[Molecular biological aspects of neuroplasticity: approaches for treating tinnitus and hearing disorders].

Peripheral and central structures are involved in the onset of tinnitus. Neuronal plasticity is of special importance for the occurrence of central tinnitus and its persistent form. Neuronal plasticity is the ability of the brain to adapt its own structure (synapses, nerve cells, or even whole areas of the brain) and its organization to modified biological requirements. Neuroplasticity is an ongoing dynamic process. Generally speaking, there are two types of plasticity: synaptic and cortical. Cortical plasticity involves activity-dependent changes in size, connectivity, or in the activation pa

Adaptive gene regulation in the Striatum of RGS9-deficient mice.

BACKGROUND: RGS9-deficient mice show drug-induced dyskinesia but normal locomotor activity under unchallenged conditions. RESULTS: Genes related to Ca2+ signaling and their functions were regulated in RGS9-deficient mice. CONCLUSION: Changes in Ca2+ signaling that compensate for RGS9 loss-of-function can explain the normal locomotor activity in RGS9-deficient mice under unchallenged conditions. SIGNIFICANCE: Identified signaling components may represent novel targets in antidyskinetic therapy. The long splice variant of the regulator of G-protein signaling 9 (RGS9-2) is enriched in striatal

Adaptive-filter models of the cerebellum: computational analysis.

Many current models of the cerebellar cortical microcircuit are equivalent to an adaptive filter using the covariance learning rule. The adaptive filter is a development of the original Marr-Albus framework that deals naturally with continuous time-varying signals, thus addressing the issue of 'timing' in cerebellar function, and it can be connected in a variety of ways to other parts of the system, consistent with the microzonal organization of cerebellar cortex. However, its computational capacities are not well understood. Here we summarise the results of recent work that has focused on two

Differential regulation of synaptic plasticity and cerebellar motor learning by the C-terminal PDZ-binding motif of GluRdelta2.

The delta2 glutamate receptor (GluRdelta2) is predominantly expressed in Purkinje cells and plays crucial roles in cerebellar functions: GluRdelta2-/- mice display ataxia and impaired motor learning. In addition, long-term depression (LTD) at parallel fiber (PF)-Purkinje cell synapses is abrogated, and synapse formation with PFs and climbing fibers (CFs) is severely disturbed in GluRdelta2-/- Purkinje cells. Recently, we demonstrated that abrogated LTD was restored in GluRdelta2-/- Purkinje cells by the virus-mediated expression of the wild-type GluRdelta2 transgene (Tg(wt)) but not by that of

β-Caryophyllene prevents cognitive impairment in mice following acute sleep deprivation by alleviating hippocampal structural and functional alterations.

Sleep deprivation (SD) has become a prevalent issue leading to suboptimal health conditions. While, there is still a lack of effective intervention methods. Cannabinoid receptor type 2 (CB2R) is reported to possess anti-inflammatory properties and facilitate synaptic plasticity, while without the risk of causing psychological addiction. Here we administered β-caryophyllene (BCP), the highly selective CB2R agonist, and observed whether cognitive impairment following acute sleep deprivation (ASD) was prevented. Our results demonstrated that administration of BCP for 3 days before ASD markedly pr

Deficient cerebellar long-term depression and impaired motor learning in mGluR1 mutant mice.

mGluR1 mutant mice are viable but show characteristic cerebellar symptoms such as ataxic gait and intention tremor. The anatomy of the cerebellum is not overtly disturbed. Excitatory synaptic transmission from parallel fibers (PFs) to Purkinje cells and that from climbing fibers (CFs) to Purkinje cells appear to be functional, and voltage-gated Ca2+ channels of Purkinje cells are normal. Both PF and CF synapses display normal short-term synaptic plasticity to paired stimuli. By marked contrast, long-term depression (LTD) is clearly deficient and conditioned eyeblink response is impaired. We co

Age-dependent evaluation of long-term depression responses in hyperthyroid rats: Possible roles of oxidative intracellular redox status.

