The progesterone blockade of the luteinizing hormone surge is overcome by RU486.

Progesterone can prevent the oestrogen-induced and spontaneous preovulatory luteinizing hormone (LH) surges but the mechanisms underlying this effect remain poorly understood. Using a follicular phase ovariectomised (OVX) ewe model and by elevating progesterone in the presence of oestrogen to inhibit the LH surge, we investigated whether the progesterone receptor antagonist, RU486, could block the inhibitory effects of progesterone. Accordingly, intravaginal progesterone implants were inserted into OVX Ile-de-France ewes (n = 18), bearing 10 mm Silastic 17beta-oestradiol implants. Ten days lat

Serotonin, a neurotransmitter involved in the regulation of luteinizing hormone release.

The involvement of serotonin (5-HT) in the regulation of LH secretion is discussed on the basis of experimental and physiological models. The role of 5-HT on low amplitude pulsatile LH release in male rats is not yet clear, in spite of the fact that recent results suggest a weak permissive role. In ovariectomized rats, 5-HT expresses a negative influence on the increased rate of LH release, which is converted into a stimulatory effect by pretreatment of the animals with E2. Moreover, in castrated female rats there is a morning/afternoon oscillatory pattern in 5-HT metabolism of brain areas ass

On the role of dihydrotestosterone in regulating luteinizing hormone secretion in man.

The effect of dihydrotestosterone [17 beta-hydroxy-5 alpha-androstone-3-one (DHT) in the negative regulation of LH was studied throughout 3 months of percutaneous administration of this steroid to six normal male volunteers and six patients with primary testicular hypogonadism. In addition, testosterone (T) was administered according to the same protocol to the six hypogonadal patients. Plasma T, DHT, estradiol, sex hormone-binding globulin, and LH levels were measured by RIA before and weekly during treatment. In normal men, plasma DHT rose regularly from 0.5 +/- 0.2 to 3.3 +/- 0.9 ng/ml afte

The problem of anti-doping control of luteinizing hormone in boxing.

Luteinizing hormone (LH) is physiologically produced by the anterior pituitary gland. Male athletes may use pharmaceutical LH for doping since it increases the production of testosterone by testes. This hormone is thus on the World Anti-Doping Agency (WADA) list of substances prohibited for males. Anti-doping laboratories perform the assay of this hormone in urine and report abnormally elevated results. We observed a highly significant prevalence of abnormal results in samples taken after a boxing match. Comparison of the descriptive statistics for 426 LH values observed in boxing and other sp

Suppression of plasma luteinizing hormone by prolactin in the male rat.

Although a direct effect of PRL on gonadotropins has been previously suggested, it has not been convincingly demonstrated. The secretion of LH and FSH was studied in response to the stimuli of castration and LH releasing hormone administration in adult male rats made hyperprolactinemic with ectopic pituitary glands. Although plasma LH and FSH levels were similar in non-castrate hyperprolactinemic rats vs. controls, LH concentrations 24 h postcastration were less in hyperprolactinemic animals as compared to controls (P less than 0.001). The level of LH achieved was inversely correlated with the

Obesity alters the estrogen-induced luteinizing hormone surge in ovariectomized ewes†.

Obesity is a leading health issue with a negative effect on fertility, but the complexity of the relationship between obesity and fertility requires further study. Santoro and colleagues have characterized the metabolic environment of obesity and the development of reprometabolic syndrome. Due to the reproductive similarities between ewes and women, we use ovariectomized ewes as a translational model of reprometabolic syndrome and tested whether obesity alters the estradiol-17β (E2)-induced luteinizing hormone (LH) surge. Each ovariectomized ewe serves as her own biological control during the

Intracerebroventricular injection of neuropeptide Y suppresses luteinizing hormone pulses in mice.

Stress impairs reproduction, at least in part, by inhibition of Kisspeptin/Neurokinin B/Dynorphin (KNDy) cells and suppression of gonadotropin secretion. However, the mechanism for this inhibition is not clear. Neuropeptide Y (NPY) is a pleiotropic molecule that is produced in neurons throughout the brain and mediates several physiological processes including energy homeostasis and stress responses. Centrally administered NPY elicits variable effects on luteinizing hormone (LH) secretion, which vary with species, gonadal status, and the site of infusion. Therefore, our objective was to determi

Diagnostic utility of basal luteinizing hormone in evaluating central precocious puberty.

BACKGROUND: Basal luteinizing hormone (bLH) is increasingly proposed as a surrogate marker of hypothalamic-pituitary-gonadal activation, potentially reducing the need for gonadotropin-releasing hormone (GnRH) testing in children with suspected central precocious puberty (CPP). However, diagnostic thresholds remain assay-dependent, and values below the detection limit complicate interpretation. OBJECTIVE: This study aims to assess the diagnostic performance of bLH in predicting GnRH test response in patients evaluated for suspected CPP, using a censoring-aware approach for values below the ass

Luteinizing hormone supplementation in controlled ovarian stimulation: the Iran Delphi consensus.

