Tumor-Infiltrating Lymphocytes as a Predictor of Adjuvant Avelumab Efficacy in Patients with High-risk Triple-Negative Breast Cancer.

PURPOSE: We aimed to investigate the prognostic and predictive value of tumor-infiltrating lymphocytes (TIL) for adjuvant immunotherapy in high-risk early triple-negative breast cancer (TNBC). PATIENTS AND METHODS: The phase III A-BRAVE trial randomized 466 patients with high-risk early TNBC to adjuvant avelumab or observation after standard therapy. Inclusion criteria allowed 2 strata: stratum A (primary surgery followed by adjuvant chemotherapy, defined at high risk based on pathologic stage) and stratum B (neoadjuvant chemotherapy followed by surgery without pathologic complete response).

Prognostic Value of Neutrophil-to-Lymphocyte Ratio and Tumor-Infiltrating Lymphocytes (TILs), for Patients with Triple-Negative Breast Cancer.

During the process of carcinogenesis, inflammation originating in the tumor or its microenvironment promotes tumor growth. An elevated neutrophil-lymphocyte ratio (NLR) has been considered a biomarker associated with reduced survival in several types of cancer. A retrospective study was conducted and the results of 49 triple-negative breast cancer (TNBC) biopsies from 2012 to 2017 were reviewed. The patients were divided into two groups: those with nonbasal subtype TNBC and those with basal-like TNBC. To differentiate TNBC tumors by subtype, cytokeratin 5/6 (CK5/6) expression was assessed. The

Expansion of IL-2-independent tumor-infiltrating lymphocytes through a feeder-free process: a preclinical study for solid tumors.

OBJECTIVE: Conventional tumor-infiltrating lymphocyte (TIL) therapy for solid tumors relies on high-dose interleukin-2 (IL-2) during expansion and post-infusion, and promotes T-cell exhaustion and toxicity. Herein, we developed a feeder-free, low-dose IL-2 TIL expansion protocol and evaluated whether hydroxychloroquine (HCQ) or programmed cell death protein 1 (PD-1) blockade might enhance therapeutic efficacy and decrease IL-2 dependence. METHODS: TILs from multiple solid tumors were expanded ex vivo with decreased-dose IL-2, IL-7, and IL-15 plus CD3/CD28 co-stimulation, without feeder cells.

Prognostic Significance of Poly (ADP-Ribose) Polymerase 1 Binding Protein Expression and CD8+ Tumor-Infiltrating Lymphocytes in Hepatocellular Carcinoma.

The role of poly (adenosine diphosphate [ADP]-ribose) polymerase 1 binding protein (PARPBP) in hepatocellular carcinoma (HCC) prognosis remains unclear. This study investigated PARPBP expression in HCC clinical samples and evaluated its clinicopathological significance in relation to CD8-positive tumor-infiltrating lymphocytes (CD8+ TILs). PARPBP expression and CD8+ TIL density were assessed in surgical specimens from 96 patients with HCC. We performed immunohistochemical analysis to determine PARPBP protein levels, which were correlated with clinicopathological features and patient outcomes.

Flow cytometry quantification of tumor-infiltrating lymphocytes to predict the survival of patients with diffuse large B-cell lymphoma.

INTRODUCTION: Our previous studies have demonstrated that tumor-infiltrating lymphocytes (TILs), including normal B cells, T cells, and natural killer (NK) cells, in diffuse large B-cell lymphoma (DLBCL) have a significantly favorable impact on the clinical outcomes of patients treated with standard chemoimmunotherapy. In this study, to gain a full overview of the tumor immune microenvironment (TIME), we assembled a flow cytometry cohort of 102 patients diagnosed with DLBCL at the Duke University Medical Center. METHODS: We collected diagnostic flow cytometry data, including the proportion of

Prognostic Value of Tumor-Infiltrating Lymphocytes and Tertiary Lymphoid Structures in Epstein-Barr Virus-Associated and -Negative Gastric Carcinoma.

