GLP-1 Analogue Liraglutide Attenuates Mutant Huntingtin-Induced Neurotoxicity by Restoration of Neuronal Insulin Signaling.

Huntington's disease (HD) is a progressive and fatal neurodegenerative disease caused by CAG repeat expansion in the coding region of huntingtin (HTT) protein. The accumulation of mutant HTT (mHTT) contributes to neurotoxicity by causing autophagy defects and oxidative stress that ultimately lead to neuronal death. Interestingly, epidemiologic studies have demonstrated that the prevalence of type-2 diabetes, a metabolic disease mainly caused by defective insulin signaling, is higher in patients with HD than in healthy controls. Although the precise mechanisms of mHTT-mediated toxicity remain u

Liraglutide Modulates Appetite and Body Weight Through Glucagon-Like Peptide 1 Receptor-Expressing Glutamatergic Neurons.

Glucagon-like peptide 1 receptor (GLP-1R) agonists are U.S. Food and Drug Administration-approved weight loss drugs. Despite their widespread use, the sites of action through which GLP-1R agonists (GLP1RAs) affect appetite and body weight are still not fully understood. We determined whether GLP-1Rs in either GABAergic or glutamatergic neurons are necessary for the short- and long-term effects of the GLP1RA liraglutide on food intake, visceral illness, body weight, and neural network activation. We found that mice lacking GLP-1Rs in vGAT-expressing GABAergic neurons responded identically to co

Liraglutide attenuates obese-associated breast cancer cell proliferation via inhibiting PI3K/Akt/mTOR signaling pathway.

This study aims to explore the anti-proliferative, pro-apoptotic, and anti-migration activities of liraglutide (LGT) in MCF-7 breast cancer (BC) cells in subjects with obesity, particularly its effects on the PI3K/Akt/mTOR/AMPK pathway. The role of AMPK/SIRT-1, an essential regulator of adipokine production, in the effect of LGT on the production of adipose-derived adipokine was also assessed. MCF-7 cells were incubated in conditioned medium (CM) generated from adipose-derived stem cells (ADSCs) of obese subjects. MCF-7 cells were then treated with LGT for 72 h. Effect of hCMSCs and liraglutide combination in ALI through cAMP/PKAc/β-catenin signaling pathway.

Источник пометил публикацию как отозванную

BACKGROUND: ALI/ARDS is the major cause of acute respiratory failure in critically ill patients. As human chorionic villi-derived MSCs (hCMSCs) could attenuate ALI in the airway injury model, and liraglutide, glucagon-like peptide 1 (GLP-1) agonist, possesses anti-inflammatory and proliferation promotion functions, we proposed to probe the potential combinatory effect of hCMSCs and liraglutide on ALI. METHODS: We examined the time- and dose-dependent manner of GLP-1R, SPC, Ang-1, and FGF-10 with LPS via western blot and qRT-PCR. Western blot and chromatin immunoprecipitation assay detected th

The GLP1R Agonist Liraglutide Reduces Hyperglucagonemia Induced by the SGLT2 Inhibitor Dapagliflozin via Somatostatin Release.

The newest classes of anti-diabetic agents include sodium-glucose cotransporter 2 (SGLT2) inhibitors and glucagon-like peptide 1 receptor (GLP1R) agonists. The SGLT2 inhibitor dapagliflozin reduces glucotoxicity by glycosuria but elevates glucagon secretion. The GLP1R agonist liraglutide inhibits glucagon; therefore, we hypothesize that the cotreatment of dapagliflozin with liraglutide could reduce hyperglucagonemia and hyperglycemia. Here we use five complementary models: human islet cultures, healthy mice, db/db mice, diet-induced obese (DIO) mice, and somatostatin receptor-2 (SSTR2) KO mice

Liraglutide Treatment Reduces Endothelial Endoplasmic Reticulum Stress and Insulin Resistance in Patients With Diabetes Mellitus.

Background Prior studies have shown that nutrient excess induces endoplasmic reticulum ( ER ) stress in nonvascular tissues from patients with diabetes mellitus ( DM ). ER stress and the subsequent unfolded protein response may be protective, but sustained activation may drive vascular injury. Whether ER stress contributes to endothelial dysfunction in patients with DM remains unknown. Methods and Results To characterize vascular ER stress, we isolated endothelial cells from 42 patients with DM and 37 subjects without DM. Endothelial cells from patients with DM displayed higher levels of ER st

A Systematic Review and Meta-Analysis of Efficacy and Safety of Liraglutide in Patients with Type 2 Diabetes Mellitus.

BACKGROUND: Obesity plays a pivotal and modifiable role in the development and progression of type 2 diabetes mellitus (T2DM). Clinicians increasingly use lower doses of liraglutide (1.2 mg and 1.8 mg) to achieve clinically meaningful weight loss while maintaining effective glycemic control in people with T2DM and obesity. In this meta-analysis, we compared the efficacy and safety of liraglutide 1.2 mg and 1.8 mg in this population. METHODS: We systematically searched PubMed, the Cochrane Central Register of Controlled Trials, LENS, ClinicalTrials.gov, and the Virtual Health Library (VHL) for

Weekly Liraglutide for the Management of Intractable Polydipsia and Interdialytic Weight Gain in a Patient on Hemodialysis: A Case Report.

Volume status management is a critical challenge in hemodialysis. Water transgression usually results from intractable polydipsia, for which there are no approved pharmacological treatments. Given the emerging evidence on the role of glucagon-like peptide-1 receptor agonists (GLP-1RAs) in modulating neurological pathways that regulate dipsogenic behavior and satiety, their use is proposed as an unprecedented therapeutic opportunity. A 66-year-old woman with end-stage renal disease (ESRD) secondary to glomerulonephritis was in a chronic hemodiafiltration program without residual diuresis. She s