INTRODUCTION: Numerous consensus documents worldwide address luteinizing hormone (LH) supplementation in controlled ovarian stimulation, yet to the best of our knowledge, only one consensus paper has been published in the Arab region. This study presents a Delphi consensus by seven Iranian infertility experts, offering real-world clinical perspectives. The aim was to develop evidence-based opinions on LH's role alongside FSH in various aspects of assisted reproductive technology (ART), including LH levels, monitoring, r-hLH use, and suggested activity. METHODS: Employing the Delphi consensus

Luteinizing hormone stimulates the expression of amphiregulin in human theca cells.

BACKGROUND: Luteinizing hormone (LH) can stimulate mural granulosa cells to produce Amphiregulin (AREG), which can induce the resumption of meiosis in oocytes. Theca cells are present in the outer layer of follicles, providing communication with the pituitary axis through the established vascular system around the follicle. As LH target cells, it is unknown whether theca cells can produce AREG after LH stimulation. METHODS: Primary cultured human theca cells were treated with LH (with or without the inhibitor of PKA, H89), or agonists of adenylate cyclase (forskolin or db-cAMP). The mRNA and

Inactivating mutations of the human luteinizing hormone receptor in both sexes.

The human luteinizing hormone/chorionic gonadotropin receptor (LHCGR) plays a fundamental role in male and female reproductive physiology. Over the past 15 years, several homozygous or compound heterozygous loss-of-function mutations in the LHCGR gene have been described in males and females. In genetic males, mutations in LHCGR were associated with distinct degrees of impairment in pre- and postnatal testosterone secretion resulting in a phenotypic spectrum. Patients with the severe form of LH resistance have predominantly female external genitalia and absence of secondary sex differentiation

Changes in luteinizing hormone and insulin secretion in polycystic ovarian syndrome.

Uncertainties regarding the pathogenetic changes underlying the polycystic ovarian syndrome (PCOS) have been reported. The aim of this study was to investigate the endocrine and metabolic features of PCOS patients in relation to luteinizing hormone (LH) secretion. Androgen assays, oral glucose tolerance tests, hyperinsulinaemic euglycaemic clamps and gonadotrophin releasing hormone (GnRH) tests were performed in 100 patients. Sixty-six patients scheduled as hyperinsulinaemic and 34 as normoinsulinaemic showed similar concentrations of LH, follicle stimulating hormone (FSH), LH/FSH ratio, and L

Comparison of morphine tolerance in body temperature and luteinizing hormone secretion.

Small doses of morphine induce a hyperthermic response that does not change with repeated injections, whereas higher doses induce a hypothermia that changes to hyperthermia with repeated treatment. Experiment 1 confirmed these results in ovariectomized rats, using 6 repeated injections of morphine at 5 or 30 mg/kg. Using the same treatment regimen. Experiment 2 showed that the low dose of morphine induced a transient suppression of LH levels followed by a hypersecretion, and that repeated injections did not affect this response. The high dose (30 mg/kg) of morphine initially induced a sustaine

Opioid modulation of tonic luteinizing hormone release in ovariectomized dairy cows.

A possible role of endogenous opioid peptides (EOP) in regulating the release of luteinizing hormone (LH) in the absence of ovarian influence was investigated. Experiments were conducted on three lactating Holstein-Friesian dairy cows, 20-27 days after ovariectomy. The cows were bled before and after a single intravenous (i.v.) injection of either 250 mg of naloxone (EOP antagonist) or 300 mg of morphine (EOP agonist) or a combination of the two in Experiments 1, 2 and 3, respectively. The mean and basal LH concentrations and the LH pulse frequency and amplitude were compared before and after

Influence of bilateral cryptorchidism on rat pituitary luteinizing hormone charge microheterogeneity.

Chromatofocusing was used to characterize the isohormones of rat luteinizing hormone (rLH) in extracts of pituitaries from intact and bilaterally cryptorchid animals. Pituitary extracts contained at least seven isoelectric variants (isohormones) or rLH: one eluted in the column void volume (Isohormone, I, pI greater than 9.8), five exhibited apparent pI's in the range of 9.25 to 8.97 (designated as Isohormones II-VI) and one bound to the column but eluted with 1.0 M NaCl (Isohormone VII, pI less than 7.0). In both intact and cryptorchid rats, a large percentage of the rLH was present as Isohor

Hypogonadism and Leydig cell hypoplasia unresponsive to human luteinizing hormone (hLH).

A 22-year-old man with hypogonadism, Leydig cell hypoplasia, high plasma LH, and normal plasma FSH was given human chorionic gonadotropin (hCG) and human luteinizing hormone (hLH) in order to assess testicular testosterone responses to exogenously administered gonadotropins. The maximal testosterone level detected after six doses of hCG, 5000 IU per dose given intramuscularly (IM) was 256 ng/dl. After eight doses of hLH, 550-1100 IU per dose given IM, the plasma testosterone level was unchanged, 100-200 ng/dl. The lack of a response to exogenously administered gonadotropins in our patient sugg
