The use of multi-species probiotic-derived vesicles to encapsulate luteolin for preventing colitis in a mouse model.

Probiotic-derived membrane vesicles (MVs) offer advantages over live probiotics, including lower immunogenicity and enhanced bioavailability of dietary compounds. However, their composition and function depend on culture conditions. In this study, MVs from a single probiotic strain (S-MVs) were compared with those from co-cultured multi-species probiotics (M-MVs). M-MVs showed superior radical scavenging ability due to characteristic antioxidant proteins. Luteolin (Lut), a hydrophobic flavonoid, was loaded into M-MVs to generate Lut-MVs. Compared with Lut or M-MVs alone, Lut-MVs had enhanced w

Luteolin as a multifaceted immunomodulator: insights into its effects on diverse immune cell populations and therapeutic implications.

Luteolin, a natural flavonoid, exerts broad immunomodulatory effects across multiple immune cell populations, positioning it as a promising candidate for treating inflammatory diseases, infections, and cancer. This review synthesizes current evidence on luteolin's effects on T cells, natural killer (NK) cells, dendritic cells (DCs), macrophages, neutrophils, eosinophils, and basophils. Luteolin promotes the differentiation of regulatory T cells (Tregs) and suppresses pro-inflammatory T helper 17 (Th17) and Th2 responses, thereby restoring immune balance in sepsis, allergies, and autoimmunity.

Effect of luteolin on oxidative stress and inflammation in the human osteoblast cell line hFOB1.19 in an inflammatory microenvironment.

BACKGROUND: Periapical lesions are characterized by periapical inflammation and damage to periapical tissues and eventually lead to bone resorption and even tooth loss. H2O2 is widely used in root canal therapy for patients with periapical inflammation. Luteolin possesses high anti-inflammatory, antioxidant, and anticancer potential. However, the underlying mechanism of the efficacy of H2O2 and luteolin on oxidative stress and inflammatory tissue has not been previously addressed. We aimed to investigate the anti-inflammatory and antioxidative effects of luteolin on H2O2-induced cellular oxida

Potentiation of luteolin cytotoxicity by flavonols fisetin and quercetin in human chronic lymphocytic leukemia cell lines.

Despite numerous studies chronic lymphocytic leukemia (CLL) still remains an incurable disease. Therefore, all new compounds and novel strategies which are able to eradicate CLL cells should be considered as valuable clues for a potential future remedy against this malignancy. In the present study, the cytotoxic profiles of natural flavonoids were described in two human CLL cell lines, HG-3 and EHEB, indicating the flavone luteolin as the most potent flavonoid with half-maximal inhibitory constants (IC50) of 37 μM and 26 μM, respectively. Luteolin significantly increased the apoptotic cell pop

Beneficial Effects of Co-Ultramicronized Palmitoylethanolamide/Luteolin in a Mouse Model of Autism and in a Case Report of Autism.

AIMS: Autism spectrum disorder (ASD) is a condition defined by social communication deficits and repetitive restrictive behaviors. Association of the fatty acid amide palmitoylethanolamide (PEA) with the flavonoid luteolin displays neuroprotective and antiinflammatory actions in different models of central nervous system pathologies. We hypothesized that association of PEA with luteolin might have therapeutic utility in ASD, and we employed a well-recognized autism animal model, namely sodium valproate administration, to evaluate cognitive and motor deficits. METHODS: Two sets of experiments

Luteolin reverses chemoresistance in colorectal cancer cells via TET1/TDG-dependent epigenetic repression of β-catenin.

Resistance to 5-fluorouracil (5-FU) and oxaliplatin (OXT) remains a major cause of treatment failure in colorectal cancer (CRC). Luteolin, a dietary flavonoid with anticancer properties, has shown therapeutic potential in various malignancies, but its effects on chemotherapy-resistant CRC remain poorly understood. In this study, we investigated whether luteolin restores chemosensitivity in 5-FU-resistant (SNUC5/5-FUR) and OXT-resistant (SNUC5/OXTR) CRC cells and explored the underlying molecular mechanisms. Luteolin reduced cell viability and induced apoptosis in both resistant cell lines. Mec

Luteolin Ameliorates Sepsis-Induced Acute Lung Injury by Targeting RPTPα to Reprogram Macrophage M1-M2 Polarization.

Dysregulated macrophage polarization is a critical pathological driver of sepsis-induced acute lung injury (ALI). However, the absence of novel therapeutic targets constitutes a significant translational barrier, underscoring the urgent need for advancements in precision medicine. In this study, we demonstrate that the natural product luteolin (LU) effectively promotes LPS-induced M1-to-M2 macrophage polarization by modulating the expression of pro-inflammatory and anti-inflammatory cytokines. Subsequently, thermal proteome profiling identifies receptor-type protein tyrosine phosphatase α (RPT

Cyclodextrin MOFs modified dry powder inhalers quadruple bioavailability of luteolin to ameliorate fibrosing interstitial lung disease.

Fibrosing interstitial lung disease (ILD) is a pathological condition that is highly heterogeneous and lethal, and has few effective treatment choices. Other than pirfenidone and nintedanib for the therapy of idiopathic pulmonary fibrosis, no medications are currently licensed for the treatment of ILD. Luteolin is a common flavonoid with multiple biological effects such as anti-inflammation but with poor solubility and absorption. In this study, we loaded luteolin into γ-cyclodextrin metal-organic frameworks (CD-MOFs) to deliver the medicine to the lungs using dry powder inhalers; in vitro pul

Luteolin Improves BBB Integrity Following Ischemic Stroke Injury by Downregulating the Expression of LRP1 and MMP9.

Ischemic stroke is a prevalent neurological disorder often resulting in brain tissue injury and blood-brain barrier (BBB) damage. Luteolin (LUT), a natural polyphenolic compound in grape skin, wine, and other plants, has neuroprotective properties against ischemic stroke. However, it remains unclear whether its protective effect involves alterations in BBB integrity. This study aimed to examine the BBB-protective impact of LUT in a photothrombotic (PT) mouse model and oxygen-glucose deprivation (OGD) cell model and investigate its underlying mechanism. First, we used neurofunctional scoring, 2

Luteolin decreases sFlt-1 and mean arterial pressure in the RUPP animal model of placental ischemia.

OBJECTIVES: Preeclampsia (PE) is a complication of pregnancy and a leading cause of maternal and fetal morbidity and mortality. Despite being prevalent and highly morbid, there are no approved treatments to relieve PE signs and symptoms. Many studies have established that elevated levels of circulating anti-angiogenic soluble fms-like tyrosine kinase 1 (sFlt-1) protein drives the maternal syndrome observed in PE, and reductions in sFlt-1 improved symptoms. We have previously reported that the bioflavonoid luteolin reduces sFlt-1 expression and the vasoconstrictor endothelin-1 (ET-1) in vitro a