BACKGROUND: According to the free radical theory, a gradual accumulation of the free radicals normally produced in the body underlies the changes associated with aging. Thyroid hormones (THs) are related to oxidative stress not only due to their stimulation of metabolism but also due to their effects on antioxidant mechanisms. Thyroid dysfunction increases with age; thus, changes in TH levels in elderly individuals could be a factor affecting the development of neurodegenerative diseases. However, the relationship is not always clear, based on current evidence regarding synaptic plasticity. M

Designable van der Waals Crystal for Artificial Neuronal Cell Mimicking.

Optogenetics employs light to regulate neuronal activity with exceptional spatiotemporal precision, thereby enabling the direct modulation of learning and memory processes in the human brain. This capability to externally control neuronal signaling via optical stimuli has provided profound insights into brain function and established a versatile strategy for engineering bio-inspired information processing. Herein, a designable van der Waals (vdW) crystal has been demonstrated for device-scale neuronal cell mimicking. The structural similarity between ion-channel in biological membranes and lay

Alteration of synaptic plasticity by neonatal seizures in rat somatosensory cortex.

Seizures in newborns are associated with a high risk for subsequent epilepsy and adverse neurodevelopmental consequences. Understanding the mechanisms by which neonatal seizures adversely disturb the immature brain is important in developing therapeutic strategies. Using the convulsant agent flurothyl to mimic repetitive neonatal seizures we show that early-life seizures result in long-term alteration in the maintenance phase of long-term potentiation (LTP) in layer IV to layer II/III synapses of the somatosensory cortex without alteration of basal synaptic transmission, the induction phase of

Intracerebroventricular administration of ouabain alters synaptic plasticity and dopamine release in rat medial prefrontal cortex.

Intracerebroventricular (ICV) administration of ouabain, a specific Na-K-ATPase inhibitor, in rats mimics the manic phenotypes of bipolar disorder and thus has been proposed as one of the best animal models of mania. Bipolar mania has been known to be associated with dysfunctions of medial prefrontal cortex (mPFC), a brain area critically involved in mental functions; however, the exact mechanism underlying these dysfunctions is not yet clear. The present study investigated synaptic transmission, synaptic plasticity, and dopamine release in Sprague-Dawley rat mPFC following ICV administration

Protective effects of sodium benzoate against toluene-induced reward enhancement, behavioral disturbances, and impaired synaptic plasticity in mice.

Toluene is one of the most commonly abused solvents. Inhibition of N-methyl-D-aspartate receptor (NMDAR) has been linked to the rewarding and behavioral effects of toluene. One of the avenues to enhance NMDAR function is to increase the levels of D-serine via inhibiting D-amino acids oxidase (DAAO) activity. The present study determined whether sodium benzoate, a DAAO inhibitor, could counteract the toluene-induced brain stimulation reward enhancement, behavioral disturbances, and hippocampal synaptic dysfunction after acute toluene exposure. Male mice received various doses of sodium benzoate

α8-integrins are required for hippocampal long-term potentiation but not for hippocampal-dependent learning.

Integrins are heterodimeric transmembrane cell adhesion receptors that are essential for a wide range of biological functions via cell-matrix and cell-cell interactions. Recent studies have provided evidence that some of the subunits in the integrin family are involved in synaptic and behavioral plasticity. To further understand the role of integrins in the mammalian central nervous system, we generated a postnatal forebrain and excitatory neuron-specific knockout of alpha8-integrin in the mouse. Behavioral studies showed that the mutant mice are normal in multiple hippocampal-dependent learni

The critical role of intracellular calcium in the mechanisms of plasticity of common snail defensive behavior command neurons LPl1 and RPl1 in nociceptive sensitization.

Studies on semi-intact common snail preparations addressed the involvement of intracellular calcium in changes in the excitability and responses to sensory stimuli of defensive behavior command neurons LPl1 and RPl1 during the acquisition of nociceptive sensitization. Application of sensitizing stimuli to the heads of control snails led to depolarization of neuron membranes, increases in neuron excitability, and depression of the responses of neurons to sensory stimuli during the short-term stage, and marked facilitation of responses in the long-term stage of sensitization. Acquisition of sens

[Critical role of intracellular calcium in plasticity mechanisms of defense behavior command neurons LPl1 and PPl1 of Helix lucorum during nociceptive sensitization].

