Assessment of the lymphatic system by indirect lymphography in patients with post-thrombotic syndrome.

OBJECTIVE: Alterations in lower extremity lymph drainage caused by chronic venous obstruction (CVO) have not been well studied, partially because of a lack of standardized imaging modalities to assess the quality of lymphatic drainage in the lower extremities of patients with post-thrombotic syndrome (PTS). However, these changes are likely to have an impact on the severity of the disease and clinical outcomes of interventions. In the present study, we investigated the feasibility and diagnostic value of preintervention indirect lymphography in patients with CVO and their pre- and postinterven

Meningeal Lymphatics: From Anatomy to Central Nervous System Immune Surveillance.

At steady state, the CNS parenchyma has few to no lymphocytes and less potent Ag-presentation capability compared with other organs. However, the meninges surrounding the CNS host diverse populations of immune cells that influence how CNS-related immune responses develop. Interstitial and cerebrospinal fluid produced in the CNS is continuously drained, and recent advances have emphasized that this process is largely taking place through the lymphatic system. To what extent this fluid process mobilizes CNS-derived Ags toward meningeal immune cells and subsequently the peripheral immune system t

Extracellular bacterial lymphatic metastasis drives Streptococcus pyogenes systemic infection.

Unassisted metastasis through the lymphatic system is a mechanism of dissemination thus far ascribed only to cancer cells. Here, we report that Streptococcus pyogenes also hijack lymphatic vessels to escape a local infection site, transiting through sequential lymph nodes and efferent lymphatic vessels to enter the bloodstream. Contrasting with previously reported mechanisms of intracellular pathogen carriage by phagocytes, we show S. pyogenes remain extracellular during transit, first in afferent and then efferent lymphatics that carry the bacteria through successive draining lymph nodes. We

The pathomechanism of posttraumatic edema of the lower limbs: II--Changes in the lymphatic system.

BACKGROUND: The peripheral lymphatic system reacts to penetrating microorganisms and self-antigens released from tissues and cells damaged by trauma or intracellular pathogens. The response of regional lymph nodes to tissue trauma has not been thoroughly studied. We investigated the changes in lower limb lymphatics and nodes after fractures and soft tissue injuries. This type of injury is frequently complicated by limb edema. Posttraumatic edema of lower limbs is characterized by long-lasting swelling of the limb, erythema, and increased skin temperature at the site of injury. This suggests th

Exploring the significance of IgM expression in the lymphatic system during breast cancer progression.

BACKGROUND: Breast cancer is a malignant tumor characterized by metastasis. It first spreads through the lymphatic system to lymph nodes of various levels and can subsequently metastasize to distant organs via the bloodstream. Therefore, identifying protein biomarkers associated with breast cancer is of great significance for predicting disease progression, implementing early drug therapy or interventions, and improving prognosis. This study aims to investigate the dynamic changes in immunoglobulin M (IgM) levels in lymphoid tissues and lymph fluid during the progression of breast cancer and to

Compensatory mechanism of the injured lymphatic system: New insights from fluorescent nanoparticle assisted lymphangiography.

Dysfunction of lymphatic system is closely associated with many diseases, including obesity, diabetes, asthma and cancer. During the diagnosis and treatment of lymph related diseases, a deep understanding of the lymphatic morphology and function is of vital importance. Although the tracing of sentinel lymph node is largely achieved in recent years, there is still a lack of systematic depictions of lymphatic drainage pathways, especially in real-time way. This study aims to perform lymphangiography on small animals using FITC@SiO2 nanoparticles (NPs) to systematically depict their lymphatic dra

Surface-Engineered HA-PEG-ICG/PLGA Nanoprobes with Vessels Targeting for Lymphatic System Visualization.

Targeted imaging of the lymphatic system is essential for the early diagnosis and management of lymphatic disorders, such as lymphedema. In this study, we developed a lymphatic-targeted fluorescent nanoprobe by encapsulating indocyanine green (ICG) within poly(lactic-co-glycolic acid) (PLGA) nanoparticles, further surface-modified with hyaluronic acid-polyethylene glycol (HA-PEG) to enhance specificity (HA-PEG-ICG/PLGA NPs). The nanoparticles were synthesized via a microemulsion technique followed by surface cross-linking, and thoroughly characterized by ultraviolet-visible (UV-vis) spectrosco

The Glymphatic System and Subarachnoid Lymphatic-Like Membrane: Recent Developments in Cerebrospinal Fluid Research.