BACKGROUND: Tumor-infiltrating lymphocytes (TILs) are considered a manifestation of the host immune response against cancer and tertiary lymphoid structures (TLS) may contribute to lymphocytes recruitment. Both of them have been reported as potential prognostic parameters in some human malignancies. However, the roles of TILs, TLS, and their correlation in Epstein-Barr Virus-associated gastric carcinoma (EBVaGC) and EBV-negative gastric carcinoma (EBVnGC) are largely unknown. METHODS: To observe the correlation among TILs, TLS, and clinicopathological characteristics and their prognostic sign

Randomized, Prospective Evaluation Comparing Intensity of Lymphodepletion Before Adoptive Transfer of Tumor-Infiltrating Lymphocytes for Patients With Metastatic Melanoma.

PURPOSE: Adoptive cell transfer, the infusion of large numbers of activated autologous lymphocytes, can mediate objective tumor regression in a majority of patients with metastatic melanoma (52 of 93; 56%). Addition and intensification of total body irradiation (TBI) to the preparative lymphodepleting chemotherapy regimen in sequential trials improved objective partial and complete response (CR) rates. Here, we evaluated the importance of adding TBI to the adoptive transfer of tumor-infiltrating lymphocytes (TIL) in a randomized fashion. PATIENTS AND METHODS: A total of 101 patients with meta

Identification of a new endoplasmic reticulum-resident protein recognized by HLA-A24-restricted tumor-infiltrating lymphocytes of lung cancer.

To help clarify the molecular basis of tumor immunology in lung cancer, we have investigated antigens recognized by HLA-A24-restricted CTLs established from T cells infiltrating into lung adenocarcinoma and report a new gene encoding tumor epitopes recognized by the CTLs. This gene was located on chromosome 4q31.22 and encoded an unreported endoplasmic reticulum-resident protein with 412 deduced amino acids. This protein had a molecular mass of 46 kDa and was expressed in the majority of malignant cells and tissues tested, with the exception of T-cell leukemia cells, but was not expressed in a

Convection-Enhanced Delivery of Tumor-Infiltrating Lymphocytes Enhances Intratumoral Distribution and Therapeutic Efficacy in an Orthotopic Rat Glioma Model.

Background: Adoptive cell therapy using tumor-infiltrating lymphocytes (TILs) is a potential strategy for glioma treatment, but effective intracranial delivery remains a major obstacle. Convection-enhanced delivery (CED) may improve local parenchymal coverage by bypassing the blood-brain barrier and using pressure-driven interstitial transport. Methods: We evaluated whether CED could improve the early intracerebral distribution and antitumor activity of ex vivo-expanded TILs in an orthotopic rat C6 glioma model. Expanded TILs were characterized as a CD3-enriched lymphocyte product with inducib

Pretreatment tumor infiltrating lymphocytes and outcome in patients with HR+/HER2- advanced breast cancer treated with CDK4/6 inhibitors.

Growing evidence suggests that enhancement of the tumor immune response contributes to the antitumor activity of CDK4/6 inhibitors (CDK4/6i), but the role of pretreatment tumor immune microenvironment is not clear. We retrospectively investigated in patients with HR+/HER2- advanced breast cancer receiving CDK4/6i and endocrine therapy whether pretreatment stromal tumor infiltrating lymphocytes (sTILs) were associated with outcome. A total of 100 patients were evaluable 53 treated with palbociclib, 44 with ribociclib and 3 with abemaciclib. Tumors were classified as sTILs positive (sTILs+) when

[Cell therapy with tumor infiltrating lymphocytes; a therapeutic alternative for malignant melanoma and soon also other types of cancer].

Therapy for advanced cutaneous melanoma with tumor infiltrating lymphocytes (TIL) has shown promising clinical results for 35 years. Owing to advanced production and treatment methods, TIL therapy has only been accessible in a few centers in Europe and the USA. Recently, following a company phase 2 trial and an academic Phase III trial with superior response rate compared to patients treated with the immune checkpoint inhibitor (ICI) ipilimumab, TIL therapy gained FDA approval. Several studies are now ongoing where patients with other tumor diagnoses such as cervical cancer and non-small-cell