The role of intracellular calcium in changes in excitability and responses of defense behavior command neurons LP11 and PP11 of Helix lucorum to sensory stimulation was investigated in semi-intact preparation of a snail during nociceptive sensitization. It was found that application of sensitizing stimuli onto the snail's head initiated membrane depolarization, increase in its excitability as well as depression of neural responses evoked by sensory stimuli in short-term period of sensitization and significant facilitation of neural responses in long-term period of sensitization. [AMPA receptors: molecular mechanisms of synaptic plasticity and the potential of Ampasse in addressing cognitive impairment].

Dysfunction of the glutamatergic system, specifically involving AMPA receptors, is recognized as a significant pathogenic mechanism underlying cognitive impairment. AMPA glutamate receptors mediate fast excitatory postsynaptic currents in the majority of brain synapses and play a crucial role in the processes associated with long-term potentiation, which is integral to memory function. Additional mechanisms of synaptic plasticity linked to AMPA receptors include long-term depression of synaptic transmission, homeostatic plasticity, and AMPA receptor trafficking. Bidirectional plasticity expressed by GABAergic synapses in the neonatal rat hippocampus.

1. Activity-dependent plasticity of GABAergic synaptic transmission was investigated in neonatal rat hippocampal slices obtained between postnatal day (P) 2-10 using intracellular recording techniques. In all experiments, AMPA receptors were blocked by continual application of CNQX (10 microM). 2. Between P2 and P4, tetanic stimulation (TS) evoked NMDA receptor-dependent long-term depression of monosynaptic GABAA EPSPS (LTDGABAA). In contrast, when NMDA receptors were blocked by D-AP5 (50 microM), the same TS evoke long-term potentiation of GABAA EPSPS (LTPGABAA). 3. Between P6 and P10, TS fai

Excess ribosomal protein production unbalances translation in a model of Fragile X Syndrome.

Dysregulated protein synthesis is a core pathogenic mechanism in Fragile X Syndrome (FX). The mGluR Theory of FX predicts that pathological synaptic changes arise from the excessive translation of mRNAs downstream of mGlu1/5 activation. Here, we use a combination of CA1 pyramidal neuron-specific TRAP-seq and proteomics to identify the overtranslating mRNAs supporting exaggerated mGlu1/5 -induced long-term synaptic depression (mGluR-LTD) in the FX mouse model (Fmr1-/y). Our results identify a significant increase in the translation of ribosomal proteins (RPs) upon mGlu1/5 stimulation that coinc

Down-regulated striatal gene expression for synaptic plasticity-associated proteins in addiction and relapse vulnerable animals.

Reducing the likelihood of relapse represents one of the greatest obstacles in the successful treatment of cocaine addiction. Dysregulation of the synaptic plasticity processes long-term potentiation (LTP) and long-term depression (LTD) is thought to be associated with protracted relapse risk. To improve our understanding of the molecular mechanisms contributing to relapse vulnerability we trained rats (n=52) to self-administer cocaine and phenotyped animals as relapse-vulnerable or relapse-resilient using procedures adapted from Deroche-Gamonet et al. (Science 2004, 305, 1014-1017). Gene expr

Electrooptic Dual-Mode Synergetic Control of Two-Dimensional Nonvolatile Floating-Gate Transistors for Artificial Synapses.

Floating-gate memories based on two-dimensional (2D) materials show great potential for mimicking synapses in artificial neural networks (ANNs), offering a promising pathway toward hardware neuromorphic computing. However, current implementations predominantly rely on electrical modulation, limiting their applications in future artificial visual systems. Here, we demonstrate an electrooptic dual-mode artificial synapse based on a van der Waals heterostructured MoS2 floating-gate transistor. This device uniquely combines electrically controlled nonvolatile programmability with light-tunable mul