BACKGROUND: Cerebrospinal fluid (CSF) circulates throughout the ventricles, cranial and spinal subarachnoid spaces, and central spinal cord canal. CSF protects the central nervous system through mechanical cushioning, regulation of intracranial pressure, regulation of metabolic homeostasis, and provision of nutrients. Recently, investigators have characterized the glial-lymphatic (glymphatic) system, the analog of the lymphatic system in the central nervous system, and described a fourth meningeal layer; the subarachnoid lymphatic-like membrane (SLYM)relevant to the CSF. METHODS: A narrative

High-frequency ultrasound imaging for monitoring the function of meningeal lymphatic system in mice.

The meningeal lymphatic system drains the cerebrospinal fluid from the subarachnoid space to the cervical lymphatic system, primarily to the deep cervical lymph nodes. Perturbations of the meningeal lymphatic system have been linked to various neurologic disorders. A method to specifically monitor the flow of meningeal lymphatic system in real time is unavailable. In the present study, we adopted the high-frequency ultrasound (HFUS) with 1,1'diocatadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI)-loaded microbubble and FePt@PLGA nanoparticle contrast agents to evaluate the flow of

The meningeal lymphatic vessels and the glymphatic system: Potential therapeutic targets in neurological disorders.

The recent discovery of the meningeal lymphatic vessels (mLVs) and glymphatic pathways has challenged the long-lasting dogma that the central nervous system (CNS) lacks a lymphatic system and therefore does not interact with peripheral immunity. This discovery has reshaped our understanding of mechanisms underlying CNS drainage. Under normal conditions, a close connection between mLVs and the glymphatic system enables metabolic waste removal, immune cell trafficking, and CNS immune surveillance. Dysfunction of the glymphatic-mLV system can lead to toxic protein accumulation in the brain, and i

Revisiting the gut-liver axis: gut lymphatic system in liver cirrhosis and portal hypertension.

The lymphatic vascular system runs parallel to the blood vascular system, comprising a network of lymphatic vessels and secondary lymphoid organs. The intestinal lymphatic capillaries (lacteals) and the associated collecting vessels in the mesentery form the gut lymphatic system. The gut lymphatic vasculature comprises the longest-studied lymphatic vessel bed and plays a significant role in the uptake and transport of dietary fat, abdominal fluid balance, and gut immunosurveillance. Gut is closely connected to liver through the portal circulation. In several experimental and clinical studies,

Dynamic contrast-enhanced magnetic resonance lymphangiography in pediatric patients with central lymphatic system disorders.

Central conducting lymphatics (CCLs) disorders represent a broad spectrum of clinical entities ranging from self-limiting traumatic leaks treated by conservative strategies, to complex lymphatic circulation abnormalities that are progressive and unresponsive to currently available treatments. Dynamic contrast-enhanced magnetic resonance lymphangiography (DCMRL) performed by intranodal injection of gadolinium-based contrast material is a recently developed technique which allows a minimally invasive evaluation of the CCL abnormalities providing a dynamic assessment of lymph flow and its pathway

Mechanisms of lymphatic system-specific viral replication and its potential role in autoimmune disease.

Viral infections can be fatal because of the direct cytopathic effects of the virus or the induction of a strong, uncontrolled inflammatory response. Virus and host intrinsic characteristics strongly modulate the outcome of viral infections. Recently we determined the circumstances under which enhanced replication of virus within the lymphoid tissue is beneficial for the outcome of a disease. This enforced viral replication promotes anti-viral immune activation and, counterintuitively, accelerates virus control. In this review we summarize the mechanisms that contribute to enforced viral repli

Surgical damage to the lymphatic system promotes tumor growth via impaired adaptive immune response.

BACKGROUND: Both lymph nodes (LNs) and lymphatic channels from primary sites to regional LNs are critical for initiation of adaptive immunity. However, as LNs are common metastatic sites in skin cancers, LN biopsies or dissections are frequently performed. In addition, reconstructive skin flaps after tumor resection may damage lymphatic flow from primary sites to regional LNs. OBJECTIVE: This study was designed to investigate the effect on tumor progression by such surgeries. METHODS: We developed a mouse model that simulates LNs dissection or skin flap that blocks lymphatic flow from primar
